    Caractérisation du protéome vascuolaire de la plante modèle Arabidopsis thaliana et étude de son rôle dans la détoxication du cadmium

    Afin de mieux comprendre les mécanismes du trafic cellulaire, les processus de transport des substrats vacuolaires à travers le tonoplaste, le stockage des métabolites et leur dégradation, une analyse globale et exhaustive du protéome vacuolaire d'Arabidopsis thaliana a été réalisée. La connaissance de la localisation subcellulaire des protéines permet de mieux comprendre la fonction des organelles et la compartimentation du métabolisme des plantes. Mais la description précise du protéome d'un organite nécessite d'identifier clairement les véritables protéines résidantes du compartiment étudié. Une tâche si précise est complexe puisqu'elle nécessite la mise en place d'une préparation d'organites purs et homogènes. Pour y parvenir, un protocole de purification de vacuoles à partir de protoplastes isolés de cellules en culture sur un gradient de densité de Ficoll a été amélioré. La combinaison de plusieurs approches de protéomique a permis d'identifier les protéines présentes dans les fractions vacuolaires soluble et membranaire de façon quantitative et fonctionnelle. Les différentes approches ont ainsi mis en évidence des associations et mécanismes moléculaires complexes qui régissent les différentes activités vacuolaires. Cette protéothèque de référence constitue une base pour étudier la dynamique du protéome vacuolaire en réponse à plusieurs stress incluant les métaux lourds. Plusieurs méthodes sans a priori et ciblée ont été proposé afin d'étudier l'impact du cadmium sur la vacuole, ce compartiment cellulaire clé de la détoxication.To better understand the mechanisms governing cellular traffic, transport process of substrates across the tonoplast, storage of various metabolites and their ultimate degradation, a comprehensive and thorough analysis of Arabidopsis thaliana vacuolar proteome was performed. Protein subcellular localization knowledge is an important step toward assigning functions of organelles and plant metabolism compartmentation. But confident description of proteome organelle content requires clear identification of the true resident proteins of the studied compartment. This task involves pitfalls and requires that either organelle preparations are free of contaminants or that techniques are used to discriminate between genuine organelle residents and contaminating proteins. To achieve this, vacuoles purification protocol from protoplasts on a Ficoll density gradient has been improved. The combination of several proteomic approaches attempt to present soluble and membrane vacuolar proteins in a quantitative and functional manner. Different approaches have thus shown associations and complex molecular mechanisms that govern the various vacuolar activities. The constitute proteins library provides references to study the vacuolar proteome dynamics in response to different stresses including heavy metals. Many methods without a priori or targeted were proposed to study the impact of cadmium on the vacuole, the key cell compartment of detoxification.     Characteristics of adolescent attempts to manage overweight

    This study evaluates the actions taken by obese adolescents in an attempt to lose weight in relation to changes in weight. Particular emphasis is put on the context surrounding the management process. Thirty one obese adolescents aged 14-18 participated in this study. The adolescents answered a written questionnaire, took a body-size dissatisfaction test and were interviewed. Their family physicians were interviewed by telephone. Quantitative data concerned changes in weight and body-size dissatisfaction scores. Qualitative analysis included cases analyses and building up of synthesis variables. Loss of weight is linked with continuity of management attempts as well as with medical follow-up. The most frequent management attempt is diet. Self-image is correlated with weight. The exclusive emphasis put on diet is discussed in relation to the importance of more psychosocial factors such as self-image, denial and attraction to discontinuity.     Processing of blood samples influences PBMC viability and outcome of cell-mediated immune responses in antiretroviral therapy-naïve HIV-1-infected patients

    AbstractIntracellular cytokine staining (ICS) assay is increasingly used in vaccine clinical trials to measure antigen-specific T-cell mediated immune (CMI) responses in cryopreserved peripheral blood mononuclear cells (PBMCs) and whole blood. However, recent observations indicate that several parameters involved in blood processing can impact PBMC viability and CMI responses, especially in antiretroviral therapy (ART)-naïve HIV-1-infected individuals.In this phase I study (NCT01610427), we collected blood samples from 22 ART-naïve HIV-1-infected adults. PBMCs were isolated and processed for ICS assay. The individual and combined effects of the following parameters were investigated: time between blood collection and PBMC processing (time-to-process: 2, 7 or 24h); time between PBMC thawing and initiation of in vitro stimulation with HIV-1 antigens (resting-time: 0, 2, 6 and 18h); and duration of antigen-stimulation in PBMC cultures (stimulation-time: 6h or overnight). The cell recovery after thawing, cell viability after ICS and magnitude of HIV-specific CD8+ T-cell responses were considered to determine the optimal combination of process conditions. The impact of time-to-process (2 or 4h) on HIV-specific CD8+ T-cell responses was also assessed in a whole blood ICS assay.A higher quality of cells in terms of recovery and viability (up to 81% and >80% respectively) was obtained with shorter time-to-process (less than 7h) and resting-time (less than 2h) intervals. Longer (overnight) rather than shorter (6h) stimulation-time intervals increased the frequency of CD8+-specific T-cell responses using ICS in PBMCs without change of the functionality. The CD8+ specific T-cell responses detected using fresh whole blood showed a good correlation with the responses detected using frozen PBMCs.Our results support the need of standardized procedures for the evaluation of CMI responses, especially in HIV-1-infected, ART-naïve patients

