    Neurobiological mechanisms of control in alcohol use disorder – Moving towards mechanism-based non-invasive brain stimulation treatments

    Alcohol use disorder (AUD) is characterized by excessive habitual drinking and loss of control over alcohol intake despite negative consequences. Both of these aspects foster uncontrolled drinking and high relapse rates in AUD patients. Yet, common interventions mostly focus on the phenomenological level, and prioritize the reduction of craving and withdrawal symptoms. Our review provides a mechanistic understanding of AUD and suggests alternative therapeutic approaches targeting the mechanisms underlying dysfunctional alcohol-related behaviours. Specifically, we explain how repeated drinking fosters the development of rigid drinking habits and is associated with diminished cognitive control. These behavioural and cognitive effects are then functionally related to the neurobiochemical effects of alcohol abuse. We further explain how alterations in fronto-striatal network activity may constitute the neurobiological correlates of these alcohol-related dysfunctions. Finally, we discuss limitations in current pharmacological AUD therapies and suggest non-invasive brain stimulation (like TMS and tDCS interventions) as a potential addition/alternative for modulating the activation of both cortical and subcortical areas to help re-establish the functional balance between controlled and automatic behaviour

    Affective styles and emotional lateralization: A promising framework for animal welfare research

    Abstract The growing recognition of animals as individuals has broader implications for farm animal welfare research. Even under highly standardized on-farm conditions, farm animals show heterogeneous but individually consistent behavioural patterns towards various stimuli, based on how they appraise these stimuli. As a result, animal welfare is likely to be highly individual as well, and studying the proximate mechanisms underlying distinct individual behaviour patterns and appraisal will improve animal welfare research. We propose to extend the framework of affective styles to bridge the gap between existing research fields on animal personality and affective states. Affective styles refer to consistent individual differences in emotional reactivity and regulation and can be predicted by baseline cerebral lateralization. Likewise, animals with consistent left or right motor biases—a proxy measure of individual patterns in cerebral lateralization—have been shown to differ in their personality, emotional reactivity, motivational tendencies or coping styles. In this paper, we present the current knowledge of the links between laterality and stable individual traits in behaviour and affect in light of hypotheses on emotional lateralization. Within our suggested framework, we make recommendations on how to investigate affective styles in non-human animals and give practical examples. This approach has the potential to promote a science of affective styles in nonhuman animals and significantly advance research on animal welfare

    The neurophysiology of continuous action monitoring.

    Monitoring actions is essential for goal-directed behavior. However, as opposed to short-lasting, and regularly reinstating monitoring functions, the neural processes underlying continuous action monitoring are poorly understood. We investigate this using a pursuit-tracking paradigm. We show that beta band activity likely maintains the sensorimotor program, while theta and alpha bands probably support attentional sampling and information gating, respectively. Alpha and beta band activity are most relevant during the initial tracking period, when sensorimotor calibrations are most intense. Theta band shifts from parietal to frontal cortices throughout tracking, likely reflecting a shift in the functional relevance from attentional sampling to action monitoring. This study shows that resource allocation mechanisms in prefrontal areas and stimulus-response mapping processes in the parietal cortex are crucial for adapting sensorimotor processes. It fills a knowledge gap in understanding the neural processes underlying action monitoring and suggests new directions for examining sensorimotor integration in more naturalistic experiments.     Investigating structural and functional neural correlates in children and adolescents with antisocial behavior

    Antisocial behavior is highly prevalent in young and adult populations worldwide and constitutes a major public health problem due to the huge burden on the individual as well as the significant economic burden on society. A better understanding of the underlying neurobiological mechanisms of antisocial behavior is warranted to improve current diagnostics (e.g. early detection of children at risk) and effective prevention/treatment programs. So far, neuroimaging studies have indicated neural atypicalities in youths with antisocial behavior; however, the direction and location of these brain alterations vary across studies. These ambiguities are most likely caused by the heterogeneity of the young samples with antisocial behavior studied, especially regarding sex, clinical diagnoses, and the presence of callous-unemotional traits. The central aim of this dissertation was to further the neuroscientific knowledge of antisocial behavior in children and adolescents by investigating the underlying structural and functional neurobiological characteristics, with an extra focus on possible sex differences and callous-unemotional traits. First, we examined the current neuroimaging literature, through meta-analyses, with the purpose of overcoming the heterogeneity of antisocial behavior and generating a common “overlapping” pattern of structural and functional atypicalities in youths with antisocial behavior. Secondly, the relation between callous-unemotional traits and brain structure was investigated separately for sex and independently of psychiatric comorbidities. Thirdly, this work investigated the white matter integrity within a homogenous group of girls with conduct disorder –the severe variant of antisocial behavior– in comparison to typically developing peers. This work expands our current knowledge on the structural and functional neural correlates in children and adolescents with antisocial behavior in several ways. For one, our meta-analytic results indicate a consistent pattern of gray matter reductions and hypoactivations in brain areas within the prefrontal and limbic cortex. These findings fit a recently proposed neurobiological model that connects alterations within similar brain regions with the behavioral dispositions of antisocial behavior (e.g. dysfunctions in empathy, emotional learning, and decision making). Secondly, we observed a positive relation between callous-unemotional traits and bilateral insula volume in a large international population of typically developing boys, but not in girls, independent of psychiatric disorders. This demonstrates that callous-unemotional traits have a sex-specific neurobiological basis beyond psychiatric samples. Thirdly, this work presents novel findings of white-matter integrity alterations in the body of the corpus callosum of girls with antisocial behavior, indicating possible reduced interhemispheric processing and consequent emotion processing abilities. In short, the present thesis provides original findings regarding the neurobiology of antisocial behavior in youths and emphasizes the importance of callous-unemotional traits and sex differences. Our results encourage future studies to further investigate the developmental trajectories and potential neural markers of antisocial behavior in order to enhance early detection and improve intervention programs, which could ultimately reduce antisocial behavior and delinquency in our society
