    Effects of P-MAPA Immunomodulator on Toll-Like Receptors and p53: Potential Therapeutic Strategies for Infectious Diseases and Cancer

The present study aimed to characterize the immunomodulatory effects of P-MAPA on TLRs in vitro and in vivo, as well as to investigate its potential as adjuvant therapy in infectious diseases and cancer. Methods: For these purposes, the activity of P-MAPA on TLRs was assayed in vitro through NF-kappa B activation in HEK293 cells expressing a given TLR, and using an in vivo animal model for bladder cancer (BC). The antimicrobial activity of P-MAPA was tested against Mycobacterium tuberculosis (TB) in vitro in an MIC assay, and in vivo using an aerosol infection model of murine tuberculosis. Antitumor effects of P-MAPA were tested in an animal model with experimentally induced BC. Moxifloxacin (MXF) and Bacillus Calmette-Guerin (BCG) were used as positive controls in the animal models. Results: The results showed that P-MAPA, administered alone or in combination with MXF, induced significant responses in vivo against TB. In contrast, the compound did not show antimicrobial activity in vitro. P-MAPA showed a significant stimulatory effect on human TLR2 and TLR4 in vitro. In BC, TLR2, TLR4 and p53 protein levels were significantly higher in the P-MAPA group than in the BCG group. The most common histopathological changes in each group were papillary carcinoma in BC group, low-grade intraepithelial neoplasia in BCG group and simple hyperplasia in P-MAPA group. Concerning the toxicological analysis performed during BC treatment, P-MAPA did not show evidence for hepatotoxicity and nephrotoxicity. Conclusions: In conclusion, P-MAPA acted as TLR ligand in vitro and improved the immunological status in BC, increasing TLR2 and TLR4 protein levels. P-MAPA immunotherapy was more effective in restoring p53 and TLRs reactivities and showed significantly greater antitumor activity than BCG. The activation of TLRs and p53 may provide a hypothetical mechanism for the therapeutic effects in both cancer and infectious diseases.     Biogeographical distribution, conservation and local use of Amburana acreana (Ducke) A. C. Sm. in the Cacoal-Rondônia region, Brazil

    Amburana acreana (Ducke) A. C. Sm., popularly known as "cerejeira" or emburana, is a tree belonging to the Fabaceae family (Leguminosae: Faboideae) and is included in the list of threatened and endangered species, according to Regulation No. 06 of the Brazilian Ministry of the Environment, dated 23 September 2008, standing out as one of the most vulnerable species. The objective of this study was to analyse the biogeographic distribution of A. acreana in the municipality of Cacoal, located in the Brazilian Amazon. The research was based on field data, complemented by herbarium records, databases, sampling and surveys of the local community to obtain information on the location and abundance of the species. The use of advanced tools such as DIVA-GIS 7.5® and Terra-i® made it possible to visualise the distribution of A. acreana in Brazil, both in its natural habitat (States of Acre, Mato Grosso and Rondônia) and in cultivated areas (Minas Gerais and Rio de Janeiro), extending also to Bolivia and Peru. In the municipality of Cacoal, the presence of the species was confirmed in ten localities, two of which hosted more than six adult individuals. The consideration of the microclimatic conditions of these localities proves to be a crucial element for proposing effective conservation strategies. The study not only provides a detailed overview of the distribution of A. acreana, but also highlights the need to address specific environmental conditions to ensure the long-term conservation of this threatened species in the Amazon region

    Modulation of the pharmacological effects of enzymatically-active PLA2 by BTL-2, an isolectin isolated from the Bryothamnion triquetrum red alga

