    Understanding and Identifying Cognitive Load in Networked Learning

    This paper considers cognitive load theory (CLT) in the context of networked learning (NL).  It aligns with NL practitioners' efforts to understand and eliminate barriers to learning in NL situations.  The ideas presented are based on the premise that by recognising and either minimising or eliminating instances of unnecessary cognitive load in NL situations educators can improve learners’ abilities to acquire and develop schema and, in doing so, educators can support learning in NL situations.  The presentation brings together current thinking in cognitive load theory and descriptions of key aspects of NL to identify and describe of potential instances of cognitive load experienced by networked learners.The paper is structured in three main sections: The first section provides the background to our exploration of CLT in the context of NL.  It includes an overview of CLT and its development; an overview of NL; and a definition of the problem this paper seeks to address, namely, that NL situations include a number of instances of cognitive load which may not be present in other (e.g., face-to-face; on-campus) learning situations.  The second section explores common features of NL and identifies potential sources of cognitive load in NL situations.  It is organised according to key features of the 'architecture' of NL:  the learning environment; learning tasks and learner activity.  By identifying potential instances of cognitive load, the presentation provides a basis for, firstly, understanding cognitive load in NL; and, secondly, addressing it. Key sources of cognitive load referenced in this paper include the presentation of information in NL situations; the use of mediating technologies; the demands of managing information in connected environments; the load associated with technology-mediated social activity, including computer-mediated communication; the presentation of learning tasks; and the demands of 'learning to learn' in NL situations.  The third section of the paper identifies a potential research agenda to guide further explorations of CLT in NL including: research into technical aspects of NL to improve the presentation of information and computer interfaces; research into the use of instructional design techniques sympathetic to CLT and specifically targeting NL and engagement tasks; research to understand learning to learn online in NL from a CLT perspective

    Slowmation: An Innovative Twenty-First Century Teaching and Learning Tool for Science and Mathematics Pre-service Teachers

    Slowmation is a twenty-first century digital literacy educational tool. This teaching and learning tool has been incorporated as an assessment strategy in the curriculum area of science and mathematics with pre-service teachers (PSTs). This paper explores two themes: developing twenty-first century digital literacy skills and modelling best practice assessment tools. In the growing debate about the impact of multi-model representations, researchers such as Hoban and Nielsen, and Brown, Murcia and Hackling emphasise the development of conceptual understandings and semiotics. This paper focuses on PSTs’ experiences of and reflections on Slowmation as an educational tool. Data was collected from a cohort of final year PSTs who created, presented and reflected on their Slowmation process

    Re-annotation and re-analysis of the Campylobacter jejuni NCTC11168 genome sequence

    BACKGROUND: Campylobacter jejuni is the leading bacterial cause of human gastroenteritis in the developed world. To improve our understanding of this important human pathogen, the C. jejuni NCTC11168 genome was sequenced and published in 2000. The original annotation was a milestone in Campylobacter research, but is outdated. We now describe the complete re-annotation and re-analysis of the C. jejuni NCTC11168 genome using current database information, novel tools and annotation techniques not used during the original annotation. RESULTS: Re-annotation was carried out using sequence database searches such as FASTA, along with programs such as TMHMM for additional support. The re-annotation also utilises sequence data from additional Campylobacter strains and species not available during the original annotation. Re-annotation was accompanied by a full literature search that was incorporated into the updated EMBL file [EMBL: AL111168]. The C. jejuni NCTC11168 re-annotation reduced the total number of coding sequences from 1654 to 1643, of which 90.0% have additional information regarding the identification of new motifs and/or relevant literature. Re-annotation has led to 18.2% of coding sequence product functions being revised. CONCLUSIONS: Major updates were made to genes involved in the biosynthesis of important surface structures such as lipooligosaccharide, capsule and both O- and N-linked glycosylation. This re-annotation will be a key resource for Campylobacter research and will also provide a prototype for the re-annotation and re-interpretation of other bacterial genomes

    Hijacking bacterial glycosylation for the production of glycoconjugates, from vaccines to humanised glycoproteins.

