    Deterministic Preparation of Individual Rubidium-85 Dimers via Photoassociation

    In this thesis, we present experimental methods and results for direct measurements of the photoassociation of two individual ultra-cold rubidium-85 atoms, each prepared in the |F = 2,m_F = −2⟩ ground state. From there two individual rubidium-85 atoms are loaded into separate far-off resonance optical tweezers which are near-deterministically prepared into |F = 2,m_F = −2⟩. The two optical tweezers are then merged and the atoms are exposed to photoassociation light. To ensure the correct frequency of photoassociation light is used a dual cavity locking scheme, capable of generating frequencies with a stability of ±425.7 kHz over a large wavelength range for use in photoassociation spectroscopy, was developed. From there, a light modulation technique is implemented to isolate the photoassociation event from unwanted far-off resonance trap effects, such as the AC Stark shift. By controlling the intensity, frequency, and duration parameters of the photoassociation pulses applied to the rubidium atoms, frequency-dependent, and time-dependent atom loss is observed for a single band of the 0^{+}_{u} series; located at 377.00067±0.00006 THz. The resonance and linewidth data exhibits the same behaviour as photoassociation performed on large atom ensembles within the many-body regime. However, two distinct fast and slow photoassociation rates are observed in contrast to the many-body regime where only a single photoassociation rate constant, K_{PA}, has been reported. This marks a novel physical process which heretofore has not been reported in the field of atomic or molecular physics

    Cetaceans sightings during research cruises in three remote Atlantic British Overseas Territories

    Marine mammal sightings were recorded during research cruises to three remote, mid-ocean British Overseas Territories in the South Atlantic and Southern Ocean. In March to April 2018 and 2019, the Exclusive Economic Zones (EEZs) of tropical St Helena and temperate Tristan da Cunha were surveyed. The sub-polar waters of South Georgia and the South Sandwich Islands (SGSSI) were surveyed in February to March 2019. At St Helena in 2018, five species were recorded during 11 sightings, and in 2019, four species, with one additional unidentified species, during seven sightings. Most of these sightings were of dolphin species, which are known to be resident around the Island and seamounts. In Tristan da Cunha in 2018, a total of five identified and one unidentified species were recorded during six sightings, half of which were associated with the Islands or seamounts. In 2019, due to rough weather, no sightings were recorded in the Tristan waters. Around SGSSI, 162 sightings of 236 cetaceans were made in 2019, mostly of baleen whales, with seven species identified with certainty. Sightings around the southern South Sandwich Islands' included beaked whales and large dolphins, whereas baleen whales dominated in the northern South Sandwich Islands. These results provide new data for rarely surveyed regions, helping to build a spatial picture of important areas for marine mammals, which will help inform marine spatial protection strategies

    Accuracy of methods for diagnosing atrial fibrillation using 12-lead ECG: a systematic review and meta-analysis

    Background: Screening for atrial fibrillation (AF) using 12-lead-electrocardiograms (ECGs) has been recommended; however, the best method for interpreting ECGs to diagnose AF is not known. We compared accuracy of methods for diagnosing AF from ECGs. Methods: We searched MEDLINE, EMBASE, CINAHL and LILACS until March 24, 2014. Two reviewers identified eligible studies, extracted data and appraised quality using the QUADAS-2 instrument. Meta-analysis, using the bivariate hierarchical random effects method, determined average operating points for sensitivities, specificities, positive and negative likelihood ratios (PLR, NLR) and enabled construction of Summary Receiver Operating Characteristic (SROC) plots. Results: 10 studies investigated 16 methods for interpreting ECGs (n = 55,376 participant ECGs). The sensitivity and specificity of automated software (8 studies; 9 methods) were 0.89 (95% C.I. 0.82–0.93) and 0.99 (95% C.I. 0.99–0.99), respectively; PLR 96.6 (95% C.I. 64.2–145.6); NLR 0.11 (95% C.I. 0.07–0.18). Indirect comparisons with software found healthcare professionals (5 studies; 7 methods) had similar sensitivity for diagnosing AF but lower specificity [sensitivity 0.92 (95% C.I. 0.81–0.97), specificity 0.93 (95% C.I. 0.76–0.98), PLR 13.9 (95% C.I. 3.5–55.3), NLR 0.09 (95% C.I. 0.03–0.22)]. Sub-group analyses of primary care professionals found greater specificity for GPs than nurses [GPs: sensitivity 0.91 (95% C.I. 0.68–1.00); specificity 0.96 (95% C.I. 0.89–1.00). Nurses: sensitivity 0.88 (95% C.I. 0.63–1.00); specificity 0.85 (95% C.I. 0.83–0.87)]. Conclusions: Automated ECG-interpreting software most accurately excluded AF, although its ability to diagnose this was similar to all healthcare professionals. Within primary care, the specificity of AF diagnosis from ECG was greater for GPs than nurses

    St Helena marine water quality: Background conditions and development of assessment levels for coastal pollutants

