    On the assimilation of SWOT type data into 2D shallow-water models

    In river hydraulics, assimilation of water level measurements at gauging stations is well controlled, while assimilation of images is still delicate. In the present talk, we address the richness of satellite mapped information to constrain a 2D shallow-water model, but also related difficulties. 2D shallow models may be necessary for small scale modelling in particular for low-water and flood plain flows. Since in both cases, the dynamics of the wet dry front is essential, one has to elaborate robust and accurate solvers. In this contribution we introduce robust second order, stable finite volume scheme [CoMaMoViDaLa]. Comparisons of real like tests cases with more classical solvers highlight the importance of an accurate flood plain modelling. A preliminary inverse study is presented in a flood plain flow case, [LaMo] [HoLaMoPu]. As a first step, a 0th order data processing model improves observation operator and produces more reliable water level derived from rough measurements [PuRa]. Then, both model and flow behaviours can be better understood thanks to variational sensitivities based on a gradient computation and adjoint equations. It can reveal several difficulties that a model designer has to tackle. Next, a 4D-Var data assimilation algorithm used with spatialized data leads to improved model calibration and potentially leads to identify river discharges. All the algorithms are implemented into DassFlow software (Fortran, MPI, adjoint) [Da]. All these results and experiments (accurate wet-dry front dynamics, sensitivities analysis, identification of discharges and calibration of model) are currently performed in view to use data from the future SWOT mission

    Transfert : les déterminants de la performance des exploitations agricoles familiales

    Le transfert des exploitations familiales est une étape cruciale du cycle de vie de la majorité des exploitations agricoles québécoises. Avec un taux élevé d’échec important de 40 à 50 % dans les cinq années suivant le transfert d’une PME, il est important de connaître les causes de ces échecs ainsi que les déterminants de la performance des exploitations survivantes après le cap des premières années. Cet article présente les déterminants financiers et technico-économiques de cette performance en s’appuyant sur trois périodes : le montage financier, l’année du transfert et l’évolution de la situation. Les résultats démontrent que ce sont les ratios technico-économiques de l’exploitation à l’année du transfert et durant la période post-transfert qui ont la plus grande influence sur la performance financière cinq années après.The transfer of family farms is a crucial step in the life cycle of the majority of farms in Quebec. Within five years following the transfer of SMEs about 40-50% fail. It is important to know the causes of their failures and the determinants of performance of farms after surviving the first course of years. This article presents the financial and technico-economic determinants of this performance based on three periods: the financing, the year of transfer and the evolution of the situation. The results show that the technico-economic ratios the year of transfer and during the post-transfer period have the greatest influence on financial performance five years after the transfer.     Complete exon sequencing of all known Usher syndrome genes greatly improves molecular diagnosis

    <p>Abstract</p> <p>Background</p> <p>Usher syndrome (USH) combines sensorineural deafness with blindness. It is inherited in an autosomal recessive mode. Early diagnosis is critical for adapted educational and patient management choices, and for genetic counseling. To date, nine causative genes have been identified for the three clinical subtypes (USH1, USH2 and USH3). Current diagnostic strategies make use of a genotyping microarray that is based on the previously reported mutations. The purpose of this study was to design a more accurate molecular diagnosis tool.</p> <p>Methods</p> <p>We sequenced the 366 coding exons and flanking regions of the nine known USH genes, in 54 USH patients (27 USH1, 21 USH2 and 6 USH3).</p> <p>Results</p> <p>Biallelic mutations were detected in 39 patients (72%) and monoallelic mutations in an additional 10 patients (18.5%). In addition to biallelic mutations in one of the USH genes, presumably pathogenic mutations in another USH gene were detected in seven patients (13%), and another patient carried monoallelic mutations in three different USH genes. Notably, none of the USH3 patients carried detectable mutations in the only known USH3 gene, whereas they all carried mutations in USH2 genes. Most importantly, the currently used microarray would have detected only 30 of the 81 different mutations that we found, of which 39 (48%) were novel.</p> <p>Conclusions</p> <p>Based on these results, complete exon sequencing of the currently known USH genes stands as a definite improvement for molecular diagnosis of this disease, which is of utmost importance in the perspective of gene therapy.</p

    Blood Signature of Pre-Heart Failure: A Microarrays Study

    International audienceBACKGROUND: The preclinical stage of systolic heart failure (HF), known as asymptomatic left ventricular dysfunction (ALVD), is diagnosed only by echocardiography, frequent in the general population and leads to a high risk of developing severe HF. Large scale screening for ALVD is a difficult task and represents a major unmet clinical challenge that requires the determination of ALVD biomarkers. METHODOLOGY/PRINCIPAL FINDINGS: 294 individuals were screened by echocardiography. We identified 9 ALVD cases out of 128 subjects with cardiovascular risk factors. White blood cell gene expression profiling was performed using pangenomic microarrays. Data were analyzed using principal component analysis (PCA) and Significant Analysis of Microarrays (SAM). To build an ALVD classifier model, we used the nearest centroid classification method (NCCM) with the ClaNC software package. Classification performance was determined using the leave-one-out cross-validation method. Blood transcriptome analysis provided a specific molecular signature for ALVD which defined a model based on 7 genes capable of discriminating ALVD cases. Analysis of an ALVD patients validation group demonstrated that these genes are accurate diagnostic predictors for ALVD with 87% accuracy and 100% precision. Furthermore, Receiver Operating Characteristic curves of expression levels confirmed that 6 out of 7 genes discriminate for left ventricular dysfunction classification. CONCLUSIONS/SIGNIFICANCE: These targets could serve to enhance the ability to efficiently detect ALVD by general care practitioners to facilitate preemptive initiation of medical treatment preventing the development of HF

    Juxtaposing BTE and ATE – on the role of the European insurance industry in funding civil litigation

    One of the ways in which legal services are financed, and indeed shaped, is through private insurance arrangement. Two contrasting types of legal expenses insurance contracts (LEI) seem to dominate in Europe: before the event (BTE) and after the event (ATE) legal expenses insurance. Notwithstanding institutional differences between different legal systems, BTE and ATE insurance arrangements may be instrumental if government policy is geared towards strengthening a market-oriented system of financing access to justice for individuals and business. At the same time, emphasizing the role of a private industry as a keeper of the gates to justice raises issues of accountability and transparency, not readily reconcilable with demands of competition. Moreover, multiple actors (clients, lawyers, courts, insurers) are involved, causing behavioural dynamics which are not easily predicted or influenced. Against this background, this paper looks into BTE and ATE arrangements by analysing the particularities of BTE and ATE arrangements currently available in some European jurisdictions and by painting a picture of their respective markets and legal contexts. This allows for some reflection on the performance of BTE and ATE providers as both financiers and keepers. Two issues emerge from the analysis that are worthy of some further reflection. Firstly, there is the problematic long-term sustainability of some ATE products. Secondly, the challenges faced by policymakers that would like to nudge consumers into voluntarily taking out BTE LEI
