    Empathetic Humanism and Multiethnic Narratives

    This dissertation uncovers how select multiethnic American literatures imagine minoritarian subjectivity that is not premised on categories of nationalism or American mythos of agency, but rather privilege non-Western humanist subject-formation processes. Given their outlying position in the American literary canon, multiethnic, interracial texts have the capacity to engage not only alternative frameworks of subject formation, but also specifically humanist frames – meaning, encounters of inclusion that occur because of a reciprocal recognition of a shared condition of being. Investigating narratives of interethnic reception, this project illuminates how some modes of being, such as suffering and joy, illustrate a new humanism predicated on radical empathy. Employing an archive of narrative works that range from Barack Obama’s Dreams of My Father (1995) to Monique Truong’s Bitter in the Mouth (2010), alongside texts such as Karen Tei Yamashita’s Through the Arc of the Rainforest (1990), this dissertation shows how minority subjectivities can be cultivated beyond the domain of settler agendas: how, for example, ethnic difference incites cross-cultural dignity, not racial subjugation; or how militaristic violence heralds guardianship amongst its victims, not reactive hatred. Ultimately, this narrative methodology works to undermine mechanisms of agency that create subjects only to control, condition, and constrain them. Exploring how multiethnic literature expresses underexamined humanist encounters between minoritized peoples, this dissertation demonstrates the potential to destabilize the exclusionist nationalism that first marginalized them, without engaging or relying on said national codes of subjectivity. To that end, this project concludes by exploring how a methodology of empathetic humanism can be put into praxis. Providing examples of partnerships with local disenfranchised groups to co-create public-facing projects that expand the frame of “who counts” as a viable subject, this dissertation closes by demonstrating how empathetic humanism as a theory and methodology can be employed to benefit the public, common good

    Dating violence: college students\u27 experiences and intervention suggestions

    The dating violence relationship experiences of students were investigated at a southeast regional university. A third of the 509 participants indicated they were victims of dating violence (n = 173), and almost 25% (n = 124) indicated they had victimized someone they had dated. Weapons included guns, knives, golf clubs, machetes, and tasers. Student participants offered three categories of interventions: Counseling, Improved Campus Security, and Educational Programs.     Genetic Ablation of CD68 Results in Mice with Increased Bone and Dysfunctional Osteoclasts

    CD68 is a member of the lysosome associated membrane protein (LAMP) family that is restricted in its expression to cells of the monocyte/macrophage lineage. This lineage restriction includes osteoclasts, and, while previous studies of CD68 in macrophages and dendritic cells have proposed roles in lipid metabolism, phagocytosis, and antigen presentation, the expression and function of CD68 in osteoclasts have not been explored. In this study, we investigated the expression and localization of CD68 in macrophages and osteoclasts in response to the monocyte/macrophage-colony stimulating factor (M-CSF) and the receptor activator of NF-ÎșB ligand (RANKL). We found that M-CSF stimulates CD68 expression and RANKL alters the apparent molecular weight of CD68 as measured by Western immunoblotting. In addition, we explored the significance of CD68 expression in osteoclasts by generating mice that lack expression of CD68. These mice have increased trabecular bone, and in vitro assessment of CD68−/− osteoclasts revealed that, in the absence of CD68, osteoclasts demonstrate an accumulation of intracellular vesicle-like structures, and do not efficiently resorb bone. These findings demonstrate a role for CD68 in the function of osteoclasts, and future studies will determine the mechanistic nature of the defects seen in CD68−/− osteoclasts

    Optimising medication data collection in a large-scale clinical trial

Objective: Pharmaceuticals play an important role in clinical care. However, in community-based research, medication data are commonly collected as unstructured free-text, which is prohibitively expensive to code for large-scale studies. The ASPirin in Reducing Events in the Elderly (ASPREE) study developed a two-pronged framework to collect structured medication data for 19,114 individuals. ASPREE provides an opportunity to determine whether medication data can be cost-effectively collected and coded, en masse from the community using this framework. Methods: The ASPREE framework of type-to-search box with automated coding and linked free text entry was compared to traditional method of free-text only collection and post hoc coding. Reported medications were classified according to their method of collection and analysed by Anatomical Therapeutic Chemical (ATC) group. Relative cost of collecting medications was determined by calculating the time required for database set up and medication coding. Results Overall, 122,910 participant structured medication reports were entered using the type-tosearch box and 5,983 were entered as free-text. Free-text data contributed 211 unique medications not present in the type-to-search box. Spelling errors and unnecessary provision of additional information were among the top reasons why medications were reported as freetext. The cost per medication using the ASPREE method was approximately USD 0.03comparedwithUSD0.03 compared with USD 0.20 per medication for the traditional method. Conclusion Implementation of this two-pronged framework is a cost-effective alternative to free-text only data collection in community-based research. Higher initial set-up costs of this combined method are justified by long term cost effectiveness and the scientific potential for analysis and discovery gained through collection of detailed, structured medication data

