    Towards Ecologically Sustainable Management of the Torres Strait Prawn Fishery. CRC Torres Strait Task T1.5 – Final Report

    This publication, which is the final report to the Torres Strait Cooperative Research Centre, provides an overview of all the research that was conducted as part of the Torres Strait CRC Task 1.5 - Towards Ecologically Sustainable Management of the Torres Strait Prawn Fishery The objectives of the task were: To develop cost-effective protocols to monitor and quantify the bycatch and environmental impacts of commercial prawn trawling. To monitor the status of target species using both fishery dependent and fishery independent data. To develop biological reference points for target species and undertake management strategy evaluation, in particular a risk assessment of fishing at various levels of fishing mortality. This report focuses on the second component of objective 1 and details a comparative analysis of bycatch samples collected from areas of the Torres Strait that were both closed and open to prawn trawl fishing. The report also reviews the research conducted in relation to objectives 2 and 3 which are detailed in a separate report, Stock Assessment of the Torres Strait Tiger Prawn Fishery (Penaeus esculentus)

    Fisheries long term monitoring program : summary of tiger and endeavour prawn survey results: 1998-2006

    There are two distinct tiger and endeavour prawn fisheries that extend along the northern Queensland east coast and into Torres Strait: the Torres Strait Prawn Fishery (TSPF) and the northern segment (north of 22° S) of the East Coast Trawl Fishery (ECTF). This document reports on the Torres Strait, and a section of the northern Queensland segment of the ECTF (defined as the section of the Queensland east coast from Cape Flattery 16° S to the tip of Cape York 10.4° S. This section will be referred to as Far North Queensland). The prawn trawl fishery in this Far North Queensland (FNQ) region mainly targets tiger, endeavour and king prawns. These three prawn groups consist of six main species: the brown tiger prawn (Penaeus esculentus), the grooved tiger prawn (Penaeus semisulcatus), the blue endeavour prawn (Metapenaeus endeavouri), the red endeavour prawn (Metapenaeus ensis), the redspot king prawn (Melicertus longistylus), and the western king prawn (Melicertus latisulcatus). The average annual catch (from 1998–2004) of the tiger and endeavour prawn sector of the northern Queensland ECTF (north of 22° S) is approximately 1700 t of tiger prawns, 1200 t of endeavour prawns and 900 t of king prawns (CFISH data, 1998–2004). For the Torres Strait the average annual catch (from 2000–04) is 675 t of tiger prawns, 1018 t of endeavour prawns and 77 t of king prawns (Taylor et al. 2006). The annual value of the northern Queensland and the Torres Strait prawn fisheries is approximately A40m(Williams2002)andA40m (Williams 2002) and A22m, respectively (McLoughlin 2002). The initial annual prawn surveys (1998–2000) were a component of the Fisheries Research and Development Corporation project 97/146, conducted by Department of Primary Industries and Fisheries (DPI&F). The project developed a fishery independent sampling procedure for tiger and endeavour prawn stocks in northern Queensland and Torres Strait waters. The DPI&F Long Term Monitoring Program continued the annual prawn surveys, using the same survey procedures and sites. The key objective of the annual prawn surveys is to monitor prawn species, size, distribution and relative abundance in order to contribute to assessments of the status of tiger, endeavour and king prawns in northern Queensland and Torres Strait waters. The surveys also document bycatch in the fishery area to enhance understanding of the fishery’s potential impacts on the ecosystem. The survey results for FNQ indicate that 1999 and 2005 were years of higher than average tiger prawn recruitment while 2000 was a year of lower than average recruitment. Similarly, for Torres Strait, 2000 was a year of low tiger prawn recruitment whereas 1998, 2005 and 2006 were years of above average recruitment. The results presented in this report, a previous summary of survey results (Turnbull et al. 2004), and a detailed analysis of both the 1998 to 2002 surveys and commercial catch data (Turnbull et al. 2005) indicate that these surveys are a cost effective tool in providing the data needed to monitor prawn stocks, especially in multi-species fisheries such as the northern Queensland east coast tiger prawn fishery. The survey catch rates can be compared with, and provide confidence in, the trends observed in the commercial harvest data (Turnbull et al. 2005, O’Neill and Turnbull 2006, O’Neill and Leigh 2006) especially for tiger prawns which are considered susceptible to overfishing. It is recommended that the LTMP prawn surveys be continued as they provide an important time-series of fishery independent data that complements the analysis of the commercial fishery data. The survey data complements the commercial harvest data and will assist with the development of species-based assessment and monitoring of the northern Queensland tiger prawn fishery

