    Heat shock protein 70 and nitric oxide concentrations in non-tumorous and neoplastic canine mammary tissues: preliminary results – Short communication

    The concentrations of heat shock protein 70 (Hsp70) and nitric oxide ions (NO), measured as nitrite, were determined in canine mammary tumours and nontumorous mammary gland tissues. The concentrations of Hsp70 and NO were significantly higher in both benign and malignant tumours than in non-tumorous mammary tissues. Hsp70 concentration decreased with the increase in the grade of histological malignancy. A strong positive correlation was found between the concentrations of Hsp70 and NO in the benign tumours as well as in grade I and grade II malignant tumours. The results indicate that the process of neoplastic transformation in the canine mammary gland is related to a significant increase in Hsp70 and NO concentration in tumour tissues, and an interdependence between Hsp70 and nitrite ion production can be observed

    Učinak odabranih makrolida na neka obilježja goveđih leukocita

    The aim of the study was to evaluate the effect of tylosin, tilmicosin and roxithromycin on viability, nitro blue tetrazolium reduction (NBT) assay, chemotaxis, apoptosis and oxidative stress in bovine leukocytes in vitro conditions. The material for the study consisted of blood collected into EDTA tubes from the external jugular vein of Holstein-Friesian cattle aged 1 week to 2.5 years, during routine veterinary examinations. In leukocytes the percent of viability, nitrate ion concentration (NO), metabolic activity (NBT, nitrotetrazolium blue reduction assay), chemotactic activity and apoptosis were determined. The results indicated a slight negative effect of these macrolides on the viability of the leukocytes, and confirmed the ability of macrolides to induce apoptosis in leukocytes in vitro. These results indicate that all of the macrolides investigated exhibit a modulatory effect on the functions of leukocytes isolated from cattle of different ages. The strongest inhibitory effect on the NBT reduction assay and chemotaxis of the leukocytes was exhibited by roxithromycin, which at the same time had the least negative effect on the leukocytes.Cilj je ovog istraživanja bio procijeniti učinak tilozina, tilmikozina i roksitromicina na preživljavanje, test redukcije “nitro-tetrazol-modrog” (NBT), kemotaksiju, apoptozu i oksidacijski stres goveđih leukocita u uvjetima in vitro. Prilikom rutinskog veterinarskog pregleda, u epruvete s EDTA bila je uzeta krv iz jugularne vene goveda holštajnsko-frizijske pasmine u dobi od tjedan dana do 2,5 godine. Određen je postotak preživljavanja leukocita, koncentracija iona nitrata (NO), metabolička aktivnost (test redukcije “nitro-tetrazol-modrog”, NBT), kemotaksijska aktivnost i apoptoza. Rezultati su pokazali blagi negativni učinak spomenutih makrolida na preživljavanje leukocita i potvrdili sposobnost makrolida da potaknu njihovu apoptozu in vitro te naznačuju da svi pretraženi makrolidi imaju modulacijski učinak na funkciju leukocita goveda različite dobi. Najjači inhibicijski učinak na test redukcije NBT i kemotaksiju leukocita pokazao je roksitromicin, koji je istodobno imao najmanje negativan učinak na leukocite

    Characterization and comparison of enterococcus spp. Isolates from feces of healthy dogs and urine of dogs with utis

    Enterococcus spp. are opportunistic pathogens of both humans and animals characterized by high resistance to antimicrobials. Dogs could be intestinal carriers or suffer from Enterococcus infections, mainly urinary tract infections (UTIs). This study aimed to analyze and compare En-terococcus spp. isolated from healthy dog stools and sick dog urine. Overall, 51 isolates (29 from stools and 22 from UTI) were characterized at species level and tested for antimicrobial resistance, biofilm production and presence of resistance and virulence genes. E. faecium and E. faecalis resulted as equally distributed in stools samples, while E. faecalis predominated among UTI isolates. HLAR phenotype was detected in 47.1% isolates; 64.7% isolates were resistant to ampicillin (47.1% with a MIC ≥ 64 µg/mL). High levels of resistance were recorded for fluoroquinolones (enrofloxacin 74.5%, ciprofloxacin 66.7%), clindamycin (84.3%), tetracycline (78.4%) and quinupristin–dalfopristin (78.4%). No vancomycin resistant strains were detected. All but one isolate were multidrug-resistant. Most detected resistance genes were tetM (70.5%), pbp4 (52.9%) and aph(3′ )-IIIa (39.2%). All isolates were able to produce biofilm, but isolates from UTIs and belonging to E. faecalis more frequently resulted in strong biofilm producers. Most detected virulence genes were asa1 (52.9%), gelE (41.2%), cylA (37.3%) and esp (35.3%); all of them resulted as more frequently associated to E. faecalis. No particular differences emerged between isolates from feces and UTI, considering all evaluated aspects. Our results confirm pet dogs as carriers of multidrug-resistant enterococci; stool microflora could be considered as the most probable source of enterococcal UTI and E. faecalis carried by dogs seems to be more virulent than E. faecium, justifying its more frequent involvement in urinary tract infections

