    Secretome of bovine amniotic and endometrial cells: application for in vitro embryo production.

    Some maternal miRNAs are involved in early stage embryos [Abd El Naby, 2011]. Microvesicles (MVs)have been suggested as carrier of miRNAs for maternal-to-embryonic communication during the firstdays of early development [Mondou, 2012]. MVs, together with soluble factors, are components ofconditioned media (CM) produced by cells during their culture. Aim of this study was to understand therole of CM, MVs and supernatant (SN, obtained from CM deprived of MVs) secreted by bovineendometrial and amnion cells on embryo development.In vitro produced embryos were cultured in SOFaa alone (CTR) or supplemented on day 5 postfertilizationwith 10% of endometrial or amniotic CM or SN or 100x106 MVs/ml.The blastocyst rate obtained culturing embryos with amniotic CM and MVs was not significantlydifferent from the CTR (34.17±3.29%, 32.82±3.26% and 35.45±2.53% respectively). The rate obtained byamniotic SN was 25.80±2.83% and statistically lower (P<0.05) than the other groups. The rate obtainedby endometrial products were lower than CTR and the other conditions. The ICM of embryos culturedin medium supplemented with amniotic components had a higher number of cells than the CTR group:30.4±1.83 and 29.42±1.27 for CM and MVs respectively compared to 27.6±1.44 for CTR (P<0.05).Our data showed that exposing the embryos to the amniotic secretome does not improve theblastocyst rate, but increases their quality. The hypothesis is that miRNAs contained into MVs maycontribute to the production of better quality embryos and that amniotic secretome supplies a morephysiological environment for the embryo development

    Microvesicles secreted from equine amniotic cells and their potential role in in vitro cell tendon repair

    The regenerative mechanisms ascribed to mesenchymal stem cells (MSCs) are classified into 3 categories: differentiating into damaged cell types, supplying nutrients, and improving survival/functions of the endogenous cells via paracrine actions. However, because of the inhospitable microenvironment of the injured tissues, a proportion of the implanted MSCs may quickly die, suggesting that other mechanisms might be present. This notion is supported by the overlapping beneficial effect (in terms of time of healing) resulted  after the injection of AMCs or of amniotic mesenchymal cells - conditioned medium (AMC-CM)  in equine spontaneous injured tendons and ligaments. Microvesicles (MVs) released by cells are an integral component of the cell-to-cell communication network involved in tissue regeneration.In the present study, MVs secreted by AMCs were investigated with Nanosigth instrument and TEM. Then, the in vitro incorporation of MVs into equine tendon cells was studied by a dose-response curve. Lastly, the ability of MVs to counteract an in vitro inflammatory process induced by lipolysaccaride on tendon cells was studied evaluating the expression of pro-inflammatory genes like metallopeptidase (MPP) 1 and 13, and prostaglandin-endoperoxide synthase 2 (COX2). Results demonstrated that AMCs secreted MVs ranging in size from 100 to 1000 nm with a prevalence of 100-200 nm large MVs. Tendon cells were able to uptake them with an inverse relationship between concentration and time. The greatest incorporation was detectable at 40x106 MVs/ml after 72h. MVs induced down-regulation of MMP1 and MMP13, suggesting that they may have contributed, along with soluble factors, to in vivo tendon regeneration

    Intervención pedagógica de la fluidez y de la comprensión lectora en el 2º grado de primaria a través de talleres de mediación entre pares

    Con el objetivo de generar mejoras en la fluidez (precisión, velocidad lectora, prosodia y automatización de la lectura) y la comprensión lectora en estudiantes, a través de un diseño cuasi experimental pre test post test con grupo control. Se realizó una intervención pedagógica basada en Talleres de Mediación Entre Pares en una muestra de estudiantes de 2º grado de Primaria de Lima (grupo experimental = 127, y grupo control = 169). Los instrumentos de medición aplicados fueron: la Rúbrica de Lectura Oral RELO (fluidez y la comprensión lectora en la lectura oral) de Medina (2013) y la Prueba de Complejidad Lingüística Progresiva Nivel 2 (comprensión lectora silenciosa) de Alliende, Condemarín y Milicic (2001). Las medidas de comparativas de post test señalan una mejora significativa (p<.001) tras la intervención en la fluidez lectora, la precisión, la automaticidad de la lectura; muy significativa (p<.01) en la prosodia, y ningún efecto significativo sobre la velocidad lectora. No obstante, se aprecian incrementos en los promedios de comprensión lectora en la lectura oral, y en la comprensión a nivel literal y a nivel reorganizativo, estos no alcanzan a ser significativos, sin embargo, los análisis sí señalan que la intervención mejoró la comprensión en el nivel inferencial (p<.001) y en el nivel crítico (p<.01); la comprensión lectora a nivel de la lectura silenciosa no observó una mejora significativa. Este estudio muestra la importancia y el impacto de la mediación entre pares para enriquecer los procesos de aprendizaje de la lectura en las primeras etapas, en especial en realidades con menor avance en el aprendizaje de la fluidez y de la comprensión lectora

