    Research of origin and work on clonal selection of Montenigrin grapevine varieties cv. vranac and cv. kratosija

    Viticulture production and winemaking of Montenegro are mostly based on growing of autochthonous grapevine varieties. Besides Vranac variety, Kratosija variety takes a significant place in Montenegrin autochthonous grapevine varieties. According to many literature data, Kratosija is an autochthonous grapevine variety whose origin and cultivation started earlier than with Vranac variety. Because of its heterogeneity, it is not so much represented in Montenegrin vineyards and it is mostly found in combination with Vranac variety. The research results about the origin of Vranac variety are shown in this paper. Furthermore, genetic identification which approved originality of Vranac variety is shown and it has also been approved that autochthonous variety Kratosija has the same genetic profile as cv. Zinfandel. The paper presents multi-annual results of examination of variability of their populations and work on clonal selection. Mother vines were selected and vineyards of pre-base and base category of Vranac variety potential clones were planted. The results achieved, in view of manifesting agro-biological, economic and technological characteristics, point to the need for further work on clonal selecton of these grapevine varieties

    Antioxidant Activity of Selected Polyphenolics in Yeast Cells: The Case Study of Montenegrin Merlot Wine

    Screens of antioxidant activity (AA) of various natural products have been a focus of the research community worldwide. This work aimed to differentiate selected samples of Merlot wines originated from Montenegro, with regard to phenolic profile and antioxidant capacity studied by survival rate, total sulfhydryl groups and activities of glutathione peroxidase (GPx), glutathione reductase and catalase in H2O2-stressed Saccharomyces cerevisiae cells. In this study, DPPH assay was also performed. Higher total phenolic content leads to an enhanced AA under both conditions. The same trend was observed for catechin and gallic acid, the most abundant phenolics in the examined wine samples. Finally, the findings of an Artificial Neural Network (ANN) model were in a good agreement (r(2) = 0.978) with the experimental data. All tested samples exhibited a protective effect in H2O2-stressed yeast cells. Pre-treatment with examined wines increased survival in H2O2-stressed cells and shifted antioxidative defense towards GPx-mediated defense. Finally, sensitivity analysis of obtained ANN model highlights the complexity of the impact that variations in the concentrations of specific phenolic components have on the antioxidant defense system

    Analysis of shared heritability in common disorders of the brain

    ience, this issue p. eaap8757 Structured Abstract INTRODUCTION Brain disorders may exhibit shared symptoms and substantial epidemiological comorbidity, inciting debate about their etiologic overlap. However, detailed study of phenotypes with different ages of onset, severity, and presentation poses a considerable challenge. Recently developed heritability methods allow us to accurately measure correlation of genome-wide common variant risk between two phenotypes from pools of different individuals and assess how connected they, or at least their genetic risks, are on the genomic level. We used genome-wide association data for 265,218 patients and 784,643 control participants, as well as 17 phenotypes from a total of 1,191,588 individuals, to quantify the degree of overlap for genetic risk factors of 25 common brain disorders. RATIONALE Over the past century, the classification of brain disorders has evolved to reflect the medical and scientific communities' assessments of the presumed root causes of clinical phenomena such as behavioral change, loss of motor function, or alterations of consciousness. Directly observable phenomena (such as the presence of emboli, protein tangles, or unusual electrical activity patterns) generally define and separate neurological disorders from psychiatric disorders. Understanding the genetic underpinnings and categorical distinctions for brain disorders and related phenotypes may inform the search for their biological mechanisms. RESULTS Common variant risk for psychiatric disorders was shown to correlate significantly, especially among attention deficit hyperactivity disorder (ADHD), bipolar disorder, major depressive disorder (MDD), and schizophrenia. By contrast, neurological disorders appear more distinct from one another and from the psychiatric disorders, except for migraine, which was significantly correlated to ADHD, MDD, and Tourette syndrome. We demonstrate that, in the general population, the personality trait neuroticism is significantly correlated with almost every psychiatric disorder and migraine. We also identify significant genetic sharing between disorders and early life cognitive measures (e.g., years of education and college attainment) in the general population, demonstrating positive correlation with several psychiatric disorders (e.g., anorexia nervosa and bipolar disorder) and negative correlation with several neurological phenotypes (e.g., Alzheimer's disease and ischemic stroke), even though the latter are considered to result from specific processes that occur later in life. Extensive simulations were also performed to inform how statistical power, diagnostic misclassification, and phenotypic heterogeneity influence genetic correlations. CONCLUSION The high degree of genetic correlation among many of the psychiatric disorders adds further evidence that their current clinical boundaries do not reflect distinct underlying pathogenic processes, at least on the genetic level. This suggests a deeply interconnected nature for psychiatric disorders, in contrast to neurological disorders, and underscores the need to refine psychiatric diagnostics. Genetically informed analyses may provide important "scaffolding" to support such restructuring of psychiatric nosology, which likely requires incorporating many levels of information. By contrast, we find limited evidence for widespread common genetic risk sharing among neurological disorders or across neurological and psychiatric disorders. We show that both psychiatric and neurological disorders have robust correlations with cognitive and personality measures. Further study is needed to evaluate whether overlapping genetic contributions to psychiatric pathology may influence treatment choices. Ultimately, such developments may pave the way toward reduced heterogeneity and improved diagnosis and treatment of psychiatric disorders