    OBJECTIVES: Glycosylation or the modification of a cellular component with a carbohydrate moiety has been demonstrated in all three domains of life as a basic post-translational process important in a range of biological processes. This review will focus on the latest studies attempting to exploit bacterial N-linked protein glycosylation for glycobiotechnological applications including glycoconjugate vaccine and humanised glycoprotein production. The challenges that remain for these approaches to reach full biotechnological maturity will be discussed. KEY FINDINGS: Oligosaccharyltransferase-dependent N-linked glycosylation can be exploited to make glycoconjugate vaccines against bacterial pathogens. Few technical limitations remain, but it is likely that the technologies developed will soon be considered a cost-effective and flexible alternative to current chemical-based methods of vaccine production. Some highlights from current glycoconjugate vaccines developed using this in-vivo production system include a vaccine against Shigella dysenteriae O1 that has passed phase 1 clinical trials, a vaccine against the tier 1 pathogen Francisella tularensis that has shown efficacy in mice and a vaccine against Staphylococcus aureus serotypes 5 and 8. Generation of humanised glycoproteins within bacteria was considered impossible due to the distinct nature of glycan modification in eukaryotes and prokaryotes. We describe the method used to overcome this conundrum to allow engineering of a eukaryotic pentasaccharide core sugar modification within Escherichia coli. This core was assembled by combining the function of the initiating transferase WecA, several Alg genes from Saccharomyces cerevisiae and the oligosaccharyltransferase function of the Campylobacter jejuni PglB. Further exploitation of a cytoplasmic N-linked glycosylation system found in Actinobacillus pleuropneumoniae where the central enzyme is known as N-linking glycosyltransferase has overcome some of the limitations demonstrated by the oligosaccharyltransferase-dependent system. SUMMARY: Characterisation of the first bacterial N-linked glycosylation system in the human enteropathogen Campylobacter jejuni has led to substantial biotechnological applications. Alternative methods for glycoconjugate vaccine production have been developed using this N-linked system. Vaccines against both Gram-negative and Gram-positive organisms have been developed, and efficacy testing has thus far demonstrated that the vaccines are safe and that robust immune responses are being detected. These are likely to complement and reduce the cost of current technologies thus opening new avenues for glycoconjugate vaccines. These new markets could potentially include glycoconjugate vaccines tailored specifically for animal vaccination, which has until today thought to be non-viable due to the cost of current in-vitro chemical conjugation methods. Utilisation of N-linked glycosylation to generate humanised glycoproteins is also close to becoming reality. This 'bottom up' assembly mechanism removes the heterogeneity seen in current humanised products. The majority of developments reported in this review exploit a single N-linked glycosylation system from Campylobacter jejuni; however, alternative N-linked glycosylation systems have been discovered which should help to overcome current technical limitations and perhaps more systems remain to be discovered. The likelihood is that further glycosylation systems exist and are waiting to be exploited

    Comparative Genomic Analysis and In Vivo Modeling of Streptococcus pneumoniae ST3081 and ST618 Isolates Reveal Key Genetic and Phenotypic Differences Contributing to Clonal Replacement of Serotype 1 in The Gambia

    Streptococcus pneumoniae serotype 1 is one of the leading causes of invasive pneumococcal disease (IPD) in West Africa, with ST618 being the dominant cause of IPD in The Gambia. Recently however, a rare example of clonal replacement was observed, where the ST3081 clone of serotype 1 replaced the predominant ST618 clone as the main cause of IPD. In the current study, we sought to find the reasons for this unusual replacement event. Using whole-genome sequence analysis and clinically relevant models of in vivo infection, we identified distinct genetic and phenotypic characteristics of the emerging ST3081 clone. We show that ST3081 is significantly more virulent than ST618 in models of invasive pneumonia, and is carried at higher densities than ST618 during nasopharyngeal carriage. We also observe sequence type-specific accessory genes and a unique sequence type-specific fixed mutation in the pneumococcal toxin pneumolysin, which is associated with increased hemolytic activity in ST3081 and may contribute to increased virulence in this clone. Our study provides evidence that, within the same serotype 1 clonal complex, biological properties differ significantly from one clone to another in terms of virulence and host invasiveness, and that these differences may be the result of key genetic differences within the genome

    Holocene glacial activity in Barilari Bay, west Antarctic Peninsula, tracked by magnetic mineral assemblages: Linking ice, ocean, and atmosphere

