    Alternative mRNA Editing in Trypanosomes Is Extensive and May Contribute to Mitochondrial Protein Diversity

    The editing of trypanosome mitochondrial mRNAs produces transcripts necessary for mitochondrial functions including electron transport and oxidative phosphorylation. Precursor-mRNAs are often extensively edited by specific uridine insertion or deletion that is directed by small guide RNAs (gRNAs). Recently, it has been shown that cytochrome c oxidase subunit III (COXIII) mRNAs can be alternatively edited to encode a novel mitochondrial membrane protein composed of a unique hydrophilic N-terminal sequence of unknown function and the C-terminal hydrophobic segment of COXIII. To extend the analysis of alternative editing in Trypanosoma brucei we have constructed libraries with over 1100 full-length mitochondrial cDNAs and the sequences of over 1200 gRNA genes. Using this data, we show that alternative editing of COXIII, ATPase subunit 6 (A6), and NADH dehydrogenase subunits 7, 8 and 9 (ND7, 8, 9) mRNAs can produce novel open reading frames (ORFs). Several gRNAs potentially responsible for the alternative editing of these mRNAs were also identified. These findings show that alternative editing of mitochondrial mRNAs is common in T. brucei and expands the diversity of mitochondrial proteins in these organisms

    Analysis of cell wall proteins regulated in stem of susceptible and resistant tomato species after inoculation with Ralstonia solanacearum: a proteomic approach

    Proteomics approach was used to elucidate the molecular interactions taking place at the stem cell wall level when tomato species were inoculated with Ralstonia solanacearum, a causative agent of bacterial wilt. Cell wall proteins from both resistant and susceptible plants before and after the bacterial inoculation were extracted from purified cell wall with salt buffers and separated with 2-D IEF/SDS–PAGE and with 3-D IEF/SDS/SDS–PAGE for basic proteins. The gels stained with colloidal Coomassie revealed varied abundance of protein spots between two species (eight proteins in higher abundance in resistant and six other in susceptible). Moreover, proteins were regulated differentially in response to bacterial inoculation in resistant (seven proteins increased and eight other decreased) as well as in susceptible plants (five proteins elevated and eight other suppressed). Combination of MALDI-TOF/TOF MS and LC-ESI-IonTrap MS/MS lead to the identification of those proteins. Plants responded to pathogen inoculation by elevating the expression of pathogenesis related, other defense related and glycolytic proteins in both species. However, cell wall metabolic proteins in susceptible, and antioxidant, stress related as well as energy metabolism proteins in resistant lines were suppressed. Most of the proteins of the comparative analysis and other randomly picked spots were predicted to have secretion signals except some classical cytosolic proteins

    Construção e validação de vídeo educativo para orientação de pais de crianças em cateterismo intermitente limpo

    RESUMO Objetivo: Construir e validar vídeo educativo para orientação de pais de crianças em cateterismo intermitente limpo. Método: Estudo metodológico, desenvolvido em duas etapas: construção e validação de vídeo com juízes expertises no período de março a dezembro de 2016. A construção da tecnologia teve como referencial teórico a Teoria de Adaptação. Para análise dos dados utilizou-se do índice de correlação intraclasse. Resultados: A primeira versão do vídeo foi composta por 12 minutos, a validação indicou a substituição de termos técnicos por linguagem coloquial, dinamismo nos diálogos e menção à lavagem das mãos antes de reunir o material. No que se refere à confiabilidade total, o coeficiente de correlação intraclasse para todas as categorias avaliadas obteve o valor de 0,768, considerado razoável. Na avaliação de clareza de linguagem, pertinência à prática e relevância teórica, os resultados foram 0,745, 0,771 e 0,777, respectivamente, considerados razoáveis, com p<0,0001. Conclusão: O vídeo educativo mostrou-se válido quanto à aparência e ao conteúdo, com potencial para mediar práticas educativas em contexto hospitalar e ambulatorial
