MAXI J1659-152 is a bright X-ray transient black-hole candidate binary system discovered in September 2010. We report here on MAXI, RXTE, Swift, and XMM-Newton observations during its 2010/2011 outburst. We find that during the first one and a half week of the outburst the X-ray light curves display drops in intensity at regular intervals, which we interpret as absorption dips. About three weeks into the outbursts, again drops in intensity are seen. These dips have, however, a spectral behaviour opposite to that of the absorption dips, and are related to fast spectral state changes (hence referred to as transition dips). The absorption dips recur with a period of 2.414+/-0.005 hrs, which we interpret as the orbital period of the system. This implies that MAXI J1659-152 is the shortest period black-hole candidate binary known to date. The inclination of the accretion disk with respect to the line of sight is estimated to be 65-80 degrees. We propose the companion to the black-hole candidate to be close to an M5 dwarf star, with a mass and radius of about 0.15-0.25 M_sun and 0.2-0.25 R_sun, respectively. We derive that the companion had an initial mass of about 1.5 M_sun, which evolved to its current mass in about 5-6 billion years. The system is rather compact (orbital separation of larger than ~1.33 R_sun), and is located at a distance of 8.6+/-3.7 kpc, with a height above the Galactic plane of 2.4+/-1.0 kpc. The characteristics of short orbital period and high Galactic scale height are shared with two other transient black-hole candidate X-ray binaries, i.e., XTE J1118+480 and Swift J1735.5-0127. We suggest that all three are kicked out of the Galactic plane into the halo, rather than being formed in a globular cluster.Comment: 20 pages, 14 figures, accepted for publication in A&

UPR Signal Activation by Luminal Sensor Domains

    Get PDF
The unfolded protein response (UPR) is a cell-signaling system that detects the accumulation of unfolded protein within the endoplasmic reticulum (ER) and initiates a number of cellular responses to restore ER homeostasis. The presence of unfolded protein is detected by the ER-luminal sensor domains of the three UPR-transducer proteins IRE1, PERK, and ATF6, which then propagate the signal to the cytosol. In this review, we discuss the various mechanisms of action that have been proposed on how the sensor domains detect the presence of unfolded protein to activate downstream UPR signaling. © 2013 by the authors; licensee MDPI, Basel, Switzerland

XMM-Newton observations reveal the disappearance of the wind in 4U1630-47

We report on XMM-Newton observations of the black hole X-ray binary 4U 1630−47 during its 2012−2013 outburst. The first five observations monitor the source as its luminosity increases across the high−soft state of accretion. In the sixth observation the source has made a transition to an “anomalous” state, characterised by a significant contribution of electron scattering. A thermally/radiatively driven disc wind is present in the first four observations, which becomes more photoionised as the luminosity increases with time. In the fifth observation, the wind is not observed any more as a consequence of strong photoionisation and the low sensitivity of this observation. This overall trend is then consistent with a fully ionised wind causing the electron scattering characteristic of the anomalous state in the sixth observation. A broad iron emission line co-exists with the absorption features from the wind in the first four observations but is not visible in the last two observations. We find that the changes in the state of the wind as measured from modelling the absorption features with a self-consistent warm absorber model are correlated to the changes in the broad iron line. When the latter is modelled with a reflection component we find that the reflection fraction decreases as the illumination increases. We propose that the changes in both the absorption and broad emission lines are caused by the increasing luminosity and temperature of the accretion disc along the soft state. Such changes ultimately enable the transition to a state where the wind is fully ionised and consequently Comptonisation plays a significant role

Optimal mathematical programming and variable neighborhood search for k-modes categorical data clustering

    Get PDF
The conventional k-modes algorithm and its variants have been extensively used for categorical data clustering. However, these algorithms have some drawbacks, e.g., they can be trapped into local optima and sensitive to initial clusters/modes. Our numerical experiments even showed that the k-modes algorithm could not identify the optimal clustering results for some special datasets regardless the selection of the initial centers. In this paper, we developed an integer linear programming (ILP) approach for the k-modes clustering, which is independent to the initial solution and can obtain directly the optimal results for small-sized datasets. We also developed a heuristic algorithm that implements iterative partial optimization in the ILP approach based on a framework of variable neighborhood search, known as IPO-ILP-VNS, to search for near-optimal results of medium and large sized datasets with controlled computing time. Experiments on 38 datasets, including 27 synthesized small datasets and 11 known benchmark datasets from the UCI site were carried out to test the proposed ILP approach and the IPO-ILP-VNS algorithm. The experimental results outperformed the conventional and other existing enhanced k-modes algorithms in literature, updated 9 of the UCI benchmark datasets with new and improved results

