Wnt/β-catenin signaling stimulates the expression and synaptic clustering of the autism-associated Neuroligin 3 gene
Indexación: Scopus.Synaptic abnormalities have been described in individuals with autism spectrum disorders (ASD). The cell-adhesion molecule Neuroligin-3 (Nlgn3) has an essential role in the function and maturation of synapses and NLGN3 ASD-associated mutations disrupt hippocampal and cortical function. Here we show that Wnt/β-catenin signaling increases Nlgn3 mRNA and protein levels in HT22 mouse hippocampal cells and primary cultures of rat hippocampal neurons. We characterized the activity of mouse and rat Nlgn3 promoter constructs containing conserved putative T-cell factor/lymphoid enhancing factor (TCF/LEF)-binding elements (TBE) and found that their activity is significantly augmented in Wnt/β-catenin cell reporter assays. Chromatin immunoprecipitation (ChIP) assays and site-directed mutagenesis experiments revealed that endogenous β-catenin binds to novel TBE consensus sequences in the Nlgn3 promoter. Moreover, activation of the signaling cascade increased Nlgn3 clustering and co-localization with the scaffold PSD-95 protein in dendritic processes of primary neurons. Our results directly link Wnt/β-catenin signaling to the transcription of the Nlgn3 gene and support a functional role for the signaling pathway in the dysregulation of excitatory/inhibitory neuronal activity, as is observed in animal models of ASD.https://www.nature.com/articles/s41398-018-0093-y.pd
Silício No Desenvolvimento In Vitro Da Orquídea Cattleya Forbesii
Addition of Silicon (Si) to culture media has been shown to improve the development of seedlings grown in vitro, and to reduce losses during the acclimatization phase. The objective of this study was to evaluate the in vitro growth of Cattleya forbesii (Orchidaceae) in MS medium containing five different concentrations of SiO2 (0.0, 0.5, 1.0, 1.5, and 2.0 g·L−1). At day 200, the following variables were measured: number of roots, average length of the root system, leaf area, number of leaves and shoots, shoot height, fresh and dry masses of roots and shoots, water content of roots and shoots, and pH of the culture medium. Most variables decreased as the concentration of Si increased, reducing the in vitro vegetative growth of C. forbesii. Accumulation of Si in leaf tissues was detected by scanning electron microscopy, confirming uptake by plants. The Si source and concentrations tested showed no beneficial effect on in vitro growth of C. forbesii. © 2016, Universidade Federal Rural do Semi-Arid. All rights reserved.291182
Estudos sobre a nutrição mineral do cafeeiro: XXXI - influência do cobre do substrato no crescimento e composição mineral do cafeeiro (Coffea arabica L.) var. Mundo Novo e Catuaí
This work was designed to study the effect of copper in the medium on growth and mineral composition of the coffee plant, bearing in mind the possibility of accumulation of toxic residues in the soil as a consequence of the treatments against rust. - Young plants of the varieties Mundo Novo and Catuaí were grown in nutrient solution in the presence of Cu concentrations from 0.02 to 200 ppm. Toxic effects were found to be darkening and cessation of root growth; wilt, necrosis and leaf drop; abnormalities in the epidermis, parenchyma and chloroplasts. The threshold level for copper toxicity in around 30 ppm in the leaves. Mundo Novo seems to be more sensitive to copper toxicity. - Under soil conditions it was found that the joint use of lime and stable manure eliminates the toxicity of an application of 200 ppm of copper.A sensibilidade das variedades Catuaí e Mundo Novo ao nível de cobre no substrato foi estudada em condições controladas. Disorder and thermally driven vortex-lattice melting in La{2-x}Sr{x}CuO{4} crystals
Magnetization measurements in La{2-x}Sr{x}CuO{4} crystals indicate vortex
order-disorder transition manifested by a sharp kink in the second
magnetization peak. The transition field exhibits unique temperature
dependence, namely a strong decrease with temperature in the entire measured
range. This behavior rules out the conventional interpretation of a
disorder-driven transition into an entangled vortex solid phase. It is shown
that the transition in La{2-x}Sr{x}CuO{4} is driven by both thermally- and
disorder-induced fluctuations, resulting in a pinned liquid state. We conclude
that vortex solid-liquid, solid-solid and solid to pinned-liquid transitions
are different manifestations of the same thermodynamic order-disorder
transition, distinguished by the relative contributions of thermal and

Circular orbits of corotating binary black holes: comparison between analytical and numerical results
We compare recent numerical results, obtained within a ``helical Killing
vector'' (HKV) approach, on circular orbits of corotating binary black holes to
the analytical predictions made by the effective one body (EOB) method (which
has been recently extended to the case of spinning bodies). On the scale of the
differences between the results obtained by different numerical methods, we
find good agreement between numerical data and analytical predictions for
several invariant functions describing the dynamical properties of circular
orbits. This agreement is robust against the post-Newtonian accuracy used for
the analytical estimates, as well as under choices of resummation method for
the EOB ``effective potential'', and gets better as one uses a higher
post-Newtonian accuracy. These findings open the way to a significant
``merging'' of analytical and numerical methods, i.e. to matching an EOB-based
analytical description of the (early and late) inspiral, up to the beginning of
the plunge, to a numerical description of the plunge and merger. We illustrate
also the ``flexibility'' of the EOB approach, i.e. the possibility of
determining some ``best fit'' values for the analytical parameters by

Impact of regulated and emerging pollutants and microplastics in marine ecosystems (IMPACTA project)
Marine ecosystems are nowadays subjected to a massive input of synthetic chemicals from everywhere. Unfortunately only a small portion of them is being monitored, and it is necessary to identify which pollutants can produce adverse impacts in the marine environment. IMPACTA project (CTM2013-48194-C3) is characterizing the distribution of regulated and emerging contaminants and microplastics in marine sediments, and evaluating the biological effects that they can cause (sing sublethal embryotoxicity tests, endocrine disruption and biomarkers). Sensitive and selective analytical methods are being developed and validated for pharmaceuticals, perfluorinated compounds, organophosphorus pesticides, triazines, personal care products, nonylphenols and alkylated PAHs in sediments. Thus, relevant pollutants present in coastal and offshore areas will be identified. Furthermore potential toxic effects of the contaminants present in coastal sediments are being assessed through embryotoxicity bioassays and also the biological effects on different marine species as a consequence of their exposition to specific compounds.
