Electric and magnetic dipolar response of Germanium spheres: Interference effects, scattering anisotropy and optical forces
We address the scattering cross sections, and their consequences, for
submicrometer Germanium spheres. It is shown that there is a wide window in the
near infrared where light scattering by these particles is fully described by
their induced electric and magnetic dipoles. In this way, we observe remarkable
anisotropic scattering angular distributions, as well as zero forward or
backward scattered intensities, which until recently was theoretically
demonstrated only for hypothetically postulated magnetodielectric spheres.
Also, interesting new effects of the optical forces exerted on these objects

Motivational analysis comparing three contexts of physical activity
El estudio analizó las diferencias motivacionales entre estudiantes de educación física, deportistas y practicantes de ejercicio físico no competitivo. Además, examinó el poder de predicción del clima motivacional percibido y de los tipos de motivación sobre la propensión a la experiencia autotélica en los tres contextos. Se utilizaron tres muestras diferentes: 895 estudiantes de educación física, 413 deportistas y 727 practicantes de ejercicio físico no competitivo. El análisis de regresión mostró que el clima tarea y la motivación intrínseca predijeron positivamente y de forma significativa la experiencia autotélica en los tres contextos, de ahí la importancia de fomentar este tipo de clima motivacional. No obstante, los resultados han mostrado diferencias motivacionales entre los contextos de educación física, deporte competitivo y ejercicio físico no competitivo. Los resultados son discutidos en relación al diferente carácter competitivo/lúdico y voluntariedad/obligatoriedad que rodea a la práctica física en cada uno de los contextos comparados en este estudioThe study analyzed the motivational differences between physical education students, athletes and non-competitive exercisers. Furthermore, the predictive power of perceived motivational climate and types of motivation on the autotelic experience propensity in the three contexts was examined. Three different samples were used: 895 physical education students, 413 athletes and 727 non-competitive exercisers. Regression analysis showed that task-involving climate and intrinsic motivation positively and significantly predicted the autotelic experience in the three contexts, hence the importance of promoting this type of motivational climate. However, the results revealed that there were motivational differences between the contexts of physical education, competitive sports and non-competitive exercise. Data-driven reconstruction of chaotic dynamical equations: the H\'enon-Heiles type system
In this study, the classical two-dimensional potential
, , is considered. At , the system is superintegrable and
integrable, respectively, whereas for it exhibits a richer chaotic
dynamics. For instance, at it coincides with the H\'enon-Heiles system.
The periodic, quasi-periodic and chaotic motions are systematically
characterized employing time series, Poincar\'e sections, symmetry lines and
the largest Lyapunov exponent as a function of the energy and the parameter
. Concrete results for the lowest cases are presented in complete
detail. This model is used as a benchmark system to estimate the accuracy of
the Sparse Identification of Nonlinear Dynamical Systems (SINDy) method, a
data-driven algorithm which reconstructs the underlying governing dynamical
equations. We pay special attention at the transition from regular motion to
chaos and how this influences the precision of the algorithm. In particular, it
is shown that SINDy is a robust and stable tool possessing the ability to
generate non-trivial approximate analytical expressions for periodic
trajectories as well
Breast cancer PAM50 signature: correlation and concordance between RNA-Seq and digital multiplexed gene expression technologies in a triple negative breast cancer series
[Background]: Full RNA-Seq is a fundamental research tool for whole transcriptome analysis. However, it is too costly and time consuming to be used in routine clinical practice. We evaluated the transcript quantification agreement between RNA-Seq and a digital multiplexed gene expression platform, and the subtype call after running the PAM50 assay in a series of breast cancer patients classified as triple negative by IHC/FISH. The goal of this study is to analyze the concordance between both expression platforms overall, and for calling PAM50 triple negative breast cancer intrinsic subtypes in particular.[Results]: The analyses were performed in paraffin-embedded tissues from 96 patients recruited in a multicenter, prospective, non-randomized neoadjuvant triple negative breast cancer trial (NCT01560663). Pre-treatment core biopsies were obtained following clinical practice guidelines and conserved as FFPE for further RNA extraction. PAM50 was performed on both digital multiplexed gene expression and RNA-Seq platforms. Subtype assignment was based on the nearest centroid classification following this procedure for both platforms and it was concordant on 96% of the cases (N = 96). In four cases, digital multiplexed gene expression analysis and RNA-Seq were discordant. The Spearman correlation to each of the centroids and the risk of recurrence were above 0.89 in both platforms while the agreement on Proliferation Score reached up to 0.97. In addition, 82% of the individual PAM50 genes showed a correlation coefficient > 0.80.[Conclusions]: In our analysis, the subtype calling in most of the samples was concordant in both platforms and the potential discordances had reduced clinical implications in terms of prognosis. GEICAM Guidelines for the Management of Patients with Breast Cancer During the COVID-19 Pandemic in Spain
Breast cancer (BC) is the most common cancer in women in
Spain. During the COVID-19 pandemic caused by the SARSCoV-2 virus, patients with BC still require timely treatment
and follow-up; however, hospitals are overwhelmed with
infected patients and, if exposed, patients with BC are at
higher risk for infection and serious complications if infected.
