    Generation and characterization of the human iPSC line PBMC1-iPS4F1 from adult peripheral blood mononuclear cells

    AbstractHere we describe the generation and characterization of the human induced pluripotent stem cell (iPSC) line PBMC1-iPS4F1 from peripheral blood mononuclear cells from a healthy female with Spanish background. We used heat sensitive, non-integrative Sendai viruses containing the reprogramming factors Oct3/4, Sox2, Klf4 and c-Myc, whose expression was silenced in the established iPSC line. Characterization of the PBMC1-iPS4F1 cell line included analysis of typical pluripotency-associated factors at mRNA and protein level, alkaline phosphatase enzymatic activity, and in vivo and in vitro differentiation studies

    iFADO project: contribution to the determination of the Good Environmental Status in the Atlantic Area through modeling and in situ monitoring approaches

    The project iFADO ( innovation in the Framework of the Atlantic Deep Ocean ) aims to combine conventional monitoring programs (with special focus on novel semi-automatic techniques that can be applied for in situ biological monitoring) with emerging technologies (such as gliders, satellite imagery, etc.) and modeling approaches to assist the implementation of the Marine Strategy Framework Directive (MSFD), providing tools for the optimization of observing strategies for better evaluation and forecasting of the Good Environmental Status (GES). Within this framework, the multidisciplinary iFADO cruise (within the RADPROF time series program, Instituto Español de Oceanografía) was carried out with transnational collaboration on summer 2020 along a standard coastal-oceanic transect off NW Iberian Peninsula, including physical, biogeochemical and biological measurements. Additionally, an effort is being made in the implementation and validation of existing and new biogeochemical models in the Atlantic margin, focusing on key variables for the implementation of MSFD Descriptors. The combination of iFADO models and in situ monitoring approaches not only might contribute data and potential indicators for MSFD Descriptors 1, 2, 3, 4, 5, 6, 7 and 10, from coastal to open-ocean environments, but they are essential tools to detect, model and predict temporal changes in marine ecosystems in the study area

    El establecimiento de las Normas Internacionales de Información Financiera (NIIF) y la NIC 41 ( Norma Internacional de Contabilidad) en la ley 1314 de 2009, busca determinar la convergencia de los principios y normas de contabilidad e información financiera hacia los estándares internacionales, permitiendo la identificación y reconocimiento de los activos biológicos; por este motivo el grupo de investigación HUTIN (Huellas Tecnológicas de Investigación), desarrolló un proyecto en el cual se diagnosticó las empresas del sector ganadero dedicadas a la cría de ganado bovino de la región del Bajo Cauca Antioqueño implementen esta norma, ya que posibilita el reconocimiento económico de los activos biológicos, valor razonable y deducción de costos que son representativos en los estados financieros. Esto se logró a través de encuestas realizadas a empresas de la subregión del Bajo Cauca Antioqueño dedicadas a la cría de ganado bovino, obteniendo la información necesaria para documentar el proceso y obtener un estándar para ser implementado en el sector siendo una herramienta eficaz para alcanzar mercados globalizados y competitivos. Se percibe un bajo nivel en las empresas ganaderas sobre el proceso de implementación de la NIIF (NIC 41). La implementación de esta norma le permite al empresario del sector ganadero identificar el estado actual de sus activos biológicos e inventarios y poder deducirse costos significativos en el proceso productivo, contando con una información económica, contable y financiera fundamentales en la toma de decisiones

    Bacterial community composition and optical signature of DOM shape empirical leucine-to-carbon conversion factors in north-eastern Atlantic waters (0-4000 m)

    Oral communicationMicrobial heterotrophic activity is a major process regulating the flux of dissolved organic matter (DOM) in the ocean. DOM quantity and quality strongly influence its microbial utilization and fate in the ocean. In order to broaden the vertical resolution of leucine-to-carbon conversion factors (CFs), needed for converting substrate incorporation into biomass production by heterotrophic bacteria, nine dilution experiments were performed in the north Atlantic. We found a very consistent depth-stratification in empirical CFs values from epipelagic to bathypelagic waters (3.95 ± 0.05 to 0.90 ± 0.51 kg C mol Leu-1). Our results demonstrated that the customarily used CF of 1.55 kg C mol Leu-1 in oceanic waters leads to an underestimation of prokaryotic heterotrophic production in epi- and mesopelagic waters, while it causes a severe overestimation in bathypelagic waters. Pearson correlations showed that CFs were related not only to hydrographic variables but also to specific phylogenetic groups and DOM quality and quantity indicators. Furthermore, a multiple linear regression model predicting CFs from relatively simple hydrographic and optical spectroscopic measurements is provided. Taken together, our results suggest that differences in CFs throughout the water column might be mostly associated to the quality of DOM affecting the response of particular phylogenetic groups.ASL

