    The Effect of BMI on Oxygen Saturation at Rest and During Mild Walking

    Eighty one subjects were examined for the relationship between BMI, body fat, arterial oxygen saturation and arterial PO2 with the subjects at rest and after 5 minutes of walking on a treadmill ergometer at 3 mph at a 3% grade. They had BMIs between 19 and 50. All subjects were free of cardiovascular disease and had normal blood pressure making it safe for them to participate in mild exercise. They were all able to walk for at least 10 minutes without cardiovascular complications and were not taking any medications that altered the autonomic nervous system. The age was between 22 and 68. The results showed that above a BMI of about 30, there was an inverse relationship between BMI and oxygen saturation in fingertip blood (correlation -0.81.

    Loss of murine Paneth cell function alters the immature intestinal microbiome and mimics changes seen in neonatal necrotizing enterocolitis

    Necrotizing enterocolitis (NEC) remains the leading cause of gastrointestinal morbidity and mortality in premature infants. Human and animal studies suggest a role for Paneth cells in NEC pathogenesis. Paneth cells play critical roles in host-microbial interactions and epithelial homeostasis. The ramifications of eliminating Paneth cell function on the immature host-microbial axis remains incomplete. Paneth cell function was depleted in the immature murine intestine using chemical and genetic models, which resulted in intestinal injury consistent with NEC. Paneth cell depletion was confirmed using histology, electron microscopy, flow cytometry, and real time RT-PCR. Cecal samples were analyzed at various time points to determine the effects of Paneth cell depletion with and without Klebsiella gavage on the microbiome. Deficient Paneth cell function induced significant compositional changes in the cecal microbiome with a significant increase in Enterobacteriacae species. Further, the bloom of Enterobacteriaceae species that occurs is phenotypically similar to what is seen in human NEC. This further strengthens our understanding of the importance of Paneth cells to intestinal homeostasis in the immature intestine

    Hope in dirt: report of the Fort Apache Workshop on Forensic Sedimentology Applications to Cultural Property Crime, 15—19 October 2018

    A 2018 workshop on the White Mountain Apache Tribe lands in Arizona examined ways to enhance investigations into cultural property crime (CPC) through applications of rapidly evolving methods from archaeological science. CPC (also looting, graverobbing) refers to unauthorized damage, removal, or trafficking in materials possessing blends of communal, aesthetic, and scientific values. The Fort Apache workshop integrated four generally partitioned domains of CPC expertise: (1) theories of perpetrators’ motivations and methods; (2) recommended practice in sustaining public and community opposition to CPC; (3) tactics and strategies for documenting, investigating, and prosecuting CPC; and (4) forensic sedimentology—uses of biophysical sciences to link sediments from implicated persons and objects to crime scenes. Forensic sedimentology served as the touchstone for dialogues among experts in criminology, archaeological sciences, law enforcement, and heritage stewardship. Field visits to CPC crime scenes and workshop deliberations identified pathways toward integrating CPC theory and practice with forensic sedimentology’s potent battery of analytic methods

We show how tangle analysis can be extended to model any stable protein-DNA complex. -- Results: We discuss a computational method for finding the topological conformation of DNA bound within a protein complex. We use an elementary invariant from knot theory called colorability to encode and search for possible DNA conformations. We apply this method to analyze the experimental results of Pathania, Jayaram, and Harshey (Cell 2002). We show that the only topological DNA conformation bound by Mu transposase which is biologically likely is the five crossing solution found by Pathania et al (although other possibilities are discussed). -- Conclusion: Our algorithm can be used to analyze the results of the experimental technique described in Pathania et al in order to determine the topological conformation of DNA bound within a stable protein-DNA [email protected]

    Evaluating the influence of candidate terrestrial protected areas on coral reef condition in Fiji

    In any given region, there are multiple options for terrestrial protected area networks that achieve goals for conservation of terrestrial biodiversity and ecosystem values. When deciding on the location of terrestrial protected areas, planners typically focus only on terrestrial conservation goals, ignoring potential linked benefits to marine ecosystems. These benefits include maintenance of downstream water quality, as forest protection can prevent changes in amount and composition of river runoff that negatively impacts coral reefs. This study aims to determine the benefit of different terrestrial reserve networks to the condition of coral reefs adjacent to the main islands of Fiji to support the work of Fiji's Protected Area Committee in expanding the national protected area estate through integrated land-sea planning. Options for terrestrial protected area networks were designed using six approaches, where the primary objective of each approach was to either achieve terrestrial conservation goals (e.g., represent 40% of each vegetation type) or maximize benefits to coral reefs by minimizing potential for land-based runoff. When achieving terrestrial conservation goals was the primary objective, the potential benefits to coral reef condition were 7.7-10.4% greater than benefits from the existing network of protected areas. When benefiting reefs was the primary objective, benefits to coral reefs were 1.1-2.8 times greater per unit area than networks designed to only achieve terrestrial conservation goals, but 31-44% of the terrestrial conservation goals were not achieved. These results are already being used by Fiji's Protected Area Committee to modify the boundaries of existing priority places to deliver outcomes that better meet terrestrial conservation goals while offering greater benefits to coral reef condition through prevention of run-off

