    A culture-independent and culture-dependent study of the bacteria community from the bedrock soil interface

    In nutrient limited soils, minerals constitute a major reservoir of bio-essential elements. Consequently, the release of nutritive elements during weathering is crucial. Bacteria have been shown to enhance weathering rates; however, there has been limited work that has focused on the bacterial weathering of bedrock or parent rock, which are the major sources of minerals, in nutrient limiting soils. In this study, both a culture-independent and culture-dependent approach was used to study the bacterial community at the interface between basaltic bedrock and nutrient limiting soil in Cadiar Idris region of Snowdonia National Park, United Kingdom. High throughput sequencing method, Ion Torrent, was used to characterise the bacterial community, which generated over 250,000 sequences. Taxonomical assignment demonstrated that approximately 50% (125,000 sequences) of the community consisted of the orders Actinomycetales, Burkholderiales, Clostridales, Bacillales, Rhizobiales and Acidobacterium, with unclassified sequences representing 44% ± 1.46% (110,000 ± 3650). Bacteria belonging to the genera Serratia, Pseudomonas, Bacillus, Paenibacillus, Chromobacterium, Janthinobacterium, Burkholderia and Arthrobacter, were isolated from the sample site. All of the isolates were able to grow in a minimal growth medium, which contained glucose, ammonium chloride with basalt as the sole source of bio-essential elements. Seventy percent of the isolates significantly enhanced basalt dissolution (p < 0.05). The rate of dissolution correlated to the production of oxalic acid and acidification of the growth medium. The findings of this work suggest that at the interface between bedrock and soil heterotrophic members of the bacterial community can enhance weathering, an essential part of biogeochemical cycling in nutrient limiting soil

    Social welfare analysis of investment public-private partnership approaches for tansportation projects

    This paper has two objectives: (1) to introduce a new approach to gaining widespread support for comprehensive road pricing; and (2) to develop a detailed social welfare analysis for road pricing schemes. We first describe a new approach to garnering support for system-wide road pricing, which we refer to as an investment public-private partnership, or IP3. This approach returns a significant portion of the economic value created by road pricing back to its citizen-owners. Next, we present a social welfare framework that estimates the benefits and costs of using the IP3 approach on an urban transportation network. Policy makers typically evaluate public-private partnership (P3) projects using Value for Money (VfM) analysis. However, a P3 project's impact on overall social welfare provides a more comprehensive evaluation criterion. Apart from several theoretical studies, a detailed social welfare analysis that includes all major P3 project stakeholders is lacking. Using Fresno City's transportation system as our case study, we show that system-optimal tolling scenarios favor average users, but that governmentÂżand consequently taxpayersÂżwould pay for costly tolling systems. In contrast, unlimited profit-maximizing tolls raise substantial profits for government, for the infrastructure's citizen-owners, and for the private sector, but the average user is worse off. From a social welfare perspective, one should search for a Pareto-improvement under which all major stakeholders are better off. Our estimates indicate that a mixed private and public tolling scheme offers such an improvement. A mixed scheme results in the highest social welfare among all scenarios unless the weight placed on motorists' (i.e., transportation users') welfare is very low or the weight placed on residents' welfare is very high relative to the weight of other stakeholders

    BET bromodomains regulate transforming growth factor-beta-induced proliferation and cytokine release in asthmatic airway smooth muscle

    Airway smooth muscle (ASM) mass is increased in asthma, and ASM cells from patients with asthma are hyperproliferative and release more IL-6 and CXCL8. The BET (bromo- and extra-terminal) family of proteins (Brd2, Brd3, and Brd4) govern the assembly of histone acetylation-dependent chromatin complexes. We have examined whether they modulate proliferation and cytokine expression in asthmatic ASM cells by studying the effect of BET bromodomain mimics JQ1/SGCBD01 and I-BET762. ASM cells from healthy individuals and nonsevere and severe asthmatics were pretreated with JQ1/SGCBD01 and I-BET762 prior to stimulation with FCS and TGF-ÎČ. Proliferation was measured by BrdU incorporation. IL-6 and CXCL8 release was measured by ELISA, and mRNA expression was measured by quantitative RT-PCR. ChIP using a specific anti-Brd4 antibody and PCR primers directed against the transcriptional start site of IL-6 and CXCL8 gene promoters was performed. Neither JQ1/SGCBD01 nor I-BET762 had any effect on ASM cell viability. JQ1/SGCBD01 and I-BET762 inhibited FCS+TGF-ÎČ-induced ASM cell proliferation and IL-6 and CXCL8 release in healthy individuals (≄ 30 nm) and in nonsevere and severe asthma patients (≄100 nm), with the latter requiring higher concentrations of these mimics. JQ1/SGCBD01 reduced Brd4 binding to IL8 and IL6 promoters induced by FCS+TGF-ÎČ. Mimics of BET bromodomains inhibit aberrant ASM cell proliferation and inflammation with lesser efficiency in those from asthmatic patients. They may be effective in reducing airway remodeling in asthma

