    The paradigm of the area law and the structure of transversal and longitudinal lightfront degrees of freedom

    It is shown that an algebraically defined holographic projection of a QFT onto the lightfront changes the local quantum properties in a very drastic way. The expected ubiquitous vacuum polarization characteristic of QFT is confined to the lightray (longitudinal) direction, whereas operators whose localization is transversely separated are completely free of vacuum correlations. This unexpected ''transverse return to QM'' combined with the rather universal nature of the strongly longitudinal correlated vacuum correlations (which turn out to be described by rather kinematical chiral theories) leads to a d-2 dimensional area structure of the d-1 dimensional lightfront theory. An additive transcription in terms of an appropriately defined entropy related to the vacuum restricted to the horizon is proposed and its model independent universality aspects which permit its interpretation as a quantum candidate for Bekenstein's area law are discussed. The transverse tensor product foliation structure of lightfront degrees of freedom is essential for the simplifying aspects of the algebraic lightcone holography.     Hysteresis phenomenon in turbulent convection

    Coherent large-scale circulations of turbulent thermal convection in air have been studied experimentally in a rectangular box heated from below and cooled from above using Particle Image Velocimetry. The hysteresis phenomenon in turbulent convection was found by varying the temperature difference between the bottom and the top walls of the chamber (the Rayleigh number was changed within the range of 10710810^7 - 10^8). The hysteresis loop comprises the one-cell and two-cells flow patterns while the aspect ratio is kept constant (A=22.23A=2 - 2.23). We found that the change of the sign of the degree of the anisotropy of turbulence was accompanied by the change of the flow pattern. The developed theory of coherent structures in turbulent convection (Elperin et al. 2002; 2005) is in agreement with the experimental observations. The observed coherent structures are superimposed on a small-scale turbulent convection. The redistribution of the turbulent heat flux plays a crucial role in the formation of coherent large-scale circulations in turbulent convection.Comment: 10 pages, 9 figures, REVTEX4, Experiments in Fluids, 2006, in pres

    Chronic Hepatitis B Finite Treatment: similar and different concerns with new drug classes

    Chronic hepatitis B, a major cause of liver disease and cancer, affects over 250 million people worldwide. Currently there is no cure, only suppressive therapies. Efforts to develop finite curative HBV therapies are underway, consisting of combinations of multiple novel agents +/- nucleos(t)ide reverse transcriptase inhibitors. The HBV Forum convened a webinar in July 2021, and subsequent working group discussions to address how and when to stop finite therapy for demonstration of sustained off-treatment efficacy and safety responses. Participants included leading experts in academia, clinical practice, pharmaceutical companies, patient representatives and regulatory agencies. This Viewpoint outlines areas of consensus within our multi-stakeholder group for stopping finite therapies in chronic Hepatitis B investigational studies, including trial design, patient selection, outcomes, biomarkers, pre-defined stopping criteria, pre-defined retreatment criteria, duration of investigational therapies, and follow up after stopping therapy. Future research of unmet needs are discussed

    Delayed Differentiation Makes Many Models Compatible with Data for CD8+ T Cell Differentiation

    Upon antigen stimulation, naïve CD8+ T cells differentiate into short-lived effectors and longer-lived memory T cells. The kinetics of expansion of antigen-specific CD8+ T cells is highly reproducible at the population level, but the fate of individual naïve cells is stochastic, as individual naïve CD8+ T cells produce different numbers of effector and memory cells. Using mathematical models to analyse experimental data on tracing the fate of individual naïve T cells, it was previously shown that a linear model where naïve CD8+ T cells first differentiate into memory precursors that subsequently differentiate into effector cells describes the data best. However, this ‘memory first’ linear model assumed that the proliferation and differentiation events were distributed exponentially, whereas several studies indicate that differentiation of CD8+ T cell subsets need not follow an exponential distribution. Here we investigate the effect of delayed differentiation by adding intermediate compartments and use similar ordinary differential equations and Gillespie simulations to evaluate alternate models of CD8+ T cell differentiation. Models where a substantial fraction of the naïve CD8+ T cells directly differentiate into effector cells, without going through a memory phase, exhibit population dynamics that are very similar to the original ‘memory first’ linear model. Because alternate models with delayed differentiation perform better than those without a delay, we conclude that non-exponential forms of cellular differentiation need to be considered when comparing models. Hence the exact pathway for the differentiation of naïve CD8+ T cells into effector and memory T cells remains an open question

We have resorted to game theory models in order to demonstrate cooperative and non-cooperative scenarios, ranging from the traditional to the evolutionary within game theory. In doing so we are able to glimpse the development of modern society and a paradigm shift regarding human control over nature and to what extent it is harmful to the sustainability of our environment and the survival of future generations. Merging different fields of knowledge, we present a theoretical-philosophical approach, combined with empirical-mathematical solutions taking into account the agent-based behaviour guided blindly by instrumental rationality

