    Puerarin attenuates pressure overload-induced cardiac hypertrophy

    AbstractBackgroundPuerarin is the most abundant isoflavonoid in kudzu root. It has been used to treat angina pectoris and myocardial infarction clinically. However, little is known about the effect of puerarin on cardiac hypertrophy.MethodsAortic banding (AB) was performed to induce cardiac hypertrophy in mice. Puerarin premixed in diets was administered to mice after one week of AB. Echocardiography and catheter-based measurements of hemodynamic parameters were performed at 7 weeks after starting puerarin treatment (8 weeks post-surgery). The extent of cardiac hypertrophy was also evaluated by pathological and molecular analyses of heart samples. Cardiomyocyte apoptosis was assessed by measuring Bax and Bcl-2 protein expression and terminal deoxynucleotidyl transferase dUTP nick end labeling staining. In addition, the inhibitory effect of puerarin (1μM, 5μM, 10μM, 20μM, 40μM) on mRNA expression of atrial natriuretic peptide (ANP) and B-type natriuretic peptide (BNP) in Ang II (1μM)-stimulated H9c2 cells was investigated using quantitative real-time reverse transcription-polymerase chain reaction.ResultsEchocardiography and catheter-based measurements of hemodynamic parameters at 7 weeks revealed the amelioration of systolic and diastolic abnormalities. Puerarin also decreased cardiac fibrosis in AB mice. Moreover, the beneficial effect of puerarin was associated with the normalization in gene expression of hypertrophic and fibrotic markers. Further studies showed that pressure overload significantly induced the activation of phosphoinositide 3-kinase (PI3K)/Akt signaling and c-Jun N-terminal kinase (JNK) signaling, which was blocked by puerarin treatment. Cardiomyocyte apoptosis and induction of Bax in response to AB were suppressed by puerarin. Furthermore, the increased mRNA expression of ANP and BNP induced by Ang II (1μM) was restrained to a different extent by different concentrations of puerarin.ConclusionPuerarin may have an ability to retard the progression of cardiac hypertrophy and apoptosis which is probably mediated by the blockade of PI3K/Akt and JNK signaling pathways

    Activating Transcription Factor 3 Deficiency Promotes Cardiac Hypertrophy, Dysfunction, and Fibrosis Induced by Pressure Overload

    Activating transcription factor 3 (ATF3), which is encoded by an adaptive-response gene induced by various stimuli, plays an important role in the cardiovascular system. However, the effect of ATF3 on cardiac hypertrophy induced by a pathological stimulus has not been determined. Here, we investigated the effects of ATF3 deficiency on cardiac hypertrophy using in vitro and in vivo models. Aortic banding (AB) was performed to induce cardiac hypertrophy in mice. Cardiac hypertrophy was estimated by echocardiographic and hemodynamic measurements and by pathological and molecular analysis. ATF3 deficiency promoted cardiac hypertrophy, dysfunction and fibrosis after 4 weeks of AB compared to the wild type (WT) mice. Furthermore, enhanced activation of the MEK-ERK1/2 and JNK pathways was found in ATF3-knockout (KO) mice compared to WT mice. In vitro studies performed in cultured neonatal mouse cardiomyocytes confirmed that ATF3 deficiency promotes cardiomyocyte hypertrophy induced by angiotensin II, which was associated with the amplification of MEK-ERK1/2 and JNK signaling. Our results suggested that ATF3 plays a crucial role in the development of cardiac hypertrophy via negative regulation of the MEK-ERK1/2 and JNK pathways

    A small-molecule inhibitor of the aberrant transcription factor CBFβ-SMMHC delays leukemia in mice

The transcription factor fusion CBFβ-SMMHC (core binding factor β and the smooth-muscle myosin heavy chain), expressed in AML with the chromosome inversion inv(16)(p13q22), outcompetes wild-type CBFβ for binding to the transcription factor RUNX1, deregulates RUNX1 activity in hematopoiesis, and induces AML. Current inv(16) AML treatment with nonselective cytotoxic chemotherapy results in a good initial response but limited long-term survival. Here, we report the development of a protein-protein interaction inhibitor, AI-10-49, that selectively binds to CBFβ-SMMHC and disrupts its binding to RUNX1. AI-10-49 restores RUNX1 transcriptional activity, displays favorable pharmacokinetics, and delays leukemia progression in mice. Treatment of primary inv(16) AML patient blasts with AI-10-49 triggers selective cell death. These data suggest that direct inhibition of the oncogenic CBFβ-SMMHC fusion protein may be an effective therapeutic approach for inv(16) AML, and they provide support for transcription factor targeted therapy in other cancers

    Pharmacologic inhibition of the Menin-MLL interaction blocks progression of MLL leukemia in vivo

    Chromosomal translocations affecting mixed lineage leukemia gene (MLL) result in acute leukemias resistant to therapy. The leukemogenic activity of MLL fusion proteins is dependent on their interaction with menin, providing basis for therapeutic intervention. Here we report the development of highly potent and orally bioavailable small-molecule inhibitors of the menin-MLL interaction, MI-463 and MI-503, and show their profound effects in MLL leukemia cells and substantial survival benefit in mouse models of MLL leukemia. Finally, we demonstrate the efficacy of these compounds in primary samples derived from MLL leukemia patients. Overall, we demonstrate that pharmacologic inhibition of the menin-MLL interaction represents an effective treatment for MLL leukemias in vivo and provide advanced molecular scaffold for clinical lead identification

