    Oxidative stress by the mitochondrial monoamine oxidase B mediates calcium pyrophosphate crystal-induced arthritis

    Objective: Calcium pyrophosphate (CPP) crystal deposition in the joints is associated with a heterogeneous set of debilitating syndromes characterized by inflammation and pain, for which no effective therapies are currently available. As we found that the mitochondrial enzyme monoamine oxidase B (MAO-B) plays a fundamental role in promoting inflammatory pathways, this study aims at assessing the efficacy of two clinical-grade inhibitors (iMAO-Bs) in preclinical models of this disease, to pave the way for a novel treatment. Methods: We tested our hypothesis in two murine models of CPP-induced arthritis, by measuring cytokine and chemokine levels, along with immune cell recruitment. iMAO-Bs (rasagiline and safinamide) were administered either before or after crystal injection. To elucidate the molecular mechanism, we challenged in vitro primed macrophages with CPP crystals and assessed the impact of iMAO-Bs in dampening proinflammatory cytokines and in preserving mitochondrial function. Results: Both in preventive and therapeutic in vivo protocols, iMAO-Bs blunted the release of proinflammatory cytokines (interleukin (IL)-6 and IL1-β) and chemokines (CXCL10, CXCL1, CCL2 and CCL5) (n>6 mice/group). Importantly, they also significantly reduced ankle swelling (50.3% vs 17.1% [P<0.001] and 23.1% [P=0.005] for rasagiline and safinamide, respectively). Mechanistically, iMAO-Bs dampened the burst of reactive oxygen species (ROS) and the mitochondrial dysfunction triggered by CPP crystals in isolated macrophages. Moreover, iMAO-Bs blunted cytokine secretion and NLRP3 inflammasome activation through inhibition of the NF-κB and STAT3 pathways. Conclusion: iMAO-Bs dampen inflammation in murine models of crystal-induced arthropathy, thereby uncovering MAO-B as a promising target to treat these diseases

    Urinary Metabolites Reveal Hyperinsulinemia and Insulin Resistance in Polycystic Ovarian Syndrome (PCOS)

    The identification of insulin resistance and hyperinsulinemia in polycystic ovary syndrome (PCOS) is not a minor issue. The homeostasis model assessment of insulin resistance index (HOMA) is the most used index of IR (Insulin Resistance), validated in overweight and obese patients but not in normal-weight PCOS subjects, who can still present with increased insulin secretion by an oral glucose tolerance test [OGTT]. The evaluation of insulin secretion and resistance represents a still unresolved problem. The aim of this study is to identify a possible yet noninvasive method to properly evaluate the insulin metabolism in young non-diabetic subjects. Girls aged 14–22 years, afferent to the center of Gynecological Diseases in Childhood and Adolescence of Cagliari (Italy), were screened for PCOS. A total of 42 subjects comprised the study group. Hormonal assays, OGTT, transabdominal (TA) or transvaginal (TV) US, and urine collection for 1H-NMR analysis were assayed in the early follicular phase. A 1H-NMR coupled multivariate statistical analysis was performed. The OPLS model indicated that the NMR profile of urine had a good fit and prediction ability for the AUC OGTT with R2 = 0.813. Metabolomics can be a promising tool to the potential identification of biomarkers of an exaggerated insulin response to OGTT and can encourage substantial progress for a more accurate and early diagnosis in PCOS

    Studio degli effetti tossici del ritardante di fiamma PBDE-47 su vongola Chamelea gallina (Linnaeus, 1758)

    Lo studio ha avuto l'obiettivo di valutare gli effetti del 2,2',4,4'-tetrabromodifeniletere (PBDE‑47) su vongola Chamelea gallina (Venus gallina secondo la normativa commerciale vigente). I PBDE, impiegati in diversi prodotti industriali come ritardanti di fiamma, sono annoverati tra le sostanze pericolose dalla Direttiva 2011/65/UE. Si tratta di composti bioaccumulabili, ritenuti interferenti endocrini, genotossici e neurotossici, praticamente ubiquitari, la cui concentrazione nell'ambiente, negli ultimi anni, è aumentata in maniera considerevole. Il presente studio ha avuto l'obiettivo di verificare gli effetti del PBDE-47 su Chamelea gallina: potere tossico ed eventuali effetti dannosi sulle gonadi, capacità di bioaccumulo nei tessuti e possibile ingresso nella catena trofica. La ricerca si è avvalsa di prove sperimentali a 96 h e a 14-21gg su esemplari di vongola stabulati in acqua marina filtrata. Le prove sono state precedute da un periodo di adattamento dei molluschi della durata di 5-7gg. Le vongole sono state alimentate con alghe marine (Dunaliella tertiolecta). La scelta del composto tossico PBDE-47 è stata effettuata in considerazione delle maggiori concentrazioni, tra i congeneri di PBDE, riscontrate in alcune specie acquatiche. Lo studio ha evidenziato che le concentrazioni impiegate del contaminante non hanno alterato le funzioni vitali, causato livelli significativi di mortalità e determinato alterazioni evidenti alle gonadi di Chamelea gallina. La ricerca ha evidenziato, comunque, il potere di bioaccumulo del mollusco bivalve, permettendo al PBDE-47 l'ingresso nella catena trofica