    Advanced glycation end products (AGEs) are a heterogeneous, complex group of compounds that are formed when reducing sugar reacts in a non-enzymatic way with amino acids in proteins and other macromolecules. This occurs both exogenously (in food) and endogenously (in humans) with greater concentrations found in older adults. While higher AGEs occur in both healthy older adults and those with chronic diseases, research is progressing to both quantify AGEs in food and in people, and to identify mechanisms that would explain why some human tissues are damaged, and others are not. In the last twenty years, there has been increased evidence that AGEs could be implicated in the development of chronic degenerative diseases of aging, such as cardiovascular disease, Alzheimer’s disease and with complications of diabetes mellitus. Results of several studies in animal models and humans show that the restriction of dietary AGEs has positive effects on wound healing, insulin resistance and cardiovascular diseases. Recently, the effect of restriction in AGEs intake has been reported to increase the lifespan in animal models. This paper will summarize the work that has been published for both food AGEs and in vivo AGEs and their relation with aging, as well as provide suggestions for future research

    Corporate philanthropy through the lens of ethical subjectivity

    The dynamic organisational processes in businesses dilute the boundaries between the individual, organisational, and societal drivers of corporate philanthropy. This creates a complex framework in which charitable project selection occurs. Using the example of European tour operators, this study investigates the mechanisms through which companies invest in charitable projects in overseas destinations. Inextricably linked to this is the increasing contestation by local communities as to how they are able to engage effectively with tourism in order to realise the benefits tourism development can bring. This research furthers such debates by exploring the processes through which tour operators facilitate community development through charitable giving. Findings show, with no formal frameworks in existence, project selection depends upon emergent strategies that connect the professional with the personal, with trust being positioned as a central driver of these informal processes. Discretionary responsibilities are reworked through business leaders’ commitment to responsible business practises and the ethical subjectivity guiding these processes

    Clinical Features, Cardiovascular Risk Profile, and Therapeutic Trajectories of Patients with Type 2 Diabetes Candidate for Oral Semaglutide Therapy in the Italian Specialist Care

    Introduction: This study aimed to address therapeutic inertia in the management of type 2 diabetes (T2D) by investigating the potential of early treatment with oral semaglutide. Methods: A cross-sectional survey was conducted between October 2021 and April 2022 among specialists treating individuals with T2D. A scientific committee designed a data collection form covering demographics, cardiovascular risk, glucose control metrics, ongoing therapies, and physician judgments on treatment appropriateness. Participants completed anonymous patient questionnaires reflecting routine clinical encounters. The preferred therapeutic regimen for each patient was also identified. Results: The analysis was conducted on 4449 patients initiating oral semaglutide. The population had a relatively short disease duration (42%  60% of patients, and more often than sitagliptin or empagliflozin. Conclusion: The study supports the potential of early implementation of oral semaglutide as a strategy to overcome therapeutic inertia and enhance T2D management

    Risk portfolio on modern finance has become increasingly technical, requiring the use of sophisticated mathematical tools in both research and practice. Since companies cannot insure themselves completely against risk, as human incompetence in predicting the future precisely that written in Al-Quran surah Luqman verse 34, they have to manage it to yield an optimal portfolio. The objective here is to minimize the variance among all portfolios, or alternatively, to maximize expected return among all portfolios that has at least a certain expected return. Furthermore, this study focuses on optimizing risk portfolio so called Markowitz MVO (Mean-Variance Optimization). Some theoretical frameworks for analysis are arithmetic mean, geometric mean, variance, covariance, linear programming, and quadratic programming. Moreover, finding a minimum variance portfolio produces a convex quadratic programming, that is minimizing the objective function ðð¥with constraintsð ð 𥠥 ðandð´ð¥ = ð. The outcome of this research is the solution of optimal risk portofolio in some investments that could be finished smoothly using MATLAB R2007b software together with its graphic analysis