    The Fisheries Co-Management Guidebook: Emerging research for the effective management of small-scale fisheries

    Small-scale fisheries account for 40% of global fish catch and employ more than 90% of the world’s fishers, defining the livelihoods, nutrition, and culture of a substantial and diverse segment of humankind. In recent decades collaborative forms of fisheries management, including co-management, have gained popularity as the most appropriate, fair, and effective form of governance for small-scale fisheries. Fisheries co-management is envisioned as a process by which to reverse the interconnected crises of hunger, poverty, and biodiversity loss, transforming small-scale fisheries into engines of prosperity, inclusion, and sustainability. Yet co-management can succeed or fail, and implementation does not mean positive impacts for food security, nutrition, livelihoods, or biodiversity. Nor does it imply programs will respect human rights, gender equality, or principles of justice and equity. Fewer management programs implemented well might achieve far more than many implemented poorly, and poorly implemented co-management can be worse than no management. This guide was designed to assist practitioners in understanding the latest research on what constitutes successful fisheries co-management, and how to reach this objective. The aim is to synthesize emerging research that, if adopted, would substantially improve impacts across both ecological and social dimensions. The guide is presented as an infographic series with each infographic summarizing a substantial body of research from a particular field. This work was undertaken through a growing partnership between the Wildlife Conservation Society and WorldFish, aiming to increase collaboration between conservation and development sectors. This partnership represents a milestone towards integrated approaches for the benefit of both ecosystems and local communities

    Sustained proliferation in cancer: mechanisms and novel therapeutic targets

    Proliferation is an important part of cancer development and progression. This is manifest by altered expression and/or activity of cell cycle related proteins. Constitutive activation of many signal transduction pathways also stimulates cell growth. Early steps in tumor development are associated with a fibrogenic response and the development of a hypoxic environment which favors the survival and proliferation of cancer stem cells. Part of the survival strategy of cancer stem cells may manifested by alterations in cell metabolism. Once tumors appear, growth and metastasis may be supported by overproduction of appropriate hormones (in hormonally dependent cancers), by promoting angiogenesis, by undergoing epithelial to mesenchymal transition, by triggering autophagy, and by taking cues from surrounding stromal cells. A number of natural compounds (e.g., curcumin, resveratrol, indole-3-carbinol, brassinin, sulforaphane, epigallocatechin-3-gallate, genistein, ellagitannins, lycopene and quercetin) have been found to inhibit one or more pathways that contribute to proliferation (e.g., hypoxia inducible factor 1, nuclear factor kappa B, phosphoinositide 3 kinase/Akt, insulin-like growth factor receptor 1, Wnt, cell cycle associated proteins, as well as androgen and estrogen receptor signaling). These data, in combination with bioinformatics analyses, will be very important for identifying signaling pathways and molecular targets that may provide early diagnostic markers and/or critical targets for the development of new drugs or drug combinations that block tumor formation and progression

    Improving social acceptability of marine protected area networks: A method for estimating opportunity costs to multiple gear types in both fished and currently unfished areas

    We present a novel method for calculating the opportunity costs to fishers from their displacement by the establishment of marine protected areas (MPAs). We used a fishing community in Kubulau District, Fiji to demonstrate this method. We modelled opportunity costs as a function of food fish abundance and probability of catch, based on gear type and market value of species. Count models (including Poisson, negative binomial and two zero-inflated models) were used to predict spatial abundance of preferred target fish species and were validated against field surveys. A profit model was used to investigate the effect of restricted access to transport on costs to fishers. Spatial distributions of fish within the three most frequently sighted food fish families (Acanthuridae, Lutjanidae, Scaridae) varied, with greatest densities of Lutjanidae and Acanthuridae on barrier forereefs and greatest densities of Scaridae on submerged reefs. Modelled opportunity cost indicated that highest costs to fishers arise from restricting access to the barrier forereefs. We included our opportunity cost model in Marxan, a decision support tool used for MPA design, to examine potential MPA configurations for Kubulau District, Fiji Islands. We identified optimum areas for protection in Kubulau with: (a) the current MPA network locked in place; and (b) a clean-slate approach. Our method of modelling opportunity cost gives an unbiased estimate for multiple gear types in a marine environment and can be applied to other regions using existing species data.