    Possible detection of phase changes from the non-transiting planet HD 46375b by CoRoT

    The present work deals with the detection of phase changes in an exoplanetary system. HD 46375 is a solar analog known to host a non-transiting Saturn-mass exoplanet with a 3.0236 day period. It was observed by the CoRoT satellite for 34 days during the fall of 2008. We attempt to identify at optical wavelengths, the changing phases of the planet as it orbits its star. We then try to improve the star model by means of a seismic analysis of the same light curve and the use of ground-based spectropolarimetric observations. The data analysis relies on the Fourier spectrum and the folding of the time series. We find evidence of a sinusoidal signal compatible in terms of both amplitude and phase with light reflected by the planet. Its relative amplitude is Delta Fp/F* = [13.0, 26.8] ppm, implying an albedo A=[0.16, 0.33] or a dayside visible brightness temperature Tb ~ [1880,2030] K by assuming a radius R=1.1 R_Jup and an inclination i=45 deg. Its orbital phase differs from that of the radial-velocity signal by at most 2 sigma_RV. However, the tiny planetary signal is strongly blended by another signal, which we attribute to a telluric signal with a 1 day period. We show that this signal is suppressed, but not eliminated, when using the time series for HD 46179 from the same CoRoT run as a reference. This detection of reflected light from a non-transiting planet should be confirmable with a longer CoRoT observation of the same field. In any case, it demonstrates that non-transiting planets can be characterized using ultra-precise photometric lightcurves with present-day observations by CoRoT and Kepler. The combined detection of solar-type oscillations on the same targets (Gaulme et al. 2010a) highlights the overlap between exoplanetary science and asteroseismology and shows the high potential of a mission such as Plato.Comment: 4 pages, 6 figure

    Conformal geometry of the supercotangent and spinor bundles

    We study the actions of local conformal vector fields X∈conf(M,g) on the spinor bundle of (M,g) and on its classical counterpart: the supercotangent bundle M of (M,g). We first deal with the classical framework and determine the Hamiltonian lift of conf(M,g) to M. We then perform the geometric quantization of the supercotangent bundle of (M,g), which constructs the spinor bundle as the quantum representation space. The Kosmann Lie derivative of spinors is btained by quantization of the comoment map. The quantum and classical actions of conf(M,g) turn, respectively, the space of differential operators acting on spinor densities and the space of their symbols into conf(M,g)-modules. They are filtered and admit a common associated graded module. In the conformally flat case, the latter helps us determine the conformal invariants of both conf(M,g)-modules, in particular the conformally odd powers of the Dirac operator.Peer reviewe

    The F4/AS01B HIV-1 Vaccine Candidate Is Safe and Immunogenic, But Does Not Show Viral Efficacy in Antiretroviral Therapy-Naive, HIV-1-Infected Adults: A Randomized Controlled Trial

    The impact of the investigational human immunodeficiency virus type 1 (HIV-1) F4/AS01(B) vaccine on HIV-1 viral load (VL) was evaluated in antiretroviral therapy (ART)-naive HIV-1 infected adults.This phase IIb, observer-blind study (NCT01218113), included ART-naive HIV-1 infected adults aged 18 to 55 years. Participants were randomized to receive 2 (F4/AS01(B)_2 group, N=64) or 3 (F4/AS01(B)_3 group, N=62) doses of F4/AS01(B) or placebo (control group, N=64) at weeks 0, 4, and 28. Efficacy (HIV-1 VL, CD4(+) T-cell count, ART initiation, and HIV-related clinical events), safety, and immunogenicity (antibody and T-cell responses) were evaluated during 48 weeks.At week 48, based on a mixed model, no statistically significant difference in HIV-1 VL change from baseline was demonstrated between F4/AS01(B)_2 and control group (0.073 log(10)copies/mL [97.5% confidence interval (CI): -0.088; 0.235]), or F4/AS01(B)_3 and control group (-0.096 log(10)copies/mL [97.5% CI: -0.257; 0.065]). No differences between groups were observed in HIV-1 VL change, CD4(+) T-cell count, ART initiation, or HIV-related clinical events at intermediate timepoints. Among F4/AS01(B) recipients, the most frequent solicited symptoms were pain at injection site (252/300 doses), fatigue (137/300 doses), myalgia (105/300 doses), and headache (90/300 doses). Twelve serious adverse events were reported in 6 participants; 1 was considered vaccine-related (F4/AS01(B)_2 group: angioedema). F4/AS01(B) induced polyfunctional F4-specific CD4(+) T-cells, but had no significant impact on F4-specific CD8(+) T-cell and anti-F4 antibody levels.F4/AS01(B) had a clinically acceptable safety profile, induced F4-specific CD4(+) T-cell responses, but did not reduce HIV-1 VL, impact CD4(+) T-cells count, delay ART initiation, or prevent HIV-1 related clinical events

