    Freqüência de toxocaríase humana numa população rural de Cajamarca, Peru, mediante o uso do teste DOT-ELISA

    The aim of this study was to estimate the frequency of human toxocariasis in Cauday district, Cajamarca, Peru, using a dot-ELISA test. From June to October 2005, a total of 256 adult subjects were studied. Blood samples were collected for serology by a dot-ELISA test and for hematological examination. Parasitological examination was also carried out in stool samples to check cross-reactions in the dot-ELISA. The frequency observed was 44.92%, with a significant higher proportion of positivity in male subjects. From subjects with positive serology, 45.6% had respiratory symptoms, 40.44% abdominal pain, 32.35% hepatic symptoms, 14.7% cutaneous signs, 13.23% ocular manifestations, 43.38% eosinophilia, and all of these were statistically associated to serology. Among the population evaluated, 90.23% (231/256) were parasitized. From subjects with positive serology, 92.17% had at least one intestinal parasite and the most frequent were: Blastocystis hominis (68.38%), Giardia lamblia (28.68%), Hymenolepis nana (20.0%), Ascaris lumbricoides (15.65%), Entamoeba histolytica/E. dispar (13.24%), Cyclospora cayetanensis (4.41%), Cryptosporidium sp. (1.47%), Enterobius vermicularis (0.87%), Strongyloides stercoralis (0.87%), Taenia sp. (0.87%), and Trichuris trichiura (0.87%). The rate of false positives in the dot-ELISA test was improved by serum absorption each with A. suum antigens, with a decrease of cross-reactions. In conclusion, human toxocariasis is highly frequent in this population and some risk factors like dog/cat ownership, presence of pets within house, and previous history of geophagia were observed in the present study.O propósito do presente estudo foi estimar a freqüência da toxocaríase no distrito de Cauday, Cajamarca, Peru, usando o dot-ELISA teste. Entre junho e outubro de 2005, um total de 256 pessoas foram avaliadas. Coletaram-se amostras de sangue para o teste de dot-ELISA e para o exame hematológico e amostras de fezes para exame parasitológico. A freqüência geral de anticorpos anti-Toxocara observada foi de 44,92%, com maior proporção significativa de positividade em pessoas do sexo masculino. Das pessoas com sorologia positiva, 45,6% apresentavam sintomas respiratórios, 40,44% dores abdominais, 32,35% moléstias hepáticas, 14,7% sinais cutâneos, 13,23% manifestações oculares, 43,38% eosinofilia e todos estes fatores foram estatisticamente associados à sorologia. Entre as pessoas avaliadas 90,23% estavam parasitadas e 92,17% das pessoas com sorologia positiva tinham algum parasito intestinal, sendo os mais freqüentes: Blastocystis hominis (68,38%), Giardia lamblia (28,68%), Hymenolepis nana (20,0%), Ascaris lumbricoides (15,65%), Entamoeba histolytica/E. dispar (13,24%), Cyclospora cayetanensis (4,41%), Cryptosporidium sp. A taxa de falsos positivos no teste dot-ELISA foi melhorada pela absorção dos soros com antígenos de A. suum, com diminuição das reações cruzadas. Em conclusão, a toxocaríase humana é altamente freqüente nesta população e fatores de risco como ter um cão/gato, presença dos animais dentro de casa e estória prévia de geofagia foram observados durante o presente estudo

    Comparación de tres pruebas serológicas para detectar infecciones por Paragonimus mexicanus en gatos infectados

