    Dogslife: A web-based longitudinal study of Labrador Retriever health in the UK

    <p>Abstract</p> <p>Background</p> <p>Dogslife is the first large-scale internet-based longitudinal study of canine health. The study has been designed to examine how environmental and genetic factors influence the health and development of a birth cohort of UK-based pedigree Labrador Retrievers.</p> <p>Results</p> <p>In the first 12 months of the study 1,407 Kennel Club (KC) registered eligible dogs were recruited, at a mean age of 119 days of age (SD 69 days, range 3 days – 504 days). Recruitment rates varied depending upon the study team’s ability to contact owners. Where owners authorised the provision of contact details 8.4% of dogs were recruited compared to 1.3% where no direct contact was possible. The proportion of dogs recruited was higher for owners who transferred the registration of their puppy from the breeder to themselves with the KC, and for owners who were sent an e-mail or postcard requesting participation in the project. Compliance with monthly updates was highly variable. For the 280 dogs that were aged 400 days or more on the 30<sup>th</sup> June 2011, we estimated between 39% and 45% of owners were still actively involved in the project. Initial evaluation suggests that the cohort is representative of the general population of the KC registered Labrador Retrievers eligible to enrol with the project. Clinical signs of illnesses were reported in 44.3% of Labrador Retrievers registered with Dogslife (median age of first illness 138 days), although only 44.1% of these resulted in a veterinary presentation (median age 316 days).</p> <p>Conclusions</p> <p>The web-based platform has enabled the recruitment of a representative population of KC registered Labrador Retrievers, providing the first large-scale longitudinal population-based study of dog health. The use of multiple different methods (e-mail, post and telephone) of contact with dog owners was essential to maximise recruitment and retention of the cohort.</p

    Genomic Expression Libraries for the Identification of Cross-Reactive Orthopoxvirus Antigens

    Increasing numbers of human cowpox virus infections that are being observed and that particularly affect young non-vaccinated persons have renewed interest in this zoonotic disease. Usually causing a self-limiting local infection, human cowpox can in fact be fatal for immunocompromised individuals. Conventional smallpox vaccination presumably protects an individual from infections with other Orthopoxviruses, including cowpox virus. However, available live vaccines are causing severe adverse reactions especially in individuals with impaired immunity. Because of a decrease in protective immunity against Orthopoxviruses and a coincident increase in the proportion of immunodeficient individuals in today's population, safer vaccines need to be developed. Recombinant subunit vaccines containing cross-reactive antigens are promising candidates, which avoid the application of infectious virus. However, subunit vaccines should contain carefully selected antigens to confer a solid cross-protection against different Orthopoxvirus species. Little is known about the cross-reactivity of antibodies elicited to cowpox virus proteins. Here, we first identified 21 immunogenic proteins of cowpox and vaccinia virus by serological screenings of genomic Orthopoxvirus expression libraries. Screenings were performed using sera from vaccinated humans and animals as well as clinical sera from patients and animals with a naturally acquired cowpox virus infection. We further analyzed the cross-reactivity of the identified immunogenic proteins. Out of 21 identified proteins 16 were found to be cross-reactive between cowpox and vaccinia virus. The presented findings provide important indications for the design of new-generation recombinant subunit vaccines

    A knowledge translation collaborative to improve the use of therapeutic hypothermia in post-cardiac arrest patients: protocol for a stepped wedge randomized trial

    <p>Abstract</p> <p>Background</p> <p>Advances in resuscitation science have dramatically improved survival rates following cardiac arrest. However, about 60% of adults that regain spontaneous circulation die before leaving the hospital. Recently it has been shown that inducing hypothermia in cardiac arrest survivors immediately following their arrival in hospital can dramatically improve both overall survival and neurological outcomes. Despite the strong evidence for its efficacy and the apparent simplicity of this intervention, recent surveys show that therapeutic hypothermia is delivered inconsistently, incompletely, and often with delay.</p> <p>Methods and design</p> <p>This study will evaluate a multi-faceted knowledge translation strategy designed to increase the utilization rate of induced hypothermia in survivors of cardiac arrest across a network of 37 hospitals in Southwestern Ontario, Canada. The study is designed as a stepped wedge randomized trial lasting two years. Individual hospitals will be randomly assigned to four different wedges that will receive the active knowledge translation strategy according to a sequential rollout over a number of time periods. By the end of the study, all hospitals will have received the intervention. The primary aim is to measure the effectiveness of a multifaceted knowledge translation plan involving education, reminders, and audit-feedback for improving the use of induced hypothermia in survivors of cardiac arrest presenting to the emergency department. The primary outcome is the proportion of eligible OHCA patients that are cooled to a body temperature of 32 to 34°C within six hours of arrival in the hospital. Secondary outcomes will include process of care measures and clinical outcomes.</p> <p>Discussion</p> <p>Inducing hypothermia in cardiac arrest survivors immediately following their arrival to hospital has been shown to dramatically improve both overall survival and neurological outcomes. However, this lifesaving treatment is frequently not applied in practice. If this trial is positive, our results will have broad implications by showing that a knowledge translation strategy shared across a collaborative network of hospitals can increase the number of patients that receive this lifesaving intervention in a timely manner.</p> <p>Trial Registration</p> <p>ClinicalTrials.gov Trial Identifier: <a href="http://www.clinicaltrials.gov/ct2/show/NCT00683683">NCT00683683</a></p

