    Leadership Styles of Park and Recreation Professionals in GRPA

    Undergraduates in the Park and Recreation program at Columbus State University conducted a temperament survey using the Keirsey Temperament Sorter II to identify the temperaments/leadership styles of 50 randomly chosen Park and Recreation professionals within GRPA (Georgia Recreation and Park Association). The Keirsey divides individuals into four main temperaments: Guardian, Artisan, Idealist, and Rational. It is the duty of every leader to evaluate himself or herself in recognizing strengths, weaknesses, capabilities, and limitations. Of the 33 respondents, 73% tested as Guardians, 12% as Artisans, 6% as Idealists, and 3% as Rational. Twenty-seven percent (27%) are more specifically described as Guardian Supervisors. According to http://www.keirsey.com, Guardian Supervisors are squarely on the side of rules and procedures, and they can be quite serious about seeing to it that others toe the mark or else face the consequences. They do not hesitate to give their stamp of approval, nor do they withhold their directions or suggestions for improvement

    Evaluation of the red seaweed Mazzaella japonica as a feed additive for beef cattle

    Supplementing ruminant diets with macroalgae is gaining interest globally because bromoform-containing seaweeds (e.g., Asparagopsis spp.) have been shown to be highly effective enteric methane (CH4) inhibitors. Some alternative seaweeds decrease in vitro CH4 production, but few have been evaluated in animals. This study examined the effects of including the red seaweed Mazzaella japonica in the diet of beef cattle on dry matter intake (DMI), rumen fermentation, digestibility, nitrogen (N) utilization, and enteric CH4 production. Six ruminally cannulated, mature beef heifers (824 ± 47.1 kg) were used in a double 3 × 3 Latin square with 35-d periods. The basal diet consisted of 52% barley silage, 44% barley straw, and 4% vitamin and mineral supplement [dry matter (DM) basis]. The treatments were (DM basis): 0% (control), 1%, and 2% M. japonica. The DMI increased quadratically (P = 0.025) with the inclusion of M. japonica, such that the DMI of heifers consuming 1% was greater (P < 0.05) than that of control heifers. The apparent total-tract digestibility of DM decreased linearly (P = 0.002) with the inclusion of M. japonica, but there were no treatment differences in the digestibility of organic matter, crude protein (CP), neutral detergent fiber, or starch. The level of M. japonica linearly (P < 0.001) increased the N intake of the heifers. Fecal N excretion linearly increased (P = 0.020) with M. japonica, but there were no differences in total urinary N excretion, N fractions (allantoin, uric acid), total purine derivatives, microbial purine derivatives absorbed, microbial N flow, or retained N. There were no treatment effects on rumen pH or total volatile fatty acids (VFAs); however, adding M. japonica to the diet quadratically (P = 0.023) decreased the proportion of acetate, whereas 1% inclusion decreased the acetate proportion. Methane production (g/day) decreased quadratically (P = 0.037), such that the heifers receiving 2% M. japonica produced 9.2% less CH4 than control animals; however, CH4 yield (g/kg DMI) did not differ among treatments. We conclude that supplementing a forage-based diet with up to 2% M. japonica failed to lower the enteric CH4 yield of beef heifers. M. japonica can be used in diets to help meet the CP requirements of cattle, but inclusion rates may be limited by high inorganic matter proportions. When comprising up to 2% of the diet, M. japonica cannot be recommended as a CH4 inhibitor for beef cattle fed on high-forage diets

    Bayesian atmospheric tomography for detection and quantification of methane emissions : application to data from the 2015 Ginninderra release experiment

    Detection and quantification of greenhouse-gas emissions is important for both compliance and environment conservation. However, despite several decades of active research, it remains predominantly an open problem, largely due to model errors and assumptions that appear at each stage of the inversion processing chain. In 2015, a controlled-release experiment headed by Geoscience Australia was carried out at the Ginninderra Controlled Release Facility, and a variety of instruments and methods were employed for quantifying the release rates of methane and carbon dioxide from a point source. This paper proposes a fully Bayesian approach to atmospheric tomography for inferring the methane emission rate of this point source using data collected during the experiment from both point-and path-sampling instruments. The Bayesian framework is designed to account for uncertainty in the parameterisations of measurements, the meteorological data, and the atmospheric model itself when performing inversion using Markov chain Monte Carlo (MCMC). We apply our framework to all instrument groups using measurements from two release-rate periods. We show that the inversion framework is robust to instrument type and meteorological conditions. From all the inversions we conducted across the different instrument groups and release-rate periods, our worst-case median emission rate estimate was within 36% of the true emission rate. Further, in the worst case, the closest limit of the 95% credible interval to the true emission rate was within 11% of this true value

