Mung Bean nuclease mapping of RNAs 3' end
A method is described that allows an accurate mapping of 3' ends of RNAs. In this method a labeled DNA probe, containing the presumed 3' end of the RNA under analysis is allowed to anneals to the RNA itself. Mung-bean nuclease is then used to digest single strands of both RNA and DNA. Electrophoretic fractionation of "protected" undigested, labeled DNA is than performed using a sequence reaction of a known DNA as length marker. This procedure was applied to the analysis of both a polyA RNA (Interleukin 10 mRNA) and non polyA RNAs (sea urchin 18S and 26S rRNAs). This method might be potentially relevant for the evaluation of the role of posttrascriptional control of IL-10 in the pathogenesis of the immune and inflammatory mediated diseases associated to ageing. This might allow to develop new strategies to approach to the diagnosis and therapy of age related diseases
RACâ3 is a NFâÎșB coactivator
It has been shown that the molecular mechanism by which cytokines and glucocorticoids mutually antagonize their functions involves a mutual glucocorticoid receptor (GR)/nuclear factorâÎșB (NFâÎșB) transrepression. Here we report a role for the nuclear receptor coactivator RAC3, in modulating NFâÎșB transactivation. We found that RAC3 functions as a coactivator by binding to the active form of NFâÎșB and that overexpression of RAC3 restores GRâdependent transcription neglecting GR/NFâÎșB transrepression. The competition between GR and NFâÎșB for binding to RAC3 may represent a general mechanism by which both transcription factors mutually antagonize their activity.Fil: Werbajh, Santiago Enrique. The GAPS Programme with HARPS-N@TNG IX. The multi-planet system KELT-6: detection of the planet KELT-6 c and measurement of the Rossiter-McLaughlin effect for KELT-6 b
Aims. For more than 1.5 years we monitored spectroscopically the star KELT-6
(BD+312447), known to host the transiting hot Saturn KELT-6b, because a
previously observed long-term trend in radial velocity time series suggested
the existence of an outer companion. Methods. We collected a total of 93 new
spectra with the HARPS-N and TRES spectrographs. A spectroscopic transit of
KELT-6b was observed with HARPS-N, and simultaneous photometry was obtained
with the IAC-80 telescope. Results. We proved the existence of an outer planet
with a mininum mass Msini=3.710.21 M and a
moderately eccentric orbit () of period P3.5
years. We improved the orbital solution of KELT-6b and obtained the first
measurement of the Rossiter-McLaughlin effect, showing that the planet has a
likely circular, prograde, and slightly misaligned orbit, with a projected
spin-orbit angle =3611 degrees. We improved the KELT-6b
transit ephemeris from photometry, and we provided new measurements of the
stellar parameters. KELT-6 appears as an interesting case to study the

The GAPS Programme with HARPS-N@TNG VI: The Curious Case of TrES-4b
We revisit the TrES-4 system parameters based on high-precision HARPS-N
radial-velocity measurements and new photometric light curves. A combined
spectroscopic and photometric analysis allows us to determine a spectroscopic
orbit with an amplitude m s. The derived mass of TrES-4b is
found to be , significantly lower than
previously reported. Combined with the large radius () inferred from our analysis, TrES-4b becomes
the second-lowest density transiting hot Jupiter known. We discuss several
scenarios to explain the puzzling discrepancy in the mass of TrES-4b in the

