    Rescue of Salivary Gland Function after Stem Cell Transplantation in Irradiated Glands

    Head and neck cancer is the fifth most common malignancy and accounts for 3% of all new cancer cases each year. Despite relatively high survival rates, the quality of life of these patients is severely compromised because of radiation-induced impairment of salivary gland function and consequential xerostomia (dry mouth syndrome). In this study, a clinically applicable method for the restoration of radiation-impaired salivary gland function using salivary gland stem cell transplantation was developed. Salivary gland cells were isolated from murine submandibular glands and cultured in vitro as salispheres, which contained cells expressing the stem cell markers Sca-1, c-Kit and Musashi-1. In vitro, the cells differentiated into salivary gland duct cells and mucin and amylase producing acinar cells. Stem cell enrichment was performed by flow cytrometric selection using c-Kit as a marker. In vitro, the cells differentiated into amylase producing acinar cells. In vivo, intra-glandular transplantation of a small number of c-Kit+ cells resulted in long-term restoration of salivary gland morphology and function. Moreover, donor-derived stem cells could be isolated from primary recipients, cultured as secondary spheres and after re-transplantation ameliorate radiation damage. Our approach is the first proof for the potential use of stem cell transplantation to functionally rescue salivary gland deficiency

    Pyoderma gangrenosum after totally implanted central venous access device insertion

    <p>Abstract</p> <p>Background</p> <p>Pyoderma gangrenosum is an aseptic skin disease. The ulcerative form of pyoderma gangrenosum is characterized by a rapidly progressing painful irregular and undermined bordered necrotic ulcer. The aetiology of pyoderma gangrenosum remains unclear. In about 70% of cases, it is associated with a systemic disorder, most often inflammatory bowel disease, haematological disease or arthritis. In 25–50% of cases, a triggering factor such as recent surgery or trauma is identified. Treatment consists of local and systemic approaches. Systemic steroids are generally used first. If the lesions are refractory, steroids are combined with other immunosuppressive therapy or to antimicrobial agents.</p> <p>Case presentation</p> <p>A 90 years old patient with myelodysplastic syndrome, seeking regular transfusions required totally implanted central venous access device (Port-a-Cath<sup>®</sup>) insertion. Fever and inflammatory skin reaction at the site of insertion developed on the seventh post-operative day, requiring the device's explanation. A rapid progression of the skin lesions evolved into a circular skin necrosis. Intravenous steroid treatment stopped the necrosis' progression.</p> <p>Conclusion</p> <p>Early diagnosis remains the most important step to the successful treatment of pyoderma gangrenosum.</p

    Public responses to CO2 storage sites: Lessons from five European cases

    Studies of the factors involved in public perceptions of CO2 storage projects reveal a level of complexity and diversity that arguably confounds a comprehensive theoretical account. To some extent, a conceptual approach that simply organises the relevant social scientific knowledge thematically, rather than seeking an integrated explanation, is as useful as any single account that fails to do justice to the contingencies involved. This paper reviews and assembles such knowledge in terms of six themes and applies these themes to five European cases of carbon capture and storage (CCS) implementation. We identify the main factors involved in community responses to CCS as relating to: the characteristics of the project; the engagement process; risk perceptions; the actions of the stakeholders; the characteristics of the community, and the socio-political context

    The future of medical diagnostics: Review paper

    While histopathology of excised tissue remains the gold standard for diagnosis, several new, non-invasive diagnostic techniques are being developed. They rely on physical and biochemical changes that precede and mirror malignant change within tissue. The basic principle involves simple optical techniques of tissue interrogation. Their accuracy, expressed as sensitivity and specificity, are reported in a number of studies suggests that they have a potential for cost effective, real-time, in situ diagnosis. We review the Third Scientific Meeting of the Head and Neck Optical Diagnostics Society held in Congress Innsbruck, Innsbruck, Austria on the 11th May 2011. For the first time the HNODS Annual Scientific Meeting was held in association with the International Photodynamic Association (IPA) and the European Platform for Photodynamic Medicine (EPPM). The aim was to enhance the interdisciplinary aspects of optical diagnostics and other photodynamic applications. The meeting included 2 sections: oral communication sessions running in parallel to the IPA programme and poster presentation sessions combined with the IPA and EPPM posters sessions. © 2011 Jerjes et al; licensee BioMed Central Ltd

    CO2 wettability of seal and reservoir rocks and the implications for carbon geo-sequestration

    We review the literature data published on the topic of CO2 wettability of storage and seal rocks. We first introduce the concept of wettability and explain why it is important in the context of carbon geo-sequestration (CGS) projects, and review how it is measured. This is done to raise awareness of this parameter in the CGS community, which, as we show later on in this text, may have a dramatic impact on structural and residual trapping of CO2. These two trapping mechanisms would be severely and negatively affected in case of CO2-wet storage and/or seal rock. Overall, at the current state of the art, a substantial amount of work has been completed, and we find that: 1. Sandstone and limestone, plus pure minerals such as quartz, calcite, feldspar, and mica are strongly water wet in a CO2-water system. 2. Oil-wet limestone, oil-wet quartz, or coal is intermediate wet or CO2 wet in a CO2-water system. 3. The contact angle alone is insufficient for predicting capillary pressures in reservoir or seal rocks. 4. The current contact angle data have a large uncertainty. 5. Solid theoretical understanding on a molecular level of rock-CO2-brine interactions is currently limited. 6. In an ideal scenario, all seal and storage rocks in CGS formations are tested for their CO2 wettability. 7. Achieving representative subsurface conditions (especially in terms of the rock surface) in the laboratory is of key importance but also very challenging