    Marion County Heritage Map Project

    With the soft opening of Pasaquan, Marion County’s visionary art environment, in summer 2016, and its grand opening by Columbus State University in fall 2016, the Marion County Chamber of Commerce requested that the Columbus Community Geography Center partner to develop a heritage tour map of the county for welcome centers across the state. This small rural county of 8,700 residences, 60% white, 30% black and 7.5% Hispanic. One fifth of the population live below the poverty level. It recently lost several hundred jobs with the closure of the local chicken processing plant. Plans to launch a tourism program is understood to be of great importance to the community. CSU’s Dr. Amanda Rees, Professor of Geography, and Professor Chuck Lawson, Department of Art, College of the Arts joined forces to create a heritage tour map of Buena Vista and Marion County. Geography students ran a community workshop to identify twenty-one county and city heritage sites for inclusion. They researched and wrote short descriptions for the map and extended histories for an accompanying web page to be accessed from the map with a QR code. Students also produced an accurate map of each site and the major roads and other primary physical features of the county and city. Graphic design students then received the map text and GIS maps of the county and the city. Students designed three “roughs” of the map for external review. The first review included Marion County leaders, state tourism representatives, and several faculty in art, GIS and geography. The roughs were then refined and presented again to a group of reviewers. This project proved to be a good fit for CSU’s QEP “Real World Problems Solving” project in its testing phase in spring 2016. This interdisciplinary “service learning” project offered high impact educational practices, fieldwork, student-led heritage workshop in Marion County, critical feedback from community members on writing, and design. This interdisciplinary project was aligned with CSU’s mission to support alternative pedagogical approaches to address the needs of millennial learners

    Queer intellectual curiosity as international relations method: developing queer international relations theoretical and methodological frameworks

    This article outlines two theoretical and methodological approaches that take a queer intellectual curiosity about figurations of “homosexuality” and “the homosexual” as their core. These offer ways to conduct international relations research on “the homosexual” and on international-relations figurations more broadly, e.g. from “the woman” to “the human rights holder.” The first approach provides a method for analyzing figurations of “the homosexual” and sexualized orders of international relations that are inscribed in IR as either normal or perverse. The second approach offers instructions on how to read plural figures and plural logics that signify as normal and/or perverse (and which might be described as queer). Together, they propose techniques, devices and research questions to investigate singular and plural IR figurations – including but not exclusively those of “the homosexual” – that map international phenomena as diverse as colonialism, human rights, and the formation of states and international communities in ways that exceed IR survey research techniques that, for example, incorporate “the homosexual” into IR research through a “sexuality variable.

    Characterization of simple sequence repeats (SSRs) from Phlebotomus papatasi (Diptera: Psychodidae) expressed sequence tags (ESTs)

    <p>Abstract</p> <p>Background</p> <p><it>Phlebotomus papatasi </it>is a natural vector of <it>Leishmania major</it>, which causes cutaneous leishmaniasis in many countries. Simple sequence repeats (SSRs), or microsatellites, are common in eukaryotic genomes and are short, repeated nucleotide sequence elements arrayed in tandem and flanked by non-repetitive regions. The enrichment methods used previously for finding new microsatellite loci in sand flies remain laborious and time consuming; <it>in silico </it>mining, which includes retrieval and screening of microsatellites from large amounts of sequence data from sequence data bases using microsatellite search tools can yield many new candidate markers.</p> <p>Results</p> <p>Simple sequence repeats (SSRs) were characterized in <it>P. papatasi </it>expressed sequence tags (ESTs) derived from a public database, National Center for Biotechnology Information (NCBI). A total of 42,784 sequences were mined, and 1,499 SSRs were identified with a frequency of 3.5% and an average density of 15.55 kb per SSR. Dinucleotide motifs were the most common SSRs, accounting for 67% followed by tri-, tetra-, and penta-nucleotide repeats, accounting for 31.1%, 1.5%, and 0.1%, respectively. The length of microsatellites varied from 5 to 16 repeats. Dinucleotide types; AG and CT have the highest frequency. Dinucleotide SSR-ESTs are relatively biased toward an excess of (AX)n repeats and a low GC base content. Forty primer pairs were designed based on motif lengths for further experimental validation.</p> <p>Conclusion</p> <p>The first large-scale survey of SSRs derived from <it>P. papatasi </it>is presented; dinucleotide SSRs identified are more frequent than other types. EST data mining is an effective strategy to identify functional microsatellites in <it>P. papatasi</it>.</p
