    Validation of Web Accessibility

    Tato práce se zabývá přístupností webových stránek a možnostmi její automatizované validace. Poskytuje základní pohled na přístupnost jako takovou a identifikuje postižení, kterými mohou uživatelé internetu trpět. Jádrem práce je analýza a srovnání současných metodik tvorby přístupných webových stránek platných v USA, Evropské unii a v České republice. Na tuto analýzu navazuje průzkum existujících validátorů přístupnosti, které jsou rozčleněny do kategorií online nástroje, toolbary a desktopové aplikace. Výsledkem práce je návrh a implementace vlastního validačního nástroje.The subject matter of this project is accessibility of web pages and possibilities of its automated validation. This thesis defines what web accessibility really is and identifies what disabilities can users of the Internet suffer from. The main goal is to analyse and to compare current web accessibility rules valid in the USA, in the European Union and in the Czech Republic and to research existing validators as well. The tools are divided into the following categories: Online tools, toolbars and desktop applications. The output of this project is in form of specification and implementation of a new validation tool.

    Import 22/07/2015Cílem diplomové práce je co možná nejpřesněji vymezit problematiku, která se týká bezpečnosti a ochrany zdraví při práci a pracovněprávní odpovědnosti zaměstnavatele. Teoretický oddíl práce obsahuje popis obecné problematiky odpovědnosti za škodu a její rozdělení na škodu a nemajetkovou újmu, dále jsou vylíčeny odpovědnosti za škodu dle zákoníku práce, její druhy a systém BOZP. Stěžejní část tvoří odpovědnost zaměstnavatele za pracovní úrazy a nemoci z povolání, kde bude vymezen rozsah této odpovědnosti, podmínky zproštění, způsoby a míra náhrady škody. V praktické části jsou aplikovány poznatky z teoretického oddílu na společnosti OKD, a.s. Dle interní směrnice firmy jsou zkoumány zásady pro bezpečnost a ochranu zdraví při práci. V závěru kapitoly je provedena analýza vzniku a počtu pracovních úrazů a nemocí z povolání z hlediska časového a prostorového.The goal of this thesis is to define as precisely as possible the issues concerning health and safety at work and labor-employer liability. Theoretical section contains a description of the general issue of liability and its distribution to damage and non-material damage, are also portrayed liability for damages according to the Labor Code, its types and OSHA system. The main topic is the employer's responsibility for accidents at work and occupational diseases, which will define the scope of this responsibility, the conditions of the waiver, and ways to measure damages. In the practical part there are the findings from a theoretical section on the company OKD, a.s. According to the company's internal guidelines are explored principles for occupational safety and health protection. The chapter concludes with an analysis and creation of occupational injuries and illnesses in terms of time and space.119 - Katedra právavýborn

    Tento diplomový projekt se zabývá systémem dolování z dat v prostředí Oracle. Jedná se o klientskou aplikaci, využívající služeb serveru Oracle Data Mining Server 10.g Release 2 (10.2). Je naprogramovaná v jazyce Java, grafické uživatelské rozhraní je po staveno na platformě NetBeans. Teoretická část práce uvede čtenáře do problematiky získávání znalostí, v praktické části je nejdříve shrnuta funkce původní verze systému, jsou popsány jeho nedostatky, zdokumentovány změny, vedoucí k jejich řešení a navržen další rozvoj. Cílem projektu je upravit dolovací systém tak, aby se zvýšila použitelnost aplikace.This MSc Project deals with the system of Knowledge Discovery in Databases. It is a client application which uses the Oracle Data Mining Server's 10.g Release 2 (10.2) services. The application is implemented in Java, the graphical user interface is built on the NetBeans Rich Client Platform. The theoretical part introduces the Knowledge Discovery in Databases, while the practical part describes functionality of the original system, it's deficiencies, documents sollutions of theese deficiencies and there are proposed improvements for further development. The goal of this project is to modify the system to increase the application usability.

