    Targets for the MalI repressor at the divergent Escherichia coliK-12malX-malI promoters

    Random mutagenesis has been used to identify the target DNA sites for the MalI repressor at the divergent Escherichia coli K-12 malX-malI promoters. The malX promoter is repressed by MalI binding to a DNA site located from position -24 to position -9, upstream of the malX promoter transcript start. The malI promoter is repressed by MalI binding from position +3 to position +18, downstream of the malI transcript start. MalI binding at the malI promoter target is not required for repression of the malX promoter. Similarly, MalI binding at the malX promoter target is not required for repression of the malI. Although the malX and malI promoters are regulated by a single DNA site for cyclic AMP receptor protein, they function independently and each is repressed by MalI binding to a different independent operator site

    AcDc - A new code for the NLTE spectral analysis of accretion discs: application to the helium CV AM CVn

    We present a recently developed code for detailed NLTE calculations of accretion disc spectra of cataclysmic variables and compact X-ray binaries. Assuming a radial structure of a standard alpha-disc, the disc is divided into concentric rings. For each disc ring the solution of the radiation transfer equation and the structure equations, comprising the hydrostatic and radiative equilibrium, the population of the atomic levels as well as charge and particle conservation, is done self-consistently. Metal-line blanketing and irradiation by the central object are taken into account. As a first application, we show the influence of different disc parameters on the disc spectrum for the helium cataclysmic variable AM CVn.Comment: 7 pages, 11 figures to be published in A&

    Nanodiamond as a vector for siRNA delivery to Ewing sarcoma cells

    We investigated the ability of diamond nanoparticles (nanodiamonds, NDs) to deliver small interfering RNA (siRNA) in Ewing sarcoma cells, in the perspective of in vivo anti-cancer nucleic acid drug delivery. siRNA was adsorbed onto NDs previously coated with cationic polymer. Cell uptake of NDs has been demonstrated by taking advantage of NDs intrinsic fluorescence coming from embedded color center defects. Cell toxicity of these coated NDs was shown to be low. Consistent with the internalization efficacy, we have shown a specific inhibition of EWS/Fli-1 gene expression at the mRNA and protein level by the ND vectorized siRNA in a serum containing medium

    Testing timed systems modeled by stream X-machines

    Stream X-machines have been used to specify real systems where complex data structures. They are a variety of extended finite state machine where a shared memory is used to represent communications between the components of systems. In this paper we introduce an extension of the Stream X-machines formalism in order to specify systems that present temporal requirements. We add time in two different ways. First, we consider that (output) actions take time to be performed. Second, our formalism allows to specify timeouts. Timeouts represent the time a system can wait for the environment to react without changing its internal state. Since timeous affect the set of available actions of the system, a relation focusing on the functional behavior of systems, that is, the actions that they can perform, must explicitly take into account the possible timeouts. In this paper we also propose a formal testing methodology allowing to systematically test a system with respect to a specification. Finally, we introduce a test derivation algorithm. Given a specification, the derived test suite is sound and complete, that is, a system under test successfully passes the test suite if and only if this system conforms to the specification

    Enhanced rock-slope failure following ice-sheet deglaciation : timing and causes

    This research was supported by NERC Cosmogenic Isotope Analysis Facility [Grant Number: 9046.0308]The temporal pattern of rock-slope failures (RSFs) following Late Pleistocene deglaciation on tectonically stable terrains is controversial: previous studies variously suggest (1) a rapid response due to removal of supporting ice (‘debuttressing’), (2) a progressive decline in RSF frequency, and (3) a millennial-scale delay before peak RSF activity. We test these competing models through beryllium-10 (10Be) exposure dating of five closely-spaced quartzite RSFs on the Isle of Jura, Scotland, to establish the relationship between timing of failure and those of deglaciation, episodes of rapid warming and periods of rapid glacio-isostatic uplift. All five dated RSFs occurred at least 720–2240 years after deglaciation, with the probability of failure peaking ~2 ka after deglaciation, consistent with millennial-scale delay model (3). This excludes debuttressing as an immediate cause of failure, though it is likely that time-dependent stress release due to deglacial unloading resulted in progressive development of failure planes within the rock. Thaw of permafrost ice in joints is unlikely to have been a prime trigger of failure as some RSFs occurred several centuries after the onset of interstadial warming. Conversely, the timespan of the RSFs coincides with the period of maximum glacio-isostatic crustal uplift, suggesting that failure was triggered by uplift-driven seismic events acting on fractured rock masses. Implications of this and related research are: (1) that retreat of the last Pleistocene ice sheets across tectonically-stable mountainous terrains was succeeded by a period of enhanced rock-slope failure due to deglacial unloading and probably uplift-driven seismicity; (2) that the great majority of RSFs in the British Isles outside the limits of Loch Lomond Stadial (= Younger Dryas) glaciation are of Lateglacial (pre-Holocene) age; and (3) numerous RSFs must also have occurred inside Loch Lomond Stadial (LLS) glacial limits, but that runout debris was removed by LLS glaciers.PostprintPeer reviewe

    A Featured-Based Strategy for Stereovision Matching in Sensors with Fish-Eye Lenses for Forest Environments

