Identification of optimal assisted aspiration conditions of oocytes for use in porcine in vitro maturation: a re-evaluation of the relationship between the cumulus oocyte complex and oocyte quality
The quality of porcine oocytes for use in IVF is commonly graded according to the number of layers of cumulus cells surrounding the oocyte; together these form the cumulus oocyte complex (COC). At least three compact layers of cumulus cells is regarded as important for efficient IVP. To test this, oocytes were scored according to cumulus investment, with grade A representing COCs with three or more cumulus layers including granulosa cell-cumulus oocyte complexes, grade B those with an intact corona radiata surrounded by another layer of cumulus cells and grades C and D representing COCs with lower cumulus cell investment. These oocytes were then monitored for in vitro maturation (IVM), as assessed by tubulin immunostaining for meiotic progression, the development of a cortical granule ring, and by glutathione levels. Results indicate that grading correlates closely with nuclear maturation and cytoplasmic maturation, suggesting that grading oocytes by cumulus investment is a reliable method to predict IVM success. Importantly, Grade A and B oocytes showed no significant differences in any measure and hence using a cut-off of two or more cumulus cell layers may be optimal. We also determined the effect of assisted aspiration for oocyte retrieval, comparing the effect of needle size and applied pressure on the retrieval rate. These data indicated that both variables affected oocyte recovery rates and the quality of recovered oocytes. In combination, these experiments indicate that grade A and B oocytes have a similar developmental potential and that the recovery of oocytes of these grades is maximised by use of an 18-gauge needle and 50mmHg aspiration pressure
Serum proteins in healthy and diseased Florida manatees (Trichechus manatus latirostris)
A major goal of this study was to determine whether serum protein fractions of healthy Florida manatees differ with age, sex, or living environments (wild versus housed). A second goal was to determine which serum protein fractions vary in diseased versus healthy manatees. Serum protein fractions were determined using agarose gel electrophoresis. Healthy adults had slightly higher total serum protein and total globulin concentrations than younger animals. This largely resulted from an increase in gamma globulins with age. Total serum protein, albumin, alpha-1 globulin, beta globulin, and total globulin concentrations were slightly higher in housed manatees compared to wild manatees, but there was no significant difference in the albumin/globulin (A/G) ratio, suggesting a difference in hydration between these groups. No significant differences were attributable to sex or pregnancy.
Serum albumin concentrations and A/G ratios were significantly lower for manatees with boat trauma, entanglements, emaciation, or cold stress compared to healthy manatees. Variable increases were seen in alpha-1globulins, alpha-2 globulins, beta globulins, and gamma globulins. These globulin fractions contain positive acute-phase proteins and immunoglobulins, and their increases may reflect acute or chronic active inflammation. Changes in serum protein fractions were not consistent enough to justify the use of serum protein electrophoresis as a routine diagnostic test for manatees. However, serum (or plasma) protein electrophoresis is required when accurate values for albumin and globulins are needed in manatees and in determining which protein fractions may account for a hyperproteinemia or hypoproteinemia reported in a clinical chemistry panel
Using the past to constrain the future: how the palaeorecord can improve estimates of global warming
Climate sensitivity is defined as the change in global mean equilibrium
temperature after a doubling of atmospheric CO2 concentration and provides a
simple measure of global warming. An early estimate of climate sensitivity,
1.5-4.5{\deg}C, has changed little subsequently, including the latest
assessment by the Intergovernmental Panel on Climate Change.
The persistence of such large uncertainties in this simple measure casts
doubt on our understanding of the mechanisms of climate change and our ability
to predict the response of the climate system to future perturbations. This has
motivated continued attempts to constrain the range with climate data, alone or
in conjunction with models. The majority of studies use data from the
instrumental period (post-1850) but recent work has made use of information
about the large climate changes experienced in the geological past.
