    Synthèse biocatalytique de macrocycles planaires chiraux

    Les macrocycles représentent une catégorie chimique unique en chimie organique et ils possèdent des applications dans les industries pharmaceutique et agrochimique, en parfumerie, et dans les matériaux. Une propriété importante des macrocycles est la possibilité de démontrer de la chiralité planaire menant à des atropoisomères distincts aux propriétés uniques. Très peu de techniques générales existent pour le contrôle de l’atropoisomérisme au sein des macrocycles, rendant leur synthèse un véritable défi. Nous avons accompli le premier exemple d’une synthèse biocatalytique énantio- et atroposélective de p-cyclophanes planaires chiraux. En utilisant une lipase immobilisée commercialement disponible (CALB) et des matériaux de départ pro-chiraux simples, nous avons été en mesure de générer 23 différentes structures avec des rendements entre 11 et 88 %. Des analyses SFC ont permis l’évaluation de l’énantioenrichissement des différents macrocycles, étant compris entre 96 et >99 % ee. Surtout, les macrocycles planaires chiraux ayant des substituants de type halogène ou borylé peuvent subir de la diversification moléculaire au-delà des limites tolérées par l’enzyme. Notre découverte ouvre la porte à l’utilisation de biocatalyseurs pour le contrôle de l’atropoisomérisme lors de la formation de structures macrocycliques.Macrocycles represent a unique chemotype in organic chemistry, with applications ranging from pharmaceuticals, agrochemicals, aromachemicals and material science. An important property of macrocycles is the possibility of displaying planar chirality yielding distinct atropisomeric structures with unique properties. Very few generalized techniques capable of controlling atropisomerism in macrocycles exist, rendering their synthesis extremely challenging. We have achieved the first example of enantio- and atroposelective biocatalytic synthesis of planar chiral p-cyclophanes. Employing a commercially available immobilized lipase (CALB) and simple pro-chiral starting materials, we were able to generate 23 different structures with yields ranging from 11 to 88 %. SFC analysis permitted evaluation of the enantioenrichment of the different macrocycles, which ranged from 96 to >99 % ee. Importantly, planar chiral macrocycles having halogen or borylated substituents are capable of molecular diversification outside the boundaries of what may be tolerated by the enzyme.     Characterizing Geo-located Tweets in Brazilian Megacities

    This work presents a framework for collecting, processing and mining geo-located tweets in order to extract meaningful and actionable knowledge in the context of smart cities. We collected and characterized more than 9M tweets from the two biggest cities in Brazil, Rio de Janeiro and S\~ao Paulo. We performed topic modeling using the Latent Dirichlet Allocation model to produce an unsupervised distribution of semantic topics over the stream of geo-located tweets as well as a distribution of words over those topics. We manually labeled and aggregated similar topics obtaining a total of 29 different topics across both cities. Results showed similarities in the majority of topics for both cities, reflecting similar interests and concerns among the population of Rio de Janeiro and S\~ao Paulo. Nevertheless, some specific topics are more predominant in one of the cities

    Illicit and Prescription Drug Misuse among Sexual Minority Women in the United States: A Protocol for a Scoping Review

      Background: The United States Institute of Medicine (IOM) published reports in 1999 and 2011 identifying drug use as a priority area for sexual minority health research, specifically focused on mechanisms contributing to drug use disparities and the development of tailored interventions. Limited research has prioritized sexual minority women (SMW) with the majority of substance use research among sexual minorities focused on adolescents and men who have sex with men. This scoping review will characterize the research literature related to illicit drug use and prescription drug misuse among SMW and sub-groups within the SMW population. Through this, we will identify: (1) specific substances used; (2) patterns of substance use; (3) risk and protective factors; (4) prevention interventions; and (5) drug treatment programs specific to SMW. Methods/Design: This review will include studies with empirical data of illicit or prescription drug misuse among sexual minority women. Peer-reviewed quantitative research conducted in the United States and published in English from 2011-Present will be included. We will search Medline, PsycINFO, and Web of Science databases for relevant articles. Two independent reviewers will screen abstracts and relevant full-text studies for eligibility. Data will be extracted from eligible articles and results will be presented in narrative and tabular form as appropriate. Discussion: This work will identify gaps of knowledge in the research pertaining to illicit or prescription drug misuse among sexual minority women since the 2011 US IOM report. As a result of this work, we will propose directions for future research to address identified gaps

Annals of Glaciology, 62(85–86), (2021): 340–352, https://doi.org/10.1017/aog.2021.10.The Subglacial Antarctic Lakes Scientific Access (SALSA) Project accessed Mercer Subglacial Lake using environmentally clean hot-water drilling to examine interactions among ice, water, sediment, rock, microbes and carbon reservoirs within the lake water column and underlying sediments. A ~0.4 m diameter borehole was melted through 1087 m of ice and maintained over ~10 days, allowing observation of ice properties and collection of water and sediment with various tools. Over this period, SALSA collected: 60 L of lake water and 10 L of deep borehole water; microbes >0.2 μm in diameter from in situ filtration of ~100 L of lake water; 10 multicores 0.32–0.49 m long; 1.0 and 1.76 m long gravity cores; three conductivity–temperature–depth profiles of borehole and lake water; five discrete depth current meter measurements in the lake and images of ice, the lake water–ice interface and lake sediments. Temperature and conductivity data showed the hydrodynamic character of water mixing between the borehole and lake after entry.     Effective DNA/RNA Co-Extraction for Analysis of MicroRNAs, mRNAs, and Genomic DNA from Formalin-Fixed Paraffin-Embedded Specimens

