    Syntheses, Structures, and Characterizations of 2,2\u27,7,7\u27-Tetrasubstituted 1,1\u27-Ethynylenedinaphthalenes.

    In order to study intramolecular enzyme catalysis, chemical models have been designed to quantify the proximity and orientation effects which occur in carboxylate catalyzed hydrolyses. As part of this broader study, various 2,2\sp\prime,7,7\sp\prime-tetrasubstituted 1,1\sp\prime-ethynylenedinaphthalenes are synthesized from 2,7-dihydroxynaphthalene. Two different strategies are used to obtain the title compounds. The first strategy involves palladium-mediated couplings between electron-poor aryl iodides and electron-rich arylethynes and allows synthesis of unsymmetrical binaphthylethynes. The second approach couples two 2,7-disubstituted naphthalenes with tetrachlorocyclopropene in the presence of aluminum chloride, but only symmetrical binaphthylethynes are obtained. During the course of this work, a number of 1,2,7-trisubstituted naphthalenes, some of them previously unknown, have been prepared. Several new or improved procedures have also been developed, most notably the monomethylation of 2,7-naphthalenediol in a two solvent system, a new three-step preparation of methyl 7-methoxy-2-naphthoate, and a new two-step synthetic method for the acyl \to ethynyl conversion in electron-rich aromatic rings. Both strategies produce several new binaphthylethynes, two of which are unsymmetrical. These two compounds represent the first examples of this type of molecule. Through NMR spectroscopic techniques and X-ray diffraction, a large amount of spectral and structural data on naphthalenes has been assembled and discussed.     Benefits of low-dose inhaled fluticasone on airway response and inflammation in mild asthma

    SummaryRationaleCurrent guidelines suggest that asthma should be controlled with the lowest dose of maintenance medication required.ObjectivesTo evaluate the effects of a low dose of inhaled corticosteroid compared to a placebo, on airway inflammation and responsiveness in patients with mild symptomatic asthma.MethodsIn this randomized double-blind, placebo-controlled, parallel group study, we looked at the influence of inhaled fluticasone propionate 250μg/day for 3 months followed by 100μg/day for 9 months on airway inflammation and methacholine responsiveness in non-smoking subjects with mild allergic asthma. Subjects were evaluated at baseline and 3, 6, 9 and 12 months after treatments; a 2-week evaluation of respiratory symptoms and peak expiratory flow measurements was done before each visit.ResultsFifty-seven subjects completed the 3-month study period. Airway responsiveness, expressed as the PC20 methacholine, increased by 0.27 and 1.14 doubling concentrations, respectively, in placebo-treated (n=33) and in fluticasone-treated (n=24) asthmatic subjects (p=0.03). An additional improvement in PC20 up to 2.16 doubling concentrations was observed in the fluticasone-treated group during the 9-month lower-dose treatment (p=0.0004, end of low-dose period compared with placebo). Sputum eosinophil counts decreased after 3 months of fluticasone 250μg/day compared with placebo (p<0.0001) and remained in the normal range during the 9-month lower-dose treatment. Respiratory symptoms and peak expiratory flows did not change significantly throughout the study in both groups.ConclusionIn mild asthma, keeping a regular minimal dose of ICS after asthma control has been achieved, may lead to a further reduction in airway responsiveness and keep sputum eosinophil count within the normal range

    Amphidromy Links a Newly Documented Fish Community of Continental Australian Streams, to Oceanic Islands of the West Pacific

    BACKGROUND: Indo-Pacific high island streams experience extreme hydrological variation, and are characterised by freshwater fish species with an amphidromous life history. Amphidromy is a likely adaptation for colonisation of island streams following stochastic events that lead to local extirpation. In the Wet Tropics of north-eastern Australia, steep coastal mountain streams share similar physical characteristics to island systems. These streams are poorly surveyed, but may provide suitable habitat for amphidromous species. However, due to their ephemeral nature, common non-diadromous freshwater species of continental Australia are unlikely to persist. Consequently, we hypothesise that coastal Wet Tropics streams are faunally more similar, to distant Pacific island communities, than to nearby faunas of large continental rivers. METHODS/PRINCIPAL FINDINGS: Surveys of coastal Wet Tropics streams recorded 26 species, 10 of which are first records for Australia, with three species undescribed. This fish community is unique in an Australian context in that it contains mostly amphidromous species, including sicydiine gobies of the genera Sicyopterus, Sicyopus, Smilosicyopus and Stiphodon. Species presence/absence data of coastal Wet Tropics streams were compared to both Wet Tropics river networks and Pacific island faunas. ANOSIM indicated the fish fauna of north-eastern Australian coastal streams were more similar to distant Pacific islands (R = 0.76), than to nearby continental rivers (R = 0.98). MAIN CONCLUSIONS/SIGNIFICANCE: Coastal Wet Tropics streams are faunally more similar to distant Pacific islands (79% of species shared), than to nearby continental fauna due to two factors. First, coastal Wet Tropics streams lack many non-diadromous freshwater fish which are common in nearby large rivers. Second, many amphidromous species found in coastal Wet Tropics streams and Indo-Pacific islands remain absent from large rivers of the Wet Tropics. The evolutionary and conservation significance of this newly discovered Australian fauna requires clarification in the context of the wider amphidromous fish community of the Pacific

