    The challenges of measuring spin Seebeck noise

    Just as electronic shot noise results from the granularity of charge and the statistical variation in the arrival times of carriers in driven conductors, there are predictions for fundamental noise in magnon currents due to angular momentum being carried by discrete excitations. The advent of the inverse spin Hall effect as a transduction mechanism to convert spin current into charge current raises the prospect of experimental investigations of such magnon shot noise. Spin Seebeck effect measurements have demonstrated the electrical detection of thermally driven magnon currents and have been suggested as an avenue for accessing spin current fluctuations. We report measurements of spin Seebeck structures made from yttrium iron garnet on gadolinium gallium garnet. While these measurements do show an increase in measured noise in the presence of a magnetic field at low temperatures, the dependence on field orientation suggests an alternative origin for this signal. We describe theoretical predictions for the expected magnitude of magnon shot noise, highlighting ambiguities that exist. Analysis in terms of the sample geometry dependence of the known inverse spin Hall transduction of spin currents into charge currents implies that magnon shot noise detection through this approach is strongly suppressed.     Exposing the Causal Effect of Body Mass Index on the Risk of Type 2 Diabetes Mellitus: A Mendelian Randomization Study

    Introduction: High body mass index (BMI) is a positive associated phenotype of type 2 diabetes mellitus (T2DM). Abundant studies have observed this from a clinical perspective. Since the rapid increase in a large number of genetic variants from the genome-wide association studies (GWAS), common SNPs of BMI and T2DM were identified as the genetic basis for understanding their associations. Currently, their causality is beginning to blur.Materials and Methods: To classify it, a Mendelian randomisation (MR), using genetic instrumental variables (IVs) to explore the causality of intermediate phenotype and disease, was utilized here to test the effect of BMI on the risk of T2DM. In this article, MR was carried out on GWAS data using 52 independent BMI SNPs as IVs. The pooled odds ratio (OR) of these SNPs was calculated using inverse-variance weighted method for the assessment of 5 kg/m2 higher BMI on the risk of T2DM. The leave-one-out validation was conducted to identify the effect of individual SNPs. MR-Egger regression was utilized to detect potential pleiotropic bias of variants.Results: We obtained the high OR (1.470; 95% CI 1.170 to 1.847; P = 0.001), low intercept (0.004, P = 0.661), and small fluctuation of ORs {from -0.039 [(1.412 – 1.470) / 1.470)] to 0.075 [(1.568– 1.470) / 1.470)] in leave-one-out validation.Conclusion: We validate the causal effect of high BMI on the risk of T2DM. The low intercept shows no pleiotropic bias of IVs. The small alterations of ORs activated by removing individual SNPs showed no single SNP drives our estimate

    ISP1-anchored Polarization of GCβ/CDC50A Complex Initiates Malaria Ookinete Gliding Motility