Lorentz breaking Effective Field Theory and observational tests

Analogue models of gravity have provided an experimentally realizable test field for our ideas on quantum field theory in curved spacetimes but they have also inspired the investigation of possible departures from exact Lorentz invariance at microscopic scales. In this role they have joined, and sometime anticipated, several quantum gravity models characterized by Lorentz breaking phenomenology. A crucial difference between these speculations and other ones associated to quantum gravity scenarios, is the possibility to carry out observational and experimental tests which have nowadays led to a broad range of constraints on departures from Lorentz invariance. We shall review here the effective field theory approach to Lorentz breaking in the matter sector, present the constraints provided by the available observations and finally discuss the implications of the persisting uncertainty on the composition of the ultra high energy cosmic rays for the constraints on the higher order, analogue gravity inspired, Lorentz violations.Comment: 47 pages, 4 figures. Rational design and synthesis of modified teixobactin analogues: in vitro antibacterial activity against Staphylococcus aureus, Propionibacterium acnes and Pseudomonas aeruginosa

Teixobactin, a recently discovered depsipeptide that binds to bacterial lipid II and lipid III, provides a promising molecular scaffold for the design of new antimicrobials. Herein, we describe the synthesis and antimicrobial evaluation of systematically modified teixobactin analogues. The replacement of Ile11 residue with aliphatic isosteres, the modification of the guanidino group at residue 10 and the introduction of a rigidifying residue, dehydroamino acid into the macrocyclic ring generated useful structure‐activity information. Extensive antimicrobial susceptibility assessment against a panel of clinically relevant Staphylococcus aureus and Propionibacterium acnes led to the identification of a new lead compound, [Arg(Me)10,Nle11]teixobactin 63, with excellent bactericidal activity (MIC 2‐4 μg/mL). Significantly, the antimicrobial activity of several of the teixobactin analogues against the pathogenic Gram‐negative Pseudomonas aeruginosa was 'restored' when combined with sub‐MIC concentration of the outer membrane‐disruptive antibiotic, colistin. The antimicrobial effectiveness of [Tfn10,Nle11]teixobactin 66 (32 μg/mL)‐colistin (2 μg/mL; 0.5x MIC) combination against P. aeruginosa PAO1 reveal, for the first time, an alternative therapeutic option in the treatment of Gram‐negative infections

Abstract

Background

Mucinous ovarian carcinoma have a poorer prognosis compared with other histological subtypes. The aim of this study was to evaluate, retrospectively, the activity of chemotherapy in patients with platinum sensitive recurrent mucinous ovarian cancer.

Methods

The SOCRATES study retrospectively assessed the pattern of care of a cohort of patients with recurrent platinum-sensitive ovarian cancer observed in the years 2000–2002 in 37 Italian centres. Data were collected between April and September 2005. Patients with recurrent ovarian cancer with > 6 months of platinum free interval were considered eligible.

Results

Twenty patients with mucinous histotype and 388 patients with other histotypes were analyzed. At baseline, mucinous tumours differed from the others for an higher number of patients with lower tumor grading (p = 0.0056) and less advanced FIGO stage (p = 0.025). At time of recurrence, a statistically significant difference was found in performance status (worse in mucinous, p = 0.024). About 20% of patients underwent secondary cytoreduction in both groups, but a lower number of patients were optimally debulked in the mucinous group (p = 0.03). Patients with mucinous cancer received more frequently single agent platinum than platinum based-combination therapy or other non-platinum schedules as second line therapy (p = 0.026), with a response rate lower than in non-mucinous group (36.4% vs 62.6%, respectively, p = 0.04). Median time to progression and overall survival were worse for mucinous ovarian cancer. Finally, mucinous cancer received a lower number of chemotherapy lines (p = 0.0023).

Conclusion

This analysis shows that platinum sensitive mucinous ovarian cancer has a poor response to chemotherapy. Studies dedicated to this histological subgroup are needed.</p

Comparative assessment of texture features for the identification of cancer in ultrasound images: a review

    Get PDF
In this paper, we review the use of texture features for cancer detection in Ultrasound (US) images of breast, prostate, thyroid, ovaries and liver for Computer-Aided Diagnosis (CAD) systems. This paper shows that texture features are a valuable tool to extract diagnostically relevant information from US images. This information helps practitioners to discriminate normal from abnormal tissues. A drawback of some classes of texture features comes from their sensitivity to both changes in image resolution and grayscale levels. These limitations pose a considerable challenge to CAD systems, because the information content of a specific texture feature depends on the US imaging system and its setup. Our review shows that single classes of texture features are insufficient, if considered alone, to create robust CAD systems, which can help to solve practical problems, such as cancer screening. Therefore, we recommend that the CAD system design involves testing a wide range of texture features along with features obtained with other image processing methods. Having such a competitive testing phase helps the designer to select the best feature combination for a particular problem. This approach will lead to practical US based cancer detection systems which de- liver real benefits to patients by improving the diagnosis accuracy while reducing health care cost
