    Perinatal exposure to 5-metoxytryptamine, behavioural-stress reactivity and functional response of 5-HT1A receptors in the adolescent rat

    Serotonin is involved in a wide range of physiological and patho-physiological mechanisms. In particular, 5-HT1A receptors are proposed to mediate stress-adaptation. The aim of this research was to investigate in adolescent rats: first, the consequences of perinatal exposure to 5- metoxytryptamine (5MT), a 5-HT1/5-HT2 serotonergic agonist, on behavioural-stress reactivity in elevated plus maze, open field and forced swim tests; secondly, whether the behavioural effects induced by perinatal exposure to 5MT on open field and forced swim tests were affected by the selective 5-HT1A receptor agonist LY 228729, a compound able to elicit a characteristic set of motor behaviours on these experimental models, and by the co-administration of the selective and silent 5-HT1A antagonist WAY 100635. Results indicate that a single daily injection of 5MT to, pregnant dams from gestational days 12 to 21 (1 mg/kg s.c.), and to the pups from postnatal days 2 to 18 (0.5 mg kg s.c.), induce in the adolescent rat offspring: an increase in the percentage of entries and time spent on the open arms in the elevated plus maze; a reduction in locomotor activity and rearing frequency, and an increase in the time spent on the central areas in the open field test; a decrease in immobility and an increase in swimming in the forced swim test. Acute administration of LY 228729 (1.5 mg/kg s.c.) strongly decreases rearing frequency and increases peripheral activity in the open field test, and decreases immobility and increases swimming in the forced swim test both in perinatally vehicle and 5MT-exposed offspring. Co-administration of WAY 100635 (0.25 mg/kg s.c.) abolishes the effects exerted by LY 228729. These results suggest that, in the adolescent rat, perinatal exposure to 5MT enhances the stress-related adaptive behavioural responses, presumably through a predominant action on presynaptic 5-HT1A receptors and does not deteriorate the functional response of 5-HT1A receptors to selective agonist and antagonist compounds

    Respiratory tract infections are the most common diseases in childhood. The respiratory tract, widely branched system of ducts, is particularly exposed to the action of microorganisms transmitted by air from here the high frequency of infections they face especially in the first years of life. It is usual distinguish: upper respiratory tract infections (URTI) and lower respiratory tract infections (LRTI). In particular, in infections of the upper airways, the inflammatory process, result of the interaction between microbes and the immune response, can be localized to the mucosa of the nose or sinuses (common cold and sinusitis), or the pharynx or larynx (pharyngotonsillitis and laryngitis) and it has predominantly a viral etiology although occasionally it may be followed by bacterial complications such as otitis media. The aim of the following article is the description of these different clinical pictures, highlighting the clinical and epidemiological features and current management guidelines

    PLASMA MEMBRANE REDOX SYSTEM IN THE ERYTHROCYTES OF ROWERS: PILOT STUDY

    Background: The oxidative stress results from a change in the physiological balance between oxidant and antioxidant species. The purpose of this study is twofold: first, to investigate the effects of long-term training in sports with high energy requirements on the redox balance which exists between the plasma vs. the erythrocytes; second, to study the activity of the PMRS (Plasma Membrane Redox System), which is a compensatory mechanism of cellular redox homeostasis, in the rowers’ erythrocytes in order to determine the rowers’ counteraction to oxidative stress. Methods: Venous blood samples was collected from rowers and control group; then FRAP (Ferric Reducing Activity Power) method has been used to determine the antioxidant capabilities both in the plasma and in the erythrocytes of 22 rowers vs. 26 sedentary subjects. For the same groups of subjects, the PMRS in erythrocytes has been also evaluated. Results: The plasmatic antioxidant activity was 21% lower in the group of rowers compared to the sedentary group (p = 0,02). In contrast, no significant differences were found in the reducing activity of the erythrocytes; however the erythrocytes of the rowers have shown values of the PMRS 35% higher than the untrained group (p < 0.0001). Conclusions: Rowing induces a significant oxidative stress in the plasma corresponding to the high intensity training, while this effect lacks in erythrocytes. At the same time an increased quantity of the PMRS has been observed in the erythrocytes. In conclusion, in well trained athletes this not lead to established an oxidative stress condition because long-term training adaptatively improves the efficiency of the antioxidant syste

    Increased functional connectivity in gambling disorder correlates with behavioural and emotional dysregulation: Evidence of a role for the cerebellum

