    L'image virtuelle comme source de connaissance pour le patrimoine technique et industriel : Comment allier Histoire et Ingénierie ?

Rejoignant cette problématique d'utilisation du savoir du passé au temps présent afin d'envisager le futur, notre travail de recherche s'oriente vers les objets patrimoniaux à vocation industrielle et à caractère technique ou scientifique. Couramment utilisées par les industries contemporaines, les nouvelles technologies créent de nouveaux modes de travail, de nouvelles représentations... par le virtuel. L'approche présentée consiste à "renverser l'axe des temps de la conception" et à proposer, à partir de l'objet technique et des informations dont on dispose (objet, photos, textes, plans 2D, vestiges archéologiques...) et grâce à l'utilisation d'outils (numérisation 3D, CAO et imagerie, réalité virtuelle...), une remise en situation d'usage virtuelle dynamique des objets techniques anciens. Destinées à des fins de vulgarisations scientifiques, muséographiques, didactiques... ces images virtuelles n'en restent pas moins qu'un point de vue qui ne remplacera jamais le réel. Cependant, tout comme les relevés archéologiques permettent de démontrer une évolution anthropologique, le dessin technique révèle les savoirs et met en évidence les savoir-faire. Cette communication fera tout d'abord un état des lieux des pratiques actuelles du virtuel pour le patrimoine puis statuera sur la méthodologie développée. De nombreux exemples menés dans le cadre de cette recherche viendront en illustrer la communication

    National audienceL'enseignement de la Conception Assistée par Ordinateur en projet tutoré doit permettre d'appréhender d'une part les notions vues pendant les heures d'enseignement mais aussi les difficultés liées à une maquette numérique de taille industrielle (gestion de projet, des fichiers, travail collaboratif). La motivation et le suivi des étudiants sont les garants d'une étude intéressante et formative mais ils sont souvent délicats avec des projets industriels. Nous proposons ici un témoignage pédagogique d'une nouvelle thématique de recherche émergente: la conservation et la valorisation du patrimoine technique et industriel. La richesse technique des "vieilles" machines étudiées, du contexte de l'époque, et la qualité des différents intervenants dans le projet (conservateurs de ce patrimoine, historiens des techniques...) ont séduits les groupes d'étudiants réalisant ce type de projet et c'est pour nous, chercheurs et enseignants, un devoir de diffuser ce type de démarche afin de la rendre reproductible dans d'autres structures universitaires

    Monitoring temperate glaciers: combined use of multi-date TerraSAR-X images and continous GPS measurements

    International audienceThis paper highlights the contribution of TerraSAR-X (TSX) High Resolution (HR) images for temperate glacier monitoring. A series of 14 images have been acquired since October 2007 on the Mont-Blanc test area. This area involves well-known temperate glaciers which have been monitored and instrumented ("stick" for annual displacement/ablation, GPS, cavitometer for basal displacement...) for more than 50 years. The combined use of in-situ measurements and multi-temporal images allows to improve the potential of HR SAR measurements. Interpretation of HR images, investigation of interferometric and correlation methods, and the first glacier displacement results are presented

    Is Impact of Statin Therapy on All-Cause Mortality Different in HIV-Infected Individuals Compared to General Population? Results from the FHDH-ANRS CO4 Cohort

    French Hospital Database on HIVInternational audienceBackgroundThe effect of statins on all-cause mortality in the general population has been estimated as 0.86 (95%CI 0.79-0.94) for primary prevention. Reported values in HIV-infected individuals have been discordant. We assessed the impact of statin-based primary prevention on all-cause mortality among HIV-infected individuals.MethodsPatients were selected among controls from a multicentre nested case-control study on the risk of myocardial infarction. Patients with prior cardiovascular or cerebrovascular disorders were not eligible. Potential confounders, including variables that were associated either with statin use and/or death occurrence and statin use were evaluated within the last 3 months prior to inclusion in the case-control study. Using an intention to continue approach, multiple imputation of missing data, Cox’s proportional hazard models or propensity based weighting, the impact of statins on the 7-year all-cause mortality was evaluated.ResultsAmong 1,776 HIV-infected individuals, 138 (8%) were statins users. During a median follow-up of 53 months, 76 deaths occurred, including 6 in statin users. Statin users had more cardiovascular risk factors and a lower CD4 T cell nadir than statin non-users. In univariable analysis, the death rate was higher in statins users (11% vs 7%, HR 1.22, 95%CI 0.53-2.82). The confounders accounted for were age, HIV transmission group, current CD4 T cell count, haemoglobin level, body mass index, smoking status, anti-HCV antibodies positivity, HBs antigen positivity, diabetes and hypertension. In the Cox multivariable model the estimated hazard ratio of statin on all-cause mortality was estimated as 0.86 (95%CI 0.34-2.19) and it was 0.83 (95%CI 0.51-1.35) using inverse probability treatment weights.ConclusionThe impact of statin for primary prevention appears similar in HIV-infected individuals and in the general population

