Revealing microbial recognition by specific antibodies
Background:
Recognition of microorganisms by antibodies is a vital component of the human immune response. However, there is currently very limited understanding of immune recognition of 50 % of the human microbiome which is made up of as yet un-culturable bacteria. We have combined the use of flow cytometry and pyrosequencing to describe the microbial composition of human samples, and its interaction with the immune system.
Results:
We show the power of the technique in human faecal, saliva, oral biofilm and breast milk samples, labeled with fluorescent anti-IgG or anti-IgA antibodies. Using Fluorescence-Activated Cell Sorting (FACS), bacterial cells were separated depending on whether they are coated with IgA or IgG antibodies. Each bacterial population was PCR-amplified and pyrosequenced, characterizing the microorganisms which evade the immune system and those which were recognized by each immunoglobulin.
Conclusions:
The application of the technique to healthy and diseased individuals may unravel the contribution of the immune response to microbial infections and polymicrobial diseases
Performance comparison of microwave cavities used for extraction operations
In the agri-food industry world, billions of tons of waste are produced every year. This represents both a direct loss (due to the failure to exploit their potential value, and their nutritional and energy content) and indirect loss, due to their necessary treatment and/or disposal. Some substances contained in the wastes, of potential high value, can be recovered by means of extraction. Conventional extraction processes involve the use of solvents, which end up requiring an additional process of separation from the solute identified as the desired product. In recent years, extraction techniques have been proposed without the use of solvents. This work compares the performance of two microwave cavities used for extraction operations, both working at 2.45 GHz.
A calorimetric analysis performed by following the heating rate and temperature evolution in rack of 25 beakers filled with 25 mL of water, coupled with the solution of the heat transfer balance in the system, allowed to build the spatial distribution of the electromagnetic power dissipated as heat in each of the beakers. Fluid-dynamics aspects related to the recovery of the vapour phase produced during the extraction were also analyzed, with particular emphasis on the mean residence time of the vapour fraction in the extraction chamber as a function of its configuration
Vaptans and hyponatremia in critical patients
Hyponatremia is the most frequent fluid and electrolyte disorder in hospitalized patients (20%), particularly in ICU, associated with an increase in morbility and mortality. While hypovolaemic hyponatremia needs to be corrected with the replacement of the lost extracellular fluid by isotonic saline, euvolaemic (SIADH) and hypervolaemic hyponatremia (oedematous states like decompensated heart failure, liver cirrhosis, i.e.) are treated by restriction of fluid intake, loop diuretics and hypertonic saline. A novel approach consists in use of vaptans, non-peptide arginine vasopressin (AVP) receptor antagonists. Vaptans cause âaquaresisâ, which results in the correction of plasma osmolality and serum sodium levels, without activation of the renin-angiotensin-aldosterone system or changes in blood pressure and renal function. In this paper we critically reviewed the results of the available randomized controlled critical trials, discussing the effectiveness and safety of vaptans in treating hypervolaemic and euvolaemic hyponatremia in critical patients
Case report: Hematologic malignancies concomitant diagnosis of hairy cell leukemia and chronic lymphocytic leukemia: A rare association
A case of concomitant hairy cell leukemia (HCL) and chronic lymphocytic leukemia (CLL) in a 50- year-old man was reported. Flow cytometry and droplet digital PCR (ddPCR) were used to detect the B-Raf proto-oncogene (BRAF) V600E mutation. The HCL population was the predominant component. The patient was first treated with cladribine and then with rituximab and achieved HCL partial remission. Importantly, the high sensitivity of our flow cytometric approach allowed the detection of a small population âP3,â in addition to the typical HCL and CLL clones. The P3 clone changed over time, from an HCL-like to a CLL-like immunophenotype. This case is added to the few other cases of synchronous HCL and CLL already reported in the literature and underlines the importance of analyzing chronic lymphoproliferative disorders by highly sensitive diagnostic techniques, like the multicolor flow cytometry and ddPCR, to evaluate the possible association between HCL and CLL at diagnosis
Contribution to the floristic knowledge of eastern Irpinia and Vulture-Melfese area (Campania and Basilicata, southern Italy)
In order to improve the floristic knowledge of the Italian territory, we report the inventory of the taxa
collected during the annual field trip of the working group for Floristics, Systematics and Evolution of
the Italian Botanical Society held in 2015 in eastern Irpinia and Vulture-Melfese area (South Italy). The
investigated territories are located in southern Apennines, along the border between the Campania and
Basilicata administrative regions. These areas are scarcely known in terms of vascular flora. The floristic
samplings were performed in 19 sites selected as representative of the local environmental diversity as
regards to climate, litho-morphology and land-use.
