    Assessing microbial community diversity using amplicon length heterogeneity polymerase chain reaction

    It is thought that a microbial community is an assemblage of organisms, genes, and gene functions. Transient, acute signals such as excessive nutrient loads or disturbance and chronic signals such as seasonal temperature or rainfall impact the total environmental system. The goal of many microbial ecologists is to determine if a finely resolved study of microbial dynamics can be used as a large-scale biosensor to follow diversity patterns in the environment. With the development of new genomic tools, community-level studies have been designed that can interrogate the finer details of the biological components of a given habitat. Amplicon length heterogeneity polymerase chain reaction (LH-PCR) interrogates the hypervariable domains of the ribosomal small-subunit genes and separates these domains based on the naturally occurring sequence lengths of DNA. The amplicons are phylogenetically relevant in that the various amplicons generated can be directly associated with specific taxonomic sequences archived in the databases. The application of the LH-PCR technique as a monitoring tool for microbial ecology has been shown to enhance and extend the current understanding of the dynamics of microbial communities in their specific environments

    Influence of irrigated agriculture on soil microbial diversity

    Organic carbon (C), bacterial biomass and structural community diversity were measured in Southern Idaho soils with long term cropping histories. The soils tested were native sagebrush vegetation (NSB), irrigated moldboard plowed crops (IMP), irrigated conservation – chisel – tilled crops (ICT) and irrigated pasture systems (IP). Organic C concentration in soils decreased in the order NSB 0–5 cm > IP 0–30 cm = ICT 0–15 cm > IMP 0–30 cm > NSB 5–15 cm = NSB 15–30 cm. Active bacterial, fungal and microbial biomass correlated with soil C as measured by the Walkely Black method in positive curvilinear relationships (r2 = 0.93, 0.80 and 0.76, respectively). Amplicon length heterogeneity (LH-PCR) DNA profiling was used to access the eubacterial diversity in all soils and at all depths. The Shannon–Weaver diversity index was used to measure the differences using the combined data from three hypervariable domains of the eubacterial 16S rRNA genes. Diversity was greatest in NSB 15–30 cm soil and lowest in the IMP soil. With the exception of IMP with the lowest diversity index, the samples highest in C (NSB 0–5 cm, IP 0–30 cm, ICT 0–15 cm) reflected lower diversity indices. However, these indices were not significantly different from each other. ICT and IP increase soil C and to some extent increase diversity relative to IMP. Since soil bacteria respond quickly to environmental changes, monitoring microbial communities may be one way to assess the impact of agricultural practices such as irrigation and tillage regime

    Kinks in the Presence of Rapidly Varying Perturbations

    Dynamics of sine-Gordon kinks in the presence of rapidly varying periodic perturbations of different physical origins is described analytically and numerically. The analytical approach is based on asymptotic expansions, and it allows to derive, in a rigorous way, an effective nonlinear equation for the slowly varying field component in any order of the asymptotic procedure as expansions in the small parameter ω−1\omega^{-1}, ω\omega being the frequency of the rapidly varying ac driving force. Three physically important examples of such a dynamics, {\em i.e.}, kinks driven by a direct or parametric ac force, and kinks on rotating and oscillating background, are analysed in detail. It is shown that in the main order of the asymptotic procedure the effective equation for the slowly varying field component is {\em a renormalized sine-Gordon equation} in the case of the direct driving force or rotating (but phase-locked to an external ac force) background, and it is {\em the double sine-Gordon equation} for the parametric driving force. The properties of the kinks described by the renormalized nonlinear equations are analysed, and it is demonstrated analytically and numerically which kinds of physical phenomena may be expected in dealing with the renormalized, rather than the unrenormalized, nonlinear dynamics. In particular, we predict several qualitatively new effects which include, {\em e.g.}, the perturbation-inducedComment: New copy of the paper of the above title to replace the previous one, lost in the midst of the bulletin board.     Volatiles in lunar felsite clasts: Impact-related delivery of hydrous material to an ancient dry lunar crust