    Search for new phenomena in final states with an energetic jet and large missing transverse momentum in pp collisions at √ s = 8 TeV with the ATLAS detector

    Results of a search for new phenomena in final states with an energetic jet and large missing transverse momentum are reported. The search uses 20.3 fb−1 of √ s = 8 TeV data collected in 2012 with the ATLAS detector at the LHC. Events are required to have at least one jet with pT > 120 GeV and no leptons. Nine signal regions are considered with increasing missing transverse momentum requirements between Emiss T > 150 GeV and Emiss T > 700 GeV. Good agreement is observed between the number of events in data and Standard Model expectations. The results are translated into exclusion limits on models with either large extra spatial dimensions, pair production of weakly interacting dark matter candidates, or production of very light gravitinos in a gauge-mediated supersymmetric model.     Mesenchymal Stem Cells Promote Mammosphere Formation and Decrease E-Cadherin in Normal and Malignant Breast Cells

    Normal and malignant breast tissue contains a rare population of multi-potent cells with the capacity to self-renew, referred to as stem cells, or tumor initiating cells (TIC). These cells can be enriched by growth as "mammospheres" in three-dimensional cultures.We tested the hypothesis that human bone-marrow derived mesenchymal stem cells (MSC), which are known to support tumor growth and metastasis, increase mammosphere formation.We found that MSC increased human mammary epithelial cell (HMEC) mammosphere formation in a dose-dependent manner. A similar increase in sphere formation was seen in human inflammatory (SUM149) and non-inflammatory breast cancer cell lines (MCF-7) but not in primary inflammatory breast cancer cells (MDA-IBC-3). We determined that increased mammosphere formation can be mediated by secreted factors as MSC conditioned media from MSC spheroids significantly increased HMEC, MCF-7 and SUM149 mammosphere formation by 6.4 to 21-fold. Mammospheres grown in MSC conditioned media had lower levels of the cell adhesion protein, E-cadherin, and increased expression of N-cadherin in SUM149 and HMEC cells, characteristic of a pro-invasive mesenchymal phenotype. Co-injection with MSC in vivo resulted in a reduced latency time to develop detectable MCF-7 and MDA-IBC-3 tumors and increased the growth of MDA-IBC-3 tumors. Furthermore, E-cadherin expression was decreased in MDA-IBC-3 xenografts with co-injection of MSC.MSC increase the efficiency of primary mammosphere formation in normal and malignant breast cells and decrease E-cadherin expression, a biologic event associated with breast cancer progression and resistance to therapy

    RAD21 Cooperates with Pluripotency Transcription Factors in the Maintenance of Embryonic Stem Cell Identity

    For self-renewal, embryonic stem cells (ESCs) require the expression of specific transcription factors accompanied by a particular chromosome organization to maintain a balance between pluripotency and the capacity for rapid differentiation. However, how transcriptional regulation is linked to chromosome organization in ESCs is not well understood. Here we show that the cohesin component RAD21 exhibits a functional role in maintaining ESC identity through association with the pluripotency transcriptional network. ChIP-seq analyses of RAD21 reveal an ESC specific cohesin binding pattern that is characterized by CTCF independent co-localization of cohesin with pluripotency related transcription factors Oct4, Nanog, Sox2, Esrrb and Klf4. Upon ESC differentiation, most of these binding sites disappear and instead new CTCF independent RAD21 binding sites emerge, which are enriched for binding sites of transcription factors implicated in early differentiation. Furthermore, knock-down of RAD21 causes expression changes that are similar to expression changes after Nanog depletion, demonstrating the functional relevance of the RAD21 - pluripotency transcriptional network association. Finally, we show that Nanog physically interacts with the cohesin or cohesin interacting proteins STAG1 and WAPL further substantiating this association. Based on these findings we propose that a dynamic placement of cohesin by pluripotency transcription factors contributes to a chromosome organization supporting the ESC expression program

    On the relations between Markov chain lumpability and reversibility

    In the literature, the notions of lumpability and time reversibility for large Markov chains have been widely used to efficiently study the functional and non-functional properties of computer systems. In this paper we explore the relations among different definitions of lumpability (strong, exact and strict) and the notion of time-reversed Markov chain. Specifically, we prove that an exact lumping induces a strong lumping on the reversed Markov chain and a strict lumping holds both for the forward and the reversed processes. Based on these results we introduce the class of λρ-reversible Markov chains which combines the notions of lumping and time reversibility modulo state renaming. We show that the class of autoreversible processes, previously introduced in Marin and Rossi (Proceedings of the IEEE 21st international symposium on modeling, analysis and simulation of computer and telecommunication systems MASCOTS, pp. 151–160, 2013), is strictly contained in the class of λρ-reversible chains