    Characterisation of human saliva as a platform for oral dissolution medium development

    Human saliva is a biological fluid of great importance in the field of dissolution testing. However, until now, no consensus has been reached on its key characteristics relevant to dissolution testing. As a result, it is difficult to select or develop an in vitro dissolution medium to best represent human saliva. In this study, the pH, buffer capacity, surface tension, viscosity and flow rate of both unstimulated (US) and stimulated (SS) human saliva were investigated in order to provide a platform of reference for future dissolution studies using simulated salivary fluids. Age and gender related differences in a sample size of 30 participants for each parameter were investigated. Significant differences were established between US and SS for all characteristics except surface tension. Therefore, the requirement for using two simulated salivary fluids should be considered when developing an oral dissolution model

    Unfractionated heparin inhibits live wild type SARS-CoV-2 cell infectivity at therapeutically relevant concentrations

    BACKGROUND AND PURPOSE: Currently, there are no licensed vaccines and limited antivirals for the treatment of COVID-19. Heparin (delivered systemically) is currently used to treat anticoagulant anomalies in COVID-19 patients. Additionally, in the United Kingdom, Brazil and Australia, nebulised unfractionated heparin (UFH) is being trialled in COVID-19 patients as a potential treatment. A systematic comparison of the potential antiviral effect of various heparin preparations on live wild type SARS-CoV-2, in vitro, is needed.EXPERIMENTAL APPROACH: Seven different heparin preparations including UFH and low MW heparins (LMWH) of porcine or bovine origin were screened for antiviral activity against live SARS-CoV-2 (Australia/VIC01/2020) using a plaque inhibition assay with Vero E6 cells. Interaction of heparin with spike protein RBD was studied using differential scanning fluorimetry and the inhibition of RBD binding to human ACE2 protein using elisa assays was examined.KEY RESULTS: All the UFH preparations had potent antiviral effects, with IC50 values ranging between 25 and 41 μg·ml-1 , whereas LMWHs were less inhibitory by ~150-fold (IC50 range 3.4-7.8 mg·ml-1 ). Mechanistically, we observed that heparin binds and destabilizes the RBD protein and furthermore, we show heparin directly inhibits the binding of RBD to the human ACE2 protein receptor.CONCLUSION AND IMPLICATIONS: This comparison of clinically relevant heparins shows that UFH has significantly stronger SARS-CoV-2 antiviral activity compared to LMWHs. UFH acts to directly inhibit binding of spike protein to the human ACE2 protein receptor. Overall, the data strongly support further clinical investigation of UFH as a potential treatment for patients with COVID-19.</p

    Psychopathic leadership a case study of a corporate psychopath CEO

    This longitudinal case study reports on a charity in the UK which gained a new CEO who was reported by two middle managers who worked in the charity, to embody (respectively) all or most of the ten characteristics within a measure of corporate psychopathy. The leadership of this CEO with a high corporate psychopathy score was reported to be so poor that the organisation was described as being one without leadership and as a lost organisation with no direction. This paper outlines the resultant characteristics of the ensuing aimlessness and lack of drive of the organisation involved. Comparisons are made to a previous CEO in the same organisation, who was reportedly an authentic, effective and transformational leader. Outcomes under the CEO with a high corporate psychopathy score were related to bullying, staff withdrawal and turnover as effective employees stayed away from and/or left the organisation. Outcomes also included a marked organisational decline in terms of revenue, employee commitment, creativity and organisational innovativeness. The paper makes a contribution to both leadership and to corporate psychopathy research as it appears to be the first reported study of a CEO with a high corporate psychopathy score

