    Results of search for magnetized quark-nugget dark matter from radial impacts on Earth

    Magnetized Quark Nuggets (MQNs) are a recently proposed dark-matter candidate consistent with the Standard Model and with Tatsumi's theory of quark-nugget cores in magnetars. Previous publications have covered their formation in the early universe, aggregation into a broad mass distribution before they can decay by the weak force, interaction with normal matter through their magnetopause, and first observation consistent MQNs, i.e. a nearly tangential impact limiting their surface-magnetic-field parameter B_o from Tatsumi's values of 0.1 to 10.0 TT to new value of 1.65 TT +/- 21%. The MQN mass distribution and interaction cross section depend strongly on B_o. Their magnetopause is much larger than their geometric dimensions and can cause sufficient energy deposition to form non-meteorite craters, which are reported approximately annually. We report computer simulations of the MQN energy deposition in water-saturated peat, soft sediments, and granite and report results from excavating such a crater. Five points of agreement between observations and hydrodynamic simulations of an MQN impact support this second observation consistent with MQN dark matter and suggest a method for qualifying additional MQN events.     CO2/pH-responsive particles with built-in fluorescence read-out

    yesA novel fluorescent monomer was synthesized to probe the state of CO2-responsive cross-linked polymeric particles. The fluorescent emission of this aminobromomaleimide-bearing monomer, being sensitive to protic environments, can provide information on the core hydrophilicity of the particles and therefore indicates the swollen state and size of the particles. The particles’ core, synthesized from DEAEMA (N,N-diethylaminoethyl methacrylate), is responsive to CO2 through protonation of the tertiary amines of DEAEMA. The response is reversible and the fluorescence emission can be recovered by simply bubbling nitrogen into the particle solution.     Synthesis and electrochemical behavior of ceramic cation-exchange membranes based on zirconium phosphate

    Cation-exchange membranes made exclusively from ceramic materials have been synthesized by means of the impregnation of microporous ceramic supports with zirconium phosphate. Changes in the pore size distribution and total pore volume of the supports were provoked by the addition of starch as pore former in the fabrication procedure. This allowed the production of supports with increased effective electrical conductivities and with larger pores available for the zirconium phosphate deposition. An improved functionality for the exchange of cations was given to the ceramic membranes by means of their impregnation with the active particles of zirconium phosphate. The ion-exchange properties of the membranes were increased with further impregnation cycles and the resulting current voltage curves showed a similar shape to that typical of commercial polymeric ion-exchange membranes.     Comparability: manufacturing, characterization and controls, report of a UK Regenerative Medicine Platform Pluripotent Stem Cell Platform Workshop, Trinity Hall, Cambridge, 14–15 September 2015

    This paper summarizes the proceedings of a workshop held at Trinity Hall, Cambridge to discuss comparability and includes additional information and references to related information added subsequently to the workshop. Comparability is the need to demonstrate equivalence of product after a process change; a recent publication states that this ‘may be difficult for cell-based medicinal products’. Therefore a well-managed change process is required which needs access to good science and regulatory advice and developers are encouraged to seek help early. The workshop shared current thinking and best practice and allowed the definition of key research questions. The intent of this report is to summarize the key issues and the consensus reached on each of these by the expert delegates

    A High Throughput Screen Identifies Nefopam as Targeting Cell Proliferation in β-Catenin Driven Neoplastic and Reactive Fibroproliferative Disorders

    Fibroproliferative disorders include neoplastic and reactive processes (e.g. desmoid tumor and hypertrophic scars). They are characterized by activation of β-catenin signaling, and effective pharmacologic approaches are lacking. Here we undertook a high throughput screen using human desmoid tumor cell cultures to identify agents that would inhibit cell viability in tumor cells but not normal fibroblasts. Agents were then tested in additional cell cultures for an effect on cell proliferation, apoptosis, and β-catenin protein level. Ultimately they were tested in Apc1638N mice, which develop desmoid tumors, as well as in wild type mice subjected to full thickness skin wounds. The screen identified Neofopam, as an agent that inhibited cell numbers to 42% of baseline in cell cultures from β-catenin driven fibroproliferative disorders. Nefopam decreased cell proliferation and β-catenin protein level to 50% of baseline in these same cell cultures. The half maximal effective concentration in-vitro was 0.5 uM and there was a plateau in the effect after 48 hours of treatment. Nefopam caused a 45% decline in tumor number, 33% decline in tumor volume, and a 40% decline in scar size when tested in mice. There was also a 50% decline in β-catenin level in-vivo. Nefopam targets β-catenin protein level in mesenchymal cells in-vitro and in-vivo, and may be an effective therapy for neoplastic and reactive processes driven by β-catenin mediated signaling

    Integrated Genomic Analysis of the Ubiquitin Pathway across Cancer Types

    Protein ubiquitination is a dynamic and reversibleprocess of adding single ubiquitin molecules orvarious ubiquitin chains to target proteins. Here,using multidimensional omic data of 9,125 tumorsamples across 33 cancer types from The CancerGenome Atlas, we perform comprehensive molecu-lar characterization of 929 ubiquitin-related genesand 95 deubiquitinase genes. Among them, we sys-tematically identify top somatic driver candidates,including mutatedFBXW7with cancer-type-specificpatterns and amplifiedMDM2showing a mutuallyexclusive pattern withBRAFmutations. Ubiquitinpathway genes tend to be upregulated in cancermediated by diverse mechanisms. By integratingpan-cancer multiomic data, we identify a group oftumor samples that exhibit worse prognosis. Thesesamples are consistently associated with the upre-gulation of cell-cycle and DNA repair pathways, char-acterized by mutatedTP53,MYC/TERTamplifica-tion, andAPC/PTENdeletion. Our analysishighlights the importance of the ubiquitin pathwayin cancer development and lays a foundation fordeveloping relevant therapeutic strategies

    Azimuthal anisotropy of charged jet production in root s(NN)=2.76 TeV Pb-Pb collisions

    We present measurements of the azimuthal dependence of charged jet production in central and semi-central root s(NN) = 2.76 TeV Pb-Pb collisions with respect to the second harmonic event plane, quantified as nu(ch)(2) (jet). Jet finding is performed employing the anti-k(T) algorithm with a resolution parameter R = 0.2 using charged tracks from the ALICE tracking system. The contribution of the azimuthal anisotropy of the underlying event is taken into account event-by-event. The remaining (statistical) region-to-region fluctuations are removed on an ensemble basis by unfolding the jet spectra for different event plane orientations independently. Significant non-zero nu(ch)(2) (jet) is observed in semi-central collisions (30-50% centrality) for 20 <p(T)(ch) (jet) <90 GeV/c. The azimuthal dependence of the charged jet production is similar to the dependence observed for jets comprising both charged and neutral fragments, and compatible with measurements of the nu(2) of single charged particles at high p(T). Good agreement between the data and predictions from JEWEL, an event generator simulating parton shower evolution in the presence of a dense QCD medium, is found in semi-central collisions.