In this review, we first outline approaches that estimate climate sensitivity
using instrumental climate observations and then summarise attempts to use the
record of climate change on geological timescales. We examine the limitations
of these studies and suggest ways in which the power of the palaeoclimate
record could be better used to reduce uncertainties in our predictions of

Ramond-Ramond Fields, Fractional Branes and Orbifold Differential K-Theory
We study D-branes and Ramond-Ramond fields on global orbifolds of Type II
string theory with vanishing H-flux using methods of equivariant K-theory and
K-homology. We illustrate how Bredon equivariant cohomology naturally realizes
stringy orbifold cohomology. We emphasize its role as the correct cohomological
tool which captures known features of the low-energy effective field theory,
and which provides new consistency conditions for fractional D-branes and
Ramond-Ramond fields on orbifolds. We use an equivariant Chern character from
equivariant K-theory to Bredon cohomology to define new Ramond-Ramond couplings
of D-branes which generalize previous examples. We propose a definition for
groups of differential characters associated to equivariant K-theory. We derive
a Dirac quantization rule for Ramond-Ramond fluxes, and study flat
Ramond-Ramond potentials on orbifolds.Comment: 46 pages; v2: typos correcte
T-Duality and Penrose limits of spatially homogeneous and inhomogeneous cosmologies
Penrose limits of inhomogeneous cosmologies admitting two abelian Killing
vectors and their abelian T-duals are found in general. The wave profiles of
the resulting plane waves are given for particular solutions. Abelian and
non-abelian T-duality are used as solution generating techniques. Furthermore,
it is found that unlike in the case of abelian T-duality, non-abelian T-duality

Search for direct production of charginos and neutralinos in events with three leptons and missing transverse momentum in √s = 7 TeV pp collisions with the ATLAS detector
A search for the direct production of charginos and neutralinos in final states with three electrons or muons and missing transverse momentum is presented. The analysis is based on 4.7 fb−1 of proton–proton collision data delivered by the Large Hadron Collider and recorded with the ATLAS detector. Observations are consistent with Standard Model expectations in three signal regions that are either depleted or enriched in Z-boson decays. Upper limits at 95% confidence level are set in R-parity conserving phenomenological minimal supersymmetric models and in simplified models, significantly extending previous results
Cryopreservation of animal oocytes and embryos: current progress and future prospects
Cryopreservation describes techniques that permit freezing and subsequent warming of biological samples without loss of viability. The application of cryopreservation in assisted reproductive technology encompasses the freezing of gametes, embryos and primordial germ cells. Whilst some protocols still rely on slow-freezing techniques, most now use vitrification, or ultra-rapid freezing, for both oocytes and embryos due to an associated decreased risk of damage caused by the lack of ice crystal formation, unlike in slow-freezing techniques. Vitrification has demonstrated its use in many applications, not only following in vitro fertilisation (IVF) procedures in human embryology clinics, but also following in vitro production (IVP) of embryos in agriculturally important, or endangered animal species, prior to embryo transfer. Here we review the various cryopreservation and vitrification technologies that are used in both humans and other animals and discuss the most recent innovations in vitrification with a particular emphasis on their applicability to animal embryology
Jet size dependence of single jet suppression in lead-lead collisions at sqrt(s(NN)) = 2.76 TeV with the ATLAS detector at the LHC
Measurements of inclusive jet suppression in heavy ion collisions at the LHC
provide direct sensitivity to the physics of jet quenching. In a sample of
lead-lead collisions at sqrt(s) = 2.76 TeV corresponding to an integrated
luminosity of approximately 7 inverse microbarns, ATLAS has measured jets with
a calorimeter over the pseudorapidity interval |eta| < 2.1 and over the
transverse momentum range 38 < pT < 210 GeV. Jets were reconstructed using the
anti-kt algorithm with values for the distance parameter that determines the
nominal jet radius of R = 0.2, 0.3, 0.4 and 0.5. The centrality dependence of
the jet yield is characterized by the jet "central-to-peripheral ratio," Rcp.
Jet production is found to be suppressed by approximately a factor of two in
the 10% most central collisions relative to peripheral collisions. Rcp varies
smoothly with centrality as characterized by the number of participating
nucleons. The observed suppression is only weakly dependent on jet radius and
transverse momentum. These results provide the first direct measurement of
inclusive jet suppression in heavy ion collisions and complement previous
measurements of dijet transverse energy imbalance at the LHC.Comment: 15 pages plus author list (30 pages total), 8 figures, 2 tables,