    <p>Abstract</p> <p>Background</p> <p>An interaction between lectins from marine algae and PLA<sub>2 </sub>from rattlesnake was suggested some years ago. We, herein, studied the effects elicited by a small isolectin (BTL-2), isolated from <it>Bryothamnion triquetrum</it>, on the pharmacological and biological activities of a PLA<sub>2 </sub>isolated from rattlesnake venom (<it>Crotalus durissus cascavella</it>), to better understand the enzymatic and pharmacological mechanisms of the PLA<sub>2 </sub>and its complex.</p> <p>Results</p> <p>This PLA<sub>2 </sub>consisted of 122 amino acids (approximate molecular mass of 14 kDa), its pI was estimated to be 8.3, and its amino acid sequence shared a high degree of similarity with that of other neurotoxic and enzymatically-active PLA<sub>2</sub>s. BTL-2 had a molecular mass estimated in approximately 9 kDa and was characterized as a basic protein. In addition, BTL-2 did not exhibit any enzymatic activity.</p> <p>The PLA<sub>2 </sub>and BTL-2 formed a stable heterodimer with a molecular mass of approximately 24–26 kDa, estimated by molecular exclusion HPLC. In the presence of BTL-2, we observed a significant increase in PLA<sub>2 </sub>activity, 23% higher than that of PLA<sub>2 </sub>alone. BTL-2 demonstrated an inhibition of 98% in the growth of the Gram-positive bacterial strain, <it>Clavibacter michiganensis michiganensis </it>(Cmm), but only 9.8% inhibition of the Gram-negative bacterial strain, <it>Xanthomonas axonopodis </it>pv <it>passiflorae </it>(Xap). PLA<sub>2 </sub>decreased bacterial growth by 27.3% and 98.5% for Xap and Cmm, respectively, while incubating these two proteins with PLA<sub>2</sub>-BTL-2 inhibited their growths by 36.2% for Xap and 98.5% for Cmm.</p> <p>PLA<sub>2 </sub>significantly induced platelet aggregation in washed platelets, whereas BTL-2 did not induce significant platelet aggregation in any assay. However, BTL-2 significantly inhibited platelet aggregation induced by PLA<sub>2</sub>. In addition, PLA<sub>2 </sub>exhibited strong oedematogenic activity, which was decreased in the presence of BTL-2. BTL-2 alone did not induce oedema and did not decrease or abolish the oedema induced by the 48/80 compound.</p> <p>Conclusion</p> <p>The unexpected results observed for the PLA<sub>2</sub>-BTL-2 complex strongly suggest that the pharmacological activity of this PLA<sub>2 </sub>is not solely dependent on the presence of enzymatic activity, and that other pharmacological regions may also be involved. In addition, we describe for the first time an interaction between two different molecules, which form a stable complex with significant changes in their original biological action. This opens new possibilities for understanding the function and action of crude venom, an extremely complex mixture of different molecules.</p

    Caminhos e Lugares: modelo de mobilidade e sistema de assentamento entre os proto-Jê meridionais na região de Campo Belo do Sul, SC

    Throughout this research, I sought to demonstrate the Southern Proto-Jê occupation process in the Campo Belo do Sul region, and how the regional settlement system could be thought of and structured in relation to landscape mobility. By articulating the information it was possible to demonstrate patterns of behavior in relation to different spaces, and how the regional occupation was extended guided by the standards of accessibility and visibility of the settlements, in a cultural landscape. Thus, by relating places, material culture and mobility, through the notion of social construction and use of space, I crossed this complex cultural landscape into four layers: environment, through observation, reconstruction and comparison of environmental characteristics to cultural standards; system, thinking about the dynamics of occupation and use of different spaces; power, relating the patterns of mobility to the political and economic control of the territory, as well as the reaffirmation of socio-cultural identities; and experience, in the form of integrating cognitive aspects into the notion of spatiality, territoriality and transitability. I point out that these relationships are multiple, and are intrinsically connected in the process of occupying 600 years of the region.Sem bolsaBusquei, ao longo dessa pesquisa, demonstrar o processo de ocupação Proto-Jê meridional na região de Campo Belo do Sul, e como o sistema de assentamento regional poderia ser pensado e estruturado em relação à mobilidade na paisagem. Articulando as informações foi possível demonstrar padrões de comportamento em relação a diferentes espaços, e como a ocupação regional se estendeu orientada pelos padrões de acessibilidade e visibilidade dos assentamentos, em uma paisagem cultural. Dessa forma, ao relacionar lugares, cultura material e mobilidade, por meio da noção de construção social e uso do espaço, cruzei essa paisagem cultural complexa em quatro camadas: ambiente, por meio da observação, reconstrução e comparação de características ambientais aos padrões culturais; sistema, pensando as dinâmicas de ocupação e uso de espaços diferentes; poder, relacionando os padrões de mobilidade ao controle político e econômico do território, bem como a reafirmação de identidades sócio-culturais; e experiência, na forma de integrar aspectos cognitivos na noção de espacialidade, territorialidade e transitabilidade. Destaco que essas relações são múltiplas, e estão intrinsecamente conectadas no processo de ocupação de 600 anos da região

    Thermorheological and textural behaviour of gluten-free gels obtained from chestnut and rice flours

