    Identification of avian Mycoplasma species in commercial broilers and layers with respiratory symptoms in Balochistan

    Among many avian mycoplasmas, only the Mycoplasmas gallisepticum (MG) and Mycoplasmas synoviae (MS) are responsible for causing respiratory disease in commercial poultry. This study reported for the very first time the serological occurrence of M. gallisepticum and M. synoviae in blood samples (n = 600) from sixty flocks (n = 42 broiler and n = 18 layer flocks) with respiratory symptoms in Quetta, Pishin and Kuchlak districts of Balochistan, Pakistan. Sera were tested for MG and MS antibody responses by serum plate agglutination (SPA) test and by enzyme-linked immunosorbant assay (ELISA) Synbiotics kit. It was found that M. gallisepticum antibodies in broiler flocks detected by SPA and by ELISA tests were 10.47 and 19.76%, whereas M. synoviae were 7.86 and 11.19%, respectively. In layer flocks the MG and MS antibodies detected by SPA and ELISA were 19.44, 31.66 and 8.8, 25%, respectively. The overall antibodies of MG and MS in both broiler and layer flocks tested by SPA and ELISA was found to be 13.16, 23.33 and 8.16, 15.33%, respectively. In broiler and layer flocks the presence of antibodies against both M. gallisepticum and M. synoviae were found in tested flocks with respiratory symptoms. Further studies on prevalence and diagnosis of both the M. gallisepticum and M. synoviae in causing respiratory disease in commercial broilers and layers in Balochistan are required.Key words: Mycoplasma, broilers, serum plate agglutination, enzyme-linked immunosorbent assay

    What are the Alternatives of PG Plus Tests for Modified Asphalt Binders?

    For characterizing the polymer modified binders, different state Departments of Transportation (DOTs) use different time consuming and empirical Performance Grade (PG) Plus test methods. Furthermore, the PG Plus tests are silent when asphalt binders are modified with chemicals such as polyphosphoric acid (PPA). But, the effects of the polymer are not accurately identified through these conventional tests such as Elastic Recovery (ER) and tenacity. The main goal of this study is to recommend alternative test method(s), which can possibly be pursued by using, a Dynamic Shear Rheometer (DSR). Thus, Multiple Stress Creep and Recovery (MSCR), ER-DSR, Frequency Sweep, and Binder Yield Energy Test (BYET) are being explored to find their effectiveness. Three PG binders (PG 64-22, PG 70-22 and PG 76-22) have been selected for this investigation. These binders have been prepared with styrene-butadiene-styrene (SBS) polymer, PPA, or a combination of both. Further, chemical tests such as SARA (Saturate, Aromatic, Resin, and Asphaltene) analysis and FTIR (Fourier-transform Infrared spectroscopy) are also being explored to fulfill the objectives. Preliminary findings suggest that the PG Plus tests deem to be obsolete. Also, a single test procedure is not sufficient to identify the presence and effectiveness of modifiers in the PG binders

    Restoration of visual function in advanced disease after transplantation of purified human pluripotent stem cell-derived cone photoreceptors

    Age-related macular degeneration and other macular diseases result in the loss of light-sensing cone photoreceptors, causing irreversible sight impairment. Photoreceptor replacement may restore vision by transplanting healthy cells, which must form new synaptic connections with the recipient retina. Despite recent advances, convincing evidence of functional connectivity arising from transplanted human cone photoreceptors in advanced retinal degeneration is lacking. Here, we show restoration of visual function after transplantation of purified human pluripotent stem cell-derived cones into a mouse model of advanced degeneration. Transplanted human cones elaborate nascent outer segments and make putative synapses with recipient murine bipolar cells (BCs), which themselves undergo significant remodeling. Electrophysiological and behavioral assessments demonstrate restoration of surprisingly complex light-evoked retinal ganglion cell responses and improved light-evoked behaviors in treated animals. Stringent controls exclude alternative explanations, including material transfer and neuroprotection. These data provide crucial validation for photoreceptor replacement therapy and for the potential to rescue cone-mediated vision

    Symptoms and quality of life in late stage Parkinson syndromes: a longitudinal community study of predictive factors