    We investigate the origin and fate of lithogenic sediments using magnetic mineral assemblages in Barilari Bay, west Antarctic Peninsula (AP) from sediment cores recovered during the Larsen Ice Shelf System, Antarctica (LARISSA) NBP10-01 cruise. To quantify and reconstruct Holocene changes in covarying magnetic mineral assemblages, we adopt an unsupervised mathematical unmixing strategy and apply it to measurements of magnetic susceptibility as a function of increasing temperature. Comparisons of the unmixed end-members with magnetic observations of northwestern AP bedrock and the spatial distribution of magnetic mineral assemblages within the fjord, allow us to identify source regions, including signatures for ‘‘inner bay,’’ ‘‘outer bay,’’ and ‘‘northwestern AP’’ sources. We find strong evidence that supports the establishment of a late Holocene ice shelf in the fjord coeval with the Little Ice Age. Additionally, we present new evidence for late Holocene sensitivity to conditions akin to positive mean Southern Annual Mode states for western AP glaciers at their advanced Neoglacial positions

    Education for citizenship in South Australian public schools: a pilot study of senior leader and teacher perceptions

    Preparing students for informed and active citizenship is a core goal of education and schooling in Australia. The ways schools educate and prepare young Australians for citizenship involves a range of processes and initiatives central to the work of schools, including school ethos, mission, extra-curricular activities and community-based participation. With regard to the formal curriculum, the recent introduction and implementation of the first ever Federal Australian curriculum includes provision for a new subject – Civics and Citizenship. Research evidence from other nations suggests that schools understand, approach and enact education for citizenship in a multitude of ways, yet how Australian schools construct this aspect of their work is currently under-researched. In this context, and drawing on data from interviews with school leaders and teachers of year six-eight (11-14 year olds) students in a small sample of South Australian primary and secondary schools, we explore perceptions and current approaches to education for citizenship. Our findings suggest (i) that while school leaders and teachers value education for citizenship, they do so for different reasons; (ii) that schools place values as central to education for citizenship; and, (iii) that community involvement is typically understood as occurring within rather than beyond the school

    Genomic signatures of human and animal disease in the zoonotic pathogen Streptococcus suis

    Streptococcus suis causes disease in pigs worldwide and is increasingly implicated in zoonotic disease in East and South-East Asia. To understand the genetic basis of disease in S. suis, we study the genomes of 375 isolates with detailed clinical phenotypes from pigs and humans from the United Kingdom and Vietnam. Here, we show that isolates associated with disease contain substantially fewer genes than non-clinical isolates, but are more likely to encode virulence factors. Human disease isolates are limited to a single-virulent population, originating in the 1920 s when pig production was intensified, but no consistent genomic differences between pig and human isolates are observed. There is little geographical clustering of different S. suis subpopulations, and the bacterium undergoes high rates of recombination, implying that an increase in virulence anywhere in the world could have a global impact over a short timescale.Peer reviewe

    TOI-257b (HD 19916b): a warm sub-saturn orbiting an evolved F-type star

    ABSTRACT We report the discovery of a warm sub-Saturn, TOI-257b (HD 19916b), based on data from NASA’s Transiting Exoplanet Survey Satellite (TESS). The transit signal was detected by TESS and confirmed to be of planetary origin based on radial velocity observations. An analysis of the TESS photometry, the Minerva-Australis, FEROS, and HARPS radial velocities, and the asteroseismic data of the stellar oscillations reveals that TOI-257b has a mass of MP = 0.138 ± 0.023 MJ\rm {M_J} (43.9 ± 7.3 M\, M_{\rm \oplus}), a radius of RP = 0.639 ± 0.013 RJ\rm {R_J} (7.16 ± 0.15 R\, \mathrm{ R}_{\rm \oplus}), bulk density of 0.650.11+0.120.65^{+0.12}_{-0.11} (cgs), and period 18.388180.00084+0.0008518.38818^{+0.00085}_{-0.00084} days\rm {days}. TOI-257b orbits a bright (V = 7.612 mag) somewhat evolved late F-type star with M* = 1.390 ± 0.046 Msun\rm {M_{sun}}, R* = 1.888 ± 0.033 Rsun\rm {R_{sun}}, Teff = 6075 ± 90 K\rm {K}, and vsin i = 11.3 ± 0.5 km s−1. Additionally, we find hints for a second non-transiting sub-Saturn mass planet on a ∼71 day orbit using the radial velocity data. This system joins the ranks of a small number of exoplanet host stars (∼100) that have been characterized with asteroseismology. Warm sub-Saturns are rare in the known sample of exoplanets, and thus the discovery of TOI-257b is important in the context of future work studying the formation and migration history of similar planetary systems