    Reconstruction of primary vertices at the ATLAS experiment in Run 1 proton–proton collisions at the LHC

    This paper presents the method and performance of primary vertex reconstruction in proton–proton collision data recorded by the ATLAS experiment during Run 1 of the LHC. The studies presented focus on data taken during 2012 at a centre-of-mass energy of √s=8 TeV. The performance has been measured as a function of the number of interactions per bunch crossing over a wide range, from one to seventy. The measurement of the position and size of the luminous region and its use as a constraint to improve the primary vertex resolution are discussed. A longitudinal vertex position resolution of about 30μm is achieved for events with high multiplicity of reconstructed tracks. The transverse position resolution is better than 20μm and is dominated by the precision on the size of the luminous region. An analytical model is proposed to describe the primary vertex reconstruction efficiency as a function of the number of interactions per bunch crossing and of the longitudinal size of the luminous region. Agreement between the data and the predictions of this model is better than 3% up to seventy interactions per bunch crossing

    Surgical site infection after gastrointestinal surgery in high-income, middle-income, and low-income countries: a prospective, international, multicentre cohort study

    Background: Surgical site infection (SSI) is one of the most common infections associated with health care, but its importance as a global health priority is not fully understood. We quantified the burden of SSI after gastrointestinal surgery in countries in all parts of the world. Methods: This international, prospective, multicentre cohort study included consecutive patients undergoing elective or emergency gastrointestinal resection within 2-week time periods at any health-care facility in any country. Countries with participating centres were stratified into high-income, middle-income, and low-income groups according to the UN's Human Development Index (HDI). Data variables from the GlobalSurg 1 study and other studies that have been found to affect the likelihood of SSI were entered into risk adjustment models. The primary outcome measure was the 30-day SSI incidence (defined by US Centers for Disease Control and Prevention criteria for superficial and deep incisional SSI). Relationships with explanatory variables were examined using Bayesian multilevel logistic regression models. This trial is registered with ClinicalTrials.gov, number NCT02662231. Findings: Between Jan 4, 2016, and July 31, 2016, 13 265 records were submitted for analysis. 12 539 patients from 343 hospitals in 66 countries were included. 7339 (58·5%) patient were from high-HDI countries (193 hospitals in 30 countries), 3918 (31·2%) patients were from middle-HDI countries (82 hospitals in 18 countries), and 1282 (10·2%) patients were from low-HDI countries (68 hospitals in 18 countries). In total, 1538 (12·3%) patients had SSI within 30 days of surgery. The incidence of SSI varied between countries with high (691 [9·4%] of 7339 patients), middle (549 [14·0%] of 3918 patients), and low (298 [23·2%] of 1282) HDI (p < 0·001). The highest SSI incidence in each HDI group was after dirty surgery (102 [17·8%] of 574 patients in high-HDI countries; 74 [31·4%] of 236 patients in middle-HDI countries; 72 [39·8%] of 181 patients in low-HDI countries). Following risk factor adjustment, patients in low-HDI countries were at greatest risk of SSI (adjusted odds ratio 1·60, 95% credible interval 1·05–2·37; p=0·030). 132 (21·6%) of 610 patients with an SSI and a microbiology culture result had an infection that was resistant to the prophylactic antibiotic used. Resistant infections were detected in 49 (16·6%) of 295 patients in high-HDI countries, in 37 (19·8%) of 187 patients in middle-HDI countries, and in 46 (35·9%) of 128 patients in low-HDI countries (p < 0·001). Interpretation: Countries with a low HDI carry a disproportionately greater burden of SSI than countries with a middle or high HDI and might have higher rates of antibiotic resistance. In view of WHO recommendations on SSI prevention that highlight the absence of high-quality interventional research, urgent, pragmatic, randomised trials based in LMICs are needed to assess measures aiming to reduce this preventable complication

    Identificación del incremento de variables incidentes en riesgos ambientales en las villas cordilleranas de Potrerillos, Mendoza. Estado actual del conocimiento

    La localidad de Potrerillos, en el departamento de Luján de Cuyo, se encuentra conformada por villas cordilleranas en un fondo de montaña a gran altura en cercanía del llamado “Cordón del Plata” perteneciente a la Cordillera Frontal de Los Andes, que fueron consideradas por sus características para este trabajo de investigación como unidad funcional. En los últimos 40 años la construcción de viviendas se fue incrementando a raíz del creciente turismo y principalmente desde el año 2005, surge la radicación de familias movilizadas por un cambio en el estilo de vida urbana por el espacio rural y la protección del ambiente. El crecimiento del asentamiento humano sin el debido control sobre los recursos naturales ha tenido efectos negativos en el ambiente frágil de dichas villas y en el incremento de los riesgos de origen natural, especialmente el aluvional, viento zonda y sísmico