    Background: Paragonimiasis is a lung disease caused by Paragonimus trematodes genus. Human infection laboratory diagnosis is usually done by detection of parasite eggs in sputum or feces; however, findings may be negative and alternative diagnostic methods are required. Objectives: To compare three serologic tests for detection of Paragonimus mexicanus somatic antigen (PmAS) antibodies in serum samples of cats infected experimentally. Design: Experimental study. Setting: Instituto de Medicina Tropical “Daniel A. Carrión” bioterium, Faculty of Medicine, Universidad Nacional Mayor de San Marcos, Lima, Peru. Biologic material: Domestic cats 4-6 years old and P. mexicanus (PmAS) crude somatic antigen. Interventions: Blood samples were collected from twelve domestic cats infected with P. mexicanus. Serum samples were evaluated by enzyme-linked immunosorbent assay (ELISA), double diffusion (DD) and counterimmunoelectrophoresis (CIE). Results: Good correlation was found between ELISA results and those of CIE or DD. Both CIE and ELISA were more sensitive than DD. With CIE and ELISA sera were respectively positive 81.8% (9/11) and 75.0% (9/12), whereas 58.3% (7/12) were positive for DD test. Sera from 2 rabbits with experimental fascioliosis had positive reaction with PmAS antigen by both ELISA and CIE tests. No cross-reactivity was observed using sera from cats with dipylidiasis or from healthy cats. Conclusions: Results showed that both ELISA and CIE were equally sensitive tests for paragonimiosis diagnosis but could not be used for differential diagnosis of P. mexicanus and F. hepatica infections due to cross-reaction.Introducción: La paragonimiasis es una enfermedad pulmonar causada por tremátodes del género Paragonimus. Generalmente, el diagnóstico de laboratorio de la infección en humanos se realiza mediante la detección de huevos del parásito en esputo o heces; sin embargo, los resultados pueden ser negativos, necesitándose métodos diagnósticos alternativos. Objetivos: Comparar tres pruebas serológicas para la detección de anticuerpos contra el antígeno somático del estadio adulto de Paragonimus mexicanus (PmAS) en muestras de suero de gatos infectados experimentalmente. Diseño: Estudio experimental. Lugar: Bioterio del Instituto de Medicina Tropical “Daniel A. Carrión” de la Facultad de Medicina, Universidad Nacional Mayor de San Marcos. Material biológico: Gatos domésticos de 4 a 6 meses de edad y antígeno somático crudo de P. mexicanus (PmAS). Intervenciones: Se obtuvo muestras de sangre de 12 gatos domésticos infectados experimentalmente con P. mexicanus. Los sueros fueron evaluados por el inmunoensayo enzimático (ELISA), la doble difusión (DD) y contrainmunoelectroforesis (CIEF). Principales medidas de resultados: Se determinó el porcentaje de gatos que revelaron resultados positivos de acuerdo a los criterios establecidos. Resultados: Se encontró una buena correlación entre los resultados obtenidos por ELISA y aquellos obtenidos por CIEF o DD. La CIEF y el ELISA fueron más sensibles que la doble difusión. Con la CIEF y el ELISA, 81,8% (9/11) y 75,0% (9/12) de los sueros evaluados dieron resultados positivos, respectivamente, mientras que 58,3% (7/12) de ellos fue positivo para la DD. Los sueros de 2 conejos con fasciolosis experimental dieron una reacción positiva con el antígeno PmAS por ELISA y CIEF. No se observó reacción cruzada cuando se usó sueros de gatos con dipilidiasis o de gatos sanos. Conclusiones: Estos resultados indican que la CIEF y el ELISA, que emplean antígeno somático crudo, son pruebas de similar sensibilidad para el diagnóstico de paragonimiosis, pero no pueden ser usadas para el diagnóstico diferencial de las infecciones por P. mexicanus y F. hepatica, debido a la ocurrencia de reacciones cruzadas

    Vagal afferents contribute to sympathoexcitation-driven metabolic dysfunctions

    Multiple crosstalk between peripheral organs and the nervous system are required to maintain physiological and metabolic homeostasis. Using Vav3-deficient mice as a model for chronic sympathoexcitation-associated disorders, we report here that afferent fibers of the hepatic branch of the vagus nerve are needed for the development of the peripheral sympathoexcitation, tachycardia, tachypnea, insulin resistance, liver steatosis and adipose tissue thermogenesis present in those mice. This neuronal pathway contributes to proper activity of the rostral ventrolateral medulla, a sympathoregulatory brainstem center hyperactive in Vav3−/− mice. Vagal afferent inputs are also required for the development of additional pathophysiological conditions associated with deregulated rostral ventrolateral medulla activity.     Regulation of Embryonic and Induced Pluripotency by Aurora Kinase-p53 Signaling

    SummaryMany signals must be integrated to maintain self-renewal and pluripotency in embryonic stem cells (ESCs) and to enable induced pluripotent stem cell (iPSC) reprogramming. However, the exact molecular regulatory mechanisms remain elusive. To unravel the essential internal and external signals required for sustaining the ESC state, we conducted a short hairpin (sh) RNA screen of 104 ESC-associated phosphoregulators. Depletion of one such molecule, aurora kinase A (Aurka), resulted in compromised self-renewal and consequent differentiation. By integrating global gene expression and computational analyses, we discovered that loss of Aurka leads to upregulated p53 activity that triggers ESC differentiation. Specifically, Aurka regulates pluripotency through phosphorylation-mediated inhibition of p53-directed ectodermal and mesodermal gene expression. Phosphorylation of p53 not only impairs p53-induced ESC differentiation but also p53-mediated suppression of iPSC reprogramming.     Search for direct production of charginos and neutralinos in events with three leptons and missing transverse momentum in √s = 7 TeV pp collisions with the ATLAS detector