    该工作历时四年,由博士毕业生高涵(第一作者)、博士生杨振科和王旭(共同第一作者)、硕士生钱鹏戈和洪仁杰完成;袁晶教授为通讯作者;厦门大学为第一完成单位。 该工作揭示了疟疾病原疟原虫通过媒介按蚊传播过程中的关键步骤---控制动合子运动的环磷酸鸟苷cGMP信号的激活机制。 疟原虫属于顶复体亚门原生动物,每年导致数亿人口感染疟疾和超过40万病人死亡。疟原虫通过雌性按蚊在哺乳动物宿主间传播。疟疾病人被按蚊叮咬吸血,疟原虫雌雄配子在按蚊消化道中受精形成合子,进一步变形发育为具有运动能力的新月形动合子。只有获得运动能力的动合子,才能穿越按蚊消化道单层上皮细胞,成功感染按蚊媒介。在本研究中,通过大量基因修饰模型,发现GCβ缺失导致动合子运动完全丢失,进而失去按蚊感染和传播能力。研究还发现,GCβ由细胞质均匀分布改变为在动合子顶体突出一侧聚集,并且GCβ聚集和动合子成熟完全同步,显示GCβ聚集可能直接激活cGMP信号。 本工作还发现GCβ结合蛋白CDC50A,后者承担分子伴侣功能,在动合子转化和成熟动合子中,稳定GCβ。此外,进一步筛选发现动合子内膜复合物蛋白ISP1,能够结合和锚定GCβ/CDC50A复合物,在成熟动合子中维持复合物的聚集。本研究为深入开展寄生性原生动物的发育转化和信号调控提供了线索和借鉴。【Abstract】Ookinete gliding motility is essential for penetration of the mosquito midgut wall and transmission of malaria parasites. Cyclic guanosine monophosphate (cGMP) signaling has been implicated in ookinete gliding. However, the upstream mechanism of how the parasites activate cGMP signaling and thus initiate ookinete gliding remains unknown. Using real-time imaging to visualize Plasmodium yoelii guanylate cyclase β (GCβ), we show that cytoplasmic GCβ translocates and polarizes to the parasite plasma membrane at “ookinete extrados site” (OES) during zygote to ookinete differentiation. The polarization of enzymatic active GCβ at OES initiates gliding of matured ookinete. Both the P4-ATPase-like domain and guanylate cyclase domain are required for GCβ polarization and ookinete gliding. CDC50A, a co-factor of P4-ATPase, binds to and stabilizes GCβ during ookinete development. Screening of inner membrane complex proteins identifies ISP1 as a key molecule that anchors GCβ/CDC50A complex at the OES of mature ookinetes. This study defines a spatial-temporal mechanism for the initiation of ookinete gliding, where GCβ polarization likely elevates local cGMP levels and activates cGMP-dependent protein kinase signaling.We thank Dr. David Baker (London School of Hygiene and Tropical Medicine) for his comments on this manuscript.     Postpartum depression in mothers and fathers: a structural equation model

    open access articleBackground Post-partum depression (PPD) is a growing mental health concern worldwide. There is little evidence in the Chinese context of the relationship between paternal PPD and maternal PPD. Given the growing global concerns this relationship requires further exploration. Methods A survey was conducted with 950 total couples from March 2017 to December 2018. The study was conducted using a standardized questionnaire that included basic demographic information, information on the relationship between the mother-in-law and daughter-in-law, marital satisfaction (both maternal and paternal), and PPD symptoms. Structural Equation Modelling (SEM) analysis was used to explore the underlying mechanism for PPD symptoms in mothers and fathers. Results In 4.4% of the couples both the wife and the husband showed depressive symptoms. Maternal marital satisfaction showed a significant mediating effect on paternal PPD (B = -0.114, p < 0.01), and there was a direct effect of maternal PPD on paternal PPD (B = 0.31, p < 0.001). Conclusions This is the first study to investigate the possible correlation between maternal PPD, mother-in-law and daughter-in-law relationship satisfaction, maternal marital satisfaction, paternal marital satisfaction, and paternal PPD. It is important for future PPD interventions to target both maternal and paternal mental health, as well as the mechanisms identified that can lead to PPD

    Enrichment and characteristics of ammonia-oxidizing archaea in wastewater treatment process

    High purity ammonia-oxidizing archaea (AOA) culture containing a single AOA strain was enriched from the filtering materials of biological aerated filter. The concentration of AOA reached 3.27\ua0×\ua010\ua0copies/mL, while its proportion was 91.40%. The AOA amoA gene sequence belonged to Nitrososphaera cluster. Ammonia concentration significantly influenced the growth of AOA in culture, while total organic carbon (TOC) concentration had no obvious effect. The optimum ammonia concentration, temperature, pH and DO concentration for growth of AOA were 1\ua0mM, 30\ua0°C, 7.5 and 2.65\ua0mg/L, respectively. Under the optimum growth conditions, the AOA abundance and ammonia oxidation rate were 3.53\ua0×\ua010\ua0copies/mL and 2.54\ua0×\ua010\ua0mg/(copies·d)

