Etude des gènes Dlx5/6 et Foxl2 dans le développement et la fonction de l'utérus chez la souris

My thesis is concerned with the involvement of Foxl2, Dlx5 and Dlx6 genes in the development and function of the uterus.In the uterus, Foxl2 expression is visible from birth in the mesenchyme and during the postnatal maturation in the stroma and the myometrium. The conditional invalidation of Foxl2 by the progesterone receptor (Pgr cre / +) causes infertility. The uterus of these mice has a decrease of the stroma thickness and the myometrial layer is hypertrophic and disorganized. The conditional mutants lack the smooth muscle cell layer surrounding the blood vessels in the stroma. The study by qPCR shows that inactivation of Foxl2 deregulates the expression of certain Wnt genes involved in the maturation of the uterus.Dlx5 and Dlx6 meanwhile, are present in the uterine epithelium from E15.5. The conditional invalidation of Dlx5 and Dlx6 by the action of Pgr cre / + induced infertility. These mice present abnormally large invaginations and a very small number of uterine glands. These defects are related to a deregulation of key genes in the formation of glands like Wnt7a and Foxa2.In conclusion, our data show that Foxl2 has a crucial role in the differentiation and organization of the stroma and myometrium. Comparative analysis of molecular signatures suggests the use of gabapentin for the management of endometriosis-associated pain

Background: It has been repetitively shown that the transcription factors DLX5 and DLX6 are drastically downregulated in endometriotic lesions when compared with eutopic endometrium. These findings suggest that regulatory cascades involving DLX5/6 might be at the origin of endometriosis symptoms such as chronic pelvic pain (CPP). We have shown that inactivation of Dlx5 and Dlx5/6 in the mouse uterus results in an endometrial phenotype reminiscent of endometriosis. Methods: We focused on genes that present a similar deregulation in endometriosis and in Dlx5/6-null mice in search of new endometriosis targets. Results: We confirmed a strong reduction of DLX5 expression in endometriosis implants. We identified a signature of 30 genes similarly deregulated in human endometriosis implants and in Dlx5/6-null mouse uteri, reinforcing the notion that the downregulation of Dlx5/6 is an early event in the progress of endometriosis. CACNA2D3, a component of the \u3b12\u3b4 family of voltagedependent calcium channel complex, was strongly overexpressed both in mutant mouse uteri and in endometriosis implants, were also CACNA2D1 and CACNA2D2, other members of the \u3b12\u3b4 family involved in nociception, are upregulated. Conclusion: Comparative analysis of gene expression signatures from endometriosis and mouse models showed that calcium channel subunits \u3b12\u3b4 involved in nociception can be targets for the treatment of endometriosis-associated pain. CACNA2D3 has been associated with pain sensitization and heat nociception in animal models. In patients, CACNA2D3 variants were associated with reduced sensitivity to acute noxious stimuli. As \u3b12\u3b4s were targets of gabapentinoid analgesics, the results suggested the use of these drugs for the treatment of endometriosis-associated pain. Indeed, recent small-scale clinical studies have shown that gabapentin could be effective in women with CPP. The findings of this study reinforce the need for a large definitive trial

In 1885 the University of Oxford invited applications for the newly created Merton Professorship of English Language and Literature. The holder of the chair was, according to the statutes, to ‘lecture and give instruction on the broad history and criticism of English Language and Literature, and on the works of approved English authors’. This was not in itself a particularly innovatory move, as the study of English vernacular literature had played some part in higher education in Britain for over a century. Oxford University had put English as a subject into its pass degree in 1873, had been participating since 1878 in extension teaching, of which literary study formed a significant part, and had since 1881 been setting special examinations in the subject for its non-graduating women students. What was new was the fact that this ancient university appeared to be on the verge of granting the solid academic legitimacy of an established chair to an institutionally marginal and often contentious intellectual pursuit, acknowledging the study of literary texts in English to be a fit subject not just for women and the educationally disadvantaged but also for university men

To define coastal amenities, the engineer has first to work out a diagnosis for hydraulic and sedimentologic processes (What ? How ?, How much ? Budget), and then examine various solutions. To reach these objectives, he uses methods of study extending from natural data to physical and/or numerical models. Contributions on the limits of these various methods are examined.Pour définir un aménagement du littoral, l'ingénieur doit, en premier lieu, établir un diagnostic quant au régime hydraulique et sédimentologique (Quoi ? French shore general view about impacts of various kinds of improvments. Rules of improvment About 1 000 structures perpendicular to shore (50 km cumulated), 500 along shore (400 km cumulated) and 300 harbor or river mouth structures. These figures illustrate the magnitude of works on the french coastline and that of their associated impacts. After examining the shore natural evolution parameters, examples of impacts due to the type of structure with respect to the local littoral regime are considered. Then protection principles are proposed.Environ 1 000 ouvrages transversaux (50 km développés), 500 longitudinaux (400 km développés) et 300 ouvrages portuaires ou de débouchés en mer. Ce bilan illustre l'importance des aménagements sur le littoral français et aussi ceux des impacts qui leur sont associés. Après avoir examiné les facteurs d'évolution naturels du littoral, il a été abordé, à l'aide d'exemples, les impacts qui découlent d'une part du régime littoral existant et d'autre part du type d'aménagement envisagé.
