    Chromosome 9p21 gene copy number and prognostic significance of p16 in ESFT

    Chromosome 9p21 gene copy number in Ewing's sarcoma family of tumour (ESFT) cell lines and primary ESFT has been evaluated using Multiplex Ligation-dependent probe amplification, and the clinical significance of CDKN2A loss and p16/p14ARF expression investigated. Homozygous deletion of CDKN2A was identified in 4/9 (44%) of ESFT cell lines and 4/42 (10%) primary ESFT; loss of one copy of CDKN2A was identified in a further 2/9 (22%) cell lines and 2/42 (5%) tumours. CDKN2B was co-deleted in three (33%) cell lines and two (5%) tumours. Co-deletion of the MTAP gene was observed in 1/9 (11%) cell lines and 3/42 (7%) tumours. No correlation was observed between CDKN2A deletion and clinical parameters. However, co-expression of high levels of p16/p14ARF mRNA predicted a poor event-free survival (P=0.046, log-rank test). High levels of p16/p14ARF mRNA did not correlate with high expression of p16 protein. Furthermore, p16 protein expression did not predict event-free or overall survival. Methylation is not a common mechanism of p16 gene silencing in ESFT. These studies demonstrate that loss (homozygous deletion or single copy) of CDKN2A was not prognostically significant in primary ESFT. However, high levels of p16/p14ARF mRNA expression were predictive of a poor event-free survival and should be investigated further

    Search for lepton flavour violating decays of the Higgs boson to eτand eμin proton–proton collisions at √s=8TeV

    A direct search for lepton flavour violating decays of the Higgs boson (H) in the H →eτand H →eμchannels is described. The data sample used in the search was collected in proton–proton collisions at √s=8TeVwith the CMS detector at the LHC and corresponds to an integrated luminosity of 19.7fb−1. No evidence is found for lepton flavour violating decays in either final state. Upper limits on the branching fractions, B(H →eτ) <0.69%and B(H →eμ) <0.035%, are set at the 95% confidence level. The constraint set on B(H →eτ)is an order of magnitude more stringent than the existing indirect limits. The limits are used to constrain the corresponding flavour violating Yukawa couplings, absent in the standard model

    Search for physics beyond the standard model in events with two leptons of same sign, missing transverse momentum, and jets in proton-proton collisions at root s=13TeV

    A data sample of events from proton–proton collisions with two isolated same-sign leptons, missing transverse momentum, and jets is studied in a search for signatures of new physics phenomena by the CMS Collaboration at the LHC. The data correspond to an integrated luminosity of 35.9 fb−1, and a center-of-mass energy of 13 TeV. The properties of the events are consistent with expectations from standard model processes, and no excess yield is observed. Exclusion limits at 95% confidence level are set on cross sections for the pair production of gluinos, squarks, and same-sign top quarks, as well as top-quark associated production of a heavy scalar or pseudoscalar boson decaying to top quarks, and on the standard model production of events with four top quarks. The observed lower mass limits are as high as 1500 GeV for gluinos, 830 GeV for bottom squarks. The excluded mass range for heavy (pseudo)scalar bosons is 350–360 (350–410) GeV. Additionally, model-independent limits in several topological regions are provided, allowing for further interpretations of the results