    Gothic Revival Architecture Before Horace Walpole's Strawberry Hill

    The Gothic Revival is generally considered to have begun in eighteenth-century Britain with the construction of Horace Walpole’s villa, Strawberry Hill, Twickenham, in the late 1740s. As this chapter demonstrates, however, Strawberry Hill is in no way the first building, domestic or otherwise, to have recreated, even superficially, some aspect of the form and ornamental style of medieval architecture. Earlier architects who, albeit often combining it with Classicism, worked in the Gothic style include Sir Christopher Wren, Nicholas Hawksmoor, William Kent and Batty Langley, aspects of whose works are explored here. While not an exhaustive survey of pre-1750 Gothic Revival design, the examples considered in this chapter reveal how seventeenth- and eighteenth-century Gothic emerged and evolved over the course of different architects’ careers, and how, by the time that Walpole came to create his own Gothic ‘castle’, there was already in existence in Britain a sustained Gothic Revivalist tradition

    A prenylated dsRNA sensor protects against severe COVID-19

    Inherited genetic factors can influence the severity of COVID-19, but the molecular explanation underpinning a genetic association is often unclear. Intracellular antiviral defenses can inhibit the replication of viruses and reduce disease severity. To better understand the antiviral defenses relevant to COVID-19, we used interferon-stimulated gene (ISG) expression screening to reveal that OAS1, through RNase L, potently inhibits SARS-CoV-2. We show that a common splice-acceptor SNP (Rs10774671) governs whether people express prenylated OAS1 isoforms that are membrane-associated and sense specific regions of SARS-CoV-2 RNAs, or only express cytosolic, nonprenylated OAS1 that does not efficiently detect SARS-CoV-2. Importantly, in hospitalized patients, expression of prenylated OAS1 was associated with protection from severe COVID-19, suggesting this antiviral defense is a major component of a protective antiviral response

    Investigation of hospital discharge cases and SARS-CoV-2 introduction into Lothian care homes

    Background The first epidemic wave of severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) in Scotland resulted in high case numbers and mortality in care homes. In Lothian, over one-third of care homes reported an outbreak, while there was limited testing of hospital patients discharged to care homes. Aim To investigate patients discharged from hospitals as a source of SARS-CoV-2 introduction into care homes during the first epidemic wave. Methods A clinical review was performed for all patients discharges from hospitals to care homes from 1st March 2020 to 31st May 2020. Episodes were ruled out based on coronavirus disease 2019 (COVID-19) test history, clinical assessment at discharge, whole-genome sequencing (WGS) data and an infectious period of 14 days. Clinical samples were processed for WGS, and consensus genomes generated were used for analysis using Cluster Investigation and Virus Epidemiological Tool software. Patient timelines were obtained using electronic hospital records. Findings In total, 787 patients discharged from hospitals to care homes were identified. Of these, 776 (99%) were ruled out for subsequent introduction of SARS-CoV-2 into care homes. However, for 10 episodes, the results were inconclusive as there was low genomic diversity in consensus genomes or no sequencing data were available. Only one discharge episode had a genomic, time and location link to positive cases during hospital admission, leading to 10 positive cases in their care home. Conclusion The majority of patients discharged from hospitals were ruled out for introduction of SARS-CoV-2 into care homes, highlighting the importance of screening all new admissions when faced with a novel emerging virus and no available vaccine

    SARS-CoV-2 Omicron is an immune escape variant with an altered cell entry pathway

    Vaccines based on the spike protein of SARS-CoV-2 are a cornerstone of the public health response to COVID-19. The emergence of hypermutated, increasingly transmissible variants of concern (VOCs) threaten this strategy. Omicron (B.1.1.529), the fifth VOC to be described, harbours multiple amino acid mutations in spike, half of which lie within the receptor-binding domain. Here we demonstrate substantial evasion of neutralization by Omicron BA.1 and BA.2 variants in vitro using sera from individuals vaccinated with ChAdOx1, BNT162b2 and mRNA-1273. These data were mirrored by a substantial reduction in real-world vaccine effectiveness that was partially restored by booster vaccination. The Omicron variants BA.1 and BA.2 did not induce cell syncytia in vitro and favoured a TMPRSS2-independent endosomal entry pathway, these phenotypes mapping to distinct regions of the spike protein. Impaired cell fusion was determined by the receptor-binding domain, while endosomal entry mapped to the S2 domain. Such marked changes in antigenicity and replicative biology may underlie the rapid global spread and altered pathogenicity of the Omicron variant