    Differential cross section measurements for the production of a W boson in association with jets in proton–proton collisions at √s = 7 TeV

    Measurements are reported of differential cross sections for the production of a W boson, which decays into a muon and a neutrino, in association with jets, as a function of several variables, including the transverse momenta (pT) and pseudorapidities of the four leading jets, the scalar sum of jet transverse momenta (HT), and the difference in azimuthal angle between the directions of each jet and the muon. The data sample of pp collisions at a centre-of-mass energy of 7 TeV was collected with the CMS detector at the LHC and corresponds to an integrated luminosity of 5.0 fb[superscript −1]. The measured cross sections are compared to predictions from Monte Carlo generators, MadGraph + pythia and sherpa, and to next-to-leading-order calculations from BlackHat + sherpa. The differential cross sections are found to be in agreement with the predictions, apart from the pT distributions of the leading jets at high pT values, the distributions of the HT at high-HT and low jet multiplicity, and the distribution of the difference in azimuthal angle between the leading jet and the muon at low values.United States.     Impacts of the Tropical Pacific/Indian Oceans on the Seasonal Cycle of the West African Monsoon

    The current consensus is that drought has developed in the Sahel during the second half of the twentieth century as a result of remote effects of oceanic anomalies amplified by local land–atmosphere interactions. This paper focuses on the impacts of oceanic anomalies upon West African climate and specifically aims to identify those from SST anomalies in the Pacific/Indian Oceans during spring and summer seasons, when they were significant. Idealized sensitivity experiments are performed with four atmospheric general circulation models (AGCMs). The prescribed SST patterns used in the AGCMs are based on the leading mode of covariability between SST anomalies over the Pacific/Indian Oceans and summer rainfall over West Africa. The results show that such oceanic anomalies in the Pacific/Indian Ocean lead to a northward shift of an anomalous dry belt from the Gulf of Guinea to the Sahel as the season advances. In the Sahel, the magnitude of rainfall anomalies is comparable to that obtained by other authors using SST anomalies confined to the proximity of the Atlantic Ocean. The mechanism connecting the Pacific/Indian SST anomalies with West African rainfall has a strong seasonal cycle. In spring (May and June), anomalous subsidence develops over both the Maritime Continent and the equatorial Atlantic in response to the enhanced equatorial heating. Precipitation increases over continental West Africa in association with stronger zonal convergence of moisture. In addition, precipitation decreases over the Gulf of Guinea. During the monsoon peak (July and August), the SST anomalies move westward over the equatorial Pacific and the two regions where subsidence occurred earlier in the seasons merge over West Africa. The monsoon weakens and rainfall decreases over the Sahel, especially in August.Peer reviewe

    Search for heavy resonances decaying to two Higgs bosons in final states containing four b quarks

    A search is presented for narrow heavy resonances X decaying into pairs of Higgs bosons (H) in proton-proton collisions collected by the CMS experiment at the LHC at root s = 8 TeV. The data correspond to an integrated luminosity of 19.7 fb(-1). The search considers HH resonances with masses between 1 and 3 TeV, having final states of two b quark pairs. Each Higgs boson is produced with large momentum, and the hadronization products of the pair of b quarks can usually be reconstructed as single large jets. The background from multijet and t (t) over bar events is significantly reduced by applying requirements related to the flavor of the jet, its mass, and its substructure. The signal would be identified as a peak on top of the dijet invariant mass spectrum of the remaining background events. No evidence is observed for such a signal. Upper limits obtained at 95 confidence level for the product of the production cross section and branching fraction sigma(gg -> X) B(X -> HH -> b (b) over barb (b) over bar) range from 10 to 1.5 fb for the mass of X from 1.15 to 2.0 TeV, significantly extending previous searches. For a warped extra dimension theory with amass scale Lambda(R) = 1 TeV, the data exclude radion scalar masses between 1.15 and 1.55 TeV