    Genomic Relationships, Novel Loci, and Pleiotropic Mechanisms across Eight Psychiatric Disorders

    Genetic influences on psychiatric disorders transcend diagnostic boundaries, suggesting substantial pleiotropy of contributing loci. However, the nature and mechanisms of these pleiotropic effects remain unclear. We performed analyses of 232,964 cases and 494,162 controls from genome-wide studies of anorexia nervosa, attention-deficit/hyper-activity disorder, autism spectrum disorder, bipolar disorder, major depression, obsessive-compulsive disorder, schizophrenia, and Tourette syndrome. Genetic correlation analyses revealed a meaningful structure within the eight disorders, identifying three groups of inter-related disorders. Meta-analysis across these eight disorders detected 109 loci associated with at least two psychiatric disorders, including 23 loci with pleiotropic effects on four or more disorders and 11 loci with antagonistic effects on multiple disorders. The pleiotropic loci are located within genes that show heightened expression in the brain throughout the lifespan, beginning prenatally in the second trimester, and play prominent roles in neurodevelopmental processes. These findings have important implications for psychiatric nosology, drug development, and risk prediction.Peer reviewe

    HPLC-DAD-ESI/MS monitoring of stilbenes content in Vranec red wines produced with traditional and modern fermentation methods

    Quantification of stilbenes was performed on Vranac wines (vintage 2008 and 2009) produced by traditional and modern fermentation methods, applying different enological additions (enzyme, oak chips and grape tannins). Stilbenes, trans-resveratrol and trans-resveratrol-3-glucoside were determined using the HPLC/DAD/ESI-MS and MS/MS technique. Trans-resveratrol ranged from 0.09 to 3.3 mg/L and the trans-resveratrol-3-glucoside was present in a range of 1.13 to 2.6 mg/L. The influence of vintage, fermentation tanks and enological additions was noticed on the content of stilbenes. Wines from vintage 2008 presented higher amount of stilbenes (on average: 1.89 mg/L) compared to the wines from 2009 (on average: 1.59 mg/L) probably because of the difference in the temperature and humidity in both years. Application of modern fermentation tanks (Sifa and Ganimede) followed by additions of enzyme, oak chips and grape tannins resulted in a higher amount of stilbenes.     The Influence of Chips on Polyphenolic Content and Sensorial Properties of Vranec Wine

    The influence of oak chips on polyphenolic content, as well as sensorial characteristics of Vranec wines was studied. Wine were produced in a traditional way in the winery a the Biotechnical Faculty in Podgorica, Ljeskopolje area, Podgorica sub-region. 1 g/l of French oak chips was added in the wines in order to study its influence. It was found that the chips improved the extraction of polyphenols and sensorial characteristics of wines, compared to the control wine produced without chips. In addition, it was noticed that the obtained wines fermented with chips, presented a higher content of total polyphenols, anthocyanins, flavan-3-ols, and better colour intensity and hue, as well as better sensorial properties of the wine.     Uncertainty of Trypsin Inhibitor Activity Measurement of Legume Crops Using Microtiter Plate Method

    Trypsin inhibitors could limit utilization of legumes in human nutrition, but they could also have beneficial health effects. The objective of this study was to measure trypsin inhibitor activity (TIA) of different legumes using microtiter plate method and to identify factors that contribute to uncertainty of TIA measurement. TIA measurements were performed on seeds of faba bean, pea, common vetch, soybean, and common bean cultivars. The significant effect of legume crop on TIA measurement uncertainty was confirmed with P = 0.045. Certain sources of measurement uncertainty were related with the content of trypsin inhibitors (Tis) in legume seeds. In respect to that, significant effect of level of sample dilution (P E, 0.001) was confirmed. Significant influence of the repeated absorbance measurement of sample reaction mixture on uncertainty of TIA measurement was identified (P E, 0.001), and it took 60% of overall TIA measurement uncertainty for soybean cultivars. TIA of soybean cultivars exceeded 90 TUI/mg. Repeated absorbance measurement of positive control reaction mixture took 70% of TIA measurement uncertainty of cultivars with TIA lesser than 4.5 TUI/mg. Graduated cylinder used for preparation of the final sample solutions took the range from 45 to 90% of overall TIA measurement uncertainty of the cultivars whose TIA were in the middle of previously mentioned. The uncertainty of TIA measurement of legume crops was not studied before; thus, this study pointed out that acquiring insight into factors contributing to uncertainty of TIA measurement could give directions for improvement of TIA testing if microtiter plate method is used