    Recommended temperature metrics for carbon budget estimates, model evaluation and climate policy

    Recent estimates of the amount of carbon dioxide that can still be emitted while achieving the Paris Agreement temperature goals are larger than previously thought. One potential reason for these larger estimates may be the different temperature metrics used to estimate the observed global mean warming for the historical period, as they affect the size of the remaining carbon budget. Here we explain the reasons behind these remaining carbon budget increases, and discuss how methodological choices of the global mean temperature metric and the reference period influence estimates of the remaining carbon budget. We argue that the choice of the temperature metric should depend on the domain of application. For scientific estimates of total or remaining carbon budgets, globally averaged surface air temperature estimates should be used consistently for the past and the future. However, when used to inform the achievement of the Paris Agreement goal, a temperature metric consistent with the science that was underlying and directly informed the Paris Agreement should be applied. The resulting remaining carbon budgets should be calculated using the appropriate metric or adjusted to reflect these differences among temperature metrics. Transparency and understanding of the implications of such choices are crucial to providing useful information that can bridge the science–policy gap

Yet despite the surge in interest in the work of artists such as Alison Bechdel, and despite the existence of a substantial online archive about lesbian comics created by artists, readers, and collectors, relatively little critical attention has been directed to this work. The chapter begins to fill this gap. Taking the Bechdel’s work as its start-and-end point, it provides an overview of major developments in lesbian comics and contextualises them including in relation to the gendered conditions of possibility that define comics culture

    Psychological mindedness and abstract reasoning in late childhood and adolescence: An exploration using new instruments

    This study introduces two new measures of psychological mindedness, applying them in a study of the growth of abstract thinking in children and adolescents in a developmental design. The capacity to achieve psychological understanding of the self and of others involves comprehension of the motives, attitudes, and characteristics of the self and others. Psychological mindedness toward the self (PS) and toward others (PO) may be seen as complex cognitive capacities that should show a pattern of related development in childhood. Three groups of 60 fifth, eighth, and twelfth graders completed two measures of formal operations and two instruments to assess the two components of psychological mindedness. We find that psychological mindedness and abstract thinking both increase significantly with age, although the relationship between them is complex and varies with gender and age. Because the development of abstract reasoning skills does not correlate with the development of psychological mindedness in a simple way, a more complex model is necessary, taking age and gender differences into account. Performance on the two measures of psychological mindedness is found to be largely unrelated, suggesting that these are two different psychological skills.     Object naming induces reliance on orientation-independent representations during longer-term, but not short-term, visual remembering.

    Naming novel objects with novel count nouns changes how the objects are drawn from memory, revealing that object categorisation induces reliance on orientation-independent visual representations during longer-term remembering, but not during short-term remembering. Serial position effects integrate this finding with a more established conceptualisation of short-term and longer-term visual remembering in which the former is identified as keeping an item in mind. Adults were shown a series of four novel objects in orientations in which they would not normally be drawn from memory. When not named (“Look at this object”), the objects were drawn in the orientations in which they had been seen. When named with a novel count noun (e.g., “Look at this dax”), the final object continued to be depicted in the orientation in which it had been seen, but all other objects were depicted in an unseen but preferred (canonical) orientation, even though participants could still remember the orientations in which they had been seen. Although orientation-dependent exemplar representations appear to be more accessible than orientation-independent generic representations during short-term remembering, the reverse is the case during longer-term remembering. How the theoretical framework emerging from these observations accommodates a broader body of evidence is discussed
