    Scientific and Technological Development of Hadrontherapy

    Hadrontherapy is a novel technique of cancer radiation therapy which employs beams of charged hadrons, protons and carbon ions in particular. Due to their physical and radiobiological properties, they allow one to obtain a more conformal treatment with respect to photons used in conventional radiation therapy, sparing better the healthy tissues located in proximity of the tumour and allowing a higher control of the disease. Hadrontherapy is the direct application of research in high energy physics, making use of specifically conceived particle accelerators and detectors. Protons can be considered today a very important tool in clinical practice due to the several hospital-based centres in operation and to the continuously increasing number of facilities proposed worldwide. Very promising results have been obtained with carbon ion beams, especially in the treatment of specific radio resistant tumours. To optimize the use of charged hadron beams in cancer therapy, a continuous technological challenge is leading to the conception and to the development of innovative methods and instruments.     Nuclear Emulsion Film Detectors for Proton Radiography: Design and Test of the First Prototype

    Proton therapy is nowadays becoming a wide spread clinical practice in cancer therapy and sophisticated treatment planning systems are routinely used to exploit at best the ballistic properties of charged particles. The information on the quality of the beams and the range of the protons is a key issue for the optimization of the treatment. For this purpose, proton radiography can be used in proton therapy to obtain direct information on the range of the protons, on the average density of the tissues for treatment planning optimization and to perform imaging with negligible dose to the patient. We propose an innovative method based on nuclear emulsion film detectors for proton radiography, a technique in which images are obtained by measuring the position and the residual range of protons passing through the patient's body. Nuclear emulsion films interleaved with tissue equivalent absorbers can be fruitfully used to reconstruct proton tracks with very high precision. The first prototype of a nuclear emulsion based detector has been conceived, constructed and tested with a therapeutic proton beam at PSI. The scanning of the emulsions has been performed at LHEP in Bern, where a fully automated microscopic scanning technology has been developed for the OPERA experiment on neutrino oscillations. After track reconstruction, the first promising experimental results have been obtained by imaging a simple phantom made of PMMA with a step of 1 cm. A second phantom with five 5 x 5 mm^2 section aluminum rods located at different distances and embedded in a PMMA structure has been also imaged.     Molecular flexibility of citrus pectins by combined sedimentation and viscosity analysis

    The flexibility/rigidity of pectins plays an important part in their structure-function relationship and therefore on their commercial applications in the food and biomedical industries. Earlier studies based on sedimentation analysis in the ultracentrifuge have focused on molecular weight distributions and qualitative and semi-quantitative descriptions based on power law and Wales-van Holde treatments of conformation in terms of "extended" conformations [Harding, S. E., Berth, G., Ball, A., Mitchell, J.R., & Garcìa de la Torre, J. (1991). The molecular weight distribution and conformation of citrus pectins in solution studied by hydrodynamics. Carbohydrate Polymers, 168, 1-15; Morris, G. A., Foster, T. J., & Harding, S.E. (2000). The effect of degree of esterification on the hydrodynamic properties of citrus pectin. Food Hydrocolloids, 14, 227-235]. In the present study, four pectins of low degree of esterification 17-27% and one of high degree of esterification (70%) were characterised in aqueous solution (0.1 M NaCl) in terms of intrinsic viscosity [η], sedimentation coefficient (s°20,w) and weight average molar mass (Mw). Solution conformation/flexibility was estimated qualitatively using the conformation zoning method [Pavlov, G.M., Rowe, A.J., & Harding, S.E. (1997). Conformation zoning of large molecules using the analytical ultracentrifuge. Trends in Analytical Chemistry, 16, 401-405] and quantitatively (persistence length Lp) using the traditional Bohdanecky and Yamakawa-Fujii relations combined together by minimisation of a target function. Sedimentation conformation zoning showed an extended coil (Type C) conformation and persistence lengths all within the range Lp=10-13 nm (for a fixed mass per unit length)

    Measurement of the inclusive and dijet cross-sections of b-jets in pp collisions at sqrt(s) = 7 TeV with the ATLAS detector