    The J2-Immortalized Murine Macrophage Cell Line Displays Phenotypical and Metabolic Features of Primary BMDMs in Their M1 and M2 Polarization State

    Macrophages are immune cells that are important for the development of the defensive front line of the innate immune system. Following signal recognition, macrophages undergo activation toward specific functional states, consisting not only in the acquisition of specific features but also of peculiar metabolic programs associated with each function. For these reasons, macrophages are often isolated from mice to perform cellular assays to study the mechanisms mediating immune cell activation. This requires expensive and time-consuming breeding and housing of mice strains. To overcome this issue, we analyzed an in-house J2-generated immortalized macrophage cell line from BMDMs, both from a functional and metabolic point of view. By assaying the intracellular and extracellular metabolism coupled with the phenotypic features of immortalized versus primary BMDMs, we concluded that classically and alternatively immortalized macrophages display similar phenotypical, metabolic and functional features compared to primary cells polarized in the same way. Our study validates the use of this immortalized cell line as a suitable model with which to evaluate in vitro how perturbations can influence the phenotypical and functional features of murine macrophages

    Postpartum depression screening through artificial intelligence: preliminary data through the Talking About algorithm

    Postpartum depression (PD) is the most widespread perinatal psy­chiatric disorder, also representing the most frequent non-obstetric birth-related complication. From an epidemiological point of view, it has an average prevalence of 17-18% worldwide. This psychiatric disorder may have long-standing effects on the health of both the mother and the child, but also on the relationship with the partner (including paternal PD). Therefore, an early diagnosis is fundamental to treat this disorder immediately and avoid such complications. Talking About, by the company GPI (Trento, Italy), is a project focused on voice analysis as a medium to access human emotions. It consists of a series of Speech Emotion Recognition (SER) algorithms. The aim of the study is to evaluate the application of the artificial intelligence (AI) algorithm Talking About on the mothers’ emotions analysis. Talking About investigates the unconscious aspects of voice that usually cannot be controlled or voluntarily modified, aiming at identifying the subjects’ emotions. Thus, all bias, characterising all classic screening questionnaires, should be neutralised, achieving a sharper overview of the mothers’ emotional state. The mother’s emotional results are displayed in 5 main categories: 2 positive, 3 negative. This study has engaged a total of 154 mothers who gave birth at the “Policlinico Universitario D. Casula” and/or carried out a pediatric examination at the “ambulatorio SOS MAMI” (which is a PostNatal Care Service). They underwent both the Edinburgh Postnatal Depression Scale (EPDS) test and the Talking About voice test. Despite this study’s sample limitations, our preliminary data related to PD symptoms identification are promising and encouraging, leading the way to further investigations related to the application of AI as a PD screening support. Indeed, further studies are needed to improve our knowledge on this topic and possibly apply this tool in clinical practice in the future, even considering PD of the father

    Digital tools for supporting climate-informed agroecological transition in beef production in Brazil

    KEY MESSAGES ◼ Despite many available digital tools in the digital ecosystem, many neglect smallholders’ ranching. Also, they focus solely on performance indicators, lacking climate-informed agroecological features. ◼ Mitigation features are included only by performance assessment tools, although only by half of them. ◼ Adaptation features in digital tools include mitigation recommendations, access to pest and disease information or early warning, and product diversification. ◼ Of the digital tools providing performance assessment (6 total), all include agroecological principles, three included adaptation, and all mitigation indicators on average per tool. ◼ Social inclusion and co-design features are related to data protection for farmers, direct farmer contributions (on the improvement of the digital tools), and safety measures, especially amongst women. ◼ Tools with socially inclusive communication features are limited. Most of the tools reviewed here have more than one way of engaging farm users (IVR, SMS, etc.), and allow the integration of other tools

    Differential cross section measurements for the production of a W boson in association with jets in proton–proton collisions at √s = 7 TeV

