    Prediction of outcome after paraquat poisoning by measurement of the plasma paraquat concentration

    Background: Paraquat is a herbicide with a good occupational safety record, but a high mortality after intentional ingestion that has proved refractory to treatment. For nearly three decades paraquat concentration-time data have been used to predict the outcome following ingestion. However, none of the published methods has been independently or prospectively validated. We aimed to use prospectively collected data to test the published predictive methods and to determine if any is superior. Methods: Plasma paraquat concentrations were measured on admission for 451 patients in 10 hospitals in Sri Lanka as part of large prospective cohort study. All deaths in hospital were recorded; patients surviving to hospital discharge were followed up after 3 months to detect delayed deaths. Five prediction methods that are based on paraquat concentration-time data were then evaluated in all eligible patients. Results: All methods showed comparable performance within their range of application. For example, between 4- and 24-h prediction of prognosis was most variable between Sawada and Proudfoot methods but these differences were relatively small [specificity 0.96 (95% CI: 0.90-0.99) vs. 0.89 (0.82-0.95); sensitivity 0.57 vs. 0.79, positive and negative likelihood ratios 14.8 vs. 7.40 and 0.44 vs. 0.23 and positive predictive values 0.96 vs. 0.92, respectively]. Conclusions: All five published methods were better at predicting death than survival. These predictions may also serve as tools to identify patients who need treatment and for some assessment to be made of new treatments that are trialled without a control grou

    Stock assessment of Australian east coast Spanish mackerel : Predictions of stock status and reference points

    The stock assessment describes Australia’s east coast Spanish mackerel fishery operating in ocean waters between Cape York Peninsula and northern New South Wales. The species are a large offshore pelagic fish that can live for up to 26 years and weigh in excess of 30 kg. Spanish mackerel reach maturity above the minimum legal size of 75 cm at between two and four years of age. The stock assessment was conducted at the whole stock level and included commercial, charter, recreational and research data from both New South Wales and Queensland. The data included estimates of Spanish mackerel harvest from logbook systems and recreational fishing surveys, catch rates from commercial logbooks and historical surveys of long term commercial fishers, and annual fish age-length compositions. The fishery for east coast Spanish mackerel commenced in the early 1900s. Annual east coast harvests taken by commercial, charter and recreational fishing steadily built to peak at 1000–1300 tonnes (t) per year during the 1970s and 1000–1150 t per year between 1998 and 2003. The estimated annual Spanish mackerel harvest since 2005 reduced to 500–760 t per year. Most fishers harvest Spanish mackerel using line-fishing techniques. Net fishing for Spanish mackerel is illegal. In recent years about 47 per cent, 6 per cent and 47 per cent of annual Spanish mackerel harvests were taken respectively by commercial, charter and recreational fishing across all east coast waters. Population modelling of the data estimated that Spanish mackerel population size in 2016 was between 30–50 per cent of original biomass estimated at the start of the fishery in 1911 (best estimate around 40 per cent biomass). Estimates of recommended sustainable annual harvest of Spanish mackerel for all fishing sectors and east-coast waters ranged between 400 and 800 t. There is presently substantial unfished commercial quota. The current Queensland total allowable commercial catch quota is 574.6 t. If this were to be largely utilised, together with current or increased charter, recreational and New South Wales commercial harvests, then the biomass of the Spanish mackerel population may decline. Such high harvests would result in overfishing and reduce average catch rates longer-term. The results suggested annual harvests of around 550 t (across all sectors) will build the biomass towards the 60 per cent level, consistent with the 2027 biomass targets set in the Queensland Government’s Sustainable Fisheries Strategy. If there is a desire to operate the fishery closer towards 60 per cent biomass for better economic yield and quality of fishing (higher catch rates), then fishing pressure will need to reduce for a period of time to build the fish population to a higher biomass

    Correlated variability of Mkn 421 at X-ray and TeV wavelengths on timescales of hours

    Mkn 421 was observed for about two days with BeppoSAX, prior to and partly overlapping the start of a 1 week continuous exposure with ASCA in April 1998, as part of a world-wide multiwavelength campaign. A pronounced, well defined, flare observed in X-rays was also observed simultaneously at TeV energies by the Whipple Observatory's 10 m gamma-ray telescope.     Measurements of Distributed Strain During Impact Pile Driving

    This paper reports the use of optical Fibre Bragg Grating (FBG) sensors to monitor the stress waves generated below ground during pile driving, combined with measurements using conventional pile driving analyzer (PDA) sensors mounted at the pile head. Fourteen tubular steel piles with a diameter of 508 mm and embedded length to diameter ratios of 6 to 20 were impact driven at an established chalk test site in Kent, UK. The pile shafts were instrumented with multiple FBG strain gauges and pile head PDA sensors, which monitored the piles’ responses under each hammer blow. A high frequency (5 kHz) fibre optic interrogator allowed a previously unseen resolution of the stress wave propagation along the pile. Estimates of the base soil resistances to driving and distributions of shaft shear resistances were found through signal matching that compared time series of pile head PDA measurements and FBG strains measured below ground surface. Numerical solutions of the one-dimensional wave equation were optimised by taking account of the data from multiple FBG gauges, leading to significant advantages that have potential for widespread application in cases where high resolution strain measurements are made

    Integrin-mediated leukocyte adhesion is a critical aspect of leukocyte function that is tightly regulated by diverse stimuli, including chemokines, antigen receptors, and adhesion receptors. How cellular signals from CD31 and other adhesion amplifiers are integrated with those from classical mitogenic stimuli to regulate leukocyte function remains poorly understood. Here, we show that the cytoplasmic tail of CD31, an important integrin adhesion amplifier, propagates signals that induce T cell adhesion via β1 (VLA-4) and β2 (LFA-1) integrins. We identify the small GTPase, Rap1, as a critical mediator of this effect. Importantly, CD31 selectively activated the small Ras-related GTPase, Rap1, but not Ras, R-Ras, or Rap2. An activated Rap1 mutant stimulated T lymphocyte adhesion to intercellular adhesion molecule (ICAM) and vascular cell adhesion molecule (VCAM), as did the Rap1 guanine nucleotide exchange factor C3G and a catalytically inactive mutant of RapGAP. Conversely, negative regulators of Rap1 signaling blocked CD31-dependent adhesion. These findings identify a novel important role for Rap1 in regulating ligand-induced cell adhesion and suggest that Rap1 may play a more general role in coordinating adhesion-dependent signals during leukocyte migration and extravasation. Our findings also suggest an alternative mechanism, distinct from interference with Ras-proximal signaling, by which Rap1 might mediate transformation reversion

    Search for direct production of charginos and neutralinos in events with three leptons and missing transverse momentum in √s = 7 TeV pp collisions with the ATLAS detector

    A search for the direct production of charginos and neutralinos in final states with three electrons or muons and missing transverse momentum is presented. The analysis is based on 4.7 fb−1 of proton–proton collision data delivered by the Large Hadron Collider and recorded with the ATLAS detector. Observations are consistent with Standard Model expectations in three signal regions that are either depleted or enriched in Z-boson decays. Upper limits at 95% confidence level are set in R-parity conserving phenomenological minimal supersymmetric models and in simplified models, significantly extending previous results
