    IgM and IgG as markers of infection transplacentary Neospora caninum in bovine fetuses

    O objetivo deste estudo foi determinar a ocorrĂȘncia de anticorpos anti-Neospora caninum em 260 amostras de soro coletadas de fetos bovinos de julho de 2007 a março de 2008, em abatedouro do municĂ­pio de Santa Maria, Rio Grande do Sul, Brasil. Para detecção de anticorpos anti-N. caninum, a tĂ©cnica de imunofluorescĂȘncia indireta foi utilizada tanto para a detecção de imunoglobulinas G e M. Amostras com tĂ­tulos e" 25 foram consideradas positivas. Das 260 amostras testadas, 15% (39/260) foram positivas para anticorpos anti-N. caninum. Destas, em 38 (97,4%) foi detectada a presença de IgG anti-N. caninum e em seis (15,4%) de IgM. Em cinco amostras (12,8%) detectaram-se ambos, IgG e IgM. Os resultados reafirmam a habilidade do N. caninum em determinar infecção fetal. A pesquisa de IgM foi de limitada importĂąncia na detecção da infecção via transplacentĂĄria em soro fetal bovino.The aim of this study was to determine the occurrence of anti-Neospora caninum antibodies in serum samples collected from 260 bovine fetuses, between July 2007 and March 2008, in an abattoir in the municipality of Santa Maria, Rio Grande do Sul, Brazil. An indirect fluorescent antibody test was used to detect anti-N. caninum immunoglobulins G (IgG) and M (IgM), using a cut-off of 1:25. Considering IgG or IgM detection, 15% (39/260) of the samples tested were found positive. Among the positive samples, 38 (97.4%) were positive for IgG and 6 (15.4%) for IgM. However 5 (12.8%) of it were positive for both IgG and IgM. These results are in agreement with the proven ability of N. caninum to fetal infection.     Uninfected dogs and those naturally infected with Leishmania chagasi exhibiting different clinical forms of disease were evaluated for the presence of anti-Neospora caninum and anti-Toxoplasma gondii antibodies. Blood samples were collected from 110 mongrel dogs. Sera were tested using the indirect fluorescent antibody test (IFAT), and the animals with visceral leishmaniasis (VL) (n=60) were classified clinically. Out of the 110 sera investigated, 5 (4.5%) were positive for N. caninum (IFAT≄50) and 36 (32.7%) for T. gondii (IFAT≄16). Anti-L. chagasi antibody titers in asymptomatic dogs (n=10) were found to be significantly lower (P<0.05) than those in oligosymptomatic ones (n=22), which were in turn significantly lower (P<0.05) than those in symptomatic ones (n=28). No association between Leishmania and N. caninum infections was observed. Among dogs infected with L. chagasi, a tendency (P=0.053) towards an association between the infection with T. gondii and the appearance of VL symptoms was observed, suggesting that the clinical manifestation of VL in dogs may enhance their susceptibility to T. gondii. The possible influence of the immunosuppressive status of canine leishmaniasis in the different clinical forms of the disease is discussed.A presença de anticorpos anti-Neospora caninum e anti-Toxoplasma gondii foi avaliada em cĂŁes nĂŁo infectados e naturalmente infectados com Leishmania chagasi manifestando diferentes formas clĂ­nicas da enfermidade. Amostras de sangue foram coletadas de 110 cĂŁes sem raça definida. Os soros foram avaliados por meio da reação de imunofluorescĂȘncia indireta (RIFI) e os animais com leishmaniose visceral (LV) (n=60) foram classificados clinicamente. Dos 110 soros analisados, 5 (4,5%) foram reativos para N. caninum (RIFI≄50) e 36 (32,7%) para T. gondii (RIFI≄16). Os tĂ­tulos de anticorpos anti-L. chagasi em cĂŁes assintomĂĄticos (n=10) foram significativamente (P<0,05) mais baixos que aqueles verificados em oligossintomĂĄticos (n=22), que por sua vez foram significativamente menores (P<0,05) que em cĂŁes sintomĂĄticos (n=28). NĂŁo foi observada associação entre infecçÔes por Leishmania e N. caninum. Entre os cĂŁes infectados com L. chagasi, verificou-se uma tendĂȘncia de associação (P=0.053) entre infecção com T. gondii e aparecimento de sinais clĂ­nicos da LV, o que sugere que a manifestação clĂ­nica da LV em cĂŁes pode aumentar sua susceptibilidade ao T. gondii. Our patient was seen as a result of having an irritating cough with wheezing, mainly at night, for more than a month. Asthma was diagnosed, and he responded favourably to the treatment given. Upon being informed that he had been examined for infertility for 5 years, alpha-1 antitrypsin (AAT) levels were requested. These confirmed that he had a phenotype SZ AAT deficiency. These findings, together with some evidence published recently, suggested that there is a need to rule out AAT deficiency in males with asthma and infertility