    Measurements are reported of differential cross sections for the production of a W boson, which decays into a muon and a neutrino, in association with jets, as a function of several variables, including the transverse momenta (pT) and pseudorapidities of the four leading jets, the scalar sum of jet transverse momenta (HT), and the difference in azimuthal angle between the directions of each jet and the muon. The data sample of pp collisions at a centre-of-mass energy of 7 TeV was collected with the CMS detector at the LHC and corresponds to an integrated luminosity of 5.0 fb[superscript −1]. The measured cross sections are compared to predictions from Monte Carlo generators, MadGraph + pythia and sherpa, and to next-to-leading-order calculations from BlackHat + sherpa. The differential cross sections are found to be in agreement with the predictions, apart from the pT distributions of the leading jets at high pT values, the distributions of the HT at high-HT and low jet multiplicity, and the distribution of the difference in azimuthal angle between the leading jet and the muon at low values.United States.     Impacts of the Tropical Pacific/Indian Oceans on the Seasonal Cycle of the West African Monsoon

    The current consensus is that drought has developed in the Sahel during the second half of the twentieth century as a result of remote effects of oceanic anomalies amplified by local land–atmosphere interactions. This paper focuses on the impacts of oceanic anomalies upon West African climate and specifically aims to identify those from SST anomalies in the Pacific/Indian Oceans during spring and summer seasons, when they were significant. Idealized sensitivity experiments are performed with four atmospheric general circulation models (AGCMs). The prescribed SST patterns used in the AGCMs are based on the leading mode of covariability between SST anomalies over the Pacific/Indian Oceans and summer rainfall over West Africa. The results show that such oceanic anomalies in the Pacific/Indian Ocean lead to a northward shift of an anomalous dry belt from the Gulf of Guinea to the Sahel as the season advances. In the Sahel, the magnitude of rainfall anomalies is comparable to that obtained by other authors using SST anomalies confined to the proximity of the Atlantic Ocean. The mechanism connecting the Pacific/Indian SST anomalies with West African rainfall has a strong seasonal cycle. In spring (May and June), anomalous subsidence develops over both the Maritime Continent and the equatorial Atlantic in response to the enhanced equatorial heating. Precipitation increases over continental West Africa in association with stronger zonal convergence of moisture. In addition, precipitation decreases over the Gulf of Guinea. During the monsoon peak (July and August), the SST anomalies move westward over the equatorial Pacific and the two regions where subsidence occurred earlier in the seasons merge over West Africa. The monsoon weakens and rainfall decreases over the Sahel, especially in August.Peer reviewe

    Risk portfolio on modern finance has become increasingly technical, requiring the use of sophisticated mathematical tools in both research and practice. Since companies cannot insure themselves completely against risk, as human incompetence in predicting the future precisely that written in Al-Quran surah Luqman verse 34, they have to manage it to yield an optimal portfolio. The objective here is to minimize the variance among all portfolios, or alternatively, to maximize expected return among all portfolios that has at least a certain expected return. Furthermore, this study focuses on optimizing risk portfolio so called Markowitz MVO (Mean-Variance Optimization). Some theoretical frameworks for analysis are arithmetic mean, geometric mean, variance, covariance, linear programming, and quadratic programming. Moreover, finding a minimum variance portfolio produces a convex quadratic programming, that is minimizing the objective function ðð¥with constraintsð ð 𥠥 ðandð´ð¥ = ð. The outcome of this research is the solution of optimal risk portofolio in some investments that could be finished smoothly using MATLAB R2007b software together with its graphic analysis

    Search for heavy resonances decaying to two Higgs bosons in final states containing four b quarks

    A search is presented for narrow heavy resonances X decaying into pairs of Higgs bosons (H) in proton-proton collisions collected by the CMS experiment at the LHC at root s = 8 TeV. The data correspond to an integrated luminosity of 19.7 fb(-1). The search considers HH resonances with masses between 1 and 3 TeV, having final states of two b quark pairs. Each Higgs boson is produced with large momentum, and the hadronization products of the pair of b quarks can usually be reconstructed as single large jets. The background from multijet and t (t) over bar events is significantly reduced by applying requirements related to the flavor of the jet, its mass, and its substructure. The signal would be identified as a peak on top of the dijet invariant mass spectrum of the remaining background events. No evidence is observed for such a signal. Upper limits obtained at 95 confidence level for the product of the production cross section and branching fraction sigma(gg -> X) B(X -> HH -> b (b) over barb (b) over bar) range from 10 to 1.5 fb for the mass of X from 1.15 to 2.0 TeV, significantly extending previous searches. For a warped extra dimension theory with amass scale Lambda(R) = 1 TeV, the data exclude radion scalar masses between 1.15 and 1.55 TeV