Thus, health care providers need to evaluate each BC treatment and in-hospital visit to minimize pandemic-associated
risks while maintaining adequate treatment efficacy. Here we
present a set of guidelines regarding available options for BC
patient management and treatment by BC subtype in the
context of the COVID-19 pandemic. Owing to the lack of evidence about COVID-19 infection, these recommendations
are mainly based on expert opinion, medical organizations’
and societies’ recommendations, and some published evidence. We consider this a useful tool to facilitate medical
decision making in this health crisis situation we are facing
Therapeutic effect of all-trans-retinoic acid (at-RA) on an autoimmune nephritis experimental model: role of the VLA-4 integrin
BACKGROUND: Mercuric chloride (HgCl(2)) induces an autoimmune nephritis in the Brown Norway (BN) rats characterized by anti-glomerular basement membrane antibodies (anti-GBM Ab) deposition, proteinuria and a severe interstitial nephritis, all evident at day 13 of the disease. We assessed the effects of all-trans retinoic acid (at-RA) in this experimental model. At-RA is a vitamin A metabolite which has shown beneficial effects on several nephropathies, even though no clear targets for at-RA were provided. METHODS: We separated animals in four different experimental groups (HgCl(2), HgCl(2)+at-RA, at-RA and vehicle). From each animal we collected, at days 0 and 13, numerous biological samples: urine, to measure proteinuria by colorimetry; blood to determine VLA-4 expression by flow citometry; renal tissue to study the expression of VCAM-1 by Western blot, the presence of cellular infiltrates by immunohistochemistry, the IgG deposition by immunofluorescence, and the cytokines expression by RT-PCR. Additionally, adhesion assays to VCAM-1 were performed using K562 α4 transfectant cells. ANOVA tests were used for statistical significance estimation. RESULTS: We found that at-RA significantly decreased the serum levels of anti-GBM and consequently its deposition along the glomerular membrane. At-RA markedly reduced proteinuria as well as the number of cellular infiltrates in the renal interstitium, the levels of TNF-α and IL-1β cytokines and VCAM-1 expression in renal tissue. Moreover, we reported here for the first time in an in vivo model that at-RA reduced, to basal levels, the expression of VLA-4 (α4β1) integrin induced by mercury on peripheral blood leukocytes (PBLs). In addition, using K562 α4 stable transfectant cells, we found that at-RA inhibited VLA-4 dependent cell adhesion to VCAM-1. CONCLUSION: Here we demonstrate a therapeutic effect of at-RA on an autoimmune experimental nephritis model in rats. We report a significant reduction of the VLA-4 integrin expression on PBLs as well as the inhibition of the VLA4/VCAM1-dependent leukocyte adhesion by at-RA treatment. Probiotic supplementation influences the diversity of the intestinal microbiota during early stages of farmed Senegalese sole (Solea senegalensis, Kaup, 1858)
Ingestion of bacteria at early stages results in establishment of a primary intestinal microbiota which likely undergoes several stages along fish life. The role of this intestinal microbiota regulating body functions is crucial for larval development. Probiotics have been proved to modulate this microbiota and exert antagonistic effects against fish pathogens. In the present study, we aimed to determine bacterial diversity along different developmental stages of farmed Senegalese sole (Solea senegalensis) after feeding probiotic (Shewanella putrefaciens Pdp11) supplemented diet for a short period (10–30 days after hatching, DAH). Intestinal lumen contents of sole larvae fed control and probiotic diets were collected at 23, 56, 87, and 119 DAH and DNA was amplified using 16S rDNA bacterial domain-specific primers. Amplicons obtained were separated by denaturing gradient gel electrophoresis (DGGE), cloned, and resulting sequences compared to sequences in GenBank. Results suggest that Shewanella putrefaciens Pdp11 induces a modulation of the dominant bacterial taxa of the intestinal microbiota from 23 DAH. DGGE patterns of larvae fed the probiotic diet showed a core of bands related to Lactobacillus helveticus, Pseudomonas acephalitica, Vibrio parahaemolyticus,and Shewanella genus, together with increased Vibri o genus presence. In addition, decreased number of clones related to Photobacterium damselae subsp piscicida at 23 and 56 DAH was observed in probiotic-fed larvae. A band corresponding to Shewanella putrefaciens Pdp11 was sequenced as predominant from 23 to 119 DAH samples, confirming the colonization by the probiotics. Microbiota modulation obtained via probiotics addition emerges as an effective tool to improve Solea senegalensis larviculture.En prens