    Spatio-temporal Models of Lymphangiogenesis in Wound Healing

    Several studies suggest that one possible cause of impaired wound healing is failed or insufficient lymphangiogenesis, that is the formation of new lymphatic capillaries. Although many mathematical models have been developed to describe the formation of blood capillaries (angiogenesis), very few have been proposed for the regeneration of the lymphatic network. Lymphangiogenesis is a markedly different process from angiogenesis, occurring at different times and in response to different chemical stimuli. Two main hypotheses have been proposed: 1) lymphatic capillaries sprout from existing interrupted ones at the edge of the wound in analogy to the blood angiogenesis case; 2) lymphatic endothelial cells first pool in the wound region following the lymph flow and then, once sufficiently populated, start to form a network. Here we present two PDE models describing lymphangiogenesis according to these two different hypotheses. Further, we include the effect of advection due to interstitial flow and lymph flow coming from open capillaries. The variables represent different cell densities and growth factor concentrations, and where possible the parameters are estimated from biological data.     The Sneeuberg: A new centre of floristic endemism on the Great Escarpment, South Africa

    The Sneeuberg mountain complex (Eastern Cape) comprises one of the most prominent sections of the Great Escarpment in southern Africa but until now has remained one of the botanically least known regions. The Sneeuberg is a discrete orographical entity, being delimited in the east by the Great Fish River valley, in the west by the Nelspoort Interval, to the south by the Plains of Camdeboo, and to the north by the Great Karoo pediplain. The highest peaks range from 2278 to 2504 m above sea level, and the summit plateaux range from 1800 to 2100 m. Following extensive literature review and a detailed collecting programme, the Sneeuberg is reported here as having a total flora of 1195 species of which 107 (9%) are alien species, 33 (2.8%) are endemic, and 13 (1.1%) near-endemic. Five species previously reported as Drakensberg Alpine Centre (DAC) endemics are now known to occur in the Sneeuberg (representing range extensions of some 300–500 km). One-hundred-and-five species (8.8%) are DAC near-endemics, with the Sneeuberg being the western limit for most of these. Ten species (0.8%) represent disjunctions across the Karoo Interval from the Cape Floristic Region (CFR) to the Sneeuberg. In all, some 23 significant range extensions, eight new species, and several rediscoveries are recorded. We conclude by recognising the Sneeuberg as a new centre of endemism along the Great Escarpment, with floristic affinities with the Albany Centre and the DAC, and links to the CFR

    Advancing an energy justice perspective of fuel poverty: Household vulnerability and domestic retrofit policy in the United Kingdom

    The concept of energy justice has brought philosophies of ethics and principles of social justice to bear on a range of contemporary energy issues. More inter-disciplinary and applied endeavours are now needed to take this field forward. One such application is to the issue of fuel poverty and the challenge of retrofitting inefficient housing stock. An energy justice perspective sees fuel poverty as a fundamentally socio-political injustice, not just one of uneven distribution. Starting from this premise, we highlight the multiple injustices faced by two groups who are regarded by policymakers as being particularly vulnerable to fuel poverty: disabled people and low-income families. In the UK, these groups are nominally prioritised within fuel poverty policy, but their complex situations are not always fully appreciated. Building on the theoretical foundations of energy justice, we present an inter-disciplinary dialogue that connects this approach with wider vulnerability research and domestic energy efficiency policy. Specifically, we discuss ‘within group’ heterogeneity (recognition justice), stakeholder engagement in policy and governance (procedural justice) and the overlap of multiple structural inequalities (distributional justice). In each section we illustrate the added value of combining justice and vulnerability conceptualisations by linking them to domestic energy efficiency schemes
