    Bodentiere bieten eine Vielfalt an ökologischen Funktionen und Dienstleistungen. Dazu zählen beispielsweise die Stimulierung von Zersetzungsprozessen und der Abbau von Pflanzenrückständen, was eine Regulierung von Pflanzenpathogenen und eine Reduzierung von Schadstoffen zur Folge haben kann. Ungeklärt blieb bislang allerdings, inwiefern bestimmte Schlüsselorganismen (Regenwürmer: Lumbricus terrestris; Collembolen: Folsomia candida und Nematoden: Aphelenchoides saprophilus) und deren Interaktion einen signifikanten Beitrag zum Abbau von phytopathogenen und toxigenen Schadpilzen der Gattung Fusarium in Ernterückständen von Getreide leisten und somit zur Lösung agrarrelevanter Umweltprobleme beitragen könnten. Aus diesem Grund wurden 2011 und 2013 Untersuchungen im Freiland durchgeführt, anhand derer folgende Hypothesen geprüft werden sollten: (1) Die eingesetzten Bodentiere fördern den Abbau von Fusarium-Biomasse im Stroh; (2) Die Interaktion zwischen den Vertretern der Makrofauna, Mesofauna und Mikrofauna verstärkt die Reduzierung der Fusarium-Biomasse. In beiden Versuchsjahren wurde nach Versuchsende (4 und 8 Wochen) in Stroh und Boden die Fusarium-Biomasse in Form von Fusarium-Protein-Äquivalenten (FPE) mittels der ELISA (Enzyme-linked immunosorbent assay)-Methodik quantitativ bestimmt. Außerdem wurde die Biomasse der Regenwürmer sowie die Individuenzahlen von Collembolen und Nematoden erfasst. Als Parameter für die Attraktivität des Strohs wurde das Verhältnis zwischen den Anteilen des verbliebenen Strohs auf der Bodenoberfläche und des von den Regenwürmern in den Boden eingearbeiteten Strohs bestimmt. Die eingesetzten Bodentiere förderten den Abbau von Fusarium-Biomasse in Weizenstroh, wobei L. terrestris in seiner Funktion als Primärzersetzer den wesentlichen Beitrag leistete. Der Einfluss von Collembolen und Nematoden stellte sich in Anwesenheit von L. terrestris als vernachlässigbar heraus, da die Interaktion nicht zu einem verstärkten Rückgang der Fusarium-Biomasse führte.     Tumour-derived and host-derived nitric oxide differentially regulate breast carcinoma metastasis to the lungs

    To study the role of nitric oxide (NO) in lung metastasis of breast carcinoma, we isolated two cell clones (H and J) from the parental EMT-6 murine breast carcinoma cell line, based on their differential NO production. In vitro, EMT-6 J cells, but not EMT-6H cells, constitutively expressed inducible NO synthase (NOS II) and secreted high levels of NO. IL-1beta increased NO production in both clones, and TNF-alpha had a synergistic effect on IL-1beta-induced NO production, but NO production by EMT-6 J cells was always higher than by EMT-6H cells. Proliferation, survival and adhesion to lung-derived endothelial cells of both clones were similar and were not affected by NO. In vivo, both clones similarly located in the lungs of syngeneic mice 48 h after injection. However, EMT-6H cells were significantly more tumorigenic than EMT-6 J cells as assessed at later time points. Injection of EMT-6 J cells and simultaneous treatment of mice with aminoguanidine (AG), a NOS II inhibitor, significantly increased tumour formation. Injection of EMT-6H and EMT-6 J cells into NOS II-deficient mice resulted in a significant survival increase as compared with wild-type animals. Simultaneous administration of AG increased the death rate of NOS II-deficient mice injected with EMT-6 J cells. These results demonstrate that: (i) NO does not influence the early stages of tumour metastasis to the lungs and (ii) NOS II expression in tumour cells reduces, while NOS II expression in host cells enhances, tumour nodule development.     We highlight some of the most important cornerstones of the long standing and very fruitful collaboration of the Austrian Diophantine Number Theory research group and the Number Theory and Cryptography School of Debrecen. However, we do not plan to be complete in any sense but give some interesting data and selected results that we find particularly nice. At the end we focus on two topics in more details, namely a problem that origins from a conjecture of Rényi and Erdős (on the number of terms of the square of a polynomial) and another one that origins from a question of Zelinsky (on the unit sum number problem).     Diastolic dysfunction in diabetes and the metabolic syndrome: promising potential for diagnosis and prognosis

