    Effect of nanoparticle morphologies on signal strength in photoacoustic sensing

    Spherical gold nanoparticles with a plasmonic extinction peak at 532 nm and two sizes of star shaped gold nanoparticles with plasmonic extinction peaks at 532 nm and 600 nm were synthesised and introduced into tissue phantoms as exogenous absorbers. The photoacoustic signals generated from the three different nanoparticle morphologies embedded in tissue the phantoms is compared. The effect of nanoparticle concentration on the generated photoacoustic signal strength was also investigated for the spherical nanoparticles.     Bomb-<sup>14</sup>C analysis of ecosystem respiration reveals that peatland vegetation facilitates release of old carbon

    The largest terrestrial-to-atmosphere carbon flux is respired CO&lt;sub&gt;2&lt;/sub&gt;. However, the partitioning of soil and plant sources, understanding of contributory mechanisms, and their response to climate change are uncertain. A plant removal experiment was established within a peatland located in the UK uplands to quantify respiration derived from recently fixed plant carbon and that derived from decomposition of soil organic matter, using natural abundance &lt;sup&gt;13&lt;/sup&gt;C and bomb-&lt;sup&gt;14&lt;/sup&gt;C as tracers. Soil and plant respiration sources were found respectively to contribute ~ 36% and between 41-54% of the total ecosystem CO&lt;sub&gt;2&lt;/sub&gt; flux. Respired CO&lt;sub&gt;2&lt;/sub&gt; produced in the clipped (‘soil’) plots had a mean age of ~ 15 years since fixation from the atmosphere, whereas the &lt;sup&gt;14&lt;/sup&gt;C content of ecosystem CO&lt;sub&gt;2&lt;/sub&gt; was statistically indistinguishable from the contemporary atmosphere. Results of carbon mass balance modelling showed that, in addition to respiration from bulk soil and plant respired CO&lt;sub&gt;2&lt;/sub&gt;, a third, much older source of CO&lt;sub&gt;2&lt;/sub&gt; existed. This source, which we suggest is CO&lt;sub&gt;2&lt;/sub&gt; derived from the catotelm constituted between ~ 10 and 23% of total ecosystem respiration and had a mean radiocarbon age of between several hundred to ~ 2000 years before present (BP). These findings show that plant-mediated transport of CO&lt;sub&gt;2&lt;/sub&gt; produced in the catotelm may form a considerable component of peatland ecosystem respiration. The implication of this discovery is that current assumptions in terrestrial carbon models need to be re-evaluated to consider the climate sensitivity of this third source of peatland CO&lt;sub&gt;2&lt;/sub&gt;

    A genome-wide association study of radiotherapy induced toxicity in head and neck cancer patients identifies a susceptibility locus associated with mucositis

    PURPOSE: A two-stage genome-wide association study was carried out in head and neck cancer (HNC) patients aiming to identify genetic variants associated with either specific radiotherapy-induced (RT) toxicity endpoints or a general proneness to develop toxicity after RT.MATERIALS AND METHODS: The analysis included 1780 HNC patients treated with primary RT for laryngeal or oro/hypopharyngeal cancers. In a non-hypothesis-driven explorative discovery study, associations were tested in 1183 patients treated within The Danish Head and Neck Cancer Group. Significant associations were later tested in an independent Dutch cohort of 597 HNC patients and if replicated, summary data obtained from discovery and replication studies were meta-analysed. Further validation of significantly replicated findings was pursued in an Asian cohort of 235 HNC patients with nasopharynx as the primary tumour site.RESULTS: We found and replicated a significant association between a locus on chromosome 5 and mucositis with a pooled OR for rs1131769*C in meta-analysis = 1.95 (95% CI 1.48-2.41; ppooled = 4.34 × 10-16).CONCLUSION: This first exploratory GWAS in European cohorts of HNC patients identified and replicated a risk locus for mucositis. A larger Meta-GWAS to identify further risk variants for RT-induced toxicity in HNC patients is warranted.</p

    Systematic uncertainties in the light-element abundances and their evolution make a rigorous statistical assessment difficult. However, using Bayesian methods we show that the following statement is robust: the predicted and measured abundances are consistent with 95\% credibility only if the baryon-to-photon ratio is between 2×10102\times 10^{-10} and 6.5×10106.5\times 10^{-10} and the number of light neutrino species is less than 3.9.     Glaucoma is a multi-factorial blinding disease in which genetic factors play an important role. Elevated intraocular pressure is a highly heritable risk factor for primary open angle glaucoma and currently the only target for glaucoma therapy. Our study helps to better understand underlying genetic and molecular mechanisms that regulate intraocular pressure, and identifies a new candidate gene, Cacna2d1, that modulates intraocular pressure and a promising therapeutic, pregabalin, which binds to CACNA2D1 protein and lowers intraocular pressure significantly.     Cross-ancestry genome-wide association analysis of corneal thickness strengthens link between complex and Mendelian eye diseases

    Central corneal thickness (CCT) is a highly heritable trait associated with complex eye diseases such as keratoconus and glaucoma. We perform a genome-wide association meta-analysis of CCT and identify 19 novel regions. In addition to adding support for known connective tissue-related pathways, pathway analyses uncover previously unreported gene sets. Remarkably, >20% of the CCT-loci are near or within Mendelian disorder genes. These included FBN1, ADAMTS2 and TGFB2 which associate with connective tissue disorders (Marfan, Ehlers-Danlos and Loeys-Dietz syndromes), and the LUM-DCN-KERA gene complex involved in myopia, corneal dystrophies and cornea plana. Using index CCT-increasing variants, we find a significant inverse correlation in effect sizes between CCT and keratoconus (r =-0.62, P = 5.30 × 10-5) but not between CCT and primary open-angle glaucoma (r =-0.17, P = 0.2). Our findings provide evidence for shared genetic influences between CCT and keratoconus, and implicate candidate genes acting in collagen and extracellular matrix regulation

    Improving Genetic Prediction by Leveraging Genetic Correlations Among Human Diseases and Traits

    Genomic prediction has the potential to contribute to precision medicine. However, to date, the utility of such predictors is limited due to low accuracy for most traits. Here theory and simulation study are used to demonstrate that widespread pleiotropy among phenotypes can be utilised to improve genomic risk prediction. We show how a genetic predictor can be created as a weighted index that combines published genome-wide association study (GWAS) summary statistics across many different traits. We apply this framework to predict risk of schizophrenia and bipolar disorder in the Psychiatric Genomics consortium data, finding substantial heterogeneity in prediction accuracy increases across cohorts. For six additional phenotypes in the UK Biobank data, we find increases in prediction accuracy ranging from 0.7 for height to 47 for type 2 diabetes, when using a multi-trait predictor that combines published summary statistics from multiple traits, as compared to a predictor based only on one trait. © 2018 The Author(s)