    This paper describes a novel feature-based stereovision matching process based on a pair of omnidirectional images in forest stands acquired with a stereovision sensor equipped with fish-eye lenses. The stereo analysis problem consists of the following steps: image acquisition, camera modelling, feature extraction, image matching and depth determination. Once the depths of significant points on the trees are obtained, the growing stock volume can be estimated by considering the geometrical camera modelling, which is the final goal. The key steps are feature extraction and image matching. This paper is devoted solely to these two steps. At a first stage a segmentation process extracts the trunks, which are the regions used as features, where each feature is identified through a set of attributes of properties useful for matching. In the second step the features are matched based on the application of the following four well known matching constraints, epipolar, similarity, ordering and uniqueness. The combination of the segmentation and matching processes for this specific kind of sensors make the main contribution of the paper. The method is tested with satisfactory results and compared against the human expert criterion

    Parental Substance Abuse As an Early Traumatic Event. Preliminary Findings on Neuropsychological and Personality Functioning in Young Drug Addicts Exposed to Drugs Early.

    open5noParental substance use is a major risk factor for child development, heightening the risk of drug problems in adolescence and young adulthood, and exposing offspring to several types of traumatic events. First, prenatal drug exposure can be considered a form of trauma itself, with subtle but long-lasting sequelae at the neuro-behavioral level. Second, parents’ addiction often entails a childrearing environment characterized by poor parenting skills, disadvantaged contexts and adverse childhood experiences (ACEs), leading to dysfunctional outcomes. Young adults born from/raised by parents with drug problems and diagnosed with a Substance Used Disorder (SUD) themselves might display a particularly severe condition in terms of cognitive deficits and impaired personality function. This preliminary study aims to investigate the role of early exposure to drugs as a traumatic event, capable of affecting the psychological status of young drug addicts. In particular, it intends to examine the neuropsychological functioning and personality profile of young adults with severe SUDs who were exposed to drugs early in their family context. The research involved three groups, each consisting of 15 young adults (aged 18–24): a group of inpatients diagnosed with SUDs and exposed to drugs early, a comparison group of non-exposed inpatients and a group of non-exposed youth without SUDs. A neuropsychological battery (Esame Neuropsicologico Breve-2), an assessment procedure for personality disorders (Shedler-Westen Assessment Procedure-200) and the Symptom CheckList-90-Revised were administered. According to present preliminary results, young drug addicts exposed to drugs during their developmental age were characterized by elevated rates of neuropsychological impairments, especially at the expense of attentive and executive functions (EF); personality disorders were also common but did not differentiate them from non-exposed youth with SUDs.     Veterinary decision making in relation to metritis - a qualitative approach to understand the background for variation and bias in veterinary medical records

    <p>Abstract</p> <p>Background</p> <p>Results of analyses based on veterinary records of animal disease may be prone to variation and bias, because data collection for these registers relies on different observers in different settings as well as different treatment criteria. Understanding the human influence on data collection and the decisions related to this process may help veterinary and agricultural scientists motivate observers (veterinarians and farmers) to work more systematically, which may improve data quality. This study investigates qualitative relations between two types of records: 1) 'diagnostic data' as recordings of metritis scores and 2) 'intervention data' as recordings of medical treatment for metritis and the potential influence on quality of the data.</p> <p>Methods</p> <p>The study is based on observations in veterinary dairy practice combined with semi-structured research interviews of veterinarians working within a herd health concept where metritis diagnosis was described in detail. The observations and interviews were analysed by qualitative research methods to describe differences in the veterinarians' perceptions of metritis diagnosis (scores) and their own decisions related to diagnosis, treatment, and recording.</p> <p>Results</p> <p>The analysis demonstrates how data quality can be affected during the diagnostic procedures, as interaction occurs between diagnostics and decisions about medical treatments. Important findings were when scores lacked consistency within and between observers (variation) and when scores were adjusted to the treatment decision already made by the veterinarian (bias). The study further demonstrates that veterinarians made their decisions at 3 different levels of focus (cow, farm, population). Data quality was influenced by the veterinarians' perceptions of collection procedures, decision making and their different motivations to collect data systematically.</p> <p>Conclusion</p> <p>Both variation and bias were introduced into the data because of veterinarians' different perceptions of and motivations for decision making. Acknowledgement of these findings by researchers, educational institutions and veterinarians in practice may stimulate an effort to improve the quality of field data, as well as raise awareness about the importance of including knowledge about human perceptions when interpreting studies based on field data. Both recognitions may increase the usefulness of both within-herd and between-herd epidemiological analyses.</p

    Enhanced formation of giant cells in common variable immunodeficiency: Relation to granulomatous disease.

    Peripheral monocytes from patients with common variable immunodeficiency (CVID) had on average a 2 fold greater tendency to form giant cells in medium without additional cytokines. Giant cell formation was faster and 3 to 5 fold higher in most CVID cells compared to normal. Addition of IL4, GMCSF, IFNγ, TNFa and both T cell and monocyte conditioned media promoted monocyte fusion of some CVID individuals over 5 fold the normal average level, with combinations of cytokines and monokines acting synergistically. The reduction of normal giant cell formation by anti-IFNγ antibody and a greater tendency of CVID cells to fuse in immunoglobulin conditioned media suggests that standard IVIg treatment contributes to granuloma formation. CVID and normal giant cells expressed similar levels of phenotypic molecules and had similar phagocytic activity. Monocytes from many CVID patients have an elevated tendency to fuse which may explain the high incidence of granulomatous complications in CVID