    The Ginninderra CH4 and CO2 release experiment: An evaluation of gas detection and quantification techniques

    A methane (CH4) and carbon dioxide (CO2) release experiment was held from April to June 2015 at the Ginninderra Controlled Release Facility in Canberra, Australia. The experiment provided an opportunity to compare different emission quantification techniques against a simulated CH4 and CO2 point source release, where the actual release rates were unknown to the participants. Eight quantification techniques were assessed: three tracer ratio techniques (two mobile); backwards Lagrangian stochastic modelling; forwards Lagrangian stochastic modelling; Lagrangian stochastic (LS) footprint modelling; atmospheric tomography using point and using integrated line sensors. The majority of CH4 estimates were within 20% of the actual CH4 release rate (5.8 g/min), with the tracer ratio technique providing the closest estimate to both the CH4 and CO2 release rates (100 g/min). Once the release rate was known, the majority of revised estimates were within 10% of the actual release rate. The study illustrates the power of measuring the emission rate using multiple simultaneous methods and obtaining an ensemble median or mean. An ensemble approach to estimating the CH4 emission rate proved successful with the ensemble median estimate within 16% for the actual release rate for the blind release experiment and within 2% once the release rate was known. The release also provided an opportunity to assess the effectiveness of stationary and mobile ground and aerial CH4 detection technologies. Sensor detection limits and sampling rates were found to be significant limitations for CH4 and CO2 detection. A hyperspectral imager\u27s capacity to image the CH4 release from 100 m, and a Boreal CH4 laser sensor\u27s ability to track moving targets suggest the future possibility to map gas plumes using a single laser and mobile aerial reflector

    This paper examines the contributions Management Accounting Research (MAR) has (and has not) made to social and critical analyses of management accounting in the twenty-five years since its launch. It commences with a personalised account of the first named author’s experiences of behavioural, social and critical accounting in the twenty-five years before MAR appeared. This covers events in the UK, especially the Management Control Workshop, Management Accounting Research conferences at Aston, the Inter-disciplinary Perspectives on Accounting Conferences; key departments and professors; and elsewhere the formation of pan-European networks, and reflections on a years’ visit to the USA. Papers published by MAR are analysed according to year of publication, country of author and research site, research method, research subject (type of organization or subject studied), data analysis method, topic, and theory. This revealed, after initial domination by UK academics, increasing Continental European influence; increasing use of qualitative methods over a wide range of topics, especially new costing methods, control system design, change and implementation, public sector transformation, and more recently risk management and creativity. Theoretical approaches have been diverse, often multi-disciplinary, and have employed surprisingly few economic theories relative to behavioural and social theories. The research spans mainly large public and private sector organisations especially in Europe. Seven themes perceived as of interest to a social and critical theory analysis are evaluated, namely: the search for ‘Relevance Lost’ and new costing; management control, the environment and the search for ‘fits’; reconstituting the public sector; change and institutional theory; post-structural, constructivist and critical contributions; social and environmental accounting; and the changing geography of time and space between European and American research. The paper concludes by assessing the contributions of MAR against the aspirations of groups identified in the opening personal historiography, which have been largely met. MAR has made substantial contributions to social and critical accounting (broadly defined) but not in critical areas endeavouring to give greater voice and influence to marginalised sectors of society worldwide. Third Sector organisations, politics, civil society involvement, development and developing countries, labour, the public interest, political economy, and until recently social and environmental accounting have been neglected

    Prevalence and architecture of de novo mutations in developmental disorders.

    The genomes of individuals with severe, undiagnosed developmental disorders are enriched in damaging de novo mutations (DNMs) in developmentally important genes. Here we have sequenced the exomes of 4,293 families containing individuals with developmental disorders, and meta-analysed these data with data from another 3,287 individuals with similar disorders. We show that the most important factors influencing the diagnostic yield of DNMs are the sex of the affected individual, the relatedness of their parents, whether close relatives are affected and the parental ages. We identified 94 genes enriched in damaging DNMs, including 14 that previously lacked compelling evidence of involvement in developmental disorders. We have also characterized the phenotypic diversity among these disorders. We estimate that 42% of our cohort carry pathogenic DNMs in coding sequences; approximately half of these DNMs disrupt gene function and the remainder result in altered protein function. We estimate that developmental disorders caused by DNMs have an average prevalence of 1 in 213 to 1 in 448 births, depending on parental age. Given current global demographics, this equates to almost 400,000 children born per year