    BACKGROUND Palliative care is increasingly offered earlier in the cancer trajectory but rarely in Idiopathic Parkinson's Disease(IPD), Progressive Supranuclear Palsy(PSP) or Multiple System Atrophy(MSA). There is little longitudinal data of people with late stage disease to understand levels of need. We aimed to determine how symptoms and quality of life of these patients change over time; and what demographic and clinical factors predicted changes. METHODS We recruited 82 patients into a longitudinal study, consenting patients with a diagnosis of IPD, MSA or PSP, stages 3-5 Hoehn and Yahr(H&Y). At baseline and then on up to 3 occasions over one year, we collected self-reported demographic, clinical, symptom, palliative and quality of life data, using Parkinson's specific and generic validated scales, including the Palliative care Outcome Scale (POS). We tested for predictors using multivariable analysis, adjusting for confounders. FINDINGS Over two thirds of patients had severe disability, over one third being wheelchair-bound/bedridden. Symptoms were highly prevalent in all conditions - mean (SD) of 10.6(4.0) symptoms. More than 50% of the MSA and PSP patients died over the year. Over the year, half of the patients showed either an upward (worsening, 24/60) or fluctuant (8/60) trajectory for POS and symptoms. The strongest predictors of higher levels of symptoms at the end of follow-up were initial scores on POS (AOR 1.30; 95%CI:1.05-1.60) and being male (AOR 5.18; 95% CI 1.17 to 22.92), both were more predictive than initial H&Y scores. INTERPRETATION The findings point to profound and complex mix of non-motor and motor symptoms in patients with late stage IPD, MSA and PSP. Symptoms are not resolved and half of the patients deteriorate. Palliative problems are predictive of future symptoms, suggesting that an early palliative assessment might help screen for those in need of earlier intervention

    Search for Gravitational Waves from Primordial Black Hole Binary Coalescences in the Galactic Halo

    We use data from the second science run of the LIGO gravitational-wave detectors to search for the gravitational waves from primordial black hole (PBH) binary coalescence with component masses in the range 0.2--1.0M1.0 M_\odot. The analysis requires a signal to be found in the data from both LIGO observatories, according to a set of coincidence criteria. No inspiral signals were found. Assuming a spherical halo with core radius 5 kpc extending to 50 kpc containing non-spinning black holes with masses in the range 0.2--1.0M1.0 M_\odot, we place an observational upper limit on the rate of PBH coalescence of 63 per year per Milky Way halo (MWH) with 90% confidence.Comment: 7 pages, 4 figures, to be submitted to Phys.     Inhibition of HCV 3a core gene through Silymarin and its fractions

    Hepatitis C is a major health problem affecting 270 million individuals in world including Pakistan. Current treatment regimen, interferon alpha and ribavirin only cure half of patients due to side effects and high cost. In the present study Silybum marianum (Milk thistle) seeds were collected, extracted and analyzed against HCV 3a core gene by transiently transfecting the liver cells with HCV core plasmid. Our results demonstrated that Silymarin (SM) dose dependently inhibit the expression or function of HCV core gene at a non toxic concentration while the GAPDH remained constant. To identify the active ingredient, SM was fractioned by thin layer chromatography (TLC), column chromatography and HPLC. Purified fractions were tested for HCV core gene and western blotting results showed that two factions of SM (S1 and S2) inhibit HCV 3a core expression or function in liver cells Our results suggest SM and its fractions (S1 and S2) inhibit HCV core gene of 3a genotype and combination of SM and its fractions with interferon will be a better option to treat HCV infection

    Search for new phenomena in final states with an energetic jet and large missing transverse momentum in pp collisions at √ s = 8 TeV with the ATLAS detector

    Results of a search for new phenomena in final states with an energetic jet and large missing transverse momentum are reported. The search uses 20.3 fb−1 of √ s = 8 TeV data collected in 2012 with the ATLAS detector at the LHC. Events are required to have at least one jet with pT > 120 GeV and no leptons. Nine signal regions are considered with increasing missing transverse momentum requirements between Emiss T > 150 GeV and Emiss T > 700 GeV. Good agreement is observed between the number of events in data and Standard Model expectations. The results are translated into exclusion limits on models with either large extra spatial dimensions, pair production of weakly interacting dark matter candidates, or production of very light gravitinos in a gauge-mediated supersymmetric model.