The research led to the identification of 4,137 specimens of vascular plants, belonging to 815 species
and subspecies, 399 genera, and 85 families. Among these taxa, 42 were endemic to Italy, 38 were included
in the IUCN Red List of the Italian Flora, 28 were alien and 5 were cryptogenic in Campania and/
or Basilicata administrative regions. Two taxa, Aquilegia coerulea (casual alien, native to North America)
and Lolium Ăboucheanum (native), were found to be new for Italy. On the basis of the available floristic
literature the first one is also to be considered new for the European flora. At regional scale, we have found
18 taxa new for the Campania and 15 new for the Basilicata region. Finally, 10 taxa were confirmed for
Campania. Data obtained during this study, confirmed the important role of a collaborative approach
among botanists and the great relevance of these territories for plant diversity
Multiphoton Quantum Optics and Quantum State Engineering
We present a review of theoretical and experimental aspects of multiphoton
quantum optics. Multiphoton processes occur and are important for many aspects
of matter-radiation interactions that include the efficient ionization of atoms
and molecules, and, more generally, atomic transition mechanisms;
system-environment couplings and dissipative quantum dynamics; laser physics,
optical parametric processes, and interferometry. A single review cannot
account for all aspects of such an enormously vast subject. Here we choose to
concentrate our attention on parametric processes in nonlinear media, with
special emphasis on the engineering of nonclassical states of photons and
atoms. We present a detailed analysis of the methods and techniques for the
production of genuinely quantum multiphoton processes in nonlinear media, and
the corresponding models of multiphoton effective interactions. We review
existing proposals for the classification, engineering, and manipulation of
nonclassical states, including Fock states, macroscopic superposition states,
and multiphoton generalized coherent states. We introduce and discuss the
structure of canonical multiphoton quantum optics and the associated one- and
two-mode canonical multiphoton squeezed states. This framework provides a
consistent multiphoton generalization of two-photon quantum optics and a
consistent Hamiltonian description of multiphoton processes associated to
higher-order nonlinearities. Finally, we discuss very recent advances that by
combining linear and nonlinear optical devices allow to realize multiphoton
entangled states of the electromnagnetic field, that are relevant for
applications to efficient quantum computation, quantum teleportation, and

Dupilumab in the treatment of severe uncontrolled chronic rhinosinusitis with nasal polyps (CRSwNP): A multicentric observational Phase IV real-life study (DUPIREAL)
Background
Chronic rhinosinusitis with nasal polyps (CRSwNP) is associated with significant morbidity and reduced health-related quality of life. Findings from clinical trials have demonstrated the effectiveness of dupilumab in CRSwNP, although real-world evidence is still limited.
Methods
This Phase IV real-life, observational, multicenter study assessed the effectiveness and safety of dupilumab in patients with severe uncontrolled CRSwNP (nâ=â648) over the first year of treatment. We collected data at baseline and after 1, 3, 6, 9, and 12âmonths of follow-up. We focused on nasal polyps score (NPS), symptoms, and olfactory function. We stratified outcomes by comorbidities, previous surgery, and adherence to intranasal corticosteroids, and examined the success rates based on current guidelines, as well as potential predictors of response at each timepoint.
Results
We observed a significant decrease in NPS from a median value of 6 (IQR 5â6) at baseline to 1.0 (IQR 0.0â2.0) at 12âmonths (pâ<â.001), and a significant decrease in Sino-Nasal Outcomes Test-22 (SNOT-22) from a median score of 58 (IQR 49â70) at baseline to 11 (IQR 6â21; pâ<â.001) at 12âmonths. Sniffin' Sticks scores showed a significant increase over 12âmonths (pâ<â.001) compared to baseline. The results were unaffected by concomitant diseases, number of previous surgeries, and adherence to topical steroids, except for minor differences in rapidity of action. An excellent-moderate response was observed in 96.9% of patients at 12âmonths based on EPOS 2020 criteria.
Conclusions
Our findings from this large-scale real-life study support the effectiveness of dupilumab as an add-on therapy in patients with severe uncontrolled CRSwNP in reducing polyp size and improving the quality of life, severity of symptoms, nasal congestion, and smell
The SeqCOVID-Spain consortium: unravelling the dynamics of the COVID-19 first epidemic wave in Spain
PĂłster presentado a la Applied Bioinformatics and Public Health Microbiology 2021 Virtual Conference, celebrada del 5 al 7 de mayo de 2021.The COVID-19 pandemic has shaken the world since the beginning of 2020. Spain is among the European countries with the highest incidence of the disease during the first pandemic wave. We established a multidisciplinary consortium to monitor and study the evolution of the epidemic, with the aim of contributing to decision making and stopping rapid spreading across the country. We present the results for 2170 sequences from the first wave of the SARS-Cov-2 epidemic in Spain, representing
12% of diagnosed cases until 14th March. This effort allows us to document at least 500 initialintroductions, between early February-March from multiple international sources. Importantly, we document the early raise of two dominant genetic variants in Spain (Spanish Epidemic Clades), named SEC7 and SEC8, likely amplified by superspreading events. In sharp contrast to other non Asian countries those two variants were closely related to the initial variants of SARS-CoV-2 described in Asia and represented 40% of the genome sequences analyzed. The two dominant SECs were widely spread across the country compared to other genetic variants with SEC8 reaching a 60% prevalence just before the lockdown. Employing Bayesian phylodynamic analysis, we inferred a reduction in the effective reproductive number of these two SECs from around 2.5 to below 0.5 after the implementation of strict public-health interventions in mid-March. The effects of lockdown on the genetic variants of the virus are reflected in the general replacement of pre-existing SECs by a new
variant at the beginning of the summer season.
