Intensification of cattle ranching production systems: Socioeconomic and environmental synergies and risks in Brazil
Intensification of Brazilian cattle ranching systems has attracted both national and international attention due to its direct relation with Amazon deforestation on the one hand and increasing demand of the global population for meat on the other. Since Brazilian cattle ranching is predominantly pasture-based, we particularly focus on pasture management. We summarize the most recurrent opportunities and risks associated with pasture intensification that are brought up within scientific and political dialogues, and discuss them within the Brazilian context. We argue that sustainable intensification of pasturelands in Brazil is a viable way to increase agricultural output while simultaneously sparing land for nature. Since environmental degradation is often associated with low-yield extensive systems in Brazil, it is possible to obtain higher yields, while reversing degradation, by adopting practices like rotational grazing, incorporation of legumes and integrated crop-livestock-forestry systems. Technical assistance is however essential, particularly for small- and medium-scale farmers. Sound complementary policies and good governance must accompany these measures so that a ‘rebound effect’ does not lead to increased deforestation and other adverse social and environmental impacts. It is also important that animal welfare is not compromised. Although the discussion is presented with respect to Brazil, some aspects are relevant to other developing countries
In vitro anti-malarial interaction and gametocytocidal activity of cryptolepine
YesBackground: Discovery of novel gametocytocidal molecules is a major pharmacological strategy in the elimination
and eradication of malaria. The high patronage of the aqueous root extract of the popular West African anti-malarial
plant Cryptolepis sanguinolenta (Periplocaceae) in traditional and hospital settings in Ghana has directed this study
investigating the gametocytocidal activity of the plant and its major alkaloid, cryptolepine. This study also investigates
the anti-malarial interaction of cryptolepine with standard anti-malarials, as the search for new anti-malarial combinations
continues.
Methods: The resazurin-based assay was employed in evaluating the gametocytocidal properties of C. sanguinolenta
and cryptolepine against the late stage (IV/V) gametocytes of Plasmodium falciparum (NF54). A fixed ratio method
based on the SYBR Green I fluorescence-based assay was used to build isobolograms from a combination of cryptolepine
with four standard anti-malarial drugs in vitro using the chloroquine sensitive strain 3D7.
Results: Cryptolepis sanguinolenta (
IC50 = 49.65 nM) and its major alkaloid, cryptolepine (
IC50 = 1965 nM), showed
high inhibitory activity against the late stage gametocytes of P. falciparum (NF54). In the interaction assays in asexual
stage, cryptolepine showed an additive effect with both lumefantrine and chloroquine with mean ΣFIC50s of
1.017 ± 0.06 and 1.465 ± 0.17, respectively. Cryptolepine combination with amodiaquine at therapeutically relevant
concentration ratios showed a synergistic effect (mean ΣFIC50 = 0.287 ± 0.10) whereas an antagonistic activity (mean
ΣFIC50 = 4.182 ± 0.99) was seen with mefloquine.
Conclusions: The findings of this study shed light on the high gametocytocidal properties of C. sanguinolenta and
cryptolepine attributing their potent anti-malarial activity mainly to their effect on both the sexual and asexual stages
of the parasite. Amodiaquine is a potential drug partner for cryptolepine in the development of novel fixed dose
combinations

Strategies to Target Tumor Immunosuppression
The tumor microenvironment is currently in the spotlight of cancer immunology research as a key factor impacting tumor development and progression. While antigen-specific immune responses play a crucial role in tumor rejection, the tumor hampers these immune responses by creating an immunosuppressive microenvironment. Recently, major progress has been achieved in the field of cancer immunotherapy, and several groundbreaking clinical trials demonstrated the potency of such therapeutic interventions in patients. Yet, the responses greatly vary among individuals. This calls for the rational design of more efficacious cancer immunotherapeutic interventions that take into consideration the “immune signature” of the tumor. Multimodality treatment regimens that aim to enhance intratumoral homing and activation of antigen-specific immune effector cells, while simultaneously targeting tumor immunosuppression, are pivotal for potent antitumor immunity

Forging connections: anthologies, arts collectives, and the politics of inclusion
The changing social and political landscape of twentieth-century Britain catalysed a remarkable rise in collaborative activity by artists and activists of black and Asian heritage. Creative communities began to gather in both local and regional contexts, with the aim of sharing resources and securing an audience. This chapter records some of these many activities, tracing the groups’ genesis, manifest objectives, and key contributions. It argues that anthologising should be understood as a specifically motivated activity. Literary anthologies of poetry and fiction served to showcase the diversity of contemporary writing, while also suggesting its coherence. Drawing on the concept of “strategic essentialism” elucidated by Gayatri Chakravorty Spivak, I show that the anthology acts to ensure the visibility of a group, bannered as a unified and singly-titled selection of texts, while also insisting on the differences within: the heterogeneous multiplicity of black and Asian British experiences and creative practices