    Cardiac disease in diabetes mellitus and in the metabolic syndrome consists of both vascular and myocardial abnormalities. The latter are characterised predominantly by diastolic dysfunction, which has been difficult to evaluate in spite of its prevalence. While traditional Doppler echocardiographic parameters enable only semiquantitative assessment of diastolic function and cannot reliably distinguish perturbations in loading conditions from altered diastolic functions, new technologies enable detailed quantification of global and regional diastolic function. The most readily available technique for the quantification of subclinical diastolic dysfunction is tissue Doppler imaging, which has been integrated into routine contemporary clinical practice, whereas cine magnetic resonance imaging (CMR) remains a promising complementary research tool for investigating the molecular mechanisms of the disease. Diastolic function is reported to vary linearly with age in normal persons, decreasing by 0.16 cm/s each year. Diastolic function in diabetes and the metabolic syndrome is determined by cardiovascular risk factors that alter myocardial stiffness and myocardial energy availability/bioenergetics. The latter is corroborated by the improvement in diastolic function with improvement in metabolic control of diabetes by specific medical therapy or lifestyle modification. Accordingly, diastolic dysfunction reflects the structural and metabolic milieu in the myocardium, and may allow targeted therapeutic interventions to modulate cardiac metabolism to prevent heart failure in insulin resistance and diabetes

    Effect of aliskiren on post-discharge outcomes among diabetic and non-diabetic patients hospitalized for heart failure: insights from the ASTRONAUT trial

    Aims The objective of the Aliskiren Trial on Acute Heart Failure Outcomes (ASTRONAUT) was to determine whether aliskiren, a direct renin inhibitor, would improve post-discharge outcomes in patients with hospitalization for heart failure (HHF) with reduced ejection fraction. Pre-specified subgroup analyses suggested potential heterogeneity in post-discharge outcomes with aliskiren in patients with and without baseline diabetes mellitus (DM). Methods and results ASTRONAUT included 953 patients without DM (aliskiren 489; placebo 464) and 662 patients with DM (aliskiren 319; placebo 343) (as reported by study investigators). Study endpoints included the first occurrence of cardiovascular death or HHF within 6 and 12 months, all-cause death within 6 and 12 months, and change from baseline in N-terminal pro-B-type natriuretic peptide (NT-proBNP) at 1, 6, and 12 months. Data regarding risk of hyperkalaemia, renal impairment, and hypotension, and changes in additional serum biomarkers were collected. The effect of aliskiren on cardiovascular death or HHF within 6 months (primary endpoint) did not significantly differ by baseline DM status (P = 0.08 for interaction), but reached statistical significance at 12 months (non-DM: HR: 0.80, 95% CI: 0.64-0.99; DM: HR: 1.16, 95% CI: 0.91-1.47; P = 0.03 for interaction). Risk of 12-month all-cause death with aliskiren significantly differed by the presence of baseline DM (non-DM: HR: 0.69, 95% CI: 0.50-0.94; DM: HR: 1.64, 95% CI: 1.15-2.33; P < 0.01 for interaction). Among non-diabetics, aliskiren significantly reduced NT-proBNP through 6 months and plasma troponin I and aldosterone through 12 months, as compared to placebo. Among diabetic patients, aliskiren reduced plasma troponin I and aldosterone relative to placebo through 1 month only. There was a trend towards differing risk of post-baseline potassium ≥6 mmol/L with aliskiren by underlying DM status (non-DM: HR: 1.17, 95% CI: 0.71-1.93; DM: HR: 2.39, 95% CI: 1.30-4.42; P = 0.07 for interaction). Conclusion This pre-specified subgroup analysis from the ASTRONAUT trial generates the hypothesis that the addition of aliskiren to standard HHF therapy in non-diabetic patients is generally well-tolerated and improves post-discharge outcomes and biomarker profiles. In contrast, diabetic patients receiving aliskiren appear to have worse post-discharge outcomes.
