    Antibacterial potential of Urtica dioica and Lavandula angustifolia extracts against methicillin resistant Staphylococcus aureus isolated from diabetic foot ulcers

    Despite the antibacterial potential of plant extracts against several bacterial infections, until now, no major studies have been published about the effect of lavender and nettle leaves against methicillin-sensitive and methicillin-resistant strains of Staphylococcus aureus (MSSA and MRSA, respectively). Thus, the authors studied their antibacterial potential against MSSA and MRSA from diabetic foot ulcers in order to find alternatives to the systematic use of antibiotics. Phenolic acids, flavonoids, flavones and flavonols were extracted from lavender and nettle leaves and characterized by HPLC-DAD/Vis. Disc diffusion assay and minimum inhibitory concentration (MIC) were used to assess their antibacterial activity. A direct association between the high content of hydroxycinnamic acids (chlorogenic acid, caffeic acid, rosmarinic acid) and flavonoids (quercetin) and decreasing bacterial growth activity was noted.     Antagonistic activities of Kefirgel and Kefirgel products against Staphylococcus aureus isolated from diabetic ulcers

    Diabetic foot ulcers are often complicated by infection and among pathogens the Gram-positive Staphylococcus aureus is the most common isolated. Also concomitantly, the high prevalence of methicillin-resistant S. aureus (MRSA) was significant impact on successful treatment of infected foot ulcers. In this context, the purpose of the present study was to evaluate the antibacterial properties of Kefigel®, a natural product composed by nettle (Urtica dioica L), lavender (Lavandula angustifolia Mill) and kefir grains, which have been reported as to having antibacterial activity against several diseases. Here, antibacterial effect of Kefigel® and its components were investigated against 20 S. aureus isolates (10 MSSA and 10 MRSA) collected from several diabetic foot ulcers. To assess antibacterial activity, the disk diffusion assay method, minimum inhibitory concentration (MIC), minimal bacterial activity and effects on specific growth rate, were applied. Results showed that by diffusion method, only etanolic extracts of nettle and lavender showed antibacterial activity and their effects were mainly bacteriostatic. Compared to the antibiotic gentamicin, the nettle and lavender extracts showed an efficacy between 50 and 100% relative to the antibiotic. Generally, the MRSA isolates sowed higher inhibition halos comparing MSSA isolates. In turns, Kefigel® (40mg.mL-1) affects the specific growth rate of S. aureus isolates, since after incubation for 9h almost MSSA isolates growth were inhibited. A bactericide effect was observed only against one isolates MRSA. These findings indicate a potential use of Kefigel® as a natural product having an effective effect against Staphylococus aureus. The topical use of this product for prevention of diabetic foot ulcers infections can be useful, however further investigation will be made, namely tests with high Kefigel® concentrations and synergetic effects with antibiotics

    Initial in vitro evaluations of antibacterial activities of glucosinolate enzymatic hydrolysis products against plant pathogenic bacteria

    Aims: The aim of the study was to evaluate the in vitro antibacterial effects of glucosinolate hydrolysis products (GHP) against plant pathogenic micro-organisms namely Agrobacterium tumefaciens, Erwinia chrysanthemi, Pseudomonas cichorii, Pseudomonas tomato, Xanthomonas campestris and Xanthomonas juglandis. Methods and Results: Using a disc diffusion assay, seven different doses of 10 GHP were tested against each bacteria. The results showed that the isothiocyanates were potent antibacterials, whilst the other GHP were much less efficient. Moreover, the antibacterial effects were dose-dependent, increasing with the dose applied; 2-phenylethylisothiocyanate and sulforaphane showed the strongest inhibitory effects. The overall results show a great potential for using the isothiocyanates as an alternative tool to control undesired bacterial growth in plants. Conclusions: Glucosinolate hydrolysis products and more specifically the isothiocyanates: benzylisothiocyanate, 2-phenylethylisothiocyanate, the isothiocyanate Mix and sulforaphane, were effective phytochemicals against the in vitro growth of the phytopathogenic bacteria. The antibacterial activity exhibited by these phytochemicals reinforces their potential as alternatives to the traditional chemical control of phytopathogenic bacteria. Significance and Impact of the Study: This current in vitro study is the first providing comparative data on GHP as potential control agents for plant pathogenic bacteria.     Screening polymeric ionic liquids for chromatography-based purification of bacteriophage M13