    Identification and verification of heat shock protein 60 as a potential serum marker for colorectal cancer

    Colorectal cancer (CRC) is a major public health issue worldwide, and novel tumor markers may contribute to its efficient management by helping in early detection, prognosis or surveillance of disease. The aim of our study was to identify new serum biomarkers for CRC, and we followed a phased biomarker discovery and validation process to obtain an accurate preliminary assessment of potential clinical utility. We compared colonic tumors and matched normal tissue from 15 CRC patients, using two-dimensional difference gel electrophoresis (2D-DIGE), and identified 17 proteins that had significant differential expression. These results were further confirmed by western blotting for heat shock protein (HSP) 60, glutathione-S-transferase Pi, α-enolase, T-complex protein 1 subunit β, and leukocyte elastase inhibitor, and by immunohistochemistry for HSP60. Using mAbs raised against HSP60, we developed a reliable (precision of 5–15%) and sensitive (0.3 ng·mL−1) immunoassay for the detection of HSP60 in serum. Elevated levels of HSP60 were found in serum from CRC patients in two independent cohorts; the receiver-operating characteristic curve obtained in 112 patients with CRC and 90 healthy controls had an area under the curve (AUC) of 0.70, which was identical to the AUC of carcinoembryonic antigen. Combination of serum markers improved clinical performance: the AUC of a three-marker logistic regression model combining HSP60, carcinoembryonic antigen and carbohydrate antigen 19-9 reached 0.77. Serum HSP60 appeared to be more specific for late-stage CRC; therefore, future studies should evaluate its utility for determining prognosis or monitoring therapy rather than early detection

    N-Acyl Homoserine Lactones in Diverse Pectobacterium and Dickeya Plant Pathogens: Diversity, Abundance, and Involvement in Virulence

    Soft-rot bacteria Pectobacterium and Dickeya use N-acyl homoserine lactones (NAHSLs) as diffusible signals for coordinating quorum sensing communication. The production of NAHSLs was investigated in a set of reference strains and recently-collected isolates, which belong to six species and share the ability to infect the potato host plant. All the pathogens produced different NAHSLs, among which the 3-oxo-hexanoyl- and the 3-oxo-octanoyl-l-homoserine lactones represent at least 90% of total produced NAHSL-amounts. The level of NAHSLs varied from 0.6 to 2 pg/cfu. The involvement of NAHSLs in tuber maceration was investigated by electroporating a quorum quenching vector in each of the bacterial pathogen strains. All the NAHSL-lactonase expressing strains produced a lower amount of NAHSLs as compared to those harboring the empty vector. Moreover, all except Dickeya dadantii 3937 induced a lower level of symptoms in potato tuber assay. Noticeably, aggressiveness appeared to be independent of both nature and amount of produced signals. This work highlights that quorum sensing similarly contributed to virulence in most of the tested Pectobacterium and Dickeya, even the strains had been isolated recently or during the past decades. Thus, these key regulatory-molecules appear as credible targets for developing anti-virulence strategies against these plant pathogens

    Long-term effects of evolocumab in participants with HIV and dyslipidemia: results from the open-label extension period

    Objectives: People with HIV (PWH) are at an increased risk of atherosclerotic cardiovascular disease. Suboptimal responses to statin therapy in PWH may result from antiretroviral therapies (ARTs). This open-label extension study aimed to evaluate the long-term safety and efficacy of evolocumab up to 52\u200aweeks in PWH. Design: This final analysis of a multinational, placebo-controlled, double-blind, randomized phase 3 trial evaluated the effect of monthly subcutaneous evolocumab 420\u200amg on low-density lipoprotein cholesterol (LDL-C) during the open-label period (OLP) following 24\u200aweeks of double-blind period in PWH with hypercholesterolemia/mixed dyslipidemia. All participants enrolled had elevated LDL-C or nonhigh-density lipoprotein cholesterol (non-HDL-C) and were on stable maximally tolerated statin and stable ART. Methods: Efficacy was assessed by percentage change from baseline in LDL-C, triglycerides, and atherogenic lipoproteins. Treatment-emergent adverse events (TEAEs) were examined. Results: Of the 467 participants randomized in the double-blind period, 451 (96.6%) received at least one dose of evolocumab during the OLP (mean age of 56.4\u200ayears, 82.5% male, mean duration with HIV of 17.4\u200ayears). By the end of the 52-week OLP, the overall mean (SD) percentage change in LDL-C from baseline was -57.8% (22.8%). Evolocumab also reduced triglycerides, atherogenic lipid parameters (non-HDL-C, apolipoprotein B, total cholesterol, very-low-density lipoprotein cholesterol, and lipoprotein[a]), and increased HDL-C. TEAEs were similar between placebo and evolocumab during the OLP. Conclusion: Long-term administration of evolocumab lowered LDL-C and non-HDL-C, allowing more PWH to achieve recommended lipid goals with no serious adverse events. Trail registration: NCT02833844.