    Methionine biosynthesis and transport are functionally redundant for the growth and virulence of Salmonella Typhimurium

    Methionine (Met) is an amino acid essential for many important cellular and biosynthetic functions, including the initiation of protein synthesis and S-adenosylmethionine-mediated methylation of proteins, RNA, and DNA. The de novo biosynthetic pathway of Met is well conserved across prokaryotes but absent from vertebrates, making it a plausible antimicrobial target. Using a systematic approach, we examined the essentiality of de novo methionine biosynthesis in Salmonella enterica serovar Typhimurium, a bacterial pathogen causing significant gastrointestinal and systemic diseases in humans and agricultural animals. Our data demonstrate that Met biosynthesis is essential for S. Typhimurium to grow in synthetic medium and within cultured epithelial cells where Met is depleted in the environment. During systemic infection of mice, the virulence of S. Typhimurium was not affected when either de novo Met biosynthesis or high-affinity Met transport was disrupted alone, but combined disruption in both led to severe in vivo growth attenuation, demonstrating a functional redundancy between de novo biosynthesis and acquisition as a mechanism of sourcing Met to support growth and virulence for S. Typhimurium during infection. In addition, our LC-MS analysis revealed global changes in the metabolome of S. Typhimurium mutants lacking Met biosynthesis and also uncovered unexpected interactions between Met and peptidoglycan biosynthesis. Together, this study highlights the complexity of the interactions between a single amino acid, Met, and other bacterial processes leading to virulence in the host and indicates that disrupting the de novo biosynthetic pathway alone is likely to be ineffective as an antimicrobial therapy against S. Typhimurium

    Finishing the euchromatic sequence of the human genome

    The sequence of the human genome encodes the genetic instructions for human physiology, as well as rich information about human evolution. In 2001, the International Human Genome Sequencing Consortium reported a draft sequence of the euchromatic portion of the human genome. Since then, the international collaboration has worked to convert this draft into a genome sequence with high accuracy and nearly complete coverage. Here, we report the result of this finishing process. The current genome sequence (Build 35) contains 2.85 billion nucleotides interrupted by only 341 gaps. It covers ∼99% of the euchromatic genome and is accurate to an error rate of ∼1 event per 100,000 bases. Many of the remaining euchromatic gaps are associated with segmental duplications and will require focused work with new methods. The near-complete sequence, the first for a vertebrate, greatly improves the precision of biological analyses of the human genome including studies of gene number, birth and death. Notably, the human enome seems to encode only 20,000-25,000 protein-coding genes. The genome sequence reported here should serve as a firm foundation for biomedical research in the decades ahead

    Heterozygous Variants in KMT2E Cause a Spectrum of Neurodevelopmental Disorders and Epilepsy.

    We delineate a KMT2E-related neurodevelopmental disorder on the basis of 38 individuals in 36 families. This study includes 31 distinct heterozygous variants in KMT2E (28 ascertained from Matchmaker Exchange and three previously reported), and four individuals with chromosome 7q22.2-22.23 microdeletions encompassing KMT2E (one previously reported). Almost all variants occurred de novo, and most were truncating. Most affected individuals with protein-truncating variants presented with mild intellectual disability. One-quarter of individuals met criteria for autism. Additional common features include macrocephaly, hypotonia, functional gastrointestinal abnormalities, and a subtle facial gestalt. Epilepsy was present in about one-fifth of individuals with truncating variants and was responsive to treatment with anti-epileptic medications in almost all. More than 70% of the individuals were male, and expressivity was variable by sex; epilepsy was more common in females and autism more common in males. The four individuals with microdeletions encompassing KMT2E generally presented similarly to those with truncating variants, but the degree of developmental delay was greater. The group of four individuals with missense variants in KMT2E presented with the most severe developmental delays. Epilepsy was present in all individuals with missense variants, often manifesting as treatment-resistant infantile epileptic encephalopathy. Microcephaly was also common in this group. Haploinsufficiency versus gain-of-function or dominant-negative effects specific to these missense variants in KMT2E might explain this divergence in phenotype, but requires independent validation. Disruptive variants in KMT2E are an under-recognized cause of neurodevelopmental abnormalities