    Rain-Use-Efficiency: What it Tells us about the Conflicting Sahel Greening and Sahelian Paradox

    Rain Use Efficiency (RUE), defined as Aboveground Net Primary Production (ANPP) divided by rainfall, is increasingly used to diagnose land degradation. Yet, the outcome of RUE monitoring has been much debated since opposite results were found about land degradation in the Sahel region. The debate is fueled by methodological issues, especially when using satellite remote sensing data to estimate ANPP, and by differences in the ecological interpretation. An alternative method which solves part of these issues relies on the residuals of ANPP regressed against rainfall (“ANPP residuals”). In this paper, we use long-term field observations of herbaceous vegetation mass collected in the Gourma region in Mali together with remote sensing data (GIMMS-3g Normalized Difference Vegetation Index) to estimate ANPP, RUE, and the ANPP residuals, over the period 1984–2010. The residuals as well as RUE do not reveal any trend over time over the Gourma region, implying that vegetation is resilient over that period, when data are aggregated at the Gourma scale. We find no conflict between field-derived and satellite-derived results in terms of trends. The nature (linearity) of the ANPP/rainfall relationship is investigated and is found to have no impact on the RUE and residuals interpretation. However, at odds with a stable RUE, an increased run-off coefficient has been observed in the area over the same period, pointing towards land degradation. The divergence of these two indicators of ecosystem resilience (stable RUE) and land degradation (increasing run-off coefficient) is referred to as the “second Sahelian paradox”. When shallow soils and deep soils are examined separately, high resilience is diagnosed on the deep soil sites. However, some of the shallow soils show signs of degradation, being characterized by decreasing vegetation cover and increasing run-off coefficient. Such results show that contrasted changes may co-exist within a region where a strong overall re-greening pattern is observed, highlighting that both the scale of observations and the scale of the processes have to be considered when performing assessments of vegetation changes and land degradation

    Antimicrobial resistance among migrants in Europe: a systematic review and meta-analysis

    BACKGROUND: Rates of antimicrobial resistance (AMR) are rising globally and there is concern that increased migration is contributing to the burden of antibiotic resistance in Europe. However, the effect of migration on the burden of AMR in Europe has not yet been comprehensively examined. Therefore, we did a systematic review and meta-analysis to identify and synthesise data for AMR carriage or infection in migrants to Europe to examine differences in patterns of AMR across migrant groups and in different settings. METHODS: For this systematic review and meta-analysis, we searched MEDLINE, Embase, PubMed, and Scopus with no language restrictions from Jan 1, 2000, to Jan 18, 2017, for primary data from observational studies reporting antibacterial resistance in common bacterial pathogens among migrants to 21 European Union-15 and European Economic Area countries. To be eligible for inclusion, studies had to report data on carriage or infection with laboratory-confirmed antibiotic-resistant organisms in migrant populations. We extracted data from eligible studies and assessed quality using piloted, standardised forms. We did not examine drug resistance in tuberculosis and excluded articles solely reporting on this parameter. We also excluded articles in which migrant status was determined by ethnicity, country of birth of participants' parents, or was not defined, and articles in which data were not disaggregated by migrant status. Outcomes were carriage of or infection with antibiotic-resistant organisms. We used random-effects models to calculate the pooled prevalence of each outcome. The study protocol is registered with PROSPERO, number CRD42016043681. FINDINGS: We identified 2274 articles, of which 23 observational studies reporting on antibiotic resistance in 2319 migrants were included. The pooled prevalence of any AMR carriage or AMR infection in migrants was 25·4% (95% CI 19·1-31·8; I2 =98%), including meticillin-resistant Staphylococcus aureus (7·8%, 4·8-10·7; I2 =92%) and antibiotic-resistant Gram-negative bacteria (27·2%, 17·6-36·8; I2 =94%). The pooled prevalence of any AMR carriage or infection was higher in refugees and asylum seekers (33·0%, 18·3-47·6; I2 =98%) than in other migrant groups (6·6%, 1·8-11·3; I2 =92%). The pooled prevalence of antibiotic-resistant organisms was slightly higher in high-migrant community settings (33·1%, 11·1-55·1; I2 =96%) than in migrants in hospitals (24·3%, 16·1-32·6; I2 =98%). We did not find evidence of high rates of transmission of AMR from migrant to host populations. INTERPRETATION: Migrants are exposed to conditions favouring the emergence of drug resistance during transit and in host countries in Europe. Increased antibiotic resistance among refugees and asylum seekers and in high-migrant community settings (such as refugee camps and detention facilities) highlights the need for improved living conditions, access to health care, and initiatives to facilitate detection of and appropriate high-quality treatment for antibiotic-resistant infections during transit and in host countries. Protocols for the prevention and control of infection and for antibiotic surveillance need to be integrated in all aspects of health care, which should be accessible for all migrant groups, and should target determinants of AMR before, during, and after migration.
