    Усовершенствованный способ получения низкомолекулярных субстратов тромбина – п-нитроанилидов пептидов

    Low molecular weight chromogenic thrombin peptide substrates, p-nitroanilides of short peptides protected at their N-terminal amino group, were prepared by solid-phase peptide synthesis on polystyrene-divinylbenzene polymer with trityl groups with preliminary attached p-phenylene diamine moiety. After the cleavage from the resin peptide p-aminoanilides were mildly oxidized to p-nitroanilides with the mixture of potassium sulfate and persulfate. Adsorption onto polymer support Bio-Beads SM-2 with further elution by acetonitrile allowed easy separating peptide p-nitroanilides from the oxidizer and obtaining the thrombin chromogenic substrate preparations with the target substance contents of not less than 95% and yields of 30-40%. Thrombin effectively catalyzed hydrolysis of the prepared substrates with KM and Vmax values of 29-134 mM and 0.03-1/16 mM/s, respectively.Низкомолекулярные хромогенные субстраты тромбина, п-нитроанилиды защищённых по N-концевой аминогруппе коротких пептидов, получали путём твердофазного пептидного синтеза на полистирол-дивинилбензольном полимере с тритильными группами с предварительно присоединенным к ним остатком п-фенилендиамина. После отщепления от смолы п-аминоанилиды пептидов мягко окисляли до п-нитроанилидов смесью сульфата и персульфата калия. Адсорбция на полимерном носителе Bio-Beads SM-2 с элюцией ацетонитрилом позволила легко отделить полученные п-нитроанилиды пептидов от окислителя и получить препараты хромогенных субстратов тромбина с содержанием целевого вещества не ниже 95% и с выходами 30-40%. Тромбин эффективно катализировал гидролиз полученных субстратов со значением KM в диапазоне 29-134 мкМ и Vmax 0.03-1.16 мкМ/с

    The article outlines the questions of pathogenesis of acne as well as clinical trial data, which indicate that the skin of patients with acne needs comprehensive care with therapeutic cosmetics. Long term use of medical cosmetics has a positive effect on the course of acne disease. The basis of the study is the use of cosmetics line A “Skinormil” including the patented complex Citobiol lRIS, zinc sulfate and vitamin A. Seventy-four patients with acne of the first, second and third severity were monitored and it was recommended them a comprehensive skin care with medical cosmetics. According to the analysis of the clinical picture, it was concluded that on the background of proper care, there is a positive dynamics in the course of acne.В статье изложены вопросы патогенеза акне, а также данные клинического исследования, которые свидетельствуют о том, что кожа больных с акне нуждается в комплексном уходе лечебными косметическими средствами. Длительное использование средств лечебной косметики оказывает положительное влияние на течение угревой болезни. В основе исследования лежит применение косметических средств линии А «Скинормил», включающей запатентованный комплекс Citobiol IRIS, сульфат цинка и витамин А. Проведено наблюдение за 74 больными с акне первой, второй и третьей степени тяжести, которым был рекомендован комплексный уход за кожей средствами лечебной косметики. По результатам анализа клинической картины сделан вывод, что на фоне правильного ухода отмечается положительная динамика в течении акне