    Seroprevalence of bovine herpesvirus 1 related alphaherpesvirus infections in free-living and captive cervids in Poland.

    To determine the occurrence of bovine herpesvirus 1 (BoHV-1) related alphaherpesvirus infections in cervids, 1194 serum samples of wild ruminants originating from 59 forest districts of Poland were tested with IBR gB ELISA and virus neutralization test (VNT) against BoHV-1 and cervid herpesvirus 1 (CvHV-1). The seroprevalence differed significantly between free-living and captive cervids (P<0.001) with a total of 89 out of 498 (17.9%) and 268 out of 696 (38.5%) seropositive animals in each type of population. In free-ranging cervids, the highest seroprevalence was found among red deer (25.6%) and in fallow deer (23.1%), while it was the lowest in roe deer (1.7%). The seroprevalence varied at the district level between 0 and 100% with the mean value of 17.4% (95% CI:10.1-24.0). Additionally, seroprevalence was associated with afforestation (chi(2)=7.5; P=0.006) and to some degree with the mean of cattle density in province (chi(2)=7.0; P=0.08). The mean antibody titre against CvHV-1 in VNT (161.8; 95%CI: 146.0-177.6) has been significantly higher (P<0.0001) than the mean titre of BoHV-1 antibodies (10.1; 95%CI: 8.9-11.4). The results showed that BoHV-1 related alphaherpesvirus infections are present in population of free-ranging and farmed cervids in Poland. Based on the VNT results and considering the low susceptibility of red deer to BoHV-1, it seems that the dominant alphaherpesvirus circulating in wild ruminants is most likely CvHV-1 and therefore it is rather unlikely that deer in Poland could play any role as a reservoir of BoHV-1 for cattle

    Wetting films on chemically heterogeneous substrates

    Based on a microscopic density functional theory we investigate the morphology of thin liquidlike wetting films adsorbed on substrates endowed with well-defined chemical heterogeneities. As paradigmatic cases we focus on a single chemical step and on a single stripe. In view of applications in microfluidics the accuracy of guiding liquids by chemical microchannels is discussed.     First narrow-band search for continuous gravitational waves from known pulsars in advanced detector data

    Spinning neutron stars asymmetric with respect to their rotation axis are potential sources of continuous gravitational waves for ground-based interferometric detectors. In the case of known pulsars a fully coherent search, based on matched filtering, which uses the position and rotational parameters obtained from electromagnetic observations, can be carried out. Matched filtering maximizes the signalto- noise (SNR) ratio, but a large sensitivity loss is expected in case of even a very small mismatch between the assumed and the true signal parameters. For this reason, narrow-band analysis methods have been developed, allowing a fully coherent search for gravitational waves from known pulsars over a fraction of a hertz and several spin-down values. In this paper we describe a narrow-band search of 11 pulsars using data from Advanced LIGO’s first observing run. Although we have found several initial outliers, further studies show no significant evidence for the presence of a gravitational wave signal. Finally, we have placed upper limits on the signal strain amplitude lower than the spin-down limit for 5 of the 11 targets over the bands searched; in the case of J1813-1749 the spin-down limit has been beaten for the first time. For an additional 3 targets, the median upper limit across the search bands is below the spin-down limit. This is the most sensitive narrow-band search for continuous gravitational waves carried out so far