The GAPS programme with HARPS-N@TNG IV: A planetary system around XO-2S
We performed an intensive radial velocity monitoring of XO-2S, the wide
companion of the transiting planet-host XO-2N, using HARPS-N at TNG in the
framework of the GAPS programme. The radial velocity measurements indicate the
presence of a new planetary system formed by a planet that is slightly more
massive than Jupiter at 0.48 au and a Saturn-mass planet at 0.13 au. Both
planetary orbits are moderately eccentric and were found to be dynamically
stable. There are also indications of a long-term trend in the radial
velocities. This is the first confirmed case of a wide binary whose components
both host planets, one of which is transiting, which makes the XO-2 system a
unique laboratory for understanding the diversity of planetary systems.Comment: 7 pages, 3 figures, accepted on A&A Lette
Transiting exoplanets from the CoRoT space mission VIII. CoRoT-7b: the first Super-Earth with measured radius
We report the discovery of very shallow (DF/F = 3.4 10-4), periodic dips in
the light curve of an active V = 11.7 G9V star observed by the CoRoT satellite,
which we interpret as due to the presence of a transiting companion. We
describe the 3-colour CoRoT data and complementary ground-based observations
that support the planetary nature of the companion. Methods. We use CoRoT color
information, good angular resolution ground-based photometric observations in-
and out- of transit, adaptive optics imaging, near-infrared spectroscopy and
preliminary results from Radial Velocity measurements, to test the diluted
eclipsing binary scenarios. The parameters of the host star are derived from
optical spectra, which were then combined with the CoRoT light curve to derive
parameters of the companion. We examine carefully all conceivable cases of
false positives, and all tests performed support the planetary hypothesis.
Blends with separation larger than 0.40 arcsec or triple systems are almost
excluded with a 8 10-4 risk left. We conclude that, as far as we have been
exhaustive, we have discovered a planetary companion, named CoRoT-7b, for which
we derive a period of 0.853 59 +/- 3 10-5 day and a radius of Rp = 1.68 +/-
0.09 REarth. Analysis of preliminary radial velocity data yields an upper limit
of 21 MEarth for the companion mass, supporting the finding.

Novel Blood Pressure Locus and Gene Discovery Using Genome-Wide Association Study and Expression Data Sets From Blood and the Kidney.
Elevated blood pressure is a major risk factor for cardiovascular disease and has a substantial genetic contribution. Genetic variation influencing blood pressure has the potential to identify new pharmacological targets for the treatment of hypertension. To discover additional novel blood pressure loci, we used 1000 Genomes Project-based imputation in 150 134 European ancestry individuals and sought significant evidence for independent replication in a further 228 245 individuals. We report 6 new signals of association in or near HSPB7, TNXB, LRP12, LOC283335, SEPT9, and AKT2, and provide new replication evidence for a further 2 signals in EBF2 and NFKBIA Combining large whole-blood gene expression resources totaling 12 607 individuals, we investigated all novel and previously reported signals and identified 48 genes with evidence for involvement in blood pressure regulation that are significant in multiple resources. Three novel kidney-specific signals were also detected. These robustly implicated genes may provide new leads for therapeutic innovation

The TrES-4 system was observed with HARPS-N on 17 individual epochs between March 2013 and July 2014. We carried out Ic-band precision photometric observations of two complete transit events of TrES-4 b with the CAHA 1.23-m on UT 2013 July 6 and UT 2014 June 30. (2 data files)
Differential cross section measurements for the production of a W boson in association with jets in protonâproton collisions at âs = 7 TeV
Measurements are reported of differential cross sections for the production of a W boson, which decays into a muon and a neutrino, in association with jets, as a function of several variables, including the transverse momenta (pT) and pseudorapidities of the four leading jets, the scalar sum of jet transverse momenta (HT), and the difference in azimuthal angle between the directions of each jet and the muon. The data sample of pp collisions at a centre-of-mass energy of 7 TeV was collected with the CMS detector at the LHC and corresponds to an integrated luminosity of 5.0 fb[superscript â1]. The measured cross sections are compared to predictions from Monte Carlo generators, MadGraph + pythia and sherpa, and to next-to-leading-order calculations from BlackHat + sherpa. The differential cross sections are found to be in agreement with the predictions, apart from the pT distributions of the leading jets at high pT values, the distributions of the HT at high-HT and low jet multiplicity, and the distribution of the difference in azimuthal angle between the leading jet and the muon at low values.United States.
