    Antibacterial effects of Eucalyptus globulus leaf extract on pathogenic bacteria isolated from specimens of patients with respiratory tract disorders

    ABSTRACTThe antibacterial activity of Eucalyptus globulus leaf extract was determined for 56 isolates of Staphylococcus aureus, 25 isolates of Streptococcus pyogenes, 12 isolates of Streptococcus pneumoniae and seven isolates of Haemophilus influenzae obtained from 200 clinical specimens of patients with respiratory tract disorders. MIC50s for these species were 64, 32, 16 and 16 mg/L, respectively; MIC90s were 128, 64, 32 and 32 mg/L, respectively; and MBCs were 512, 128, 64 and 64 mg/L, respectively. These results suggest that further studies to clarify the possible therapeutic role of E. globulus leaf extract in the treatment of respiratory tract infection are warranted

    Optimization and efficient purification in production of Brucella melitensis recombinant HSP and TF proteins with low endotoxin contents

    Background: The development of an effective subunit vaccine against brucellosis is a research area of intense. But optimization of recombinant proteins production in Escherichia coli and content of endotoxins associated with final recombinant proteins are very important. Objectives: In the present study, expression and purification of Brucella melitensis rHSP and rTF were optimized to reduce endotoxin contaminants. Materials and Methods: pDEST-tf and pDEST-hsp were transformed into E. coli BL21 (DE3), and then B. melitensis recombinant HSPA and TF proteins were overexpressed. Purification of these proteins was optimized to remove most of endotoxin contaminants from the end product using 0.1 Triton X-114 in washing buffers. Results: An endotoxin reduction of less than 0.05 EUmg/1 was achieved with protein recovery close to an 80 yield. Conclusions: As this new protocol requires only one step to simultaneously purify tagged proteins and eliminate endotoxins, it represents a substantial advantage in time, effort, and expense. © 2013, Ahvaz Jundishapur University of Medical Sciences; Licensee Kowsar Ltd

    The Combination of Soft System and Quality Function Deployment Methodologies in the Design and Development of the Comprehensive Model for World Class Manufacturing Processes

    The present paper attempts to design and develop a comprehensive model based on which the manufacturing processes can be promoted toward a world class manufacturing level. In this paper, it is believed that the Soft System Methodologies (SSM) can be utilized in a synthesized fashion to attain the world – class manufacturing status and the Quality Function Deployment (QFD) to analyze and assess customer needs and requirements and to design production processes used for the achievement of high–quality products. It is believed that such a combination can lead to the successful design of a model of key success criteria for the purpose of achieving the above mentioned goal.     A Comprehensive Model for Status Assessments’ Gap Evaluation in the World Class Manufacturing -- Based on Modifications’ Development of ESCAP Approach

    In this paper we generalize and develop a comprehensive model upon which we can determine the position of and gap in the world class manufacturing processes. This paper is based on the belief that for evaluation of world class manufacturing processes, we can use determination of gap in world class manufacturing processes, generalized from ESCAP (United Nations Economic and Social Commission for Asia and the Pacific). In order to achieve the above objective, and to determine the total position and gap in question, ESCAP model was used to generate a formula called Situation Contribution Coefficient (SCC) for Manufacturing Position at the global level.Key words: Manufacturing processes; Determining the position of world class processes; ESCAP mode

    Efficacy of compound therapy by ginseng and ciprofloxacin on bacterial prostatitis

    Objective: Genitourinary tract infections play a significant role in male infertility. Infections of reproductive sex glands, such as the prostate, impair function and indirectly affect male fertility. The general aim of this study is to investigate the protective effect of Korean red ginseng (KRG) on prostatitis in male rats treated with ciprofloxacin (CIPX). Materials and Methods: In this experimental study, we randomly divided 72 two male Wistar rats into 9 groups. The groups were treated as follows for 10 days: i. Control (no medication), ii. Sham (normal saline injection into the vas deferens and oral administration of phosphate-buffered saline (PBS), iii. Ginseng, iv. CPIX, v. CIPX+ginseng, vi. Uropathogenic Escherichia coli (E. coli) (UPEC), vii. UPEC+ginseng, viii. UPEC+CIPX, and ix. UPEC+ginseng+CIPX. The rats were killed 14 days after the last injection and the prostate glands were removed. After sample preparation, routine histology was performed using hematoxylin and eosin staining. The terminal deoxynucleotidyl transferase mediated dUTP-biotin nick end labeling (TUNEL) method was used to determine the presence of apoptotic cells. Results: The severity score for acinar changes and inflammatory cell infiltration in the UPEC+CIPX group did not significantly different from the UPEC group. However this score significantly decreased in the UPEC+CIPX+ginseng group compared to the UPEC group. Apoptotic index of all ginseng treated groups significantly decreased compared to the UPEC and CPIX groups. Conclusion: These results suggested that ginseng might be an effective adjunct in CIPX treatment of prostatitis. The combined use ginseng and CIPX was more effective than ginseng or CIPX alone