    Endothelial dysfunction and vascular disease

    The endothelium can evoke relaxations (dilatations) of the underlying vascular smooth muscle, by releasing vasodilator substances. The best characterized endothelium-derived relaxing factor (EDRF) is nitric oxide (NO). The endothelial cells also evoke hyperpolarization of the cell membrane of vascular smooth muscle (endothelium-dependent hyperpolarizations, EDHF-mediated responses). Endothelium-dependent relaxations involve both pertussis toxin-sensitive G i (e.g. responses to serotonin and thrombin) and pertussis toxin-insensitive G q (e.g. adenosine diphosphate and bradykinin) coupling proteins. The release of NO by the endothelial cell can be up-regulated (e.g. by oestrogens, exercise and dietary factors) and down-regulated (e.g. oxidative stress, smoking and oxidized low-density lipoproteins). It is reduced in the course of vascular disease (e.g. diabetes and hypertension). Arteries covered with regenerated endothelium (e.g. following angioplasty) selectively loose the pertussis toxin-sensitive pathway for NO release which favours vasospasm, thrombosis, penetration of macrophages, cellular growth and the inflammatory reaction leading to atherosclerosis. In addition to the release of NO (and causing endothelium-dependent hyperpolarizations), endothelial cells also can evoke contraction (constriction) of the underlying vascular smooth muscle cells by releasing endothelium-derived contracting factor (EDCF). Most endothelium-dependent acute increases in contractile force are due to the formation of vasoconstrictor prostanoids (endoperoxides and prostacyclin) which activate TP receptors of the vascular smooth muscle cells. EDCF-mediated responses are exacerbated when the production of NO is impaired (e.g. by oxidative stress, ageing, spontaneous hypertension and diabetes).     Trends in SAVR with biological vs. mechanical valves in middle-aged patients: results from a French large multi-centric survey

    Background/introductionCurrently, despite continued issues with durability ( 1), biological prosthetic valves are increasingly chosen over mechanical valves for surgical aortic valve replacement (SAVR) in adult patients of all ages, at least in Western countries. For younger patients, this choice means assuming the risks associated with a redo SAVR or valve-in-valve procedure.PurposeTo assess the use of mechanical vs. biological valve prostheses for SAVR relative to patient's age and implant time in a large population extracted from the French National Database EPICARD.MethodsPatients in EPICARD undergoing SAVR from 2007 to 2022 were included from 22 participating public or private centers chosen to represent a balanced representation of centre sizes and geographical discrepancies. Patients with associated pathology of the aorta (aneurysm or dissection) and requiring a vascular aortic prosthesis were excluded. Comparisons were made amongst centers, valve choice, implant date range, and patient age.ResultsWe considered 101,070 valvular heart disease patients and included 72,375 SAVR (mean age 71.4 ± 12.2 years). We observed a mechanical vs. biological prosthesis ratio (MBPR) of 0.14 for the overall population. Before 50 years old (y-o), MBPR was >1.3 (p < 0.001) while patients above 60 years-old received principally biological SAVR (p < 0.0001). Concerning patients between 50 and 60 years-old patients, MPVR was 1.04 (p = 0.03). Patients 50–60 years-old from the first and second study duration quartile (before August 2015) received preferentially mechanical SAVR (p < 0.001). We observed a shift towards more biological SAVR (p < 0.001) for patients from the third and fourth quartile to reach a MBPR at 0.43 during the last years of the series. Incidentally, simultaneous mitral valve replacement were more common in case of mechanical SAVR (p < 0.0001), while associated CABGs were more frequent in case of biological SAVR (p < 0.0001).ConclusionIn a large contemporary French patient population, real world practice showed a recent shift towards a lower age-threshold for biological SAVR as compared to what would suggest contemporary guidelines