    Early immune responses and parasite tissue distribution in mice experimentally infected with oocysts of either archetypal or non-archetypal genotypes of Toxoplasma gondii

    In most of the world Toxoplasma gondii is comprised of archetypal types (types I, II and III); however, South America displays several non-archetypal strains. This study used an experimental mouse model to characterize the immune response and parasite kinetics following infection with different parasite genotypes. An oral inoculation of 50 oocysts per mouse from T. gondii M4 type II (archetypal, avirulent), BrI or BrIII (non-archetypal, virulent and intermediate virulent, respectively) for groups (G)2, G3 and G4, respectively was used. The levels of mRNA expression of cytokines, immune compounds, cell surface markers and receptor adapters [interferon gamma (IFNγ), interleukin (IL)-12, CD8, CD4, CD25, CXCR3 and MyD88] were quantified by SYBR green reverse transcription-quantitative polymerase chain reaction. Lesions were characterized by histology and detection by immunohistochemistry established distribution of parasites. Infection in G2 mice was mild and characterized by an early MyD88-dependent pathway. In G3, there were high levels of expression of pro-inflammatory cytokines IFNγ and IL-12 in the mice showing severe clinical symptoms at 8–11 days post infection (dpi), combined with the upregulation of CD25, abundant tachyzoites and tissue lesions in livers, lungs and intestines. Significant longer expression of IFNγ and IL-12 genes, with other Th1-balanced immune responses, such as increased levels of CXCR3 and MyD88 in G4, resulted in survival of mice and chronic toxoplasmosis, with the occurrence of tissue cysts in brain and lungs, at 14 and 21 dpi. Different immune responses and kinetics of gene expression appear to be elicited by the different strains and non-archetypal parasites demonstrated higher virulence

    Detection model based on representation of quantum particles by classical random fields: Born's rule and beyond

    Recently a new attempt to go beyond quantum mechanics (QM) was presented in the form of so called prequantum classical statistical field theory (PCSFT). Its main experimental prediction is violation of Born's rule which provides only an approximative description of real probabilities. We expect that it will be possible to design numerous experiments demonstrating violation of Born's rule. Moreover, recently the first experimental evidence of violation was found in the triple slits interference experiment, see \cite{WWW}. Although this experimental test was motivated by another prequantum model, it can be definitely considered as at least preliminary confirmation of the main prediction of PCSFT. In our approach quantum particles are just symbolic representations of "prequantum random fields," e.g., "electron-field" or "neutron-field"; photon is associated with classical random electromagnetic field. Such prequantum fields fluctuate on time and space scales which are essentially finer than scales of QM, cf. `t Hooft's attempt to go beyond QM \cite{H1}--\cite{TH2}. In this paper we elaborate a detection model in the PCSFT-framework. In this model classical random fields (corresponding to "quantum particles") interact with detectors inducing probabilities which match with Born's rule only approximately. Thus QM arises from PCSFT as an approximative theory.     Measurement of the View the tt production cross-section using eÎŒ events with b-tagged jets in pp collisions at √s = 13 TeV with the ATLAS detector