    St Helena is an isolated oceanic island located in the tropical South Atlantic, and knowledge of broadscale oceanography and productivity in its surrounding waters is limited. This study used model outputs (2007-2017), remote sensing data (1998-2017) and survey measurements (April 2018 and 2019) to determine background conditions for nutrients, chlorophyll and suspended particulate matter (SPM) in offshore waters and propose standards (thresholds) for assessing inshore water quality based on 50% deviation from seasonal (usually June to November) or annual averages. Seasonal thresholds were proposed for surface nitrate (average 0.18 mu M), phosphate (average 0.26 mu M), silicate (average 2.60 mu M), chlorophyll (average 0.45 mu g chl l(-1)), and SPM (average 0.96 mg l(-1)). Associated background values for most surface parameters (phosphate 0.17 mu M, silicate 1.57 mu M, chlorophyll 0.30 mu g chl l(-1); from model outputs and remote sensing) were slightly higher than offshore observations (April 2019). For nitrate, the average background value (0.12 mu M) was lower than the observed average (0.24 mu M). At depth (150-500 m), annual background values from model outputs were high (nitrate 26.8 mu M, phosphate 1.8 mu M, silicate 17.3 mu M). Observed water masses at depths >150 m, identified to be of Antarctic and Atlantic origin, were nutrient-rich (e.g., 16 mu M for nitrate, April 2019) and oxygen deficient (<4-6 mg l(-1)). A thermocline layer (between ca. 10 and 230 m), characterized by a sub-surface chlorophyll maximum (average 0.3-0.5 mu g chl l(-1)) near the bottom of the euphotic zone (ca. 100 m), is likely to sustain primary and secondary production at St Helena. For assessing inshore levels of chemical contaminants and fecal bacteria estimated from survey measurements, standards were derived from the literature. A preliminary assessment of inshore observations using proposed thresholds from surface offshore waters and relevant literature standards indicated concerns regarding levels of nutrients and fecal bacteria at some locations. More detailed modeling and/or field-based studies are required to investigate seasonal trends and nutrient availability to inshore primary producers and to establish accurate levels of any contaminants of interest or risk to the marine environment

    Search for dark matter produced in association with bottom or top quarks in √s = 13 TeV pp collisions with the ATLAS detector

    A search for weakly interacting massive particle dark matter produced in association with bottom or top quarks is presented. Final states containing third-generation quarks and miss- ing transverse momentum are considered. The analysis uses 36.1 fb−1 of proton–proton collision data recorded by the ATLAS experiment at √s = 13 TeV in 2015 and 2016. No significant excess of events above the estimated backgrounds is observed. The results are in- terpreted in the framework of simplified models of spin-0 dark-matter mediators. For colour- neutral spin-0 mediators produced in association with top quarks and decaying into a pair of dark-matter particles, mediator masses below 50 GeV are excluded assuming a dark-matter candidate mass of 1 GeV and unitary couplings. For scalar and pseudoscalar mediators produced in association with bottom quarks, the search sets limits on the production cross- section of 300 times the predicted rate for mediators with masses between 10 and 50 GeV and assuming a dark-matter mass of 1 GeV and unitary coupling. Constraints on colour- charged scalar simplified models are also presented. Assuming a dark-matter particle mass of 35 GeV, mediator particles with mass below 1.1 TeV are excluded for couplings yielding a dark-matter relic density consistent with measurements

    Identification of genetic variants associated with Huntington's disease progression: a genome-wide association study

    Background Huntington's disease is caused by a CAG repeat expansion in the huntingtin gene, HTT. Age at onset has been used as a quantitative phenotype in genetic analysis looking for Huntington's disease modifiers, but is hard to define and not always available. Therefore, we aimed to generate a novel measure of disease progression and to identify genetic markers associated with this progression measure. Methods We generated a progression score on the basis of principal component analysis of prospectively acquired longitudinal changes in motor, cognitive, and imaging measures in the 218 indivduals in the TRACK-HD cohort of Huntington's disease gene mutation carriers (data collected 2008–11). We generated a parallel progression score using data from 1773 previously genotyped participants from the European Huntington's Disease Network REGISTRY study of Huntington's disease mutation carriers (data collected 2003–13). We did a genome-wide association analyses in terms of progression for 216 TRACK-HD participants and 1773 REGISTRY participants, then a meta-analysis of these results was undertaken. Findings Longitudinal motor, cognitive, and imaging scores were correlated with each other in TRACK-HD participants, justifying use of a single, cross-domain measure of disease progression in both studies. The TRACK-HD and REGISTRY progression measures were correlated with each other (r=0·674), and with age at onset (TRACK-HD, r=0·315; REGISTRY, r=0·234). The meta-analysis of progression in TRACK-HD and REGISTRY gave a genome-wide significant signal (p=1·12 × 10−10) on chromosome 5 spanning three genes: MSH3, DHFR, and MTRNR2L2. The genes in this locus were associated with progression in TRACK-HD (MSH3 p=2·94 × 10−8 DHFR p=8·37 × 10−7 MTRNR2L2 p=2·15 × 10−9) and to a lesser extent in REGISTRY (MSH3 p=9·36 × 10−4 DHFR p=8·45 × 10−4 MTRNR2L2 p=1·20 × 10−3). The lead single nucleotide polymorphism (SNP) in TRACK-HD (rs557874766) was genome-wide significant in the meta-analysis (p=1·58 × 10−8), and encodes an aminoacid change (Pro67Ala) in MSH3. In TRACK-HD, each copy of the minor allele at this SNP was associated with a 0·4 units per year (95% CI 0·16–0·66) reduction in the rate of change of the Unified Huntington's Disease Rating Scale (UHDRS) Total Motor Score, and a reduction of 0·12 units per year (95% CI 0·06–0·18) in the rate of change of UHDRS Total Functional Capacity score. These associations remained significant after adjusting for age of onset. Interpretation The multidomain progression measure in TRACK-HD was associated with a functional variant that was genome-wide significant in our meta-analysis. The association in only 216 participants implies that the progression measure is a sensitive reflection of disease burden, that the effect size at this locus is large, or both. Knockout of Msh3 reduces somatic expansion in Huntington's disease mouse models, suggesting this mechanism as an area for future therapeutic investigation