    In this detailed geochemical, petrological, and microstructural study of felsite clast materials contained in Apollo breccia samples 12013, 14321, and 15405, little evidence was found for relatively enriched reservoirs of endogenic lunar volatiles. NanoSIMS measurements have revealed very low volatile abundances (≀2–18 ppm hydrogen) in nominally anhydrous minerals (NAMS) plagioclase, potassic alkali feldspar, and SiO2 that make up a majority of these felsic lithologies. Yet these mineral assemblages and clast geochemistries on Earth would normally yield relatively high volatiles contents in their NAMS (∌20 to ≄80 ppm hydrogen). This difference is particularly notable in felsite 14321,1062 that exhibits extremely low volatile abundances (≀2 ppm hydrogen) and a relatively low amount of microstructural evidence for shock metamorphism given that it is a clast of the most evolved (∌74 wt.% SiO2) rock-type returned from the Moon. If taken at face value, ‘wet’ felsic magmas (∌1.2–1.7 wt.% water) are implied by the relatively high hydrogen contents of feldspar in felsite clasts in Apollo samples 12013 and 15405, but these results are likely misleading. These felsic clasts have microstructural features indicative of significantly higher shock stress than 14321,1062. These crustal lithologies likely obtained no more water from the lunar interior than the magma body producing 14321,1062. Rather, we suggest hydrogen was enriched in samples 12013 and 15405 by impact induced exchange, and/or partial assimilation of volatiles added to the surface of the Moon by a hydrated impactor (asteroid or comet) or the solar wind. Thus, the best estimate for magmatic water contents of felsic lunar magmas comes from 14321,1062 that leads to a calculated magmatic water content of ≀0.2 wt.%. This dry felsic magma has a slightly greater, but comparable water content to the ancient mafic magmas implied by the other lithologies that we have studied. Based on this and expanding evidence for a significantly dry ancient or early degassed Moon it is likely that some recent estimates (100's ppm) of the water abundances in the lunar parental magma ocean have been overestimated

    Double agents: gendered organizational culture, control and resistance

I suggest the values which senior managers encourage consultants to embody prioritize so-called`masculine' attributes over `feminine' ones. I attempt to demonstrate the limits of cultural control by outlining three ways in which the consultants engage with this imposed culture: defiance, parody and ritual. These activities contain gendered assumptions similar to those embedded in corporate culture. I discuss the potential such practices have for resisting corporate culture and the gender within it, suggesting that one source of ambiguity within workplace `control' and `resistance' practices is that they employ overlapping cultural resources and assumptions

Design/methodology/approach – The methodology is based on a series of case studies to develop and test the decision process. Findings – A decision process that leads the practitioner through an analytical process to decide which manufacturing activities they should carryout themselves. Practical implications – Strategic positioning is concerned with choosing those production related activities that an organisations should carry out internally, and those that should be external and under the ownership and control of suppliers, partners, distributors and customers. Originality/value – This concept extends traditional decision paradigms, such as those associated with “make versus buy” and “outsourcing”, by looking at the interactions between manufacturing operations and the wider supply chain networks associated with the organisation

    Search for direct production of charginos and neutralinos in events with three leptons and missing transverse momentum in √s = 7 TeV pp collisions with the ATLAS detector

    A search for the direct production of charginos and neutralinos in final states with three electrons or muons and missing transverse momentum is presented. The analysis is based on 4.7 fb−1 of proton–proton collision data delivered by the Large Hadron Collider and recorded with the ATLAS detector. Observations are consistent with Standard Model expectations in three signal regions that are either depleted or enriched in Z-boson decays. Upper limits at 95% confidence level are set in R-parity conserving phenomenological minimal supersymmetric models and in simplified models, significantly extending previous results

    Jet size dependence of single jet suppression in lead-lead collisions at sqrt(s(NN)) = 2.76 TeV with the ATLAS detector at the LHC

    Measurements of inclusive jet suppression in heavy ion collisions at the LHC provide direct sensitivity to the physics of jet quenching. In a sample of lead-lead collisions at sqrt(s) = 2.76 TeV corresponding to an integrated luminosity of approximately 7 inverse microbarns, ATLAS has measured jets with a calorimeter over the pseudorapidity interval |eta| < 2.1 and over the transverse momentum range 38 < pT < 210 GeV. Jets were reconstructed using the anti-kt algorithm with values for the distance parameter that determines the nominal jet radius of R = 0.2, 0.3, 0.4 and 0.5. The centrality dependence of the jet yield is characterized by the jet "central-to-peripheral ratio," Rcp. Jet production is found to be suppressed by approximately a factor of two in the 10% most central collisions relative to peripheral collisions. Rcp varies smoothly with centrality as characterized by the number of participating nucleons. The observed suppression is only weakly dependent on jet radius and transverse momentum. These results provide the first direct measurement of inclusive jet suppression in heavy ion collisions and complement previous measurements of dijet transverse energy imbalance at the LHC.Comment: 15 pages plus author list (30 pages total), 8 figures, 2 tables, submitted to Physics Letters B.     Toward a Multifaceted Heuristic of Digital Reading to Inform Assessment, Research, Practice, and Policy

    In this commentary, the author explores the tension between almost 30 years of work that has embraced increasingly complex conceptions of digital reading and recent studies that risk oversimplifying digital reading as a singular entity analogous with reading text on a screen. The author begins by tracing a line of theoretical and empirical work that both informs and complicates our understanding of digital literacy and, more specifically, digital reading. Then, a heuristic is proposed to systematically organize, label, and define a multifaceted set of increasingly complex terms, concepts, and practices that characterize the spectrum of digital reading experiences. Research that informs this heuristic is used to illustrate how more precision in defining digital reading can promote greater clarity across research methods and advance a more systematic study of promising digital reading practices. Finally, the author discusses implications for assessment, research, practice, and policy