    A search for the direct production of charginos and neutralinos in final states with three electrons or muons and missing transverse momentum is presented. The analysis is based on 4.7 fb−1 of proton–proton collision data delivered by the Large Hadron Collider and recorded with the ATLAS detector. Observations are consistent with Standard Model expectations in three signal regions that are either depleted or enriched in Z-boson decays. Upper limits at 95% confidence level are set in R-parity conserving phenomenological minimal supersymmetric models and in simplified models, significantly extending previous results

    Aislamiento de Candida albicans de hisopado nasal y faríngeo en alumnos de secundaria

    Objective: To determine the presence of Candida albicans in throat and nasal secretion in high school students. Design: Cross-sectional study. Setting: Institute of Tropical Medicine, Universidad Nacional Mayor de San Marcos, Lima, Peru. Participants: High school students from San Juan Macias School in Santa Anita, Lima, Peru. Interventions: Nasal and throat samples were collected from 102 14-17 year-old students. Samples were grown on sabouraud agar and Candida CHROMOagar and identified by chlamydospores study and metabolic tests. Main outcome measures: Identification of C. albicans yeast. Results: Candida yeast was isolated from 11 students (10.8%). A significant percentage of yeast (36.4%) developed moderate resistance to fluconazole. Conclusions: Continuous surveillance of medically important yeasts in nasopharyngeal carriers is suggested in order to be prepared for eventual infectious conditions.Introducción: Candida sp puede encontrarse como comensal en equilibrio en la cavidad bucal humana; pero, en la población pediátrica y adolescente con un sistema inmune inmaduro las condiciones de la levadura se tornarían favorables para su patogenia. Objetivo: Determinar la presencia de Candida albicans en secreción faríngea y nasal en alumnos de educación secundaria. Diseño: Estudio descriptivo transversal. Lugar: Instituto de Medicina Tropical, Universidad Nacional Mayor de San Marcos, Lima, Perú. Participantes: Alumnos del 4° (52) y 5° (50) años de secundaria. Intervenciones: En octubre del 2007, las muestras nasales y faríngeas de 102 estudiantes de 14 a 17 años fueron colectadas en medios de transporte y luego cultivadas en los laboratorios del Instituto de Medicina Tropical, en agar sabouraud y CHROMOagar Candida. Se identificó las colonias sospechosas de Candida sp mediante el estudio de clamiodoconidias, tubo germinativo y pruebas metabólicas. Principales medidas de resultados: Identificación de levaduras de Candida sp. Resultados: Se aisló levaduras del género Cándida en 11 de los escolares (10,8%). El 36,4% de las levaduras presentó resistencia moderada al antimicótico fluconazol. Conclusiones: Es recomendable continuar con estudios de vigilancia epidemiológica sobre las levaduras de importancia médica en portadores nasofaríngeos, con el fin estar preparados ante eventuales cuadros infecciosos

    Host galaxy identification for supernova surveys

    Host galaxy identification is a crucial step for modern supernova (SN) surveys such as the Dark Energy Survey (DES) and the Large Synoptic Survey Telescope (LSST), which will discover SNe by the thousands. Spectroscopic resources are limited, so in the absence of real-time SN spectra these surveys must rely on host galaxy spectra to obtain accurate redshifts for the Hubble diagram and to improve photometric classification of SNe. In addition, SN luminosities are known to correlate with host-galaxy properties. Therefore, reliable identification of host galaxies is essential for cosmology and SN science. We simulate SN events and their locations within their host galaxies to develop and test methods for matching SNe to their hosts. We use both real and simulated galaxy catalog data from the Advanced Camera for Surveys General Catalog and MICECATv2.0, respectively. We also incorporate "hostless" SNe residing in undetected faint hosts into our analysis, with an assumed hostless rate of 5%. Our fully automated algorithm is run on catalog data and matches SNe to their hosts with 91% accuracy. We find that including a machine learning component, run after the initial matching algorithm, improves the accuracy (purity) of the matching to 97% with a 2% cost in efficiency (true positive rate). Although the exact results are dependent on the details of the survey and the galaxy catalogs used, the method of identifying host galaxies we outline here can be applied to any transient survey