    Karpatian and Lower Badenian deposits were newly exposed by a series of wells in the area of Černovice. Lower Badenian deposits (“Brno sands”) were interpreted as deposits of coarse-grained delta. The source area was the “passive” basin margin (the European platform, Brno Massif). The Karpatian strata represent deposits of the inner and outer shelf zone.     Rapid changes in gene expression: DNA determinants of promoter regulation by the concentration of the transcription initiating NTP in Bacillus subtilis

    In bacteria, rapid changes in gene expression can be achieved by affecting the activity of RNA polymerase with small molecule effectors during transcription initiation. An important small molecule effector is the initiating nucleoside triphosphate (iNTP). At some promoters, an increasing iNTP concentration stimulates promoter activity, while a decreasing concentration has the opposite effect. Ribosomal RNA (rRNA) promoters from Gram-positive Bacillus subtilis are regulated by the concentration of their iNTP. Yet, the sequences of these promoters do not emulate the sequence characteristics of [iNTP]-regulated rRNA promoters of Gram-negative Escherichia coli. Here, we identified the 3′-promoter region, corresponding to the transcription bubble, as key for B. subtilis rRNA promoter regulation via the concentration of the iNTP. Within this region, the conserved −5T (3 bp downstream from the −10 hexamer) is required for this regulation. Moreover, we identified a second class of [iNTP]-regulated promoters in B. subtilis where the sequence determinants are not limited to the transcription bubble region. Overall, it seems that various sequence combinations can result in promoter regulation by [iNTP] in B. subtilis. Finally, this study demonstrates how the same type of regulation can be achieved with strikingly different promoter sequences in phylogenetically distant species

    ArelA-dependent regulatory cascade for auto-induction of microbisporicin production inMicrobispora corallina

    Microbisporicin is a potent type I lantibiotic produced by the rare actinomycete M icrobispora corallina that is in preclinical trials for the treatment of infections caused by methicillin‐resistant isolates of S taphylococcus aureus (MRSA). Analysis of the gene cluster for the biosynthesis of microbisporicin, which contains two unique post‐translationally modified residues (5‐chlorotryptophan and 3, 4‐dihydroxyproline), has revealed an unusual regulatory mechanism that involves a pathway‐specific extracytoplasmic function sigma factor (MibX)/anti‐sigma factor (MibW) complex and an additional transcriptional regulator MibR. A model for the regulation of microbisporicin biosynthesis derived from transcriptional, mutational and quantitative reverse transcription polymerase chain reaction analyses suggests that MibR, which contains a C‐terminal DNA‐binding domain found in the LuxR family of transcriptional activators, functions as an essential master regulator to trigger microbisporicin production while MibX and MibW induce feed‐forward biosynthesis and producer immunity. Moreover, we demonstrate that initial expression of mib R, and thus microbisporicin production, is dependent on the ppGpp synthetase gene (relA) of M . corallina. In addition, we show that constitutive expression of either of the two positively acting regulatory genes, mib R or mib X, leads to precocious and enhanced microbisporicin production

    High dimensional and high resolution pulse sequences for backbone resonance assignment of intrinsically disordered proteins

    Four novel 5D (HACA(N)CONH, HNCOCACB, (HACA)CON(CA)CONH, (H)NCO(NCA)CONH), and one 6D ((H)NCO(N)CACONH) NMR pulse sequences are proposed. The new experiments employ non-uniform sampling that enables achieving high resolution in indirectly detected dimensions. The experiments facilitate resonance assignment of intrinsically disordered proteins. The novel pulse sequences were successfully tested using δ subunit (20 kDa) of Bacillus subtilis RNA polymerase that has an 81-amino acid disordered part containing various repetitive sequences

    Influence of organic molecules on the aggregation of TiO2 nanoparticles in acidic conditions

    Engineered nanoparticles released into the environment may interact with natural organic matter (NOM). Surface complexation affects the surface potential, which in turn may lead to aggregation of the particles. Aggregation of synthetic TiO2 (anatase) nanoparticles in aqueous suspension was investigated at pH 2.8 as a function of time in the presence of various organic molecules and Suwannee River fulvic acid (SRFA), using dynamic light scattering (DLS) and high-resolution transmission electron microscopy (TEM). Results showed that the average hydrodynamic diameter and ?-potential were dependent on both concentration and molecular structure of the organic molecule. Results were also compared with those of quantitative batch adsorption experiments. Further, a time study of the aggregation of TiO2 nanoparticles in the presence of 2,3-dihydroxybenzoic acid (2,3-DHBA) and SRFA, respectively, was performed in order to observe changes in ?-potential and particle size over a time period of 9 months. In the 2,3-DHBA-TiO2 system, ?-potentials decreased with time resulting in charge neutralization and/or inversion depending on ligand concentration. Aggregate sizes increased initially to the micrometer size range, followed by disaggregation after several months. No or very little interaction between SRFA and TiO2 occurred at the lowest concentrations tested. However, at the higher concentrations of SRFA, there was an increase in both aggregate size and the amount of SRFA adsorbed to the TiO2 surface. This was in correlation with the ?-potential that decreased with increased SRFA concentration, leading to destabilization of the system. These results stress the importance of performing studies over both short and long time periods to better understand and predict the long-term effects of nanoparticles in the environment