    Phase II of the LAMOST-Kepler/K2 survey. I. Time series of medium-resolution spectroscopic observations

    Phase \RNum{2} of the LAMOST-{\sl Kepler/K}2 survey (LK-MRS), initiated in 2018, aims at collecting medium-resolution spectra (R7,500R\sim7,500; hereafter MRS) for more than 50,00050,000 stars with multiple visits (60\sim60 epochs) over a period of 5 years (2018 September to 2023 June). We selected 20 footprints distributed across the {\sl Kepler} field and six {\sl K}2 campaigns, with each plate containing a number of stars ranging from 2,000\sim2,000 to 3,000\sim 3,000. During the first year of observations, the LK-MRS has already collected 280,000\sim280,000 and 369,000\sim369,000 high-quality spectra in the blue and red wavelength range, respectively. The atmospheric parameters and radial velocities for 259,000\sim259,000 spectra of 21,05321,053 targets were successfully calculated by the LASP pipeline. The internal uncertainties for the effective temperature, surface gravity, metallicity, and radial velocity are found to be 100100\,K, 0.150.15\,dex, 0.090.09\,dex, and 1.001.00\,km\,s1^{-1}, respectively. We found 14,99714,997, 20,09120,091, and 1,5141,514 stars in common with the targets from the LAMOST low-resolution survey (LRS), GAIA and APOGEE, respectively, corresponding to a fraction of 70%\sim70\%, 95%\sim95\% and 7.2%\sim7.2\%. In general, the parameters derived from LK-MRS spectra are consistent with those obtained from the LRS and APOGEE spectra, but the scatter increases as the surface gravity decreases when comparing with the measurements from APOGEE. A large discrepancy is found with the GAIA values of the effective temperature. The comparisons of radial velocities of LK-MRS to GAIA and LK-MRS to APOGEE nearly follow an Gaussian distribution with a mean μ1.10\mu\sim1.10 and 0.730.73\,km\,s1^{-1}, respectively.Comment: 24 pages, 15 figures, 4 tables, ApJS, accepte

    Characterization and source identification of fine particulate matter in urban Beijing during the 2015 Spring Festival

    The Spring Festival (SF) is the most important holiday in China for family reunion and tourism. During the 2015 SF an intensive observation campaign of air quality was conducted to study the impact of the anthropogenic activities and the dynamic characteristics of the sources. During the study period, pollution episodes frequently occurred with 12 days exceeding the Chinese Ambient Air Quality Standards for 24-h average PM2.5 (75 μg/m3), even 8 days with exceeding 150 μg/m3. The daily maximum PM2.5 concentration reached 350 μg/m3 while the hourly minimum visibility was <0.8 km. Three pollution episodes were selected for detailed analysis including chemical characterization and diurnal variation of the PM2.5 and its chemical composition, and sources were identified using the Positive Matrix Factorization model. The first episode occurring before the SF was characterized by more formation of SO42− and NO3− and high crustal enrichment factors for Ag, As, Cd, Cu, Hg, Pb, Se and Zn and seven categories of pollution sources were identified, whereby vehicle emission contributed 38% to the PM2.5. The second episode occurring during the SF was affected heavily by large-scale firework emissions, which led to a significant increase in SO42−, Cl−, OC, K and Ba; these emissions were the largest contributor to the PM2.5 accounting for 36%. During the third episode occurring after the SF, SO42−, NO3−, NH4+ and OC were the major constituents of the PM2.5 and the secondary source was the dominant source with a contribution of 46%. The results provide a detailed understanding on the variation in occurrence, chemical composition and sources of the PM2.5 as well as of the gaseous pollutants affected by the change in anthropogenic activities in Beijing throughout the SF. They highlight the need for limiting the firework emissions during China's most important traditional festival

    Juxtaposing BTE and ATE – on the role of the European insurance industry in funding civil litigation

    One of the ways in which legal services are financed, and indeed shaped, is through private insurance arrangement. Two contrasting types of legal expenses insurance contracts (LEI) seem to dominate in Europe: before the event (BTE) and after the event (ATE) legal expenses insurance. Notwithstanding institutional differences between different legal systems, BTE and ATE insurance arrangements may be instrumental if government policy is geared towards strengthening a market-oriented system of financing access to justice for individuals and business. At the same time, emphasizing the role of a private industry as a keeper of the gates to justice raises issues of accountability and transparency, not readily reconcilable with demands of competition. Moreover, multiple actors (clients, lawyers, courts, insurers) are involved, causing behavioural dynamics which are not easily predicted or influenced. Against this background, this paper looks into BTE and ATE arrangements by analysing the particularities of BTE and ATE arrangements currently available in some European jurisdictions and by painting a picture of their respective markets and legal contexts. This allows for some reflection on the performance of BTE and ATE providers as both financiers and keepers. Two issues emerge from the analysis that are worthy of some further reflection. Firstly, there is the problematic long-term sustainability of some ATE products. Secondly, the challenges faced by policymakers that would like to nudge consumers into voluntarily taking out BTE LEI