    Gambling disorder (GD) is a psychiatric disease that has been recently classified as a behavioural addiction. So far, a very few studies have investigated the alteration of functional connectivity in GD patients, thus the concrete interplay between relevant function-dependent circuitries in such disease has not been comprehensively assessed. The aim of this research was to investigate resting-state functional connectivity in GD patients, searching for a correlation with GD symptoms severity. GD patients were assessed for gambling behaviour, impulsivity, cognitive distortions, anxiety and depression, in comparison with healthy controls (HC). Afterwards, they were assessed for resting-state functional magnetic resonance imaging; functional connectivity was assessed through a data-driven approach, by using independent component analysis. The correlation between gambling severity and the strength of specific resting-state networks was also investigated. Our results show that GD patients displayed higher emotional and behavioural impairment than HC, together with an increased resting state functional connectivity in the network including anterior cingulate cortex, the caudate nucleus and nucleus accumbens, and within the cerebellum, in comparison with the control group. Moreover, a significant correlation between behavioural parameters and the strength of the resting-state cerebellar network was found. Overall, the functional alterations in brain connectivity involving the cerebellum observed in this study underpin the emotional and behavioural impairment recorded in GD patients. This evidence suggests the employment of novel neuromodulatory therapeutic approaches involving specific and salient targets such as the cerebellum in addictive disorders

    I patriarchi verdi dei Monti Sicani

    Gli alberi vetusti costituiscono un segno della capacità e plasticità di adattamento di alcune specie ma sono anche spesso frutto del caso o di una considerazione particolare da parte di alcune comunità locali rispetto ad alcune piante. Sui Monti Sicani, situati nel settore centro-occidentale della Sicilia, si rileva la presenza di numerosi alberi monumentali, la cui conservazione è stata assicurata, nonostante le trasformazioni del paesaggio avvenute nel corso di un lunghissimo periodo, dalla positiva influenza delle conoscenze, tradizioni e buone pratiche delle popolazioni locali (De Gregorio, 2008). Tuttavia, ad eccezione del censimento dei grandi alberi di Sicilia (Schicchi e Raimondo, 2007), si dispone di scarsi riferimenti storico-scientifici per l’area, per cui si è ritenuto opportuno realizzare un nuovo lavoro di catalogazione e censimento degli alberi monumentali ricadenti nell’area dei Monti Sicani. Tale censimento ha preso avvio anche da segnalazioni da parte di cittadini ed istituzioni coinvolte, a diversi livelli, nella gestione e fruizione del territorio, e ha consentito di individuare le diverse tipologie di alberi monumentali al fine di identificare, oltre agli alberi isolati, ambiti in cui il concetto di monumentalità si estende non ad un elemento puntuale ma ad un raggruppamento/popolamento di individui che caratterizzano una parte del paesaggio naturale. Una volta stabilito il carattere di monumentalità, sulla base dello schema proposto da Raimondo e Schicchi (2009), si sono realizzate per ciascun patriarca verde specifiche schede comprendenti denominazione, localizzazione e parametri stazionali, descrizione e parametri dendrologici dell’albero, stato vegetativo e fitosanitario dell’esemplare, note e curiosità (Schicchi e Raimondo, 2006). Sono stati individuati 27patriarchi verdi, per la maggior parte di nuova acquisizione; inoltre sono stati raccolti ulteriori dati anche per gli individui già inseriti nel registro degli alberi monumentali della Regione Sicilia (Schicchi e Raimondo, 2007). Gli alberi censiti appartengono a specie selvatiche o coltivate da antica data, afferenti ai generi Olea, Quercus, Prunus, Pyrus, Populus, Pistacia, Sorbus, Fraxinus e Pinus, che vegetano nell'ambito di formazioni forestali a differente grado di naturalità o costituiscono “relitti” all'interno degli agro-ecosistemi tradizionali. Sulla base delle schede è stato possibile valutare gli eventuali interventi conservativi necessari, oltre il 40 % degli alberi monumentali necessita di specifici interventi di conservazione, per le loro precarie condizioni generali, spesso aggravate da rischi di origine antropica. In mancanza di una periodica attività di monitoraggio e di idonee azioni di conservazione, nell’arco di pochi decenni alcuni di questi alberi potrebbero subire danni significativi per sbrancamenti, cedimenti strutturali, attacchi parassitari ed incendi

    Association between sleep-disordered breathing and hearing disorders: Clinical observation in Sicilian patients

    Introduction: To examine the putative association between Sleep-Disordered breathing and hearing disorders Material and methods: 120 Sicilian subjects ranging from 14 to 85 years of age who were divided in 46 cases suffering from tinnitus (G1 group) and 74 controls (G2 group) were evaluated through STOP BANG screening questionnaire and Four-Variable Screening Tool; after Data collecting each subject underwent Audiological assessment by multi-frequency audiometry (PTA) and Transient-evoked otoacoustic emissions (TEOAE-diagnostic) for each ear. Results: Cases showed: PTA significantly severe than the control group (58.70% vs. 16.89% hearing loss; P<0.001), such as a lower signal-to-noise ratio (SNRs) (P<0.05). Moreover Tinnitus subjects had a higher risk to develop Sleep-Disordered breathing respect to controls (P<0.001). OSA risk population - subjects positive to both screening questionnaires - had tinnitus, wore hearing threshold mean values and, lower SNRs values than total cohort (P<0.01). The relative risk of Sleep-Disordered breathing and tinnitus was 4.83 (P<0.0001). Conclusions: Our results stress a probable association between tinnitus, hearing loss and Sleep-Disordered breathing even if further studies will be needed to confirm our findings

    Early Lung Function Abnormalities in Acromegaly.