    A gravitational-wave standard siren measurement of the Hubble constant

    On 17 August 2017, the Advanced LIGO 1 and Virgo 2 detectors observed the gravitational-wave event GW170817-a strong signal from the merger of a binary neutron-star system 3 . Less than two seconds after the merger, a γ-ray burst (GRB 170817A) was detected within a region of the sky consistent with the LIGO-Virgo-derived location of the gravitational-wave source 4-6 . This sky region was subsequently observed by optical astronomy facilities 7 , resulting in the identification 8-13 of an optical transient signal within about ten arcseconds of the galaxy NGC 4993. This detection of GW170817 in both gravitational waves and electromagnetic waves represents the first 'multi-messenger' astronomical observation. Such observations enable GW170817 to be used as a 'standard siren' 14-18 (meaning that the absolute distance to the source can be determined directly from the gravitational-wave measurements) to measure the Hubble constant. This quantity represents the local expansion rate of the Universe, sets the overall scale of the Universe and is of fundamental importance to cosmology. Here we report a measurement of the Hubble constant that combines the distance to the source inferred purely from the gravitational-wave signal with the recession velocity inferred from measurements of the redshift using the electromagnetic data. In contrast to previous measurements, ours does not require the use of a cosmic 'distance ladder' 19 : the gravitational-wave analysis can be used to estimate the luminosity distance out to cosmological scales directly, without the use of intermediate astronomical distance measurements. We determine the Hubble constant to be about 70 kilometres per second per megaparsec. This value is consistent with existing measurements 20,21 , while being completely independent of them. Additional standard siren measurements from future gravitationalwave sources will enable the Hubble constant to be constrained to high precision

    Multi-messenger observations of a binary neutron star merger

    On 2017 August 17 a binary neutron star coalescence candidate (later designated GW170817) with merger time 12:41:04 UTC was observed through gravitational waves by the Advanced LIGO and Advanced Virgo detectors. The Fermi Gamma-ray Burst Monitor independently detected a gamma-ray burst (GRB 170817A) with a time delay of ~1.7 s with respect to the merger time. From the gravitational-wave signal, the source was initially localized to a sky region of 31 deg2 at a luminosity distance of 40+8-8 Mpc and with component masses consistent with neutron stars. The component masses were later measured to be in the range 0.86 to 2.26 Mo. An extensive observing campaign was launched across the electromagnetic spectrum leading to the discovery of a bright optical transient (SSS17a, now with the IAU identification of AT 2017gfo) in NGC 4993 (at ~40 Mpc) less than 11 hours after the merger by the One- Meter, Two Hemisphere (1M2H) team using the 1 m Swope Telescope. The optical transient was independently detected by multiple teams within an hour. Subsequent observations targeted the object and its environment. Early ultraviolet observations revealed a blue transient that faded within 48 hours. Optical and infrared observations showed a redward evolution over ~10 days. Following early non-detections, X-ray and radio emission were discovered at the transient’s position ~9 and ~16 days, respectively, after the merger. Both the X-ray and radio emission likely arise from a physical process that is distinct from the one that generates the UV/optical/near-infrared emission. No ultra-high-energy gamma-rays and no neutrino candidates consistent with the source were found in follow-up searches. These observations support the hypothesis that GW170817 was produced by the merger of two neutron stars in NGC4993 followed by a short gamma-ray burst (GRB 170817A) and a kilonova/macronova powered by the radioactive decay of r-process nuclei synthesized in the ejecta

    Search for Tensor, Vector, and Scalar Polarizations in the Stochastic Gravitational-Wave Background

    The detection of gravitational waves with Advanced LIGO and Advanced Virgo has enabled novel tests of general relativity, including direct study of the polarization of gravitational waves. While general relativity allows for only two tensor gravitational-wave polarizations, general metric theories can additionally predict two vector and two scalar polarizations. The polarization of gravitational waves is encoded in the spectral shape of the stochastic gravitational-wave background, formed by the superposition of cosmological and individually unresolved astrophysical sources. Using data recorded by Advanced LIGO during its first observing run, we search for a stochastic background of generically polarized gravitational waves. We find no evidence for a background of any polarization, and place the first direct bounds on the contributions of vector and scalar polarizations to the stochastic background. Under log-uniform priors for the energy in each polarization, we limit the energy densities of tensor, vector, and scalar modes at 95% credibility to Ω0T<5.58×10-8, Ω0V<6.35×10-8, and Ω0S<1.08×10-7 at a reference frequency f0=25 Hz. © 2018 American Physical Society