    The article discusses results of the structural and functional analysis of molecular genetic abnormalities in various malignant tumors. Investigations have discovered more than 20 new markers for sporadic breast cancer. Several of them formed the test system, allowing the diagnosis with a specificity of 100%. Appearance of TMPRSS2/ERG4 chimeric gene is a frequent tumor-specific event, its expression is correlated with more aggressive forms of prostate cancer, may serve as a molecular marker for tumor cells and androgen assessment of tumor response to hormonal therapy. The effective systems for the early diagnosis of cervix and endometrium cancer were developed as well. Mutations in the VHL, deletions of chromosome 3 and methylation of several genes can predict the course and selection of effective therapy of clear cell kidney cancer. a number of molecular markers were identified for early diagnosis and prognosis of recurrence of bladder cancer. For diagnosis, prognosis and treatment of brain tumors we developed an effective complex system of markers. Protocol of molecular genetics investigation reveals the cause of the disease by more than 90% of patients with retinoblastoma. In order to study abnormal methylation in tumor genomes an innovative technology AFLOAT has been developed that allows to efficiently identify new markers with diagnostic value. Test systems of molecular genetic and epigenetic markers for early diagnosis and prognosis as well as for cancer therapy optimization have shown to be effective, have been approved for use in clinical practice and are being introduced into practical healthcare.Рассматриваются результаты структурно-функционального анализа молекулярно-генетических нарушений при различных злокачественных опухолях. Исследования позволили обнаружить более 20 новых маркеров для спорадического рака молочной железы, из которых сформированы тест-системы, позволяющие проводить диагностику со специфичностью 100%. Образование химерного гена TMPRSS2/ERG4 является частым опухоль-специфическим событием, его экспрессия коррелирует с более агрессивными формами рака предстательной железы может служить молекулярным маркером  андрогенчувствительности опухолевых клеток и оценки ответа опухоли на гормональную терапию. Разработаны эффективные системы для ранней диагностики рака шейки матки и эндометрия. Мутации гена VHL, делеции хромосомы 3 и метилирование ряда генов позволяет прогнозировать течение и подбор эффективной терапии светлоклеточного рака почки. Для ранней диагностики, прогноза течения и рецидивирования рака мочевого пузыря определен целый ряд молекулярных маркеров. Для диагностики, прогноза и лечения опухолей мозга разработана эффективная комплексная система маркеров. Протокол молекулярно-генетического  исследования позволяет обнаружить причину заболевания более чем у 90% пациентов с ретинобластомой.  Для исследования аномального метилирования в опухолевых геномах разработана инновационная технология AFLOAT, позволяющая эффективно выявлять новые маркеры, имеющие диагностическое значение. Тест-системы молекулярно-генетических и эпигенетических маркеров для ранней диагностики, прогноза течения и оптимизации терапии злокачественных новообразований показали свою эффективность, получили разрешение на использование в клинической практике и в настоящее время активно внедряются в практическое здравоохранение.

    General Relativistic MHD Jets

    Magnetic fields connecting the immediate environs of rotating black holes to large distances appear to be the most promising mechanism for launching relativistic jets, an idea first developed by Blandford and Znajek in the mid-1970s. To enable an understanding of this process, we provide a brief introduction to dynamics and electromagnetism in the spacetime near black holes. We then present a brief summary of the classical Blandford-Znajek mechanism and its conceptual foundations. Recently, it has become possible to study these effects in much greater detail using numerical simulation. After discussing which aspects of the problem can be handled well by numerical means and which aspects remain beyond the grasp of such methods, we summarize their results so far. Simulations have confirmed that processes akin to the classical Blandford-Znajek mechanism can launch powerful electromagnetically-dominated jets, and have shown how the jet luminosity can be related to black hole spin and concurrent accretion rate. However, they have also shown that the luminosity and variability of jets can depend strongly on magnetic field geometry.     Measurement of the B0-anti-B0-Oscillation Frequency with Inclusive Dilepton Events

    The B0B^0-Bˉ0\bar B^0 oscillation frequency has been measured with a sample of 23 million \B\bar B pairs collected with the BABAR detector at the PEP-II asymmetric B Factory at SLAC. In this sample, we select events in which both B mesons decay semileptonically and use the charge of the leptons to identify the flavor of each B meson. A simultaneous fit to the decay time difference distributions for opposite- and same-sign dilepton events gives Δmd=0.493±0.012(stat)±0.009(syst)\Delta m_d = 0.493 \pm 0.012{(stat)}\pm 0.009{(syst)} ps1^{-1}.Comment: 7 pages, 1 figure, submitted to Physical Review Letter

    Measurement of the polarisation of W bosons produced with large transverse momentum in pp collisions at sqrt(s) = 7 TeV with the ATLAS experiment