Another relevant contribution of this project will be the assessment of the impact of micro-plastics, first time in the Spanish coastal areas. Their potential toxic effects and their role in the transference of pollutants in the marine environment are being assessed. The concentration and composition of microplastics in sediments and demersal fish stomachs are being characterized, and their potential biological effects on marine invertebrates are also being investigated
Profile of micronucleus frequencies and DNA damage in different species of fish in a eutrophic tropical lake
Lake Paranoá is a tropical reservoir for the City of Brasilia, which became eutrophic due to inadequate sewage treatment associated with intensive population growth. At present, two wastewater treatment plants are capable of processing up to 95% of the domestic sewage, thereby successfully reducing eutrophization. We evaluated both genotoxic and cytotoxic parameters in several fish species (Geophagus brasiliensis, Cichla temensis, Hoplias malabaricus, Astyanax bimaculatus lacustres, Oreochromis niloticus, Cyprinus carpio and Steindachnerina insculpita) by using the micronucleus (MN) test, the comet assay and nuclear abnormality assessment in peripheral erythrocytes. The highest frequencies of MN were found in Cichla temensis and Hoplias malabaricus, which were statistically significant when compared to the other species. However, Steindachnerina insculpita (a detritivorous and lake-floor feeder species) showed the highest index of DNA damage in the comet assay, followed by C. temensis (piscivorous). Nuclear abnormalities, such as binucleated, blebbed, lobed and notched cells, were used as evidence of cytotoxicity. Oreochromis niloticus followed by Hoplias malaricus, ominivorous/detritivotous and piscivorous species, respectively, presented the highest frequency of nuclear abnormalities, especially notched cells, while the herbivorous Astyanax bimaculatus lacustres showed the lowest frequency compared to the other species studied. Thus, for biomonitoring aquatic genotoxins under field conditions, the food web should also be considered
Search for direct production of charginos and neutralinos in events with three leptons and missing transverse momentum in √s = 7 TeV pp collisions with the ATLAS detector
A search for the direct production of charginos and neutralinos in final states with three electrons or muons and missing transverse momentum is presented. The analysis is based on 4.7 fb−1 of proton–proton collision data delivered by the Large Hadron Collider and recorded with the ATLAS detector. Observations are consistent with Standard Model expectations in three signal regions that are either depleted or enriched in Z-boson decays. Upper limits at 95% confidence level are set in R-parity conserving phenomenological minimal supersymmetric models and in simplified models, significantly extending previous results
Jet size dependence of single jet suppression in lead-lead collisions at sqrt(s(NN)) = 2.76 TeV with the ATLAS detector at the LHC
Measurements of inclusive jet suppression in heavy ion collisions at the LHC
provide direct sensitivity to the physics of jet quenching. In a sample of
lead-lead collisions at sqrt(s) = 2.76 TeV corresponding to an integrated
luminosity of approximately 7 inverse microbarns, ATLAS has measured jets with
a calorimeter over the pseudorapidity interval |eta| < 2.1 and over the
transverse momentum range 38 < pT < 210 GeV. Jets were reconstructed using the
anti-kt algorithm with values for the distance parameter that determines the
nominal jet radius of R = 0.2, 0.3, 0.4 and 0.5. The centrality dependence of
the jet yield is characterized by the jet "central-to-peripheral ratio," Rcp.
Jet production is found to be suppressed by approximately a factor of two in
the 10% most central collisions relative to peripheral collisions. Rcp varies
smoothly with centrality as characterized by the number of participating
nucleons. The observed suppression is only weakly dependent on jet radius and
transverse momentum. These results provide the first direct measurement of
inclusive jet suppression in heavy ion collisions and complement previous
measurements of dijet transverse energy imbalance at the LHC.Comment: 15 pages plus author list (30 pages total), 8 figures, 2 tables,