    This paper describes a measurement of the inclusive top quark pair production cross-section (σttÂŻ) with a data sample of 3.2 fb−1 of proton–proton collisions at a centre-of-mass energy of √s = 13 TeV, collected in 2015 by the ATLAS detector at the LHC. This measurement uses events with an opposite-charge electron–muon pair in the final state. Jets containing b-quarks are tagged using an algorithm based on track impact parameters and reconstructed secondary vertices. The numbers of events with exactly one and exactly two b-tagged jets are counted and used to determine simultaneously σttÂŻ and the efficiency to reconstruct and b-tag a jet from a top quark decay, thereby minimising the associated systematic uncertainties. The cross-section is measured to be: σttÂŻ = 818 ± 8 (stat) ± 27 (syst) ± 19 (lumi) ± 12 (beam) pb, where the four uncertainties arise from data statistics, experimental and theoretical systematic effects, the integrated luminosity and the LHC beam energy, giving a total relative uncertainty of 4.4%. The result is consistent with theoretical QCD calculations at next-to-next-to-leading order. A fiducial measurement corresponding to the experimental acceptance of the leptons is also presented

    Variation in Structure and Process of Care in Traumatic Brain Injury: Provider Profiles of European Neurotrauma Centers Participating in the CENTER-TBI Study.

    INTRODUCTION: The strength of evidence underpinning care and treatment recommendations in traumatic brain injury (TBI) is low. Comparative effectiveness research (CER) has been proposed as a framework to provide evidence for optimal care for TBI patients. The first step in CER is to map the existing variation. The aim of current study is to quantify variation in general structural and process characteristics among centers participating in the Collaborative European NeuroTrauma Effectiveness Research in Traumatic Brain Injury (CENTER-TBI) study. METHODS: We designed a set of 11 provider profiling questionnaires with 321 questions about various aspects of TBI care, chosen based on literature and expert opinion. After pilot testing, questionnaires were disseminated to 71 centers from 20 countries participating in the CENTER-TBI study. Reliability of questionnaires was estimated by calculating a concordance rate among 5% duplicate questions. RESULTS: All 71 centers completed the questionnaires. Median concordance rate among duplicate questions was 0.85. The majority of centers were academic hospitals (n = 65, 92%), designated as a level I trauma center (n = 48, 68%) and situated in an urban location (n = 70, 99%). The availability of facilities for neuro-trauma care varied across centers; e.g. 40 (57%) had a dedicated neuro-intensive care unit (ICU), 36 (51%) had an in-hospital rehabilitation unit and the organization of the ICU was closed in 64% (n = 45) of the centers. In addition, we found wide variation in processes of care, such as the ICU admission policy and intracranial pressure monitoring policy among centers. CONCLUSION: Even among high-volume, specialized neurotrauma centers there is substantial variation in structures and processes of TBI care. This variation provides an opportunity to study effectiveness of specific aspects of TBI care and to identify best practices with CER approaches

    Search for TeV-scale gravity signatures in high-mass final states with leptons and jets with the ATLAS detector at sqrt [ s ] = 13TeV

    A search for physics beyond the Standard Model, in final states with at least one high transverse momentum charged lepton (electron or muon) and two additional high transverse momentum leptons or jets, is performed using 3.2 fb−1 of proton–proton collision data recorded by the ATLAS detector at the Large Hadron Collider in 2015 at √s = 13 TeV. The upper end of the distribution of the scalar sum of the transverse momenta of leptons and jets is sensitive to the production of high-mass objects. No excess of events beyond Standard Model predictions is observed. Exclusion limits are set for models of microscopic black holes with two to six extra dimensions

    A search is presented for dark matter produced in association with a hadronically decaying W or Z boson using 3.2 fb−1 of pp collisions at View the MathML sources=13 TeV recorded by the ATLAS detector at the Large Hadron Collider. Events with a hadronic jet compatible with a W or Z boson and with large missing transverse momentum are analysed. The data are consistent with the Standard Model predictions and are interpreted in terms of both an effective field theory and a simplified model containing dark matter