    The inclusive and dijet production cross-sections have been measured for jets containing b-hadrons (b-jets) in proton-proton collisions at a centre-of-mass energy of sqrt(s) = 7 TeV, using the ATLAS detector at the LHC. The measurements use data corresponding to an integrated luminosity of 34 pb^-1. The b-jets are identified using either a lifetime-based method, where secondary decay vertices of b-hadrons in jets are reconstructed using information from the tracking detectors, or a muon-based method where the presence of a muon is used to identify semileptonic decays of b-hadrons inside jets. The inclusive b-jet cross-section is measured as a function of transverse momentum in the range 20 < pT < 400 GeV and rapidity in the range |y| < 2.1. The bbbar-dijet cross-section is measured as a function of the dijet invariant mass in the range 110 < m_jj < 760 GeV, the azimuthal angle difference between the two jets and the angular variable chi in two dijet mass regions. The results are compared with next-to-leading-order QCD predictions. Good agreement is observed between the measured cross-sections and the predictions obtained using POWHEG + Pythia. MC@NLO + Herwig shows good agreement with the measured bbbar-dijet cross-section. However, it does not reproduce the measured inclusive cross-section well, particularly for central b-jets with large transverse momenta.Comment: 10 pages plus author list (21 pages total), 8 figures, 1 table, final version published in European Physical Journal

    Inactivation of class II PI3K-C2 alpha induces leptin resistance, age-dependent insulin resistance and obesity in male mice

    AIMS/HYPOTHESIS: While the class I phosphoinositide 3-kinases (PI3Ks) are well-documented positive regulators of metabolism, the involvement of class II PI3K isoforms (PI3K-C2α, -C2β and -C2γ) in metabolic regulation is just emerging. Organismal inactivation of PI3K-C2β increases insulin signalling and sensitivity, whereas PI3K-C2γ inactivation has a negative metabolic impact. In contrast, the role of PI3K-C2α in organismal metabolism remains unexplored. In this study, we investigated whether kinase inactivation of PI3K-C2α affects glucose metabolism in mice. METHODS: We have generated and characterised a mouse line with a constitutive inactivating knock-in (KI) mutation in the kinase domain of the gene encoding PI3K-C2α (Pik3c2a). RESULTS: While homozygosity for kinase-dead PI3K-C2α was embryonic lethal, heterozygous PI3K-C2α KI mice were viable and fertile, with no significant histopathological findings. However, male heterozygous mice showed early onset leptin resistance, with a defect in leptin signalling in the hypothalamus, correlating with a mild, age-dependent obesity, insulin resistance and glucose intolerance. Insulin signalling was unaffected in insulin target tissues of PI3K-C2α KI mice, in contrast to previous reports in which downregulation of PI3K-C2α in cell lines was shown to dampen insulin signalling. Interestingly, no metabolic phenotypes were detected in female PI3K-C2α KI mice at any age. CONCLUSIONS/INTERPRETATION: Our data uncover a sex-dependent role for PI3K-C2α in the modulation of hypothalamic leptin action and systemic glucose homeostasis. In order to measure the signal produced by the passage of a gravitational wave, they aim to reach unprecedent sensitivities in measuring the relative displacements of the mirrors. One of them , the 3-km-long Virgo gravitational wave antenna, which will be particularly sensitive in the low frequency range (10-100 Hz), is presently in its commissioning phase.     Measurement of charged-particle event shape variables in inclusive root(s)=7 TeV proton-proton interactions with the ATLAS detector

    The measurement of charged-particle event shape variables is presented in inclusive inelastic pp collisions at a center-of-mass energy of 7 TeV using the ATLAS detector at the LHC. The observables studied are the transverse thrust, thrust minor, and transverse sphericity, each defined using the final-state charged particles' momentum components perpendicular to the beam direction. Events with at least six charged particles are selected by a minimum-bias trigger. In addition to the differential distributions, the evolution of each event shape variable as a function of the leading charged-particle transverse momentum, charged-particle multiplicity, and summed transverse momentum is presented. Predictions from several Monte Carlo models show significant deviations from data

    All-sky search for gravitational-wave bursts in the second joint LIGO-Virgo run

    We present results from a search for gravitational-wave bursts in the data collected by the LIGO and Virgo detectors between July 7, 2009 and October 20, 2010: data are analyzed when at least two of the three LIGO-Virgo detectors are in coincident operation, with a total observation time of 207 days. The analysis searches for transients of duration < 1 s over the frequency band 64-5000 Hz, without other assumptions on the signal waveform, polarization, direction or occurrence time. All identified events are consistent with the expected accidental background. We set frequentist upper limits on the rate of gravitational-wave bursts by combining this search with the previous LIGO-Virgo search on the data collected between November 2005 and October 2007. The upper limit on the rate of strong gravitational-wave bursts at the Earth is 1.3 events per year at 90% confidence. We also present upper limits on source rate density per year and Mpc^3 for sample populations of standard-candle sources. As in the previous joint run, typical sensitivities of the search in terms of the root-sum-squared strain amplitude for these waveforms lie in the range 5 10^-22 Hz^-1/2 to 1 10^-20 Hz^-1/2.