Polygenic Risk Scores for Prediction of Breast Cancer and Breast Cancer Subtypes
Stratification of women according to their risk of breast cancer based on polygenic risk scores (PRSs) could improve screening and prevention strategies. Our aim was to develop PRSs, optimized for prediction of estrogen receptor (ER)-specific disease, from the largest available genome-wide association dataset and to empirically validate the PRSs in prospective studies. The development dataset comprised 94,075 case subjects and 75,017 control subjects of European ancestry from 69 studies, divided into training and validation sets. Samples were genotyped using genome-wide arrays, and single-nucleotide polymorphisms (SNPs) were selected by stepwise regression or lasso penalized regression. The best performing PRSs were validated in an independent test set comprising 11,428 case subjects and 18,323 control subjects from 10 prospective studies and 190,040 women from UK Biobank (3,215 incident breast cancers). For the best PRSs (313 SNPs), the odds ratio for overall disease per 1 standard deviation in ten prospective studies was 1.61 (95%CI: 1.57-1.65) with area under receiver-operator curve (AUC) = 0.630 (95%CI: 0.628-0.651). The lifetime risk of overall breast cancer in the top centile of the PRSs was 32.6%. Compared with women in the middle quintile, those in the highest 1% of risk had 4.37- and 2.78-fold risks, and those in the lowest 1% of risk had 0.16- and 0.27-fold risks, of developing ER-positive and ER-negative disease, respectively. Goodness-of-fit tests indicated that this PRS was well calibrated and predicts disease risk accurately in the tails of the distribution. Circadian Rhythm and Sleep Disruption: Causes, Metabolic Consequences and Countermeasures.
Circadian (∼ 24 hour) timing systems pervade all kingdoms of life, and temporally optimize behaviour and physiology in humans. Relatively recent changes to our environments, such as the introduction of artificial lighting, can disorganize the circadian system, from the level of the molecular clocks that regulate the timing of cellular activities to the level of synchronization between our daily cycles of behaviour and the solar day. Sleep/wake cycles are intertwined with the circadian system, and global trends indicate that these too are increasingly subject to disruption. A large proportion of the world's population is at increased risk of environmentally-driven circadian rhythm and sleep disruption, and a minority of individuals are also genetically predisposed to circadian misalignment and sleep disorders. The consequences of disruption to the circadian system and sleep are profound and include myriad metabolic ramifications, some of which may be compounded by adverse effects on dietary choices. If not addressed, the deleterious effects of such disruption will continue to cause widespread health problems; therefore, implementation of the numerous behavioural and pharmaceutical interventions that can help restore circadian system alignment and enhance sleep will be important
Transcriptome-wide association study of breast cancer risk by estrogen-receptor status
Previous transcriptome-wide association studies (TWAS) have identified breast cancer risk genes by integrating data from expression quantitative loci and genome-wide association studies (GWAS), but analyses of breast cancer subtype-specific associations have been limited. In this study, we conducted a TWAS using gene expression data from GTEx and summary statistics from the hitherto largest GWAS meta-analysis conducted for breast cancer overall, and by estrogen receptor subtypes (ER+ and ER-). We further compared associations with ER+ and ER- subtypes, using a case-only TWAS approach. We also conducted multigene conditional analyses in regions with multiple TWAS associations. Two genes, STXBP4 and HIST2H2BA, were specifically associated with ER+ but not with ER- breast cancer. We further identified 30 TWAS-significant genes associated with overall breast cancer risk, including four that were not identified in previous studies. Conditional analyses identified single independent breast-cancer gene in three of six regions harboring multiple TWAS-significant genes. Our study provides new information on breast cancer genetics and biology, particularly about genomic differences between ER+ and ER- breast cancer.Peer reviewe