    BACKGROUND: Acromegaly is an insidious disorder caused by a pituitary growth hormone (GH)-secreting adenoma resulting in high circulating levels of GH and insulin-like growth factor I (IGF-I). Respiratory disorders are common complications in acromegaly, and can severely impact on quality of life, eventually affecting mortality. OBJECTIVES: The present study aimed to explore structural and functional lung alterations of acromegalic subjects. METHODS: We enrolled 10 consecutive patients (M/F: 5/5) affected by acromegaly. In all patients, magnetic resonance imaging (MRI) revealed the presence of pituitary tumor. All patients underwent clinical, lung functional, biological, and radiological assessments. Ten healthy age-matched subjects also served as controls. RESULTS: No statistically significant differences in lung function were detected between acromegalic and healthy subjects (p ≥ 0.05 for all analyses). However, the diffusing capacity for CO (TLCO) was significantly lower in the acromegalic group than in healthy subjects (TLCO% predicted: 78.1 ± 16 vs. 90 ± 6 %, respectively, p = 0.04; KCO% predicted: 77 ± 16 vs. 93 ± 5 %, p = 0.02, respectively). None of the lung function parameters correlated with duration of the disease, or with inflammatory marker of the airways. In acromegalics, biological (exhaled NO concentrations) and imaging (total lung volume, TLV, and mean lung density, MLD) evaluations were within normal values. The TLV measured by HRCT was 3540 ± 1555 ml in acromegalics, and the MLD was -711 ± 73 HU. None of the lung functional, radiological, and biological findings correlated with GH or IGF-I levels, and no correlation was found with duration of disease. CONCLUSIONS: In the current study, lung function evaluation allowed to detect early involvement of lung parenchyma, as assessed by TLCO and KCO, even in the absence of parenchymal density alterations of the lung by HRCT. These findings suggest to routinely include the carbon monoxide diffusing capacity in the lung function assessment for an early intervention in acromegaly

    Widespread Hypomethylation Occurs Early and Synergizes with Gene Amplification during Esophageal Carcinogenesis

    Although a combination of genomic and epigenetic alterations are implicated in the multistep transformation of normal squamous esophageal epithelium to Barrett esophagus, dysplasia, and adenocarcinoma, the combinatorial effect of these changes is unknown. By integrating genome-wide DNA methylation, copy number, and transcriptomic datasets obtained from endoscopic biopsies of neoplastic progression within the same individual, we are uniquely able to define the molecular events associated progression of Barrett esophagus. We find that the previously reported global hypomethylation phenomenon in cancer has its origins at the earliest stages of epithelial carcinogenesis. Promoter hypomethylation synergizes with gene amplification and leads to significant upregulation of a chr4q21 chemokine cluster and other transcripts during Barrett neoplasia. In contrast, gene-specific hypermethylation is observed at a restricted number of loci and, in combination with hemi-allelic deletions, leads to downregulatation of selected transcripts during multistep progression. We also observe that epigenetic regulation during epithelial carcinogenesis is not restricted to traditionally defined “CpG islands,” but may also occur through a mechanism of differential methylation outside of these regions. Finally, validation of novel upregulated targets (CXCL1 and 3, GATA6, and DMBT1) in a larger independent panel of samples confirms the utility of integrative analysis in cancer biomarker discovery

    Multi-messenger observations of a binary neutron star merger

    On 2017 August 17 a binary neutron star coalescence candidate (later designated GW170817) with merger time 12:41:04 UTC was observed through gravitational waves by the Advanced LIGO and Advanced Virgo detectors. The Fermi Gamma-ray Burst Monitor independently detected a gamma-ray burst (GRB 170817A) with a time delay of ~1.7 s with respect to the merger time. From the gravitational-wave signal, the source was initially localized to a sky region of 31 deg2 at a luminosity distance of 40+8-8 Mpc and with component masses consistent with neutron stars. The component masses were later measured to be in the range 0.86 to 2.26 Mo. An extensive observing campaign was launched across the electromagnetic spectrum leading to the discovery of a bright optical transient (SSS17a, now with the IAU identification of AT 2017gfo) in NGC 4993 (at ~40 Mpc) less than 11 hours after the merger by the One- Meter, Two Hemisphere (1M2H) team using the 1 m Swope Telescope. The optical transient was independently detected by multiple teams within an hour. Subsequent observations targeted the object and its environment. Early ultraviolet observations revealed a blue transient that faded within 48 hours. Optical and infrared observations showed a redward evolution over ~10 days. Following early non-detections, X-ray and radio emission were discovered at the transient’s position ~9 and ~16 days, respectively, after the merger. Both the X-ray and radio emission likely arise from a physical process that is distinct from the one that generates the UV/optical/near-infrared emission. No ultra-high-energy gamma-rays and no neutrino candidates consistent with the source were found in follow-up searches. These observations support the hypothesis that GW170817 was produced by the merger of two neutron stars in NGC4993 followed by a short gamma-ray burst (GRB 170817A) and a kilonova/macronova powered by the radioactive decay of r-process nuclei synthesized in the ejecta