    M13 bacteriophage is a key instrument in phage display applications, as well as a possible antibacterial therapeutic agent due to its highly restrictive bacterial pathogenesis, and other applications. The traditional phage purification process is usually achieved by gradient ultracentrifugation or a combination of precipitation, centrifugation and microfiltration. These approaches easily lead to long process times, high operational costs, phage aggregation and consequent product loss (approximately 60%). This work is thus focused on an alternative potential large-scale process to achieve high yield and purity while minimizing the operational costs. Electrostatic-based separation processes are also common biomolecules purification techniques. Although anion exchange chromatography has been used before to purify several viral particles, this technique has been poorly reported for the purification of M13 phage. In a recent work, our group has demonstrated the use of a predominant anion exchange process, where a polymeric ionic liquid (PIL) was used as an alternative separation matrix for M13 bacteriophage. In this work, a variety of system parameters was studied, including chemical structure of the cation and the anion, the crosslinker nature and its concentration, either in batch adsorption/elution or chromatographic operation mode. The PIL-based chromatographic operation mode revealed to be a suitable separation process for M13 from directly filtered E. coli supernatant, reaching over 70% M13 recovery and 4.6 purification factor in a single step.     Influence of dietary Chlorella vulgaris and carbohydrate-active enzymes on growth performance, meat quality and lipid composition of broiler chickens

    Article in pressHerein, we investigated the effect of Chlorella vulgaris as ingredient (10% of incorporation) in broiler diets, supplemented or not with 2 formulations of Carbohydrate-Active enZymes (CAZymes; Rovabio Excel AP and a mixture of recombinant CAZymes, composed by an exo-b-glucosaminidase, an alginate lyase, a peptidoglycan N-acetylmuramic acid deacetylase and a lysozyme), on growth performance, meat quality, fatty acid composition, oxidative stability, and sensory traits. One hundred twenty 1-day-old Ross 308 male birds were randomly assigned to one of the 4 experimental diets (n 5 30): corn-soybean meal–basal diet (control), basal diet with 10% C. vulgaris (CV), CV supplemented with 0.005% of a commercialCAZymecocktail (Rovabio Excel AP), (CV1R), and CV supplemented with 0.01% of a 4- CAZyme mixture previously selected (CV 1 M) during the experimental period lasted from day 21 to day 35. Body weight gain and feed conversion rate of broilers were not affected by C. vulgaris but digesta viscosity increased more than 2-fold (P , 0.001) relative to the control. In addition, neither cooking loss, shear force, juiciness, flavor nor off-flavor was impaired by dietary treatments (P.0.05). By contrast, the dietary C. vulgaris increased tenderness, yellowness (b*) and total carotenoids in breast and thigh meats. However, no additional protective effect against lipid oxidation was observed in meat with the inclusion of microalga. Chlorella vulgaris, independently of CAZymes, had a minor impact on meat fatty acid composition but improved the proportion of some beneficial fatty acids. In summary, our data indicate a slight improvement of broiler meat quality and lipid nutritional value, without impairment of broilers’ growth performance, thus supporting the usefulness of this microalga in poultry diets, up to this high level of incorporation.     Se han combinado diferentes estrategias de innovación docente para la creación de material de uso en el aula invertida. Grupos de alumnos han realizado micro-videotutoriales para la resolución de problemas básicos de cortes geológicos, realizando ellos mismos desde el guion hasta la edición, pasando por la grabación. Mediante esta actividad han tenido que desarrollar un proyecto y trabajar de forma colaborativa. El resultado de este trabajo ha sido una colección de microvideos que están siendo empleados en la formación de otros alumnos mediante el aula invertida en varias asignaturas de Ciencias de la Tierra. Los videos se han puesto a disposición de los alumnos a través de la plataforma Moodle de la UPM para que puedan aprender y repasar las técnicas de resolución de problemas de forma autónoma

