    GFIT2: an experimental algorithm for vertical profile retrieval from near-IR spectra

    An algorithm for retrieval of vertical profiles from ground-based spectra in the near IR is described and tested. Known as GFIT2, the algorithm is primarily intended for CO₂, and is used exclusively for CO₂ in this paper. Retrieval of CO₂ vertical profiles from ground-based spectra is theoretically possible, would be very beneficial for carbon cycle studies and the validation of satellite measurements, and has been the focus of much research in recent years. GFIT2 is tested by application both to synthetic spectra and to measurements at two Total Carbon Column Observing Network (TCCON) sites. We demonstrate that there are approximately 3° of freedom for the CO2 profile, and the algorithm performs as expected on synthetic spectra. We show that the accuracy of retrievals of CO₂ from measurements in the 1.61μ (6220 cm⁻¹) spectral band is limited by small uncertainties in calculation of the atmospheric spectrum. We investigate several techniques to minimize the effect of these uncertainties in calculation of the spectrum. These techniques are somewhat effective but to date have not been demonstrated to produce CO₂ profile retrievals with sufficient precision for applications to carbon dynamics. We finish by discussing ongoing research which may allow CO₂ profile retrievals with sufficient accuracy to significantly improve the scientific value of the measurements from that achieved with column retrievals

    Reporting and Comparing School Performances

    This report provides advice on the collection and reporting of information about the performances of Australian schools. The focus is on the collection of nationally comparable data. Two purposes are envisaged: use by education authorities and governments to monitor school performances and, in particular, to identify schools that are performing unusually well or unusually poorly given their circumstances; and use by parents/caregivers and the public to make informed judgements about, and meaningful comparisons of, schools and their offerings. Our advice is based on a review of recent Australian and international research and experience in reporting on the performances of schools. This is an area of educational practice in which there have been many recent developments, much debate and a growing body of relevant research

    The 1989 Basic Skills Testing Program in New South Wales provides the most comprehensive picture yet compiled of literacy and numeracy learning in Australian primary schools. In 1989, some 53,800 Year 6 students in NSW government schools were tested in five aspects of literacy and numeracy. Another 2,300 Year 3 students took part in a pilot study. This book discusses the writing of the tests, the analysis of results, and the reporting of results to parents, teachers and schools. The aim of the basic skills tests is to describe, in positive terms, the skills that students have mastered, to identify areas in which students have special strengths and weaknesses, and to provide guides to further learning. The picture that emerges from this study is of widespread success in learning. The results point to much good teaching and a fine start in learning for most primary students. But this generally optimistic picture is over shadowed by the performances of some students who have not yet mastered essential Year 6 skills. Part I of the book describes the skills typical of students performing at each of five skill levels in each of five areas of learning (Reading, Language, Number, Measurement, and Space) on the tests. Part II shows how different subgroups of students performed on the tests. Results on each aspect of literacy and numeracy are reported separately for girls and boys, students with non-English-speaking backgrounds, Aboriginal and Torres Strait Islander students, and several age groupings. Part III explains and gives examples of reports mailed to parents, more detailed reports given to teachers, and summary tables generated for each school. Part IV describes procedures used to develop BSTP tests and to analyze students\u27 results in preparation for reporting. Numerous test items are presented

    Retrospective evaluation of whole exome and genome mutation calls in 746 cancer samples

    Funder: NCI U24CA211006Abstract: The Cancer Genome Atlas (TCGA) and International Cancer Genome Consortium (ICGC) curated consensus somatic mutation calls using whole exome sequencing (WES) and whole genome sequencing (WGS), respectively. Here, as part of the ICGC/TCGA Pan-Cancer Analysis of Whole Genomes (PCAWG) Consortium, which aggregated whole genome sequencing data from 2,658 cancers across 38 tumour types, we compare WES and WGS side-by-side from 746 TCGA samples, finding that ~80% of mutations overlap in covered exonic regions. We estimate that low variant allele fraction (VAF < 15%) and clonal heterogeneity contribute up to 68% of private WGS mutations and 71% of private WES mutations. We observe that ~30% of private WGS mutations trace to mutations identified by a single variant caller in WES consensus efforts. WGS captures both ~50% more variation in exonic regions and un-observed mutations in loci with variable GC-content. Together, our analysis highlights technological divergences between two reproducible somatic variant detection efforts

    The Solar Spectrum: An Atmospheric Remote Sensing Perspective

    The solar spectrum not only contains information about the composition and structure of the sun, it also provides a bright and stable continuum source for earth remote sensing (atmosphere and surface). Many types of remote sensors use solar radiation. While high-resolution spaceborne sensors (e.g. ACE) can largely remove the effects of the solar spectrum by exo-atmospheric calibration, this isn't an option for sub-orbital sensors, such as the FTIR spectrometers used in the NDACC and TCCON networks. In this case the solar contribution must be explicitly included in the spectral analysis. In this talk the methods used to derive the solar spectrum are presented, and the underlying solar physics are discussed. Implication for remote sensing are described

    Impact of mesospheric intrusions on ozone tracer-relations in the stratospheric polar vortex

    [1] Ozone-tracer relations are used to quantify chemical ozone loss in the polar vortices. The underlying assumptions for the application of this technique were extensively discussed in recent years. However, the impact intrusions of mesospheric air into the polar stratosphere have on estimates of chemical ozone loss based on the ozone-tracer technique has not hitherto been studied. Here, we revisit observations of an intrusion of mesospheric air down to altitudes of similar to 25 km (similar to 600 K potential temperature) in the Arctic vortex in 2003. The mesospheric intrusion was identified in three balloon profiles in January and March 2003 as a strong enhancement in CO. In contrast, NOy was not enhanced in the mesospheric air relative to surrounding air masses as shown by the measurement in late March 2003. The measurements influenced by mesospheric air show ozone mixing ratios ranging between 3.6 and 5.6 ppm, which are clearly greater than those found in the "early vortex" reference relation employed to deduce chemical ozone loss. Thus the impact of intrusions of mesospheric air into the polar vortex on chemical ozone loss estimates based on ozone-tracer relations are likely small; the correlations cannot be affected in a way that would lead to an overestimate of ozone depletion. Therefore ozone-tracer relations may be used for deducing chemical ozone loss in Arctic winter 2002-2003. Here we use ILAS-II satellite measurements to deduce an average chemical ozone loss in the vortex core for the partial column 380-550 K of 37 +/- 11 Dobson units in March and of 50 +/- 10 Dobson units in April 2003

    Sex differences have been observed in multiple facets of cancer epidemiology, treatment and biology, and in most cancers outside the sex organs. Efforts to link these clinical differences to specific molecular features have focused on somatic mutations within the coding regions of the genome. Here we report a pan-cancer analysis of sex differences in whole genomes of 1983 tumours of 28 subtypes as part of the ICGC/TCGA Pan-Cancer Analysis of Whole Genomes (PCAWG) Consortium. We both confirm the results of exome studies, and also uncover previously undescribed sex differences. These include sex-biases in coding and non-coding cancer drivers, mutation prevalence and strikingly, in mutational signatures related to underlying mutational processes.