    This paper describes an analysis of the angular distribution of W->enu and W->munu decays, using data from pp collisions at sqrt(s) = 7 TeV recorded with the ATLAS detector at the LHC in 2010, corresponding to an integrated luminosity of about 35 pb^-1. Using the decay lepton transverse momentum and the missing transverse energy, the W decay angular distribution projected onto the transverse plane is obtained and analysed in terms of helicity fractions f0, fL and fR over two ranges of W transverse momentum (ptw): 35 < ptw < 50 GeV and ptw > 50 GeV. Good agreement is found with theoretical predictions. For ptw > 50 GeV, the values of f0 and fL-fR, averaged over charge and lepton flavour, are measured to be : f0 = 0.127 +/- 0.030 +/- 0.108 and fL-fR = 0.252 +/- 0.017 +/- 0.030, where the first uncertainties are statistical, and the second include all systematic effects.Comment: 19 pages plus author list (34 pages total), 9 figures, 11 tables, revised author list, matches European Journal of Physics C versio

    Observation of a new chi_b state in radiative transitions to Upsilon(1S) and Upsilon(2S) at ATLAS

    The chi_b(nP) quarkonium states are produced in proton-proton collisions at the Large Hadron Collider (LHC) at sqrt(s) = 7 TeV and recorded by the ATLAS detector. Using a data sample corresponding to an integrated luminosity of 4.4 fb^-1, these states are reconstructed through their radiative decays to Upsilon(1S,2S) with Upsilon->mu+mu-. In addition to the mass peaks corresponding to the decay modes chi_b(1P,2P)->Upsilon(1S)gamma, a new structure centered at a mass of 10.530+/-0.005 (stat.)+/-0.009 (syst.) GeV is also observed, in both the Upsilon(1S)gamma and Upsilon(2S)gamma decay modes.     Search for displaced vertices arising from decays of new heavy particles in 7 TeV pp collisions at ATLAS

    We present the results of a search for new, heavy particles that decay at a significant distance from their production point into a final state containing charged hadrons in association with a high-momentum muon. The search is conducted in a pp-collision data sample with a center-of-mass energy of 7 TeV and an integrated luminosity of 33 pb^-1 collected in 2010 by the ATLAS detector operating at the Large Hadron Collider. Production of such particles is expected in various scenarios of physics beyond the standard model. We observe no signal and place limits on the production cross-section of supersymmetric particles in an R-parity-violating scenario as a function of the neutralino lifetime.     Measurement of the inclusive isolated prompt photon cross-section in pp collisions at sqrt(s)= 7 TeV using 35 pb-1 of ATLAS data

    A measurement of the differential cross-section for the inclusive production of isolated prompt photons in pp collisions at a center-of-mass energy sqrt(s) = 7 TeV is presented. The measurement covers the pseudorapidity ranges |eta|<1.37 and 1.52<=|eta|<2.37 in the transverse energy range 45<=E_T<400GeV. The results are based on an integrated luminosity of 35 pb-1, collected with the ATLAS detector at the LHC. The yields of the signal photons are measured using a data-driven technique, based on the observed distribution of the hadronic energy in a narrow cone around the photon candidate and the photon selection criteria. The results are compared with next-to-leading order perturbative QCD calculations and found to be in good agreement over four orders of magnitude in cross-section.Comment: 7 pages plus author list (18 pages total), 2 figures, 4 tables, final version published in Physics Letters

    Measurement of D*+/- meson production in jets from pp collisions at sqrt(s) = 7 TeV with the ATLAS detector

    This paper reports a measurement of D*+/- meson production in jets from proton-proton collisions at a center-of-mass energy of sqrt(s) = 7 TeV at the CERN Large Hadron Collider. The measurement is based on a data sample recorded with the ATLAS detector with an integrated luminosity of 0.30 pb^-1 for jets with transverse momentum between 25 and 70 GeV in the pseudorapidity range |eta| < 2.5. D*+/- mesons found in jets are fully reconstructed in the decay chain: D*+ -> D0pi+, D0 -> K-pi+, and its charge conjugate. The production rate is found to be N(D*+/-)/N(jet) = 0.025 +/- 0.001(stat.) +/- 0.004(syst.) for D*+/- mesons that carry a fraction z of the jet momentum in the range 0.3 < z < 1. Monte Carlo predictions fail to describe the data at small values of z, and this is most marked at low jet transverse momentum.Comment: 10 pages plus author list (22 pages total), 5 figures, 1 table, matches published version in Physical Review