    Enhancement of stress tolerance in transgenic tobacco plants constitutively expressing AtIpk2β, an inositol polyphosphate 6-/3-kinase from Arabidopsis thaliana

    Inositol phosphates (IPs) and their turnover products have been implicated to play important roles in stress signaling in eukaryotic cells. In higher plants genes encoding inositol polyphosphate kinases have been identified previously, but their physiological functions have not been fully resolved. Here we expressed Arabidopsis inositol polyphosphate 6-/3-kinase (AtIpk2β) in two heterologous systems, i.e. the yeast Saccharomycescerevisiae and in tobacco (Nicotiana tabacum), and tested the effect on abiotic stress tolerance. Expression of AtIpk2β rescued the salt-, osmotic- and temperature-sensitive growth defects of a yeast mutant strain (arg82Δ) that lacks inositol polyphosphate multikinase activity encoded by the ARG82/IPK2 gene. Transgenic tobacco plants constitutively expressing AtIpk2β under the control of the Cauliflower Mosaic Virus 35S promoter were generated and found to exhibit improved tolerance to diverse abiotic stresses when compared to wild type plants. Expression patterns of various stress responsive genes were enhanced, and the activities of anti-oxidative enzymes were elevated in transgenic plants, suggesting a possible involvement of AtIpk2β in plant stress responses

    Search for dark matter produced in association with bottom or top quarks in √s = 13 TeV pp collisions with the ATLAS detector

    A search for weakly interacting massive particle dark matter produced in association with bottom or top quarks is presented. Final states containing third-generation quarks and miss- ing transverse momentum are considered. The analysis uses 36.1 fb−1 of proton–proton collision data recorded by the ATLAS experiment at √s = 13 TeV in 2015 and 2016. No significant excess of events above the estimated backgrounds is observed. The results are in- terpreted in the framework of simplified models of spin-0 dark-matter mediators. For colour- neutral spin-0 mediators produced in association with top quarks and decaying into a pair of dark-matter particles, mediator masses below 50 GeV are excluded assuming a dark-matter candidate mass of 1 GeV and unitary couplings. For scalar and pseudoscalar mediators produced in association with bottom quarks, the search sets limits on the production cross- section of 300 times the predicted rate for mediators with masses between 10 and 50 GeV and assuming a dark-matter mass of 1 GeV and unitary coupling. Constraints on colour- charged scalar simplified models are also presented. Assuming a dark-matter particle mass of 35 GeV, mediator particles with mass below 1.1 TeV are excluded for couplings yielding a dark-matter relic density consistent with measurements

    Numerical Investigation on Hydrodynamic Performance of a Portable AUV

    This paper conducts a numerical investigation on the hydrodynamic performance of a portable autonomous underwater vehicle (AUV). The portable AUV is designed to cruise and perform some tasks autonomously in the underwater world. However, its dynamic performance is strongly affected by hydrodynamic effects. Therefore, it is crucial to investigate the hydrodynamic performance of the portable AUV for its accurate dynamic modeling and control. In this work, based on the designed portable AUV, a comprehensive hydrodynamic performance investigation was conducted by adopting the computational fluid dynamics (CFD) method. Firstly, the mechanical structure of the portable AUV was briefly introduced, and the dynamic model of the AUV, including the hydrodynamic term, was established. Then, the unknown hydrodynamic coefficients in the dynamic model were estimated through the towing experiment and the plane-motion-mechanism (PMM) experiment simulation. In addition, considering that the portable AUV was affected by wave forces when cruising near the water surface, the influence of surface waves on the hydrodynamic performance of the AUV under different wave conditions and submerged depths was analyzed. Finally, the effectiveness of our method was verified by experiments on the standard models, and a physical experiment platform was built in this work to facilitate hydrodynamic performance investigations of some portable small-size AUVs