    Nowadays, as celiac disease is becoming more common the consumers’ demand for gluten-free products with high nutritional and taste quality is increasing. This work deals with the study of the impact of four novelty gluten-free sources: chestnut flour (Cf), whole rice flour (Rw), Carolino rice flour (Rc) and Agulha rice flour (Ra). Textural, thermorheological and stability performance of gluten-free gels using different experimental techniques were evaluated. Mixed gels were also produced for comparison. Texture parameters were determined from the texture profile analysis using a texturometer. Thermorheological oscillatory measurements were conducted in a stresscontrolled rheometer in order to clarify the kinetics of gel formation and to characterise the structure of the matured gels. The stability of the gels was evaluated using transmittance profiling of the gels under gravitational fields (LUMiSizer®). Texture studies suggested that gels from mixtures of chestnut flour at 30 % and rice flour at 20 % showed the right texture to develop gel-based new desserts. Rheological results showed that the thermal profiles on heating of Cf gels were similar to those obtained for Rw and Ra, whereas Rc gels exhibited a particular pattern. Once the final gelatinisation temperature was achieved, no significant differences on the viscoelastic properties were noticed for all the tested gels. Stability tests showed that gels with Rc should present an industrial advantage over the other assayed formulations, since the stability of these gels is of the order of four times larger

    Observation of associated near-side and away-side long-range correlations in √sNN=5.02  TeV proton-lead collisions with the ATLAS detector

    Two-particle correlations in relative azimuthal angle (Δϕ) and pseudorapidity (Δη) are measured in √sNN=5.02  TeV p+Pb collisions using the ATLAS detector at the LHC. The measurements are performed using approximately 1  μb-1 of data as a function of transverse momentum (pT) and the transverse energy (ΣETPb) summed over 3.1<η<4.9 in the direction of the Pb beam. The correlation function, constructed from charged particles, exhibits a long-range (2<|Δη|<5) “near-side” (Δϕ∼0) correlation that grows rapidly with increasing ΣETPb. A long-range “away-side” (Δϕ∼π) correlation, obtained by subtracting the expected contributions from recoiling dijets and other sources estimated using events with small ΣETPb, is found to match the near-side correlation in magnitude, shape (in Δη and Δϕ) and ΣETPb dependence. The resultant Δϕ correlation is approximately symmetric about π/2, and is consistent with a dominant cos⁡2Δϕ modulation for all ΣETPb ranges and particle pT

    Evaluation of epidermal growth factor receptor mutation status in serum DNA as a predictor of response to gefitinib (IRESSA)

    The aim of this study was to evaluate the usefulness of EGFR mutation status in serum DNA as a means of predicting a benefit from gefitinib (IRESSA) therapy in Japanese patients with non-small cell lung cancer (NSCLC). We obtained pairs of tumour and serum samples from 42 patients treated with gefitinib. EGFR mutation status was determined by a direct sequencing method and by Scorpion Amplification Refractory Mutation System (ARMS) technology. EGFR mutations were detected in the tumour samples of eight patients and in the serum samples of seven patients. EGFR mutation status in the tumours and serum samples was consistent in 39 (92.9%) of the 42 pairs. EGFR mutations were strong correlations between both EGFR mutation status in the tumour samples and serum samples and objective response to gefitinib (P<0.001). Median progression-free survival time was significantly longer in the patients with EGFR mutations than in the patients without EGFR mutations (194 vs 55 days, P=0.016, in tumour samples; 174 vs 58 days, P=0.044, in serum samples). The results suggest that it is feasible to use serum DNA to detect EGFR mutation, and that it's potential as a predictor of response to, and survival on gefitinib is worthy of further evaluation

    Determinants of high residual post-PCV13 pneumococcal vaccine-type carriage in Blantyre, Malawi:a modelling study

    Background In November 2011, Malawi introduced the 13-valent pneumococcal conjugate vaccine (PCV13) into the routine infant schedule. Four to 7 years after introduction (2015–2018), rolling prospective nasopharyngeal carriage surveys were performed in the city of Blantyre. Carriage of Streptococcus pneumoniae vaccine serotypes (VT) remained higher than reported in high-income countries, and impact was asymmetric across age groups. Methods A dynamic transmission model was fit to survey data using a Bayesian Markov-chain Monte Carlo approach, to obtain insights into the determinants of post-PCV13 age-specific VT carriage. Results Accumulation of naturally acquired immunity with age and age-specific transmission potential were both key to reproducing the observed data. VT carriage reduction peaked sequentially over time, earlier in younger and later in older age groups. Estimated vaccine efficacy (protection against carriage) was 66.87% (95% CI 50.49–82.26%), similar to previous estimates. Ten-year projected vaccine impact (VT carriage reduction) among 0–9 years old was lower than observed in other settings, at 76.23% (CI 95% 68.02–81.96%), with sensitivity analyses demonstrating this to be mainly driven by a high local force of infection. Conclusions There are both vaccine-related and host-related determinants of post-PCV13 pneumococcal VT transmission in Blantyre with vaccine impact determined by an age-specific, local force of infection. These findings are likely to be generalisable to other Sub-Saharan African countries in which PCV impact on carriage (and therefore herd protection) has been lower than desired, and have implications for the interpretation of post-PCV carriage studies and future vaccination programs.</p