    Jet size dependence of single jet suppression in lead-lead collisions at sqrt(s(NN)) = 2.76 TeV with the ATLAS detector at the LHC

    Measurements of inclusive jet suppression in heavy ion collisions at the LHC provide direct sensitivity to the physics of jet quenching. In a sample of lead-lead collisions at sqrt(s) = 2.76 TeV corresponding to an integrated luminosity of approximately 7 inverse microbarns, ATLAS has measured jets with a calorimeter over the pseudorapidity interval |eta| < 2.1 and over the transverse momentum range 38 < pT < 210 GeV. Jets were reconstructed using the anti-kt algorithm with values for the distance parameter that determines the nominal jet radius of R = 0.2, 0.3, 0.4 and 0.5. The centrality dependence of the jet yield is characterized by the jet "central-to-peripheral ratio," Rcp. Jet production is found to be suppressed by approximately a factor of two in the 10% most central collisions relative to peripheral collisions. Rcp varies smoothly with centrality as characterized by the number of participating nucleons. The observed suppression is only weakly dependent on jet radius and transverse momentum. These results provide the first direct measurement of inclusive jet suppression in heavy ion collisions and complement previous measurements of dijet transverse energy imbalance at the LHC.Comment: 15 pages plus author list (30 pages total), 8 figures, 2 tables, submitted to Physics Letters B.     Measurement of the polarisation of W bosons produced with large transverse momentum in pp collisions at sqrt(s) = 7 TeV with the ATLAS experiment

    This paper describes an analysis of the angular distribution of W->enu and W->munu decays, using data from pp collisions at sqrt(s) = 7 TeV recorded with the ATLAS detector at the LHC in 2010, corresponding to an integrated luminosity of about 35 pb^-1. Using the decay lepton transverse momentum and the missing transverse energy, the W decay angular distribution projected onto the transverse plane is obtained and analysed in terms of helicity fractions f0, fL and fR over two ranges of W transverse momentum (ptw): 35 < ptw < 50 GeV and ptw > 50 GeV. Good agreement is found with theoretical predictions. For ptw > 50 GeV, the values of f0 and fL-fR, averaged over charge and lepton flavour, are measured to be : f0 = 0.127 +/- 0.030 +/- 0.108 and fL-fR = 0.252 +/- 0.017 +/- 0.030, where the first uncertainties are statistical, and the second include all systematic effects.Comment: 19 pages plus author list (34 pages total), 9 figures, 11 tables, revised author list, matches European Journal of Physics C versio

    Observation of a new chi_b state in radiative transitions to Upsilon(1S) and Upsilon(2S) at ATLAS

    The chi_b(nP) quarkonium states are produced in proton-proton collisions at the Large Hadron Collider (LHC) at sqrt(s) = 7 TeV and recorded by the ATLAS detector. Using a data sample corresponding to an integrated luminosity of 4.4 fb^-1, these states are reconstructed through their radiative decays to Upsilon(1S,2S) with Upsilon->mu+mu-. In addition to the mass peaks corresponding to the decay modes chi_b(1P,2P)->Upsilon(1S)gamma, a new structure centered at a mass of 10.530+/-0.005 (stat.)+/-0.009 (syst.) GeV is also observed, in both the Upsilon(1S)gamma and Upsilon(2S)gamma decay modes.     Search for displaced vertices arising from decays of new heavy particles in 7 TeV pp collisions at ATLAS

    We present the results of a search for new, heavy particles that decay at a significant distance from their production point into a final state containing charged hadrons in association with a high-momentum muon. The search is conducted in a pp-collision data sample with a center-of-mass energy of 7 TeV and an integrated luminosity of 33 pb^-1 collected in 2010 by the ATLAS detector operating at the Large Hadron Collider. Production of such particles is expected in various scenarios of physics beyond the standard model. We observe no signal and place limits on the production cross-section of supersymmetric particles in an R-parity-violating scenario as a function of the neutralino lifetime. Limits are presented for different squark and neutralino masses, enabling extension of the limits to a variety of other models.Comment: 8 pages plus author list (20 pages total), 8 figures, 1 table, final version to appear in Physics Letters