    Background: Retrospective studies of archived human specimens, with known clinical follow-up, are used to identify predictive and prognostic molecular markers of disease. Due to biochemical differences, however, formalin-fixed paraffinembedded (FFPE) DNA and RNA have generally been extracted separately from either different tissue sections or from the same section by dividing the digested tissue. The former limits accurate correlation whilst the latter is impractical when utilizing rare or limited archived specimens. Principal Findings: For effective recovery of genomic DNA and total RNA from a single FFPE specimen, without splitting the proteinase-K digested tissue solution, we optimized a co-extraction method by using TRIzol and purifying DNA from the lower aqueous and RNA from the upper organic phases. Using a series of seven different archived specimens, we evaluated the total amounts of genomic DNA and total RNA recovered by our TRIzol-based co-extraction method and compared our results with those from two commercial kits, the Qiagen AllPrep DNA/RNA FFPE kit, for co-extraction, and the Ambion RecoverAll TM Total Nucleic Acid Isolation kit, for separate extraction of FFPE-DNA and-RNA. Then, to accurately assess the quality of DNA and RNA co-extracted from a single FFPE specimen, we used qRT-PCR, gene expression profiling and methylation assays to analyze microRNAs, mRNAs, and genomic DNA recovered from matched fresh and FFPE MCF10A cells.     Development of an amplicon-based sequencing approach in response to the global emergence of mpox

    The 2022 multicountry mpox outbreak concurrent with the ongoing Coronavirus Disease 2019 (COVID-19) pandemic further highlighted the need for genomic surveillance and rapid pathogen whole-genome sequencing. While metagenomic sequencing approaches have been used to sequence many of the early mpox infections, these methods are resource intensive and require samples with high viral DNA concentrations. Given the atypical clinical presentation of cases associated with the outbreak and uncertainty regarding viral load across both the course of infection and anatomical body sites, there was an urgent need for a more sensitive and broadly applicable sequencing approach. Highly multiplexed amplicon-based sequencing (PrimalSeq) was initially developed for sequencing of Zika virus, and later adapted as the main sequencing approach for Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2). Here, we used PrimalScheme to develop a primer scheme for human monkeypox virus that can be used with many sequencing and bioinformatics pipelines implemented in public health laboratories during the COVID-19 pandemic. We sequenced clinical specimens that tested presumptively positive for human monkeypox virus with amplicon-based and metagenomic sequencing approaches. We found notably higher genome coverage across the virus genome, with minimal amplicon drop-outs, in using the amplicon-based sequencing approach, particularly in higher PCR cycle threshold (Ct) (lower DNA titer) samples. Further testing demonstrated that Ct value correlated with the number of sequencing reads and influenced the percent genome coverage. To maximize genome coverage when resources are limited, we recommend selecting samples with a PCR Ct below 31 Ct and generating 1 million sequencing reads per sample. To support national and international public health genomic surveillance efforts, we sent out primer pool aliquots to 10 laboratories across the United States, United Kingdom, Brazil, and Portugal. These public health laboratories successfully implemented the human monkeypox virus primer scheme in various amplicon sequencing workflows and with different sample types across a range of Ct values. Thus, we show that amplicon-based sequencing can provide a rapidly deployable, cost-effective, and flexible approach to pathogen whole-genome sequencing in response to newly emerging pathogens.     Search for dark matter produced in association with bottom or top quarks in √s = 13 TeV pp collisions with the ATLAS detector

    A search for weakly interacting massive particle dark matter produced in association with bottom or top quarks is presented. Final states containing third-generation quarks and miss- ing transverse momentum are considered. The analysis uses 36.1 fb−1 of proton–proton collision data recorded by the ATLAS experiment at √s = 13 TeV in 2015 and 2016. No significant excess of events above the estimated backgrounds is observed. The results are in- terpreted in the framework of simplified models of spin-0 dark-matter mediators. For colour- neutral spin-0 mediators produced in association with top quarks and decaying into a pair of dark-matter particles, mediator masses below 50 GeV are excluded assuming a dark-matter candidate mass of 1 GeV and unitary couplings. For scalar and pseudoscalar mediators produced in association with bottom quarks, the search sets limits on the production cross- section of 300 times the predicted rate for mediators with masses between 10 and 50 GeV and assuming a dark-matter mass of 1 GeV and unitary coupling. Constraints on colour- charged scalar simplified models are also presented. Assuming a dark-matter particle mass of 35 GeV, mediator particles with mass below 1.1 TeV are excluded for couplings yielding a dark-matter relic density consistent with measurements
